Journal of Biotechnology. 1989;10(3-4):285-291.Genetic engineering methods were used to enhance the substrate spectrum of Alcaligenes eutrophus H16, a poly-[beta]-hydroxybutyric acid (PHB) producer. Using parts of the vector pMMB33 and a 2.5 kb DNA fragment of the Bacillus subtilis chromosome a plasmid was constructed bearing the gene for levanase, an enzyme able to hydrolyze various saccharides. After transfer of the levanase gene by triparental conjugation, the gene, controlled by its own Bacillus subtilis promoter, is expressed in Alcaligenes eutrophus H16 and enables the strain to hydrolyze sucrose. However, growth on sucrose is limited; i.e. the sucrose is not transported efficiently into the cell and/or the levanase is not secreted into the medium

    One hundred and five nulliparous rabbit does were fed ad libitum one month before and during pregnancy with two isofibrous, isoenergetic and isoproteic diets supplemented with two different fat sources: 0.75% lard for control diet (C group: n=53) or 1.5% of a supplement (Optomega-50; Optivite International Ltd., España) containing a 50% of ether extract and 38% of n-3 polyunsaturated fatty acids (PUFA) for experimental diet (group P n-3; n=52). Fertility after artificial insemination and induction of ovulation with 20 μg of Gonadorelina (Inducel-GnRH, Ovejero) at 4.5 months of age was assessed. At parturition, pregnancy length and prolificacy (number and weight of born alive and number of stillborn) were observed. A total of 8 pregnant does from each experimental group were chosen at random to study embryo and placenta dimensions by means of ultrasonography at 8, 15 and 22 days of pregnancy. PUFA n-3 supplementation did not affect any productive results due the success on reproductive features of nulliparous does which are difficult to improve. Neither, any differences in ultrasonographic meausrements were obtained; nevertheless we have defined useful physiological parameters of rabbit embryo and placenta to future studies in this specie.Un total de 105 conejas nulíparas se alimentaron ad libitum un mes antes y durante su primera gestación con dos piensos isofibrosos, isoenergéticos e isoproteicos suplementados con dos fuentes de grasa diferentes: 0,75% de manteca para la dieta control (grupo C; n=53) ó 1,5% de un suplemento (Optomega-50; Optivite International Ltd., España) que contenía un 50% de extracto etéreo y 38% de ácidos grasos poli-insaturados (AG n-3) para la dieta experimental (grupo P; n=52). A los 4,5 meses de edad se determinó la fertilidad después de ser inseminadas artificialmente y tratadas con 20 μg de Gonadorelina (Inducel-GnRH, Ovejero) para inducirles la ovulación. Al parto se determinó la duración de la gestación, el número y peso de los gazapos nacidos vivos y muertos. Se escogieron 16 conejas al azar, 8 de cada grupo, y se realizaron ecografías a los 8, 15 y 22 días de gestación en las que se determinó las dimensiones del embrión y de los anejos fetales. La suplementación con AG n-3 no afectó a los resultados productivos determinados ya que se trata de parámetros que en nulíparas son difíciles de mejorar y suelen ser altos. Tampoco se observaron diferencias en las determinaciones ecográficas pero se han podido definir medidas fisiológicas de los fetos y anejos placentarios de gran utilidad para futuros estudios en esta especie

    Pion-Muon Asymmetry Revisited

    Long ago an unexpected and unexplainable phenomena was observed. The distribution of muons from positive pion decay at rest was anisotropic with an excess in the backward direction relative to the direction of the proton beam from which the pions were created. Although this effect was observed by several different groups with pions produced by different means, the result was not accepted by the physics community, because it is in direct conflict with a large set of other experiments indicating that the pion is a pseudoscalar particle. It is possible to satisfy both sets of experiments if helicity-zero vector particles exist and the pion is such a particle. Helicity-zero vector particles have direction but no net spin. For the neutral pion to be a vector particle requires an additional modification to conventional theory as discussed herein. An experiment is proposed which can prove that the asymmetry in the distribution of muons from pion decay is a genuine physical effect because the asymmetry can be modified in a controllable manner. A positive result will also prove that the pion is NOT a pseudoscalar particle.Comment: 9 pages, 3 figure

    Predator control programmes are generally implemented in an attempt to increase prey population sizes. However, predator removal could prove harmful to prey populations that are regulated primarily by parasitic infections rather than by predation. We develop models for microparasitic and macroparasitic infection that specify the conditions where predator removal will (a) increase the incidence of parasitic infection, (b) reduce the number of healthy individuals in the prey population and (c) decrease the overall size of the prey population. In general, predator removal is more likely to be harmful when the parasite is highly virulent, macroparasites are highly aggregated in their prey, hosts are long-lived and the predators select infected prey

    Measurement of the partial widths of the Z into up- and down-type quarks

    Using the entire OPAL LEP1 on-peak Z hadronic decay sample, Z -> qbarq gamma decays were selected by tagging hadronic final states with isolated photon candidates in the electromagnetic calorimeter. Combining the measured rates of Z -> qbarq gamma decays with the total rate of hadronic Z decays permits the simultaneous determination of the widths of the Z into up- and down-type quarks. The values obtained, with total errors, were Gamma u = 300 ^{+19}_{-18} MeV and Gamma d = 381 ^{+12}_{-12} MeV.     Genuine Correlations of Like-Sign Particles in Hadronic Z0 Decays

    Correlations among hadrons with the same electric charge produced in Z0 decays are studied using the high statistics data collected from 1991 through 1995 with the OPAL detector at LEP. Normalized factorial cumulants up to fourth order are used to measure genuine particle correlations as a function of the size of phase space domains in rapidity, azimuthal angle and transverse momentum. Both all-charge and like-sign particle combinations show strong positive genuine correlations. One-dimensional cumulants initially increase rapidly with decreasing size of the phase space cells but saturate quickly. In contrast, cumulants in two- and three-dimensional domains continue to increase. The strong rise of the cumulants for all-charge multiplets is increasingly driven by that of like-sign multiplets. This points to the likely influence of Bose-Einstein correlations. Some of the recently proposed algorithms to simulate Bose-Einstein effects, implemented in the Monte Carlo model PYTHIA, are found to reproduce reasonably well the measured second- and higher-order correlations between particles with the same charge as well as those in all-charge particle multiplets.Comment: 26 pages, 6 figures, Submitted to Phys.     Measurement of the Strong Coupling alpha s from Four-Jet Observables in e+e- Annihilation

    Data from e+e- annihilation into hadrons at centre-of-mass energies between 91 GeV and 209 GeV collected with the OPAL detector at LEP, are used to study the four-jet rate as a function of the Durham algorithm resolution parameter ycut. The four-jet rate is compared to next-to-leading order calculations that include the resummation of large logarithms. The strong coupling measured from the four-jet rate is alphas(Mz0)= 0.1182+-0.0003(stat.)+-0.0015(exp.)+-0.0011(had.)+-0.0012(scale)+-0.0013(mass) in agreement with the world average. Next-to-leading order fits to the D-parameter and thrust minor event-shape observables are also performed for the first time.     A measurement of the tau mass and the first CPT test with tau leptons

    We measure the mass of the tau lepton to be 1775.1+-1.6(stat)+-1.0(syst.) MeV using tau pairs from Z0 decays. To test CPT invariance we compare the masses of the positively and negatively charged tau leptons.     Measurement of Rb in e+e- Collisions at 182 - 209 GeV

    Measurements of Rb, the ratio of the bbbar cross-section to the qqbar cross- section in e+e- collisions, are presented. The data were collected by the OPAL experiment at LEP at centre-of-mass energies between 182 GeV and 209 GeV. Lepton, lifetime and event shape information is used to tag events containing b quarks with high efficiency. The data are compatible with the Standard Model expectation. The mean ratio of the eight measurements reported here to the Standard Model prediction is 1.055+-0.031+-0.037, where the first error is statistical and the second systematic.Comment: 21 pages, 5 figures, Submitted to Phys.     First Measurement of Z/gamma* Production in Compton Scattering of Quasi-real Photons

    We report the first observation of Z/gamma* production in Compton scattering of quasi-real photons. This is a subprocess of the reaction e+e- to e+e-Z/gamma*, where one of the final state electrons is undetected. Approximately 55 pb-1 of data collected in the year 1997 at an e+e- centre-of-mass energy of 183 GeV with the OPAL detector at LEP have been analysed. The Z/gamma* from Compton scattering has been detected in the hadronic decay channel. Within well defined kinematic bounds, we measure the product of cross-section and Z/gamma* branching ratio to hadrons to be (0.9+-0.3+-0.1) pb for events with a hadronic mass larger than 60 GeV, dominated by (e)eZ production. In the hadronic mass region between 5 GeV and 60 GeV, dominated by (e)egamma* production, this product is found to be (4.1+-1.6+-0.6) pb. Our results agree with the predictions of two Monte Carlo event generators, grc4f and PYTHIA.Comment: 18 pages, LaTeX, 5 eps figures included, submitted to Physics Letters
