Impact of Impaired Glucose Metabolism on Periodontitis Progression over Three Years
We evaluated the relationship between glucose abnormalities and periodontitis in overweight/obese individuals. Eight hundred and seventy (870) diabetes-free participants aged 40â65 years completed the three-year follow-up in the San Juan Overweight Adults Longitudinal Study. The ADA thresholds for fasting and 2-h post-load glucose and HbA1c were used to define prediabetes. The NHANES methods were used to assess periodontitis. Multivariable linear regression was used to evaluate the relationship between baseline glucose metabolism measures and periodontitis at follow-up, adjusting for potential confounders. There was no association between impaired glucose measures and mean pocket depth (PD), mean clinical attachment loss (CAL), or mean percent of sites â„5 mm PD. Impaired glucose tolerance (IGT) was associated with a lower mean percent of sites â„5 mm CAL (ÎČ = â1.6, p = 0.037). Prediabetes and impaired fasting glucose (IFG) were associated with improvement in mean percent of sites â„5 mm PD (ÎČ = â1.4, p = 0.022; ÎČ = â1.6, p = 0.032; respectively). IFG and IGT were associated with improvement in mean percent of sites with â„5 mm CAL (ÎČ = â1.6, p = 0.038; ÎČ = â1.9, p = 0.020; respectively). In conclusion, there were no consistent associations between baseline prediabetes or insulin resistance and periodontitis progression over a three-year period
COVID-19 and Children With Diabetes-Updates, Unknowns, and Next Steps: First, Do No Extrapolation
We are in a place where the only certainty is continued uncertainty about the course of this pandemic. We thank our international pediatric diabetes colleagues and hope their work spurs others to collaborate internationally to gather more evidence. We call on our community to articulate needs and refine recommended actions. As federal diabetes funding is uncertain and many not-for-profit organizations, including the American Diabetes Association and JDRF, announce cuts in funding opportunities and staffing, we must galvanize the pediatric diabetes volunteer community to join our efforts. We must continue to be humble and patient about what we know and advocate strenuously for coordinated, expanded, and responsive public health systems to support youth with both type 1 and type 2 diabetes
A novel isoform of the Ly108 gene ameliorates murine lupus
Studies of human systemic lupus erythematosus patients and of murine congenic mouse strains associate genes in a DNA segment on chromosome 1 with a genetic predisposition for this disease. The systematic analysis of lupus-prone congenic mouse strains suggests a role for two isoforms of the Ly108 receptor in the pathogenesis of the disease. In this study, we demonstrate that Ly108 is involved in the pathogenesis of lupus-related autoimmunity in mice. More importantly, we identified a third protein isoform, Ly108-H1, which is absent in two lupus-prone congenic animals. Introduction of an Ly108-H1âexpressing transgene markedly diminishes T cellâdependent autoimmunity in congenic B6.Sle1b mice. Thus, an immune responseâsuppressing isoform of Ly108 can regulate the pathogenesis of lupus.Peer Reviewe

Cerebral small vessel disease genomics and its implications across the lifespan
White matter hyperintensities (WMH) are the most common brain-imaging feature of cerebral small vessel disease (SVD), hypertension being the main known risk factor. Here, we identify 27 genome-wide loci for WMH-volume in a cohort of 50,970 older individuals, accounting for modification/confounding by hypertension. Aggregated WMH risk variants were associated with altered white matter integrity (pâ=â2.5Ă10-7) in brain images from 1,738 young healthy adults, providing insight into the lifetime impact of SVD genetic risk. Mendelian randomization suggested causal association of increasing WMH-volume with stroke, Alzheimer-type dementia, and of increasing blood pressure (BP) with larger WMH-volume, notably also in persons without clinical hypertension. Transcriptome-wide colocalization analyses showed association of WMH-volume with expression of 39 genes, of which four encode known drug targets. Finally, we provide insight into BP-independent biological pathways underlying SVD and suggest potential for genetic stratification of high-risk individuals and for genetically-informed prioritization of drug targets for prevention trials.Peer reviewe
Rare coding variants in PLCG2, ABI3, and TREM2 implicate microglial-mediated innate immunity in Alzheimer's disease
We identified rare coding variants associated with Alzheimerâs disease (AD) in a 3-stage case-control study of 85,133 subjects. In stage 1, 34,174 samples were genotyped using a whole-exome microarray. In stage 2, we tested associated variants (P<1Ă10-4) in 35,962 independent samples using de novo genotyping and imputed genotypes. In stage 3, an additional 14,997 samples were used to test the most significant stage 2 associations (P<5Ă10-8) using imputed genotypes. We observed 3 novel genome-wide significant (GWS) AD associated non-synonymous variants; a protective variant in PLCG2 (rs72824905/p.P522R, P=5.38Ă10-10, OR=0.68, MAFcases=0.0059, MAFcontrols=0.0093), a risk variant in ABI3 (rs616338/p.S209F, P=4.56Ă10-10, OR=1.43, MAFcases=0.011, MAFcontrols=0.008), and a novel GWS variant in TREM2 (rs143332484/p.R62H, P=1.55Ă10-14, OR=1.67, MAFcases=0.0143, MAFcontrols=0.0089), a known AD susceptibility gene. These protein-coding changes are in genes highly expressed in microglia and highlight an immune-related protein-protein interaction network enriched for previously identified AD risk genes. These genetic findings provide additional evidence that the microglia-mediated innate immune response contributes directly to AD development

Ambos proyectos parten de la metodologĂa interdisciplinaria y colaborativa con miembros de la comunidad CucapĂĄ para crear representaciones audiovisuales no estigmatizantes, que detonen el sentido de comunidad y refuercen su acervo cultural. El videojuego A Orillas del rĂo es del gĂ©nero point and click, de vista isomĂ©trica, el cual estĂĄ inspirado en el cuento oral tradicional El zorro y el coyote, que busca ser una herramienta lĂșdica de aprendizaje para reforzar el aprendizaje de la lengua CucapĂĄ en los niños. Los resultados obtenidos fueron el demo del nivel uno (escenas uno y dos); colorimetrĂa, arcos de personajes principales; programaciĂłn de minijuegos. Dentro
