    The challenges and benefits of analyzing feedback comments in surveys: Lessons from a cross-national online survey of small-scale cannabis growers

    It is common practice in survey questionnaires to include a general open and non-directive feedback question at the end, but the analysis of this type of data is rarely discussed in the methodological literature. While these open-ended comments can be useful, most researchers fail to report on this issue. The aim of this article is to illustrate and reflect upon the benefits and challenges of analyzing responses to open-ended feedback questions. The article describes the experiences of coding and analyzing data generated through a feedback question at the end of an international online survey with small-scale cannabis cultivators carried out by the Global Cannabis Cultivation Research Consortium. After describing the design and dataset of the web survey, the analytical approach and coding frame are presented. The analytical strategies chosen in this study illustrate the diversity and complexity of feedback comments which pose methodological challenges to researchers wishing to use them for data analyses. In this article, three types of feedback comments (political/policy comments, general comments of positive and negative appreciation, and methodological comments) are used to illustrate the difficulties and advantages of analyzing this type of data. The advantages of analyzing feedback comments are well known, but they seem to be rarely exploited. General feedback questions at the end of surveys are typically non-directive. If researchers want to use these data for research and analyses, they need a clear strategy. They ought to give enough thought to why they are including this type of question, and develop an analytical strategy at the design stage of the study

    How and where to find NPS users: a cross national survey among current users of new psychoactive substances.

    Use of new psychoactive substances (NPS) across Europe remains a public health challenge. The study describes potentials and limitations of methods in a transnational survey of recent marginalized, nightlife and online community NPS users in Germany, Hungary, Ireland, the Netherlands, Poland and Portugal (n=3023). In terms of demographic profile, drug use history and type of NPS, different methods reached different segments of the NPS-using population. Last year use of different NPS varied across countries and groups. Respondents used NPS in a variety of settings, with public spaces most common in marginalized group. The study suggests that prevalence rates can reveal a picture of the NPS market that significantly deviates from what law enforcement seizures indicate. Outreach in nightlife settings and peer education are recommended to inform users about health risks and to improve access to drug services and care

    Purpose Phenotype information is crucial for the interpretation of genomic variants. So far it has only been accessible for bioinformatics workflows after encoding into clinical terms by expert dysmorphologists. Methods Here, we introduce an approach driven by artificial intelligence that uses portrait photographs for the interpretation of clinical exome data. We measured the value added by computer-assisted image analysis to the diagnostic yield on a cohort consisting of 679 individuals with 105 different monogenic disorders. For each case in the cohort we compiled frontal photos, clinical features, and the disease-causing variants, and simulated multiple exomes of different ethnic backgrounds. Results The additional use of similarity scores from computer-assisted analysis of frontal photos improved the top 1 accuracy rate by more than 20–89% and the top 10 accuracy rate by more than 5–99% for the disease-causing gene. Conclusion Image analysis by deep-learning algorithms can be used to quantify the phenotypic similarity (PP4 criterion of the American College of Medical Genetics and Genomics guidelines) and to advance the performance of bioinformatics pipelines for exome analysis

    L'afflux remarquable des entrepreneurs migrants chinois dans différents pays d'Afrique occidentale au cours des dernières années a été heurtée à une résistance de plus en plus farouche par des entrepreneurs locaux établis. Que le premiers ont un avantage concurrentiel sur ce dernier en raison de traits socio-culturels distinctifs, ou si l'efficacité supposée chinoise est juste une caractéristique de toutes les diasporas mercantiles, est ouvert à la question. Cette étude exploratoire des migrants entrepreneuriales chinois et nigérians au Ghana et au Bénin tente de répondre à cette question. Apparemment, les forces culturels des agents du changement migrants ne sont pas limités à des systèmes de valeurs héritées ou religions, comme une éthique protestante ou le confucianisme, mais ils sont adaptés en permanence et ont inventé de nouveau par des réseaux transnationaux de la migration dans un monde globalisé. Il n'y a aucune preuve d'une prétendue supériorité de la culture d’innovation chinois par rapport aux cultures d’innovation africains des migrants entrepreneuriales. Plutôt, il existe une capacité accrue d'innovation d'une diaspora mercantile en général vis à vis des entrepreneurs locaux, indépendamment de l'origine de la culture nationale dans lequel il est intégré. En outre, la rivalité des entrepreneurs migrants chinois et nigérians dans les marchés africains ne conduit pas nécessairement à la concurrence coupe-gorge souvent suspectée sous l'impact de la mondialisation. Souvent, les deux groupes agissent plutôt complémentaires. Cela contribue, sous certaines conditions, même à la réduction de la pauvreté dans le pays d'accueil

    The Sail-Backed Reptile Ctenosauriscus from the Latest Early Triassic of Germany and the Timing and Biogeography of the Early Archosaur Radiation

    Background Archosaurs (birds, crocodilians and their extinct relatives including dinosaurs) dominated Mesozoic continental ecosystems from the Late Triassic onwards, and still form a major component of modern ecosystems (>10,000 species). The earliest diverse archosaur faunal assemblages are known from the Middle Triassic (c. 244 Ma), implying that the archosaur radiation began in the Early Triassic (252.3–247.2 Ma). Understanding of this radiation is currently limited by the poor early fossil record of the group in terms of skeletal remains. Methodology/Principal Findings We redescribe the anatomy and stratigraphic position of the type specimen of Ctenosauriscus koeneni (Huene), a sail-backed reptile from the Early Triassic (late Olenekian) Solling Formation of northern Germany that potentially represents the oldest known archosaur. We critically discuss previous biomechanical work on the ‘sail’ of Ctenosauriscus, which is formed by a series of elongated neural spines. In addition, we describe Ctenosauriscus-like postcranial material from the earliest Middle Triassic (early Anisian) Röt Formation of Waldhaus, southwestern Germany. Finally, we review the spatial and temporal distribution of the earliest archosaur fossils and their implications for understanding the dynamics of the archosaur radiation. Conclusions/Significance Comprehensive numerical phylogenetic analyses demonstrate that both Ctenosauriscus and the Waldhaus taxon are members of a monophyletic grouping of poposauroid archosaurs, Ctenosauriscidae, characterised by greatly elongated neural spines in the posterior cervical to anterior caudal vertebrae. The earliest archosaurs, including Ctenosauriscus, appear in the body fossil record just prior to the Olenekian/Anisian boundary (c. 248 Ma), less than 5 million years after the Permian–Triassic mass extinction. These earliest archosaur assemblages are dominated by ctenosauriscids, which were broadly distributed across northern Pangea and which appear to have been the first global radiation of archosaurs

    Heterozygous ANKRD17 loss-of-function variants cause a syndrome with intellectual disability, speech delay, and dysmorphism

    ANKRD17 is an ankyrin repeat-containing protein thought to play a role in cell cycle progression, whose ortholog in Drosophila functions in the Hippo pathway as a co-factor of Yorkie. Here, we delineate a neurodevelopmental disorder caused by de novo heterozygous ANKRD17 variants. The mutational spectrum of this cohort of 34 individuals from 32 families is highly suggestive of haploinsufficiency as the underlying mechanism of disease, with 21 truncating or essential splice site variants, 9 missense variants, 1 in-frame insertion-deletion, and 1 microdeletion (1.16 Mb). Consequently, our data indicate that loss of ANKRD17 is likely the main cause of phenotypes previously associated with large multi-gene chromosomal aberrations of the 4q13.3 region. Protein modeling suggests that most of the missense variants disrupt the stability of the ankyrin repeats through alteration of core structural residues. The major phenotypic characteristic of our cohort is a variable degree of developmental delay/intellectual disability, particularly affecting speech, while additional features include growth failure, feeding difficulties, non-specific MRI abnormalities, epilepsy and/or abnormal EEG, predisposition to recurrent infections (mostly bacterial), ophthalmological abnormalities, gait/balance disturbance, and joint hypermobility. Moreover, many individuals shared similar dysmorphic facial features. Analysis of single-cell RNA-seq data from the developing human telencephalon indicated ANKRD17 expression at multiple stages of neurogenesis, adding further evidence to the assertion that damaging ANKRD17 variants cause a neurodevelopmental disorder.Neurolog

    Es wurden 80 Straßentauben und Kotproben aus zwei Straßentaubenschlägen der „Basler Taubenaktion“ (Schweiz) auf das Vorhandensein verschiedener Krankheitserreger untersucht. Dabei wurden vier auf den Menschen übertragbare pathogene Mikroorganismen gefunden (Chlamydia spp., Salmonella spp., Campylobacter jejuni und Cryptococcus neoformans). Darüberhinaus fanden sich auch Krankheitserreger ohne zoonotisches Potential wie Trichomonas gallinae, Kokzidien, Helminthen, Ektoparasiten und Pilze, die allerdings für die Tauben selbst potenziell pathogen sind. Die Verteilung der pathogenen Mikroorganismen und Parasiten zeigte signifikante Unterschiede zwischen beiden Herkunftsschlägen. Die Tauben der beiden Brutkolonien unterschieden sich in Bezug auf den Ernährungszustand und den Befall mit zwei Federlingsarten, Columbicola columbae und Campanulotes bidentatus compar. Die Prävalenz für Trichomonas gallinae differierte nicht signifikant zwischen Jung- und Alttauben, die Jungtauben zeigten aber, sofern sie Trichomonaden beherbergten, einen signifikant stärkeren Befall. Tiere mit gutem Ernährungszustand zeigten im Vergleich zu Tieren mit mäßigem oder schlechtem Ernährungszustand einen stärkeren Befall mit Trichomonas gallinae. Auf dem Gefieder von Tauben mit schlechtem Ernährungszustand ließen sich größere Mengen der Federlingsart Columbicola columbae nachweisen während Tiere mit gutem Ernährungszustand einen signifikant höheren Befall mit Campanulotes bidentatus compar aufwiesen. Bemerkenswert war, dass eine der beiden untersuchten Brutkolonien die Brutaktivität aufgrund des Verlustes einer angestammten Nahrungsquelle seit zwei Jahren nahezu eingestellt hatte. Trotzdem wies diese Brutkolonie einen besseren Ernährungszustand auf, als diejenige Kolonie, der ihr normales Nahrungsangebot zur Verfügung stand. Eine mögliche Erklärung für dieses Resultat könnte das Vorliegen eines biologischen Kontrollmechanismus zur Reduktion der Fortpflanzungsrate unter verschlechterten Umweltbedingungen sein, um damit das Überleben der adulten Tiere zu sichern. Sollte diese These zutreffen, kann daraus gefolgert werden, dass die Fütterung von Straßentauben durch Tierfreunde möglicherweise infolge der dadurch aufrechterhaltenen Brutaktivität zu einer Verschlechterung des Gesundheitszustandes der Straßentauben führt. [English Abstract] We examined 80 feral pigeons and their fecal samples from two feral pigeon lofts of the "Pigeon Action of Basel" (Switzerland) for different pathogens. The tested material harbored four pathogenic agents transmissible to humans (Chlamydia spp., Salmonella spec., Campylobacter jejuni, Cryptococcus neoformans) In addition several pathogens were found which are no zoonotic agents but potentially pathogenic for the pigeons themselves, such as Trichomonas gallinae, coccidia, helminths, ectoparasites and fungi. The number of pathogens and parasites detected in the fecal samples varied significantly between the two localities. The pigeons of the two investigated breeding flocks differed in nutritional status and the incidence of two species of feather lice, Columbicola columbae and Campanulotes bidentatus compar. The prevalence of Trichomonas gallinae between juveniles and adults was not significantly different but juveniles exhibited significantly heavier infestation if infected. Individuals with a good nutritional status tend to show heavier infestation with Trichomonas gallinae compared to birds with moderate or poor nutritional status. Birds with a poor nutritional status tend to suffer from a heavier infestation with the feather louse C. columbae, and birds with a good nutritional status show significant heavier infestation with C. bidentatus compar. It was remarkable that one of the two investigated breeding populations almost gave up its breeding activity for two years because of the loss of its familiar food source. Nevertheless, this population showed a better nutritional status than the population without restrictions in the acquisition of food. This fact could be interpreted by the existence of a biological control mechanism for suppression of the reproduction in degraded environmental conditions to ensure the survival of the adults. If this assumption is correct, the feeding of feral pigeons by animal lovers possibly causes impairment of pigeon's health in consequence of continuation of the breeding activity in spite of declined living conditions in the city

    The wrapped corpse. Retrospective analysis of forensic autopsies from 2009 to 2016 with respect to different case constellations and motivations

    Background. Although corpse disposal itself is not a rare phenomenon, the wrapping of a corpse is less common, and its facets can be very varied. Objective. The cases available enabled different constellations to be determined and analyzed with respect to the method, motivation and type of wrapping. Material and methods. A retrospective analysis of forensic post-mortem examinations from 2009-2016 at the Institute of Forensic Medicine at the Cologne Medical School was conducted. Cases and results of the postmortem examinations were scientifically reviewed. The evaluation included only cases of corpses that had been fully or partly wrapped. Results. These cases represent a broad range of constellations including seven cases of completed homicide, three cases of completed suicide, one case of an autoerotic fatality and one case of natural death. In cases of homicide the corpse is generally wrapped for transportation and concealment but the placement and wrapping of the corpse vary. Just as varied and imaginative are the wrappings of corpses resulting from suicide, autoerotic accidents and more harmless cases
