Monoxenic culture of the slug parasite Phasmarhabditis hermaphrodita (Nematoda : Rhabditidae) with different bacteria in liquid and solid phase
Seize isolats bactériens, représentant treize espèces, ont été testés pour leur capacité à maintenir la croissance de #Phasmarhabditis hermaphrodita en culture monoxénique sur agar, en boîte de Petri. Tous les isolats permettent la croissance de #P. hermaphrodita, mais la capacité de reproduction varie considérablement suivant les différentes bactéries. Cinq espèces de bactéries ayant permis une croissance vigoureuse de #P. hermaphrodita ont été utilisées pour son élevage en milieu liquide. Toutes permettent la croissance et la reproduction du nématode mais des différences significatives se produisent en ce qui concerne le nombre total de nématodes et celui des #dauer larvae formées. Les différences entre les chiffres totaux ne sont pas significativement différentes de celles observées dans le cas de culture en boîtes de Petri. En milieu liquide, #Providencia rettgeri paraît produire les meilleurs rendements en #dauer larvae. Tant en milieu solide que liquide, la croissance monoxénique de #P. hermaphrodita$ est meilleure sur des milieux à base de rognon de porc que sur des milieux ne comportant que des éléments entièrement solubles. Thermodynamics of the Anisotropic Spin-1/2 Heisenberg Chain and Related Quantum Chains
The free energy and correlation lengths of the spin-1/2 chain are
studied at finite temperature. We use the quantum transfer matrix approach and
derive non-linear integral equations for all eigenvalues. Analytic results are
presented for the low-temperature asymptotics, in particular for the critical
chain in an external magnetic field. These results are compared to
predictions by conformal field theory. The integral equations are solved
numerically for the non-critical chain and the related spin-1 biquadratic

Last Glacial Period Cryptotephra Deposits in an Eastern North Atlantic Marine Sequence: Exploring Linkages to the Greenland Ice-Cores
The establishment of a tephra framework for the Greenland ice-cores spanning the last glacial period, particularly between 25 and 45 ka b2k, provides strong potential for precisely correlating other palaeoclimatic records to these key archives. Tephra-based synchronisation allows the relative timing of past climatic changes recorded within different depositional environments and potential causal mechanisms to be assessed. Recent studies of North Atlantic marine records have demonstrated the potential of tracing cryptotephra horizons in these sequences and the development of protocols now allows a careful assessment of the isochronous nature of such horizons. Here we report on tephrochronological investigations of a marine sequence retrieved from the Goban Spur, Eastern North Atlantic, covering ?25–60 ka b2k. Density and magnetic separation techniques and an assessment of potential transport and depositional mechanisms have identified three previously unknown isochronous tephra horizons along with deposits of the widespread North Atlantic Ash Zone II and Faroe Marine Ash Zone III. Correlations between the new horizons and the Greenland ice-core tephra framework are explored and despite no tie-lines being identified the key roles that high-resolution climatostratigraphy and shard-specific trace element analysis can play within the assessment of correlations is demonstrated. The previously unknown horizons are new additions to the overall North Atlantic tephra framework for the last glacial period and could be key horizons for future correlations
Statistical Models for Solar Flare Interval Distribution in Individual Active Regions
This article discusses statistical models for solar flare interval
distribution in individual active regions. We analyzed solar flare data in 55
active regions that are listed in the GOES soft X-ray flare catalog. We discuss
some problems with a conventional procedure to derive probability density
functions from any data set and propose a new procedure, which uses the maximum
likelihood method and Akaike Information Criterion (AIC) to objectively compare
some competing probability density functions. We found that lognormal and
inverse Gaussian models are more likely models than the exponential model for
solar flare interval distribution in individual active regions. The results
suggest that solar flares do not occur randomly in time; rather, solar flare
intervals appear to be regulated by solar flare mechanisms. We briefly mention
a probabilistic solar flare forecasting method as an application of a solar

Consumer knowledge of and engagement with traditional takeaway and dark kitchen food outlets
Background
Dark kitchens – delivery-only food outlets operating through digital technology platforms – are a contemporary addition to the food environment. Some concerns have been raised the ability for local authorities to identify and regulate these businesses, with growing concern around the nutritional quality of foods, food safety practices and the impact on the local food environment. The present work explores consumer understanding of and engagement with dark kitchen establishments.
Methods
Healthy adults living in the United Kingdom completed an online survey comprising of questions measuring participant demographics, engagement with takeaways and dark kitchens, purchasing behaviours and decision making, and knowledge and understanding around dark kitchens. Questions were primarily closed-ended and quantitative. Data were analysed using descriptive statistics.
Results
In total, 2,023 participants (46.3 ± 16.7 years) completed the survey. Forty percent purchased a takeaway at least weekly, often through aggregator applications (e.g., Just Eat, Deliveroo). Food was mainly purchased as a treat (79.3%), for enjoyment of the food or taste (60.8%) and for convenience (58.2%). When ordering, consumers considered the taste (88.1%), quality (83.5%), value for money (77.8%), and familiarity with (68.1%) and reputation of the business (60.0%). Only 24.7% of participants had heard of dark kitchens and 9.1% had knowingly purchased from one. After reading a working definition, 54.9% of consumers said they would purchase from a dark kitchen but most would want to know explicitly that they were ordering from these businesses. A major concern was trust in the food safety and hygiene standards.
Conclusion
Consumers are unfamiliar with dark kitchens and are not aware of or confident in identifying these businesses. This confusion and concerns around food safety means dark kitchens are often viewed negatively. Consumers would prefer more transparency in where their foods are being prepared to allow for more informed decision-making
The value of carbon sequestration and storage in coastal habitats
Coastal margin habitats are globally significant in terms of their capacity to sequester and store carbon, but their continuing decline, due to environmental change and human land use decisions, is reducing their capacity to provide this ecosystem service. In this paper the UK is used as a case study area to develop methodologies to quantify and value the ecosystem service of blue carbon sequestration and storage in coastal margin habitats. Changes in UK coastal habitat area between 1900 and 2060 are documented, the long term stocks of carbon stored by these habitats are calculated, and the capacity of these habitats to sequester CO2 is detailed. Changes in value of the carbon sequestration service of coastal habitats are then projected for 2000–2060 under two scenarios, the maintenance of the current state of the habitat and the continuation of current trends of habitat loss. If coastal habitats are maintained at their current extent, their sequestration capacity over the period 2000–2060 is valued to be in the region of £1 billion UK sterling (3.5% discount rate). However, if current trends of habitat loss continue, the capacity of the coastal habitats both to sequester and store CO2 will be significantly reduced, with a reduction in value of around £0.25 billion UK sterling (2000–2060; 3.5% discount rate). If loss-trends due to sea level rise or land reclamation worsen, this loss in value will be greater. This case study provides valuable site specific information, but also highlights global issues regarding the quantification and valuation of carbon sequestration and storage. Whilst our ability to value ecosystem services is improving, considerable uncertainty remains. If such ecosystem valuations are to be incorporated with confidence into national and global policy and legislative frameworks, it is necessary to address this uncertainty. Identification and characterization of mechanistically distinct inducers of gamma-globin transcription
Inhibition of HbS polymerization is a major target for therapeutic approaches in sickle cell anemia. Toward this goal, initial efforts at pharmacological elevation of fetal hemoglobin (HbF) has shown therapeutic efficacy. In order to identify well-tolerated, novel agents that induce HbF in patients, we developed a high-throughput screening approach based on induction of gamma-globin gene expression in erythroid cells. We measured gamma-globin transcription in K562 cells transfected with either gamma promoter elements fused with the locus control region hypersensitivity site 2 and luciferase reporter gene (HS2 gamma) or a beta-yeast artificial chromosome in which the luciferase reporter gene was recombined into the gamma-globin coding sequences (gamma YAC). Corresponding pharmacological increases in HbF protein were confirmed in both K562 cells and in human primary erythroid progenitor cells. Approximately 186,000 defined chemicals and fungal extracts were evaluated for their ability to increase gamma gene transcription in either HS2 gamma or gamma YAC models. Eleven distinct classes of compounds were identified, the majority of which were active within 24-48 hr. The short chain hydroxamate-containing class generally exhibited delayed maximal activity, which continued to increase transcription up to 120 hr. The cyclic tetrapeptide OSI-2040 and the hydroxamates were shown to have histone deacetylase inhibitory activity. In primary hematopoietic progenitor cell cultures, OSI-2040 increased HbF by 4.5-fold at a concentration of only 40 nM, comparable to the effects of hydroxyurea at 100 microM. Promotion of testa rupture during garden cress germination involves seed compartment-specific expression and activity of pectin methylesterases
Pectin methylesterase (PME) controls the methylesterification status of pectins and thereby determines the biophysical properties of plant cell walls, which are important for tissue growth and weakening processes. We demonstrate here that tissue-specific and spatiotemporal alterations in cell wall pectin methylesterification occur during the germination of garden cress (Lepidium sativum). These cell wall changes are associated with characteristic expression patterns of PME genes and resultant enzyme activities in the key seed compartments CAP (micropylar endosperm) and RAD (radicle plus lower hypocotyl). Transcriptome and quantitative real-time reverse transcription-polymerase chain reaction analysis as well as PME enzyme activity measurements of separated seed compartments, including CAP and RAD, revealed distinct phases during germination. These were associated with hormonal and compartment-specific regulation of PME group 1, PME group 2, and PME inhibitor transcript expression and total PME activity. The regulatory patterns indicated a role for PME activity in testa rupture (TR). Consistent with a role for cell wall pectin methylesterification in TR, treatment of seeds with PME resulted in enhanced testa permeability and promoted TR. Mathematical modeling of transcript expression changes in germinating garden cress and Arabidopsis (Arabidopsis thaliana) seeds suggested that group 2 PMEs make a major contribution to the overall PME activity rather than acting as PME inhibitors. It is concluded that regulated changes in the degree of pectin methylesterification through CAP- and RAD-specific PME and PME inhibitor expression play a crucial role during Brassicaceae seed germination
The T2K Side Muon Range Detector
The T2K experiment is a long baseline neutrino oscillation experiment aiming
to observe the appearance of {\nu} e in a {\nu}{\mu} beam. The {\nu}{\mu} beam
is produced at the Japan Proton Accelerator Research Complex (J-PARC), observed
with the 295 km distant Super- Kamiokande Detector and monitored by a suite of
near detectors at 280m from the proton target. The near detectors include a
magnetized off-axis detector (ND280) which measures the un-oscillated neutrino
flux and neutrino cross sections. The present paper describes the outermost
component of ND280 which is a side muon range detector (SMRD) composed of
scintillation counters with embedded wavelength shifting fibers and Multi-Pixel
Photon Counter read-out. The components, performance and response of the SMRD
