    Functional connectivity and amplitude of low-frequency fluctuations changes in people with complete subacute and chronic spinal cord injury.

    After spinal cord injury (SCI), reorganization processes and changes in brain connectivity occur. Besides the sensorimotor cortex, the subcortical areas are strongly involved in motion and executive control. This exploratory study focusses on the cerebellum and vermis. Resting-state functional magnetic resonance imaging (fMRI) was performed. Between-group differences were computed using analysis of covariance and post-hoc tests for the seed-based connectivity measure with vermis and cerebellum as regions of interest. Twenty participants with complete SCI (five subacute SCI, 15 with chronic SCI) and 14 healthy controls (HC) were included. Functional connectivity (FC) was lower in all subjects with SCI compared with HC in vermis IX, right superior frontal gyrus (pFDR = 0.008) and right lateral occipital cortex (pFDR = 0.036). In addition, functional connectivity was lower in participants with chronic SCI compared with subacute SCI in bilateral cerebellar crus I, left precentral- and middle frontal gyrus (pFDR = 0.001). Furthermore, higher amplitude of low-frequency fluctuations (ALFF) was found in the left thalamus in individuals with subacute SCI (pFDR = 0.002). Reduced FC in SCI indicates adaptation with associated deficit in sensory and motor function. The increased ALFF in subacute SCI might reflect reorganization processes in the subacute phase

    Estado de hidratação em atletas de Futebol americano: uma análise pré e pós-treino

    O presente estudo tem por objetivo analisar o estado de hidratação dos atletas da equipe de futebol americano (na modalidade Full Pads) pré e pós-treino pelos marcadores: massa corporal, coloração e gravidade específica (GE) da urina; vez que o estado de hidratação pode ser um fator determinante para a garantia de um ideal desempenho dos praticantes de atividade física. Peso e urina coletados para análise nos momentos pré e pós-treino, não tendo a comissão técnica qualquer regulação/controle sobre o consumo de líquidos durante o exercício. Os resultados da GE demonstraram pré-treino que 35% dos atletas estavam em condições de desidratação mínima e 65% com desidratação grave (≥1.030 g. ml-1) e no período pós-treino 13% e 87% respectivamente, havendo pela análise estatística diferença significativa (p=0.001) na GE pós-treino. A coloração da urina apresentou pré-treino 87% dos atletas no grupo hidratado, mínimo ou significativo e 13% no grupo grave, pós-treino 22% e 78% respectivamente, com diferença significativa neste segundo momento (p=0.000). As médias de massa corporal não apresentaram variação significativa (p=0.229). Concluiu-se que a inexistência de um prévio controle do estado de hidratação dos atletas, pode ser causa direta de déficit de desempenho para importantes valências físicas como capacidade anaeróbica e força. ABSTRACTHydration status in american football athletes: a pre and post-workout analysisThis study aims to analyze the hydration status of athletes of american football team (in Full Pads mode) pre and post workout by markers: body mass, staining and urine specific gravity (SG); Since the hydration status can be a determining factor for ensuring optimal performance of physical activity practitioners. Weight and urine collected for analysis in the pre and post-workout, not having the technical committee any regulation / control of fluid intake during exercise. The results showed SG pre-training 35% of athletes were minimal dehydrating conditions to 65% with severe dehydration (≥1,030 g. ml-1) and the post-training period 13% and 87% respectively, with the analysis statistical significant difference (p=0.001) in the post-workout SG. The color of urine presented pre-training 87% of the athletes in the hydrated group, minimal or significant, and 13% in the severe group, after training 22% and 78% respectively, with significant difference in this second stage (p=0.000). The body mass averages showed no significant variation (p=0.229). It was concluded that the lack of a previous control of hydration status of athletes, can be a direct cause of performance deficit to important physical valences as anaerobic capacity and strength

    HSPVdb—the Human Short Peptide Variation Database for improved mass spectrometry-based detection of polymorphic HLA-ligands

    T cell epitopes derived from polymorphic proteins or from proteins encoded by alternative reading frames (ARFs) play an important role in (tumor) immunology. Identification of these peptides is successfully performed with mass spectrometry. In a mass spectrometry-based approach, the recorded tandem mass spectra are matched against hypothetical spectra generated from known protein sequence databases. Commonly used protein databases contain a minimal level of redundancy, and thus, are not suitable data sources for searching polymorphic T cell epitopes, either in normal or ARFs. At the same time, however, these databases contain much non-polymorphic sequence information, thereby complicating the matching of recorded and theoretical spectra, and increasing the potential for finding false positives. Therefore, we created a database with peptides from ARFs and peptide variation arising from single nucleotide polymorphisms (SNPs). It is based on the human mRNA sequences from the well-annotated reference sequence (RefSeq) database and associated variation information derived from the Single Nucleotide Polymorphism Database (dbSNP). In this process, we removed all non-polymorphic information. Investigation of the frequency of SNPs in the dbSNP revealed that many SNPs are non-polymorphic “SNPs”. Therefore, we removed those from our dedicated database, and this resulted in a comprehensive high quality database, which we coined the Human Short Peptide Variation Database (HSPVdb). The value of our HSPVdb is shown by identification of the majority of published polymorphic SNP- and/or ARF-derived epitopes from a mass spectrometry-based proteomics workflow, and by a large variety of polymorphic peptides identified as potential T cell epitopes in the HLA-ligandome presented by the Epstein–Barr virus cells

    An Anomalous Type IV Secretion System in Rickettsia Is Evolutionarily Conserved

    Bacterial type IV secretion systems (T4SSs) comprise a diverse transporter family functioning in conjugation, competence, and effector molecule (DNA and/or protein) translocation. Thirteen genome sequences from Rickettsia, obligate intracellular symbionts/pathogens of a wide range of eukaryotes, have revealed a reduced T4SS relative to the Agrobacterium tumefaciens archetype (vir). However, the Rickettsia T4SS has not been functionally characterized for its role in symbiosis/virulence, and none of its substrates are known.Superimposition of T4SS structural/functional information over previously identified Rickettsia components implicate a functional Rickettsia T4SS. virB4, virB8 and virB9 are duplicated, yet only one copy of each has the conserved features of similar genes in other T4SSs. An extraordinarily duplicated VirB6 gene encodes five hydrophobic proteins conserved only in a short region known to be involved in DNA transfer in A. tumefaciens. virB1, virB2 and virB7 are newly identified, revealing a Rickettsia T4SS lacking only virB5 relative to the vir archetype. Phylogeny estimation suggests vertical inheritance of all components, despite gene rearrangements into an archipelago of five islets. Similarities of Rickettsia VirB7/VirB9 to ComB7/ComB9 proteins of epsilon-proteobacteria, as well as phylogenetic affinities to the Legionella lvh T4SS, imply the Rickettsiales ancestor acquired a vir-like locus from distantly related bacteria, perhaps while residing in a protozoan host. Modern modifications of these systems likely reflect diversification with various eukaryotic host cells.We present the rvh (Rickettsiales vir homolog) T4SS, an evolutionary conserved transporter with an unknown role in rickettsial biology. This work lays the foundation for future laboratory characterization of this system, and also identifies the Legionella lvh T4SS as a suitable genetic model

    An estimate of the number of tropical tree species

    The high species richness of tropical forests has long been recognized, yet there remains substantial uncertainty regarding the actual number of tropical tree species. Using a pantropical tree inventory database from closed canopy forests, consisting of 657,630 trees belonging to 11,371 species, we use a fitted value of Fisher’s alpha and an approximate pantropical stem total to estimate the minimum number of tropical forest tree species to fall between ∼40,000 and ∼53,000, i.e. at the high end of previous estimates. Contrary to common assumption, the Indo-Pacific region was found to be as species-rich as the Neotropics, with both regions having a minimum of ∼19,000–25,000 tree species. Continental Africa is relatively depauperate with a minimum of ∼4,500–6,000 tree species. Very few species are shared among the African, American, and the Indo-Pacific regions.
