    Effective detection of proteins following electrophoresis using extracts of locally available food species

    Procedures in life sciences research laboratories often require chemicals and plasticware that are costly, toxic or pose a risk to the environment. Therefore, sustainable alternatives would be of interest, provided that they generate suitable data quality. Coomassie blue and silver staining are the most widely used methods for detecting proteins following electrophoresis in the laboratory. However, their use presents challenges in terms of safety and waste management. In the current study, aqueous extracts were prepared from a series of common food species and evaluated as alternative stains for protein detection. Beets, blueberries, purple cabbage, raspberries and strawberries were employed to stain identical proteins separated under the same conditions in electrophoresis gels. Extracts of the first two species resulted in protein bands that were detectable through visible light transillumination, whereas extracts from all five species generated specific protein bands under ultraviolet light. The raspberry-derived extract was selected for further study based on the brightness of the fluorescent protein bands and minimal background staining. For both bovine serum albumin and lysozyme at 2.5 μg and 0.5 μg protein per band, the mean signal intensities obtained with raspberry extract staining were just below half of those obtained with Coomassie blue. Furthermore, the mean intensities using raspberry extract were equivalent to those obtained using Coomassie blue in the detection of 0.1 μg protein. Therefore, raspberry could be used to produce an effective stain for the routine laboratory analysis of proteins

    Closing in on the sources of cosmic reionization: first results from the GLASS-JWST program

    The escape fraction of Lyman-continuum (LyC) photons (fescf_{esc}) is a key parameter for determining the sources of cosmic reionization at z6z\geq 6. At these redshifts, owing to the opacity of the intergalactic medium, the LyC emission cannot be measured directly. However, LyC leakers during the epoch of reionization could be identified using indirect indicators that have been extensively tested at low and intermediate redshifts. These include a high [OIII]/[OII] flux ratio, high star-formation surface density, and compact sizes. In this work, we present observations of 29 4.5z84.5 \leq z \leq 8 gravitationally lensed galaxies in the Abell 2744 cluster field. From a combined analysis of JWST-NIRSpec and NIRCam data, we accurately derived their physical and spectroscopic properties: our galaxies have low masses (log(M)8.5)(\log(M_\star)\sim 8.5), blue UV spectral slopes (β2.1\beta \sim -2.1), compact sizes (re0.30.5r_e \sim 0.3-0.5 kpc), and high [OIII]/[OII] flux ratios. We confirm that these properties are similar to those characterizing low-redshift LyC leakers. Indirectly inferring the fraction of escaping ionizing photons, we find that more than 80% of our galaxies have predicted fescf_{esc} values larger than 0.05, indicating that they would be considered leakers. The average predicted fescf_{esc} value of our sample is 0.12, suggesting that similar galaxies at z6z\geq 6 have provided a substantial contribution to cosmic reionization.Comment: Accepted for publication in the 4.     Building leaders for the UN Ocean Science Decade : a guide to supporting early career women researchers within academic marine research institutions

    Diverse and inclusive marine research is paramount to addressing ocean sustainability challenges in the 21st century, as envisioned by the UN Decade of Ocean Science for Sustainable Development. Despite increasing efforts to diversify ocean science, women continue to face barriers at various stages of their career, which inhibits their progression to leadership within academic institutions. In this perspective, we draw on the collective experiences of thirty-four global women leaders, bolstered by a narrative review, to identify practical strategies and actions that will help empower early career women researchers to become the leaders of tomorrow. We propose five strategies: (i) create a more inclusive culture, (ii) ensure early and equitable career development opportunities for women ECRs, (iii) ensure equitable access to funding for women ECRs, (iv) offer mentoring opportunities and, (v) create flexible, family-friendly environments. Transformational, meaningful, and lasting change will only be achieved through commitment and collaborative action across various scales and by multiple stakeholders.Peer reviewe

    Detection and long-term quantification of methane emissions from an active landfill

    Landfills are a significant source of fugitive methane (CH4) emissions, which should be precisely and regularly monitored to reduce and mitigate net greenhouse gas emissions. In this study, we present long-term, in situ, near-surface, mobile atmospheric CH4 mole fraction measurements (complemented by meteorological measurements from a fixed station) from 21 campaigns that cover approximately 4 years from September 2016 to December 2020. These campaigns were utilized to regularly quantify the total CH4 emissions from an active landfill in France. We use a simple atmospheric inversion approach based on a Gaussian plume dispersion model to derive CH4 emissions. Together with the measurements near the soil surface, mainly dedicated to the identification of sources within the landfill, measurements of CH4 made on the landfill perimeter (near-field) helped us to identify the main emission areas and to provide some qualitative insights about the rank of their contributions to total emissions from the landfill. The two main area sources correspond, respectively, to a covered waste sector with infrastructure with sporadic leakages (such as wells, tanks, pipes, etc.) and to the last active sector receiving waste during most of the measurement campaigns. However, we hardly managed to extract a signal representative of the overall landfill emissions from the near-field measurements, which limited our ability to derive robust estimates of the emissions when assimilating them in the atmospheric inversions. The analysis shows that the inversions based on the measurements from a remote road further away from the landfill (far-field) yielded reliable estimates of the total emissions but provided less information on the spatial variability of emissions within the landfill. This demonstrates the complementarity between the near- and far-field measurements. According to these inversions, the total CH4 emissions have a large temporal variability and range from ∼ 0.4 to ∼ 7 t CH4 d−1, with an average value of ∼ 2.1 t CH4 d−1. We find a weak negative correlation between these estimates of the CH4 emissions and atmospheric pressure for the active landfill. However, this weak emission–pressure relationship is based on a relatively small sample of reliable emission estimates with large sampling gaps. More frequent robust estimations are required to better understand this relationship for an active landfill.</p

    Drosophila evolution over space and time (DEST):A new population genomics resource

    Drosophila melanogaster is a leading model in population genetics and genomics, and a growing number of whole-genome datasets from natural populations of this species have been published over the last years. A major challenge is the integration of disparate datasets, often generated using different sequencing technologies and bioinformatic pipelines, which hampers our ability to address questions about the evolution of this species. Here we address these issues by developing a bioinformatics pipeline that maps pooled sequencing (Pool-Seq) reads from D. melanogaster to a hologenome consisting of fly and symbiont genomes and estimates allele frequencies using either a heuristic (PoolSNP) or a probabilistic variant caller (SNAPE-pooled). We use this pipeline to generate the largest data repository of genomic data available for D. melanogaster to date, encompassing 271 previously published and unpublished population samples from over 100 locations in > 20 countries on four continents. Several of these locations have been sampled at different seasons across multiple years. This dataset, which we call Drosophila Evolution over Space and Time (DEST), is coupled with sampling and environmental meta-data. A web-based genome browser and web portal provide easy access to the SNP dataset. We further provide guidelines on how to use Pool-Seq data for model-based demographic inference. Our aim is to provide this scalable platform as a community resource which can be easily extended via future efforts for an even more extensive cosmopolitan dataset.     Home parenteral nutrition provision modalities for chronic intestinal failure in adult patients:An international survey

    Background & aims: The safety and effectiveness of a home parenteral nutrition (HPN) program depends both on the expertise and the management approach of the HPN center. We aimed to evaluate both the approaches of different international HPN-centers in their provision of HPN and the types of intravenous supplementation (IVS)-admixtures prescribed to patients with chronic intestinal failure (CIF). Methods: In March 2015, 65 centers from 22 countries enrolled 3239 patients (benign disease 90.1%, malignant disease 9.9%), recording the patient, CIF and HPN characteristics in a structured database. The HPN-provider was categorized as health care system local pharmacy (LP) or independent home care company (HCC). The IVS-admixture was categorized as fluids and electrolytes alone (FE) or parenteral nutrition, either commercially premixed (PA) or customized to the individual patient (CA), alone or plus extra FE (PAFE or CAFE). Doctors of HPN centers were responsible for the IVS prescriptions. Results: HCC (66%) was the most common HPN provider, with no difference noted between benign-CIF and malignant-CIF. LP was the main modality in 11 countries; HCC prevailed in 4 European countries: Israel, USA, South America and Oceania (p < 0.001). IVS-admixture comprised: FE 10%, PA 17%, PAFE 17%, CA 38%, CAFE 18%. PA and PAFE prevailed in malignant-CIF while CA and CAFE use was greater in benign-CIF (p < 0.001). PA + PAFE prevailed in those countries where LP was the main HPN-provider and CA + CAFE prevailed where the main HPN-provider was HCC (p < 0.001). Conclusions: This is the first study to demonstrate that HPN provision and the IVS-admixture differ greatly among countries, among HPN centers and between benign-CIF and cancer-CIF. As both HPN provider and IVS-admixture types may play a role in the safety and effectiveness of HPN therapy, criteria to homogenize HPN programs are needed so that patients can have equal access to optimal CIF care

    An intelligent interactive system for delivering individualized information to patients

    This paper is a report on the first phase of a long-term, interdisciplinary project whose goal is to increase the overall effectiveness of physicians ’ time, and thus the quality of health care, by improving the information exchange between physicians and patients in clinical settings. We are focusing on patients with long-term and chronic conditions, initially on migraine patients, who require periodic interaction with their physicians for effective management of their condition. We are using medical informatics to focus on the information needs of patients, as well as of physicians, and to address problems of information exchange. This requires understanding patients’ concerns to design an appropriate system, and using state-of-the-art artificial intelligence techniques to build an interactive explanation system. In contrast to many other knowledge-based systems, our system’s design is based on empirical data on actual information needs. We used ethnographic techniques to observe explanations actually given in clinic settings, and to conduct interviews with migraine sufferers and physicians.     Amyloids - A functional coat for microorganisms

    Amyloids are filamentous protein structures ~10 nm wide and 0.1–10 µm long that share a structural motif, the cross-β structure. These fibrils are usually associated with degenerative diseases in mammals. However, recent research has shown that these proteins are also expressed on bacterial and fungal cell surfaces. Microbial amyloids are important in mediating mechanical invasion of abiotic and biotic substrates. In animal hosts, evidence indicates that these protein structures also contribute to colonization by activating host proteases that are involved in haemostasis, inflammation and remodelling of the extracellular matrix. Activation of proteases by amyloids is also implicated in modulating blood coagulation, resulting in potentially life-threatening complications.
