    In this study, the authors used a qualitative method in which there were no calculations that required going directly to the location, simply by looking for references and several journals that were used as a basis. Then this method is motivated by library management governance, developing book collections, supporting and inhibiting factors, and increasing reading interest. Then also in terms of human resources, if not managed properly, these natural resources will decline compared to other resources. So both librarians and visitors and human resources (HR) must support each other in improving library management. That is why the quality level of human consciousness is important because school libraries and university libraries are an integral part of supporting the educational process. With the synergy of intellectual quality and rationality, there is no degradation of education, elements that must be fulfilled such as service, quality, abilities, and good attitudes from librarians and visitors, and the existence of library management functions with the fulfillment of Human Resources (HR) and collections that meet library standards based on Law No. 43 of 2007

    ABSTRAK Berdasarkan analisis penulis bahwasanya kita tahu bahwa perpustakaan merupakan sumber rujukan pengetahuan bisa dikatakan sebagai mother of knowledge yang kali ini sistem yang makin maju dan mudah untuk menemukan solusinya dengan kejanggihan teknologi, maka perpustakaan ini yang mempunyai sistem SLiMS akan diimpelemntasikan dalam perpustakaan khusus yang berada pada lembaga kampus UIN Raden Intan Lampung yaitu “perpustakaan fakultas ushuluddin ” sistem penelitian kali ini adalah menggunakan model SDLC ( Software Development Life Cycle ) , adalah sebuah pengubahan dan pembuatan sistem serta dalam model metodologinya digunakan untuk mengembangkan sistem, terdiri dari beberapa tahap : rencana (Planning), analisis( analysis), desain (design), implementasi ( implementation), uji coba ( testing) dan pengelolaan ( maintenance)pada hasil uji coba dengaan estimasi waktu dua bulan adalah pemasangan aplikasi SLiMS Bulian 9.4.1 yang sudah dipasangkannya di perpustakaan ushuluddin dengan menggunakan komputer,dan juga memiliki hasil bahwasanya pada pemasangan aplikasi SLiMS Bulian 9.4.1 pada pengembangannya ada beberapa yang error tapi tidak menghambat pada kinerja sistem kemduian melakukan uji coba pemanfaatan aplikasi pada pengunjung dengan respon yang cukup baik.tahap develop di bab 3 itu berbedan dengan bab 4 walaupun terdapat tulisan yang sama ,yang membedakan dari hal tersebut yaitu jika pdada bab 3 pada tahap develop yaitu perjalanan pemasangan web SLiMS pada laptop user atau penulis sedangkan develop bab 4 itu diuji cobakan pada komputer yang berada dikomputer lokasi perpustakaan ushuluddin tersebut.Berdasarkan pada hasil penelitian kali ini pada pengembangan dan pembahasan maka dapat kita simpulkan bahwa rancang bangun aplikasi SLiMS Bulian 9.4.1 yang sudah dikembangkan sangat layak digunakan sebagai alat aplikasi digital,yang dikembangkan telah memenuhi kriteria kelayakan baik dan mendapat respon positif baik bagi pustakawan maupun pemustaka iii meningkatkan layanan perpustakaan dan jawabannya tentu dapat meningkatkan dengan hasil responden tersebut.kemudian penulis mengujicobakan SLiMS menggunanakan usability yang terdapat 30 pertanyaan yang menyangkut pada pemanfaatan web SLiMS,pada 10 responden itu menghasilkan nilai 85,6% itu merupakan sangat layak maka dalam hal ini dapat meningkatkan layanan perpustakaan tersebut. Kemudian pada web SLiMS memiliki keterbatasan yaitu hanya bisa menjalankan pada satu perangkat laptop admin jika keaadan offline,dan juga dalam memasukan database masih menggunakan manual. Tapi SLiMS tersebut sangat berguna dan bermanfaat dikarenakan FOSS free open source. Kata Kunci : Perpustakaan,SLiMS iv ABSTRACT Based on the writer’s analysis that we know that the library is a reference source of knowledge which can be said to be the mother of knowledge which this time is a more advanced system and it is easy to find solutions with technological sophistication, then this library which has the SLiMS system will be implemented in a special library located at the UIN Raden Intan Lampung campus institution, namely "the ushuluddin faculty library". : planning, analysis, design, implementation, testing and maintenance on the results of the trial with an estimated time of two months is the installation of the SLiMS Bulian 9.4.1 application which has been installed in the ushuluddin library using a computer, and also has the result that in installing the SLiMS Bulian 9.4.1 application in its development there were several errors but did not hinder system performance then tested the use of the application on visitors with a fairly good response. Chapter 3 is different from chapter 4, although there is the same writing, what distinguishes this is that in chapter 3 at the develop stage, namely the journey of installing the SLiMS web on the user's or writer's laptop, while develop chapter 4 was tested on a computer that is on the computer located at the ushuluddin library. both librarians and users improve library services and the answers can certainly improve with the results of the respondents. Then on the SLiMS web it has limitations, namely that it can only run on one admin laptop device if it is offline, and also in entering the database it still uses manual. Dengan rumusan masalah yaitu : (1) Bagaimana penggunaan percepatan Surat Penyataan Tanggung Jawab Mutlak (SPTJM) sebagai pengganti akta nikah dalam pembuatan akta kelahiran di capil Bangkinang, (2) Bagaimana Analisa terhadap penggunaan Surat Penyataan Tanggung Jawab Mutlak (SPTJM) sebagai pengganti akta nikah dalam pembuatan akta kelahiran bagi pelaku nikah sirri perspektif maqashid syari’ah. Penelitian ini menggunakan metode penelitian lapangan (field research), dengan langsung datang ke lokasi penelitian di kantor kependudukan dan catatan sipil bangkinang. Dalam penelitian digunakan pendekatan deskriptif kualitatif. Pengumpulan data dilakukan dengan cara wawancara bebas terpimpin, observasi, dan dokumentasi. Dari penelitian yang telah dilaksanakan maka penulis dapat menyimpulkan bahwa Penggunaan Surat Pernyataan Tanggung Jawab Mutlak bagi pelaku nikah sirri boleh berdasarkan PERMENDAGRI No 9 tahun 2016 pasal 4 ayat 2 menyebutkan bahwasannya ketika ada seorang pasangan suami istri yang ingin membuatkan Akta Kelahiran anaknya tetapi tidak memiliki buku atau Akta Nikah maka dapat diganti dengan menggunakan Surat Pernyataan Tanggung Jawab Mutlak kebenaran sebagai pasangan suami istri, kebenaran pengunaan SPTJM sebagai pasangan suami isteri sebagai pengganti akta nikah untuk sebagai inovasi upaya percepatan dalam proses pembuatan akta kelahiran dan peningkatan kepemilikan akta kelahiran orang dewasa yang status hubungan orang tua pada KK menunjukkan status hubungan perkawinan sebagai pasangan suami isteri dan ditinjau dari perspektif maqashid al-syariah sebagai unsur pokok tujuan hukum islam penggunaan SPTJM sebagai solusi menjadi cara perkembangan nilai-nilai yang terkandung dalam hukum islam untuk mengatasi anak hasil pernikahan sirri demi kemaslahatan anak dan istri yang terdapat pada pasal 4 ayat 2.     Consistent imprints of elevation, soil temperature and moisture on plant and arthropod communities across two subarctic landscapes

    1. Factors shaping arthropod and plant community structure at fine spatial scales are poorly understood. This includes microclimate, which likely plays a large role in shaping local community patterns, especially in heterogeneous landscapes characterised by high microclimatic variability in space and in time.2. We explored differences in local microclimatic conditions and regional species pools in two subarctic regions: Kilpisj & auml;rvi in north-west Finland and Varanger in north-east Norway. We then investigated the relationship between fine-scale climatic variation and local community characteristics (species richness and abundance) among plants and arthropods, differentiating the latter into two groups: flying and ground-dwelling arthropods collected by Malaise and pitfall traps, respectively. Arthropod taxa were identified through DNA metabarcoding. Finally, we examined if plant richness can be used to predict patterns in arthropod communities.3. Variation in soil temperature, moisture and snow depth proved similar between regions, despite differences in absolute elevation. For each group of organisms, we found that about half of the species were shared between Kilpisj & auml;rvi and Varanger, with a quarter unique to each region.4. Plants and arthropods responded largely to the same drivers. The richness and abun-dance of both groups decreased as elevation increased and were positively correlated with higher soil moisture and temperature values. Plant species richness was a poor predictor of local arthropod richness, in particular for ground-dwelling arthropods.5. Our results reveal how microclimatic variation within each region carves pro-nounced, yet consistent patterns in local community richness and abundance out of a joint species pool

    Low antioxidant concentrations impact on multiple signalling pathways in Arabidopsis thaliana partly through NPR1

    Production of reactive oxygen species (ROS) is linked to signalling in both developmental and stress responses. The level of ROS is controlled by both production and removal through various scavengers including ascorbic acid and glutathione. Here, the role of low ascorbic acid or glutathione concentrations was investigated on ozone-induced cell death, defence signalling, and developmental responses. Low ascorbic acid concentrations in vtc1 activated expression of salicylic acid (SA)-regulated genes, a response found to be dependent on the redox-regulated transcriptional co-regulator NPR1. In contrast, low glutathione concentrations in cad2 or pad2 reduced expression of SA-regulated genes. Testing different responses to jasmonic acid (JA) revealed the presence of at least two separate JA signalling pathways. Treatment of the vtc1 mutant with JA led to hyper-induction of MONODEHYDROASCORBATE REDUCTASE3, indicating that low ascorbic acid concentrations prime the response to JA. Furthermore, NPR1 was found to be a positive regulator of JA-induced expression of MDHAR3 and TAT3. The vtc1 and npr1 mutants were sensitive to glucose inhibition of seed germination; an opposite response was found in cad2 and pad2. Overall, low ascorbic acid concentrations mostly led to opposite phenotypes to low glutathione concentrations, and both antioxidants interacted with SA and JA signalling pathways

    Isolation and characterization of low-sulphur-tolerant mutants of Arabidopsis

    Sulphur is an essential element for plant growth and development as well as for defence against biotic and abiotic stresses. Increasing sulphate utilization efficiency (SUE) is an important issue for crop improvement. Little is known about the genetic determinants of sulphate utilization efficiency. No gain-of-function mutants with improved SUE have been reported to date. Here the isolation and characterization of two low-sulphur-tolerant mutants, sue3 and sue4 are reported using a high-throughput genetic screen where a ‘sulphur-free’ solid medium was devised to give the selection pressure necessary to suppress the growth of the wild-type seedlings. Both mutants showed improved tolerance to low sulphur conditions and well-developed root systems. The mutant phenotype of both sue3 and sue4 was specific to sulphate deficiency and the mutants displayed enhanced tolerance to heavy metal and oxidative stress. Genetic analysis revealed that sue3 was caused by a single recessive nuclear mutation while sue4 was caused by a single dominant nuclear mutation. The recessive locus in sue3 is the previously identified VirE2-interacting Protein 1. The dominant locus in sue4 is a function-unknown locus activated by the four enhancers on the T-DNA. The function of SUE3 and SUE4 in low sulphur tolerance was confirmed either by multiple mutant alleles or by recapitulation analysis. Taken together, our results demonstrate that this genetic screen is a reasonable approach to isolate Arabidopsis mutants with improved low sulphur tolerance and potentially with enhanced sulphate utilization efficiency. The two loci identified in sue3 and sue4 should assist in understanding the molecular mechanisms of low sulphur tolerance

    Subcellular compartmentation of glutathione in dicotyledonous plants

    This study describes the subcellular distribution of glutathione in roots and leaves of different plant species (Arabidopsis, Cucurbita, and Nicotiana). Glutathione is an important antioxidant and redox buffer which is involved in many metabolic processes including plant defense. Thus information on the subcellular distribution in these model plants especially during stress situations provides a deeper insight into compartment specific defense reactions and reflects the occurrence of compartment specific oxidative stress. With immunogold cytochemistry and computer-supported transmission electron microscopy glutathione could be localized in highest contents in mitochondria, followed by nuclei, peroxisomes, the cytosol, and plastids. Within chloroplasts and mitochondria, glutathione was restricted to the stroma and matrix, respectively, and did not occur in the lumen of cristae and thylakoids. Glutathione was also found at the membrane and in the lumen of the endoplasmic reticulum. It was also associated with the trans and cis side of dictyosomes. None or only very little glutathione was detected in vacuoles and the apoplast of mesophyll and root cells. Additionally, glutathione was found in all cell compartments of phloem vessels, vascular parenchyma cells (including vacuoles) but was absent in xylem vessels. The specificity of this method was supported by the reduction of glutathione labeling in all cell compartments (up to 98%) of the glutathione-deficient Arabidopsis thaliana rml1 mutant. Additionally, we found a similar distribution of glutathione in samples after conventional fixation and rapid microwave-supported fixation. Thus, indicating that a redistribution of glutathione does not occur during sample preparation. Summing up, this study gives a detailed insight into the subcellular distribution of glutathione in plants and presents solid evidence for the accuracy and specificity of the applied method

    Identification of genes required for Cf-dependent hypersensitive cell death by combined proteomic and RNA interfering analyses

    Identification of hypersensitive cell death (HCD) regulators is essential to dissect the molecular mechanisms underlying plant disease resistance. In this study, combined proteomic and RNA interfering (RNAi) analyses were employed to identify genes required for the HCD conferred by the tomato resistance gene Cf-4 and the Cladosporium fulvum avirulence gene Avr4. Forty-nine proteins differentially expressed in the tomato seedlings mounting and those not mounting Cf-4/Avr4-dependent HCD were identified through proteomic analysis. Among them were a variety of defence-related proteins including a cysteine protease, Pip1, an operative target of another C. fulvum effector, Avr2. Additionally, glutathione-mediated antioxidation is a major response to Cf-4/Avr4-dependent HCD. Functional analysis through tobacco rattle virus-induced gene silencing and transient RNAi assays of the chosen 16 differentially expressed proteins revealed that seven genes, which encode Pip1 homologue NbPip1, a SIPK type MAP kinase Nbf4, an asparagine synthetase NbAsn, a trypsin inhibitor LeMir-like protein NbMir, a small GTP-binding protein, a late embryogenesis-like protein, and an ASR4-like protein, were required for Cf-4/Avr4-dependent HCD. Furthermore, the former four genes were essential for Cf-9/Avr9-dependent HCD; NbPip1, NbAsn, and NbMir, but not Nbf4, affected a nonadaptive bacterial pathogen Xanthomonas oryzae pv. oryzae-induced HCD in Nicotiana benthamiana. These data demonstrate that Pip1 and LeMir may play a general role in HCD and plant immunity and that the application of combined proteomic and RNA interfering analyses is an efficient strategy to identify genes required for HCD, disease resistance, and probably other biological processes in plants

    The Lectin Receptor Kinase LecRK-I.9 Is a Novel Phytophthora Resistance Component and a Potential Host Target for a RXLR Effector

    In plants, an active defense against biotrophic pathogens is dependent on a functional continuum between the cell wall (CW) and the plasma membrane (PM). It is thus anticipated that proteins maintaining this continuum also function in defense. The legume-like lectin receptor kinase LecRK-I.9 is a putative mediator of CW-PM adhesions in Arabidopsis and is known to bind in vitro to the Phytophthora infestans RXLR-dEER effector IPI-O via a RGD cell attachment motif present in IPI-O. Here we show that LecRK-I.9 is associated with the plasma membrane, and that two T-DNA insertions lines deficient in LecRK-I.9 (lecrk-I.9) have a ‘gain-of-susceptibility’ phenotype specifically towards the oomycete Phytophthora brassicae. Accordingly, overexpression of LecRK-I.9 leads to enhanced resistance to P. brassicae. A similar ‘gain-of-susceptibility’ phenotype was observed in transgenic Arabidopsis lines expressing ipiO (35S-ipiO1). This phenocopy behavior was also observed with respect to other defense-related functions; lecrk-I.9 and 35S-ipiO1 were both disturbed in pathogen- and MAMP-triggered callose deposition. By site-directed mutagenesis, we demonstrated that the RGD cell attachment motif in IPI-O is not only essential for disrupting the CW-PM adhesions, but also for disease suppression. These results suggest that destabilizing the CW-PM continuum is one of the tactics used by Phytophthora to promote infection. As countermeasure the host may want to strengthen CW-PM adhesions and the novel Phytophthora resistance component LecRK-I.9 seems to function in this process

    (Homo)glutathione Deficiency Impairs Root-knot Nematode Development in Medicago truncatula

    Root-knot nematodes (RKN) are obligatory plant parasitic worms that establish and maintain an intimate relationship with their host plants. During a compatible interaction, RKN induce the redifferentiation of root cells into multinucleate and hypertrophied giant cells essential for nematode growth and reproduction. These metabolically active feeding cells constitute the exclusive source of nutrients for the nematode. Detailed analysis of glutathione (GSH) and homoglutathione (hGSH) metabolism demonstrated the importance of these compounds for the success of nematode infection in Medicago truncatula. We reported quantification of GSH and hGSH and gene expression analysis showing that (h)GSH metabolism in neoformed gall organs differs from that in uninfected roots. Depletion of (h)GSH content impaired nematode egg mass formation and modified the sex ratio. In addition, gene expression and metabolomic analyses showed a substantial modification of starch and γ-aminobutyrate metabolism and of malate and glucose content in (h)GSH-depleted galls. Interestingly, these modifications did not occur in (h)GSH-depleted roots. These various results suggest that (h)GSH have a key role in the regulation of giant cell metabolism.
