    Update on the fluorometric measurement of enzymatic activities for Lysosomal Storage Disorder detection: The example of MPS VI

    Lysosomal Storage Disorders (LSD) are rare diseases that as a whole havea combined incidence ranging from 1:1500 to 1:7000 live births. One of suchdiseases is Mucopolysaccharidosis VI (MPS VI), or Maroteaux Lamy Syndrome.MPS VI patients undergo devastating and irreversible skeletal alterations andmultisystemic failure as from early childhood due to reduced Arylsulfatse B(ARSB) enzyme activity.Reaching a final diagnosis is not always a short cut path, but rather a yearslongbattle against uncertainty and unnecessary medical interventions. Ouraim is to contribute from the bench table with different approaches that couldserve as alternatives to pre-existing assays for screening and diagnosing MPSVI and other LSD.The present work is based on our research article authored by Franco etal.1 where we studied the effect of blood-derived hemoglobin, and other bloodcomponents, on the fluorescence of 4-Methylumbelliferone when measuringARSB enzyme activity from dried blood spot (DBS) samples.Our experience indicates that to date there are plenty of differentapproaches for measuring ARSB enzyme activity, although the sample typerequired or the assay in itself often make them more adaptable for either highthroughput screening or small scale diagnostics.As a whole, the fluorometric determinations seem to be the mostaccessible to low budget laboratories with equally valuable performancesas a sophisticated mass spectrometry analysis for this disease.     The general purpose of this work aims to contribute to the knowledge of specificities in the institutional processes linked to gender issues that occur in Olavarría. When we mention the idea of institutional processes, we refer in principle to a sum of activities that interact and articulate to influence a specific population set; but we also mention the ways -which institutions have to do with the treatment of gender violence- through which the behaviors of people can be regulated and even controlled. These social institutions are those that implement public policies in the local order. Although the gender problems that affect us should not be reduced to an understanding associated only with the existence of physical violence, the truth is that this particular cut is the one that is most represented in the institutional failures of the Argentine State in matters of Human rights. In this sense, it also verifies the appearance of a device forattention of violence that arises as a correlate of the demand, from the municipal State.Resumen  El propósito general de nuestro trabajo pretende contribuir al conocimiento de especificidades en los procesos institucionales vinculados a problemáticas de género que se dan en Olavarría. Cuando mencionamos la idea de procesos institucionales, nos referimos en principio a una suma de actividades que interactúan y se articulan para incidir en un conjunto poblacional determinado; pero también hacemos mención a los modos -que tienen las instituciones relacionadas con el tratamiento de la violencia de género- mediante los que se pueden regular e incluso controlar los comportamientos de las personas. Esas instituciones sociales son las que implementan políticas públicas en el orden local. Si bien los problemas de género que nos afectan no han de reducirse a un entendimiento asociado únicamente con la existencia de la violencia física, lo cierto es que este recorte particular es el que más se halla representado en los incumplimientos institucionales del Estado argentino en temas de Derechos Humanos.     In vitro and in vivo characterization of a typical and a high pathogenic bovine viral diarrhea virus type ii strains

    Non-cytopathic (ncp) type 2 bovine viral diarrhea virus (BVDV-2) is widely prevalent in Argentina causing high mortality rates in cattle herds. In this study, we characterized an Argentinean ncp BVDV-2 field isolate (98-124) compared to a high-virulence reference strain (NY-93), using in silico analysis, in vitro assays, and in vivo infections of colostrum-deprived calves (CDC) to compare pathogenic characters and virulence. In vitro infection of bovine peripheral blood mononuclear cells (PBMC) with BVDV 98-124 induced necrosis shortly after infection while NY-93 strain increased the apoptotic rate in infected cells. Experimental infection of CDC (n = 4 each) with these strains caused an enteric syndrome. High pyrexia was detected in both groups. Viremia and shedding were more prolonged in the CDC infected with the NY-93 strain. In addition, NY-93 infection elicited a severe lymphopenia that lasted for 14 days, whereas 98-124 strain reduced the leukocyte counts for 5 days. All infected animals had a diminished lymphoproliferation activity in response to a mitogen. Neutralizing and anti-NS3 antibodies were detected 3 weeks after infection in all infected calves. Virulence was associated with a more severe clinical score, prolonged immune-suppression, and a greater window for transmission. Studies of apoptosis/necrosis performed after in vitro PBMC infection also revealed differences between both strains that might be correlated to the in vivo pathogenesis.     Characterizing ecoregions in Argentinian patagonia using extant continental ostracods

    In order to characterize Patagonian (Argentina) ecoregions using non-marine ostracods, their associations in 69 environments were assessed. Twenty eight taxa were recorded, including 12 endemic of the Neotropical region. Our results indicate that Patagonian ostracods are mainly influenced by electrical conductivity (EC), altitude, pH, and temperature; and shows a correlation with Argentinian ecoregions. Assemblage I is composed of sites located at high altitude in the Andean Patagonian forest ecoregion. Host waters have low temperature, EC and pH, and support as representative species Cypris pubera, Eucypris virens, Bradleystrandesia fuscata, Tonacypris lutaria and Amphicypris nobilis. Assemblage II, related to mid-altitude environments in the Patagonian Steppe ecoregion, thrived in waters with moderate to high EC, and alkaline pH values. Dominant species includes Limnocythere rionegroensis, L. patagonica, E. virgata, Riocypris whatleyi, Riocypris sarsi, Newnhamia patagonica, Kapcypridopsis megapodus, Ilyocypris ramirezi and Penthesinelula incae. Assemblage III inhabited environments within Monte and Espinal ecoregions, situated in the eastern part of the study area at low altitude, EC moderate and temperate waters, supporting Heterocypris hyalinus, Amphicypris argentinensis, Sarscypridopsis aculeata, Cypridopsis vidua, Herpetocypris intermedia and Chlamidotheca incisa. Our results indicates that Argentinian Patagonia hosts a diverse ostracod fauna and highlights their capacity as proxies in ecological and palaeoenvironmental studies.Fil: Cusminsky, Gabriela Catalina.     Absence of the proapoptotic Bax protein extends fertility and alleviates age-related health complications in female mice

    The menopausal transition in human females, which is driven by a loss of cyclic ovarian function, occurs around age 50 and is thought to underlie the emergence of an array of health problems in aging women. Although mice do not undergo a true menopause, female mice exhibit ovarian failure long before death because of chronological age and subsequently develop many of the same age-associated health complications observed in postmenopausal women. Here we show in mice that inactivation of the proapoptotic Sax gene, which sustains ovarian lifespan into advanced age, extends fertile potential and minimizes many age-related health problems, including bone and muscle loss, excess fat deposition, alopecia, cataracts, deafness, increased anxiety, and selective attention deficit. Further, ovariectomy studies show that the health benefits gained by aged females from Bax deficiency reflect a complex interplay between ovary-dependent and -independent pathways. Importantly, and contrary to popular belief, prolongation of ovarian function into advanced age by Bax deficiency did not lead to an increase in tumor incidence. Thus, the development of methods for postponing ovarian failure at menopause may represent an attractive option for improving the quality of life in aging females. © 2007 by The National Academy of Sciences of the USA

    La imagen y la narrativa como herramientas para el abordaje psicosocial en escenarios de violencia. Departamento Santander y Antioquia.En el diplomado de profundización acompañamiento escenario de violencia, trae temáticas para razonar como futuros psicólogos el actuar con cada tema, también en el diplomado se encontraron diferentes estrategias que se pueden implementar como futuros psicólogos para un adecuado abordaje de las víctimas en escenarios de violencia. Cada una de las unidades planteadas por el diplomado se ejecutaron de acuerdo con unas fechas, donde se puso en práctica todo lo leído y aprendido en las unidades; en la primera parte con la que nos encontramos en el diplomado, fue ver como desde la perspectiva de la psicología abordamos a las víctimas del conflicto sociopolítico que estrategias psicosociales contribuyen para la disminución de la discriminación y marginación de las víctimas. Por otro lado, también se trabajó desde la imagen y narrativa como instrumentos de acción psicosocial presentando un informe analítico de las fotos voz realizadas en diferentes contextos, logrando identificar subjetividades e intersubjetividades que emergen en los escenarios de violencia y el rol que se cumple dentro de estos. Por último se trabajó desde los enfoques narrativos con los relatos del Libro: Voces que se trata de casos de violencia y esperanza en Colombia, como grupo se escogió el caso de Ana Ligia resaltando los impactos psicosociales, las voces que revelan un posicionamiento subjetivo ya sea víctima o sobreviviente y la emancipación discursiva frente a las imágenes de horror, también en la parte final se trabajó con la Comunidad de Cacarica y los emergentes psicosociales latentes por el hostigamiento militar, creando acciones de apoyo y estrategias psicosociales para los pobladores de Cacarica facilitando la potenciación del afrontamiento a la situación que vivieron. 4 Este diplomado ha marcado nuestro futuro como psicólogos con cada una de las enseñanzas que ha dejado en cuanto a los temas vistos, teniendo bases para nuestra etapa laboral y profesional.In the diploma of deepening accompaniment of the violence scenario, it brings themes to reason as future psychologists acting with each topic, also in the diploma course, different strategies were found that can be implemented as future psychologists for an adequate approach to victims in violence scenarios. Each of the units proposed by the diploma were executed according to dates, where everything read and learned in the units was put into practice; In the first part we encountered in the diploma course, it was to see how from the perspective of psychology we approached the victims of the sociopolitical conflict that psychosocial strategies contribute to the reduction of discrimination and marginalization of the victims. On the other hand, we also worked from the image and narrative as instruments of psychosocial action presenting an analytical report of the voice photos taken in different contexts, managing to identify subjectivities and intersubjectives that emerge in the scenarios of violence and the role that is fulfilled within these. On the latter he worked from the narrative approaches the stories of the Book: Voices that are cases of violence and hope in Colombia, as a group chose the case of Ana Ligia highlighting the psychosocial impacts, voices that reveal a subjective positioning either victim or survivor and discursive emancipation in the face of horror images, also in the final part they worked with the Community of Cacarica and the latent psychosocial emergencies for military harassment, creating support actions and psychosocial strategies for the inhabitants of Cacarica facilitating the Empowerment of coping with the situation they lived. This diploma has marked our future as psychologists with each one of the lessons that it has left regarding the subjects seen, having bases for our labor and professional stage. 6 Keywords: scenarios of violence, subjectivity, psychosocial strategies, psychosocial action, community, population, victims, survivors

    CCR7 as a novel therapeutic target in t-cell PROLYMPHOCYTIC leukemia

    T-cell prolymphocytic leukemia (T-PLL) is a poor prognostic disease with very limited options of efficient therapies. Most patients are refractory to chemotherapies and despite high response rates after alemtuzumab, virtually all patients relapse. Therefore, there is an unmet medical need for novel therapies in T-PLL. As the chemokine receptor CCR7 is a molecule expressed in a wide range of malignancies and relevant in many tumor processes, the present study addressed the biologic role of this receptor in T-PLL. Furthermore, we elucidated the mechanisms of action mediated by an anti-CCR7 monoclonal antibody (mAb) and evaluated whether its anti-tumor activity would warrant development towards clinical applications in T-PLL. Our results demonstrate that CCR7 is a prognostic biomarker for overall survival in T-PLL patients and a functional receptor involved in the migration, invasion, and survival of leukemic cells. Targeting CCR7 with a mAb inhibited ligand-mediated signaling pathways and induced tumor cell killing in primary samples. In addition, directing antibodies against CCR7 was highly effective in T-cell leukemia xenograft models. Together, these findings make CCR7 an attractive molecule for novel mAb-based therapeutic applications in T-PLL, a disease where recent drug screen efforts and studies addressing new compounds have focused on chemotherapy or small molecules.Peer reviewe

    LGBT+ Training needs for health and social care professionals: a cross-cultural comparison among seven European countries

    Introduction Research suggests that specific training on LGBT+ issues may improve the competencies and skills of health and social care (HSC) professionals, which reduces the negative attitudes toward LGBT+ people. Despite this, there seems to be a lack of coverage of LGBT+ needs in HSC education. The present study aims to explore the specific LGBT+ training needs of HSC professionals and to examine the relationship between these training needs and the four dimensions of the Papadopoulos model, i.e. cultural awareness, cultural knowledge, cultural sensitivity, and cultural competence. Methods The research used data from a cross-cultural project, “Intercultural Education for Nurses in Europe (IENE9),” which was administered to 412 HSC academics and workers (62% females; Mage=46.06, SDage=10.48) between February 2020 and July 2020, in seven European countries: UK (coordinator), Denmark, Spain, Germany, Cyprus, Italy, and Romania. Results Hierarchical multiple regression showed that higher training needs were associated with cultural awareness, cultural knowledge, and cultural competence. The need for training on LGBT+ issues was higher for Cyprus, Romania, Spain, Italy, and the UK, compared with Denmark (no differences between Germany and Denmark were found). Conclusions We believe that there has been a lack of focus on the LGBT+ training needs of HSC professionals: Greater efforts are required to develop a culturally competent and compassionate LGBT+ curriculum. Social Policy Implications Findings from the present study will inform the development of a free, Massive Open Online Course (MOOC), for culturally competent and compassionate HSC professionals in Europe to improve the quality of their car
