Determining the barriers to the use of post-match fatigue monitoring in the rugby codes: A Concept Mapping study.
Topics in Exercise Science and Kinesiology Volume 5: Issue 1, Article 3, 2024. The rugby codes (i.e., rugby union, rugby league, rugby sevens [termed ârugbyâ]) are team-sports that impose complex physical demands upon players which in-turn, leads to domain-specific fatigue (e.g., neuromuscular, cardio-autonomic). Quantifying post-match fatigue through various methods and metrics is important to monitor player fatigue status, which influences training readiness. The specific and general barriers limiting the use of post-match fatigue monitoring in rugby are not presently known. Therefore, the aims of this study were to identify specific and general barriers (clusters of specific barriers) to the use of post-match fatigue monitoring methods and metrics in rugby across the domains of neuromuscular performance, cardio-autonomic, tissue biomarker, and self-reported fatigue, and which of these specific barriers were considered important to overcome and feasible to overcome. An international cohort of subject matter experts (SME) in rugby completed a two-round online questionnaire survey (Round One; n = 42, Round Two; n = 13), with the responses collected and analysed using Concept Mapping. Specific barrier statements were generated based on the SME responses to Round One, which were structured and then rated by the SME for importance to overcome and feasibility to overcome in Round Two. Five clusters of specific barriers (representing the general barriers) were determined based on analyses of the SME responses: 1. âBudget and Equipmentâ, 2. âData and Testing Considerationsâ, 3. âPlayer and Coach Perceptionsâ, 4. âTest Appropriatenessâ, and 5. âTime and Spaceâ. For both importance to overcome and feasibility to overcome, the âData and Testing Considerationsâ had the highest overall rating and contained the largest number of specific barriers which rated highly. These findings should be considered when identifying which post-match fatigue monitoring methods and metrics to implement in rugby, and potentially other sports

A novel NGS library preparation method to characterize native termini of fragmented DNA.
Biological and chemical DNA fragmentation generates DNA molecules with a variety of termini, including blunt ends and single-stranded overhangs. We have developed a Next Generation Sequencing (NGS) assay, XACTLY, to interrogate the termini of fragmented DNA, information traditionally lost in standard NGS library preparation methods. Here we describe the XACTLY method, showcase its sensitivity and specificity, and demonstrate its utility in in vitro experiments. The XACTLY assay is able to report relative abundances of all lengths and types (5' and 3') of single-stranded overhangs, if present, on each DNA fragment with an overall accuracy between 80-90%. In addition, XACTLY retains the sequence of each native DNA molecule after fragmentation and can capture the genomic landscape of cleavage events at single nucleotide resolution. The XACTLY assay can be applied as a novel research and discovery tool for fragmentation analyses and in cell-free DNA
Long-Toed Salamanders in Harvested and Intact Douglas-Fir Forests of Western Montana
There is little known about how timber harvest practices have affected terrestrial amphibians in the northern Rocky Mountains. Especially lacking is information on the effects of revised harvest methods that fall within the framework of environmental or New Forestry. We estimated the relative abundance of a common forest amphibian, the long-toed salamander (Ambystoma macrodactylum) captured in pitfall arrays on intact, environmentally harvested, and overstory-removal harvested sites in mixed-conifer forests of western Montana. Pitfall data from 1994 through 1996 showed that previously logged sites contained significantly fewer long-toed salamanders regardless of harvest method used. The number of salamanders captured on intact sites (3.1 salamanders·[array]â1·[100 d]â1) was nearly three times the number captured on logged sites (1.2 salamanders·[array]â1·[100 d]â1). Habitat conditions measured in conjunction with trapping efforts indicated that lower amphibian abundance was associated with decreased numbers of large live trees. Declines in amphibian abundance occurred in the absence of changes in understory vegetation that typically occur when forest canopy is reduced. Our findings suggest that long-toed salamanders responded to changes in the physical environment, probably increased temperatures and decreased moisture. That salamanders should respond so dramatically indicates that immediate changes in physical conditions may profoundly alter habitat quality even when other components of the habitat are unaffected

Epidemiology and Molecular Relationships of Cryptosporidium spp. in People, Primates, and Livestock from Western Uganda
Cryptosporidium is a common gastrointestinal parasite known for its zoonotic potential. We found Cryptosporidium in 32.4% of people, 11.1% of non-human primates, and 2.2% of livestock in the region of Kibale National Park, Uganda. In people, infection rates were higher in one community than elsewhere, and fetching water from an open water source increased the probability of infection. Phylogenetic analyses identified clusters of Cryptosporidium with mixed host origins in people, primates, and livestock outside the park; however, parasites from primates inside the park were genetically divergent, suggesting a separate sylvatic transmission cycle. Infection was not associated with clinical disease in people, even in the case of co-infection with the gastrointestinal parasite Giardia duodenalis. Parasites such as Cryptosporidium may be maintained through frequent cross-species transmission in tropical settings where people, livestock, and wildlife interact frequently, but the parasite may undergo more host-specific transmission where such interactions do not occur. Persistent low-level shedding and immunity may limit the clinical effects of infection in such settings
Effectiveness, safety and cost-effectiveness of vaporized nicotine products versus nicotine replacement therapy for tobacco smoking cessation in a low-socioeconomic status Australian population: a study protocol for a randomized controlled trial
Background: In Australia, tobacco smoking rates have declined but inequalities remain with significantly higher smoking prevalence among low-socioeconomic populations. Clinical trial data suggest vaporized nicotine products (VNPs) aid smoking cessation. Most VNP trials have used refillable tank systems, but newer generation (pod) devices now comprise the largest market share yet have limited clinical trial evidence on safety and effectiveness. This study evaluates the effectiveness, safety and cost-effectiveness of VNPs (pod and tank device) compared with nicotine replacement therapy ([NRT]âgum or lozenge) for smoking cessation. Methods: This is a two-arm, open-label, superiority, parallel group, randomized controlled trial (RCT) with allocation concealment and blinded outcome assessment. The RCT is conducted at the National Drug and Alcohol Research Centre at the University of New South Wales, Sydney, Australia. Participants are people who smoke daily, are interested in quitting and receive a government pension or allowance (N = 1058). Participants will be randomized (1:1 ratio) to receive 8 weeks of free: VNPs, with pod (40 mg/mL nicotine salt) and tank device (18 mg/mL freebase nicotine) in mixed flavours; or NRT (gum or lozenge; 4 mg). All participants will receive daily text message behavioural support for 5 weeks. Assessments will be undertaken by telephone at baseline, with three follow-up calls (two check-in calls within the first month and final follow-up at 7 months post randomization) to ascertain smoking status, treatment adherence and adverse events. The primary outcome is 6-month continuous abstinence verified by carbon monoxide breath test of â€5ppm at 7-month follow-up. Safety and cost-effectiveness of VNPs versus NRT will also be evaluated. Discussion: Further data are required to strengthen certainty of evidence for VNPs aiding smoking cessation, particularly for newer generation pod devices. To our knowledge, this trial is the first to offer choice of VNPs and no comparative effectiveness trial data exists for new pod devices. If effective, the findings can inform wider implementation of VNPs to aid smoking cessation in a priority group. Patterns and Perceptions of Climate Change in a Biodiversity Conservation Hotspot
Quantifying local people's perceptions to climate change, and their assessments of which changes matter, is fundamental to addressing the dual challenge of land conservation and poverty alleviation in densely populated tropical regions To develop appropriate policies and responses, it will be important not only to anticipate the nature of expected changes, but also how they are perceived, interpreted and adapted to by local residents. The Albertine Rift region in East Africa is one of the world's most threatened biodiversity hotspots due to dense smallholder agriculture, high levels of land and resource pressures, and habitat loss and conversion. Results of three separate household surveys conducted in the vicinity of Kibale National Park during the late 2000s indicate that farmers are concerned with variable precipitation. Many survey respondents reported that conditions are drier and rainfall timing is becoming less predictable. Analysis of daily rainfall data for the climate normal period 1981 to 2010 indicates that total rainfall both within and across seasons has not changed significantly, although the timing and transitions of seasons has been highly variable. Results of rainfall data analysis also indicate significant changes in the intra-seasonal rainfall distribution, including longer dry periods within rainy seasons, which may contribute to the perceived decrease in rainfall and can compromise food security. Our results highlight the need for fine-scale climate information to assist agro-ecological communities in developing effective adaptive management
The ACER pollen and charcoal database: a global resource to document vegetation and fire response to abrupt climate changes during the last glacial period
Quaternary records provide an opportunity to examine the nature of the vegetation and fire responses to rapid past climate changes comparable in velocity and magnitude to those expected in the 21st-century. The best documented examples of rapid climate change in the past are the warming events associated with the DansgaardâOeschger (DâO) cycles during the last glacial period, which were sufficiently large to have had a potential feedback through changes in albedo and greenhouse gas emissions on climate. Previous reconstructions of vegetation and fire changes during the DâO cycles used independently constructed age models, making it difficult to compare the changes between different sites and regions. Here, we present the ACER (Abrupt Climate Changes and Environmental Responses) global database, which includes 93 pollen records from the last glacial period (73â15âŻka) with a temporal resolution better than 1000âŻyears, 32 of which also provide charcoal records. A harmonized and consistent chronology based on radiometric dating (14C, 234Uâ230Th, optically stimulated luminescence (OSL), 40Arâ39Ar-dated tephra layers) has been constructed for 86 of these records, although in some cases additional information was derived using common control points based on event stratigraphy. The ACER database compiles metadata including geospatial and dating information, pollen and charcoal counts, and pollen percentages of the characteristic biomes and is archived in Microsoft AccessTM at https://doi.org/10.1594/PANGAEA.870867
