    The HPLC/DAD Fingerprints and Chemometric Analysis of Flavonoid Extracts from the Selected Sage (Salvia) Species

    The results of spectrophotometric and HPLC/DAD analysis are discussed, and a comparison is made of selectively extracted flavonoid fractions derived from twenty six sage species belonging to the Salvia genus. The sage samples were harvested in the vegetation seasons 2007, 2008, and 2009. It was a goal of this study to find out which species contain the highest yields of flavonoids (recognized for their free-radical-scavenging activity), as those with the highest yields could be applied in official medicine. It was spectrophotometrically established that the four sage species can be recognized for their highest flavonoid levels, while the HPLC/DAD analysis pointed out to the four other species. The source of the discrepancy between the two evaluation approaches was discussed.Moreover, the HPLC/DAD fingerprints of the flavonoid fraction underwent a chemometric pre-treatment, and then the purified fingerprints were analyzed by means of Principal Component Analysis (PCA) for the differences in the harvesting period. A difference was revealed between the herbs harvested in the 2007 season, and those harvested in 2008 and 2009. The main source of this difference could be the seasonal weather variation and the relatively longest storage period with the plants harvested in 2007

These antioxidant compounds play an important role in human health and are of interest for the food, pharmaceutical and cosmeceutical industries. Modern chromatographic and spectrometric techniques have made analysis easier than ever before, but their success depends on the extraction method used. In fact, the different antioxidants are identifified using chromatographic techniques coupled to diff erent specififi c detectors according to the characteristics of each molecule. Beyond their well-known health-promoting effects, antioxidant molecules can also be used to functionalize or preserve the freshness, nutritive value, flflavor and color of foodstuff s, which justify their incorporation into several matrices. In this chapter, the most common antioxidant compounds in foodstuff will be described, as well as the methodologies involved in their extraction, separation, identifification and quantifification.     The extracts of seven Lavandulae species (Lavandula stoechas, Lavandula lanata, Lavandula viridis, Lavandula angustifolia "Rosea", Lavandula angustifolia "Afropurpurea", Lavandula angustifolia and one unknown) were analyzed using the reversed-phase-high performance liquid chromatography-diode array detection (RP-HPLC-DAD) with gradient elution technique to obtain the chromatographic fingerprint profiles. The HPLC analysis was performed using the Kinetex RP18 chromatographic column and eluent consisting of methanol-water-0.1% formic acid (5-100% (v/v)) at 30 °C with the run time of 60 min. and the detection wavelength 280 nm. The chromatograms were preliminary processed with the smoothing, noise reduction, background subtraction and alignment using the SpecAlign program (version 2.4.1). The presence of selected standards (apigenin, myricetin, luteolin, luteolin 7-glucoside, chlorogenic acid, caffeic acid, ferulic acid) in the extracts was confirmed. The chemical similarity between studied plants was evaluated using the Cluster Analysis (Pearson correlation coefficient, r, and Euclidean) and PCA. The preliminary antioxidant activity of studied extracts was evaluated based on the total phenolic content (Folin-Ciocalteu method), ferric ion reducing antioxidant parameter (FRAP) and α,α-diphenyl-β-picrylhydrazyl (DPPH) free radical scavenging method using the spectrophotometric technique

    Analiza materiału roślinnego jest ważnym zadaniem w poszukiwaniu roślin o działaniu farmakologicznym a także w standaryzacji leków roślinnych. Celem każdego procesu ekstrakcji jest szybka i skuteczna izolacja związków z matrycy przy użyciu minimalnej ilości rozpuszczalnika. Celem przedstawionej pracy był wybór optymalnych warunków do analizy materiału roślinnego oraz zbadanie wpływu metody ekstrakcji na wydajność izolacji niektórych furanokumaryn z owoców gorysza wyniosłego. Furanokumaryny mają istotne zastosowanie w lecznictwie. Są między innymi wykorzystywane w terapii bielactwa i łuszczycy oraz leczeniu skórnego T-komórkowego chłoniaka z erytmodermią. Zastosowano następujące metody ekstrakcyjne: wyczerpująca ekstrakcja w aparacie Soxhleta, ekstrakcja wspomagana ultradźwiękami w temperaturze 20 i 60◦C oraz ekstrakcja wspomagana mikrofalami w układzie otwartym i zamkniętym. Analiza została przeprowadzona metodą wysokosprawnej chromatografii cieczowej z detektorem UV/VIS. Wyniki wskazują, że najwyższą wydajność imperatoryny uzyskano w wyniku ekstrakcji ultradźwiękowej 60◦C, natomiast metodą najskuteczniejszą do izolacji umbeliferonu okazała się ekstrakcja mikrofalowa w układzie zamkniętym.Analysis of plant material is an important task in chemotaxonomical investigations, particularly, in search of plants with pharmacological activity or in standardisation of plant drugs. The goal of every extraction process is rapid and effective isolation of compounds from a matrix by use of minimum amount of solvent. The aim of this paper was the choice of optimal condi-tions for the analysis of plant material and effect of extraction method on the yield of some furanocoumarins from Peucedanum verticillare fruits. Furanocoumarins have important uses in human medicine and exhibit toxicity against a wide range of organisms. This group of com-pounds reveal pharmacological activity. They are important drugs in vitiligo and psoriaris therapy and are also used in therapy erythrodermic variants of cutaneous T-cell lymphoma and chronic graft-versus-host disease. The following extraction methods were used in our experiments: exhaustive extraction in Soxh-let apparatus, ultrasound- assisted solvent extraction (USAE) at 20 and 60◦C as well as micro-wave-assisted solvent extraction in open and closed system (MASE). Target compounds analysis was performed by high performance liquid chromatography with UV/VIS detector. The results indicated that, the highest yield of imperatorin can be obtained by ultrasonification at 60◦C, while microwave-assisted solvent extraction in closed system is very efficient in the extraction of umbelliferone
