Stratosphere-troposphere transport in a numerical simulation of midlatitude convection
The transport of stratospheric air deep into the troposphere via convection is
investigated numerically using the UK Met Office Unified Model. A convective system
that formed on 27 June 2004 near southeast England, in the vicinity an upper level
potential vorticity anomaly and a lowered tropopause, provides the basis for analysis.
Transport is diagnosed using a stratospheric tracer that can either be passed through or
withheld from the model’s convective parameterization scheme. Three simulations are
performed at increasingly finer resolutions, with horizontal grid lengths of 12, 4, and 1 km.
In the 12 and 4 km simulations, tracer is transported deeply into the troposphere by the
parameterized convection. In the 1 km simulation, for which the convective
parameterization is disengaged, deep transport is still accomplished but with a much
smaller magnitude. However, the 1 km simulation resolves stirring along the tropopause
that does not exist in the coarser simulations. In all three simulations, the concentration of
the deeply transported tracer is small, three orders of magnitude less than that of the
shallow transport near the tropopause, most likely because of the efficient dilution of
parcels in the lower troposphere
Major flaws in conflict prevention policies towards Africa : the conceptual deficits of international actors’ approaches and how to overcome them
Current thinking on African conflicts suffers from misinterpretations oversimplification, lack of focus, lack of conceptual clarity, state-centrism and lack of vision). The paper analyses a variety of the dominant explanations of major international actors and donors, showing how these frequently do not distinguish with sufficient clarity between the ‘root causes’ of a conflict, its aggravating factors and its triggers. Specifically, a correct assessment of conflict prolonging (or sustaining) factors is of vital importance in Africa’s lingering confrontations. Broader approaches (e.g. “structural stability”) offer a better analytical framework than familiar one-dimensional explanations. Conversation therapy with people with aphasia and conversation partners using video feedback: a group and case series investigation of changes in interaction
Conversation therapies employing video for feedback and to facilitate outcome measurement are increasingly used with people with post-stroke aphasia and their conversation partners; however the evidence base for change in everyday interaction remains limited. We investigated the effect of Better Conversations with Aphasia (BCA), an intervention that is freely available online at https://extend.ucl.ac.uk/. Eight people with chronic agrammatic aphasia, and their regular conversation partners participated in the tailored 8 week program involving significant video feedback. We explored changes in: (i) conversation facilitators (such as multi-modal turns by people with aphasia); and (ii) conversation barriers (such as use of test questions by conversation partners). The outcome of intervention was evaluated directly by measuring change in video-recorded everyday conversations. The study employed a pre-post design with multiple 5 minute samples of conversation before and after intervention, scored by trained raters blind to the point of data collection. Group level analysis showed no significant increase in conversation facilitators. There was, however, a significant reduction in the number of conversation barriers. The case series data revealed variability in conversation behaviors across occasions for the same dyad and between different dyads. Specifically, post-intervention there was a significant increase in facilitator behaviors for two dyads, a decrease for one and no significant change for five dyads. There was a significant decrease in barrier behaviors for five dyads and no significant change for three dyads. The reduction in barrier behaviors was considerable; on average change from over eight to fewer than three barrier behaviors in 5 minutes of conversation. The pre-post design has the limitation of no comparison group. However, change occurs in targeted conversational behaviors and in people with chronic aphasia and their partners. The findings suggest change can occur after eight therapy sessions and have implications for clinical practice. A reduction in barrier behaviors may be easier to obtain, although the controlled case series results demonstrate a significant increase in conversation facilitators is also possible. Convective-scale perturbation growth across the spectrum of convective regimes
Convection-permitting ensembles have led to improved forecasts of many atmospheric phenomena. However, to fully utilize these forecasts the dependence of predictability on synoptic conditions needs to be understood. In this study, convective regimes are diagnosed based on a convective timescale which identifies the degree to which convection is in equilibrium with the large-scale forcing. Six convective cases are examined in a convection-permitting ensemble constructed using the Met Office Unified Model. The
ensemble members were generated using small-amplitude buoyancy perturbations added into the boundary layer, which can be considered to represent turbulent fluctuations close to the gridscale. Perturbation growth is shown to occur on different scales with an order of magnitude difference between the regimes (O(1 km) for cases closer to non-equilibrium convection and O(10 km) for cases closer to equilibrium convection). This difference reflects the fact that cell locations are essentially random in the equilibrium events after the first 12 h of the forecast, indicating a more rapid upscale perturbation growth compared to the non-equilibrium events. Furthermore, large temporal
variability is exhibited in all perturbation growth diagnostics for the nonequilibrium regime. Two boundary condition driven cases are also considered and show similar characteristics to the non-equilibrium cases, implying that caution is needed to interpret the timescale when initiation is not within the domain. Further understanding of perturbation growth within the different regimes could lead to a better understanding of where ensemble design improvements can be made beyond increasing the model resolution and could improve interpretation of forecasts
Geometry sensing by dendritic cells dictates spatial organization and PGE2-induced dissolution of podosomes
Assembly and disassembly of adhesion structures such as focal adhesions (FAs) and podosomes regulate cell adhesion and differentiation. On antigen-presenting dendritic cells (DCs), acquisition of a migratory and immunostimulatory phenotype depends on podosome dissolution by prostaglandin E2 (PGE2). Whereas the effects of physico-chemical and topographical cues have been extensively studied on FAs, little is known about how podosomes respond to these signals. Here, we show that, unlike for FAs, podosome formation is not controlled by substrate physico-chemical properties. We demonstrate that cell adhesion is the only prerequisite for podosome formation and that substrate availability dictates podosome density. Interestingly, we show that DCs sense 3-dimensional (3-D) geometry by aligning podosomes along the edges of 3-D micropatterned surfaces. Finally, whereas on a 2-dimensional (2-D) surface PGE2 causes a rapid increase in activated RhoA levels leading to fast podosome dissolution, 3-D geometric cues prevent PGE2-mediated RhoA activation resulting in impaired podosome dissolution even after prolonged stimulation. Our findings indicate that 2-D and 3-D geometric cues control the spatial organization of podosomes. More importantly, our studies demonstrate the importance of substrate dimensionality in regulating podosome dissolution and suggest that substrate dimensionality plays an important role in controlling DC activation, a key process in initiating immune responses
Finishing the euchromatic sequence of the human genome
The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
