    Pectinase-hyperproducing mutants of Aspergillus niger C28B25 for solid-state fermentation of coffee pulp

    The aim of this study was to improve mold strains for the production of pectinases by solid-state fermentation (SSF) of coffee pulp. A screening of 248 strains, isolated in Mexico's coffee-growing areas, permitted us to select a wild strain of #Aspergillus niger$ which in 72 h attains a peak production of 27.7 U ml-1(138 U g-1 dry pulp) of pectinase measured by viscosimetry. Though the use of a selective culture medium with low water activity (aw = 0.954) with 2-deoxy-glucose (2DG) it was possible to isolate pectinase-hyperproductive mutant strains for SSF(44.5 U ml-1, 228 U g-1 dry pulp). Derepressed mutant strains that hyperproduced pectinase by submerged fermentation (SmF) were also obtained using a classic selective medium with high water activity (pectin + 2DG) with aw = 0.999. A comparison between both classes of mutants, called AW96 and AW99, respectively, points out the need to design special selective media in order to obtain strains adapted either to SSF or Smf in which the aw level would be a key selecting factor.     Escleritos aislados de Chancelloria eros Walcott, 1920, Diagoniella sp. y asociaciones de espículas hexactinéllidas-estauractinas y hexactina-pentactinas son descritas para el Cámbrico Medio de San José de Gracia, Sonora, México. El material procede de una secuencia sedimentaria marina de plataforma exterior, constituida principalmente por lutita intercalada con caliza, en asociación con fósiles representativos de la Biozona de Albertella como lo es Peronopsis bonnerensis Resser, 1938; Oryctocephalites walcotti Reeser, 1938; Elrathina antiqua Palmer y Halley, 1979 y Ogygopsis typicalis Resser, 1939; así como braquiópodos inarticulados. Chancelloria eros Walcott, 1920 ha sido reportada anteriormente para el Estado de Sonora, México, en el Cámbrico Medio de la Formación Arrojos, así como Chancelloria sp. en el Cámbrico Temprano de la Formación Puerto Blanco, en asociación con Kiwetinokia en Cerro Rajón en la región de Caborca.     Fucoidan-degrading fungal strains: screening, morphometric evaluation, and influence of medium composition

    Ten different fungal strains from the genus Aspergillus, Penicillium, and Mucor were screened for fucoidan hydrolyzing ability aiming to find microorganisms able to produce sulfated fucan-degrading enzymes. Screening was carried out by measuring the strains kinetic and morphometric behavior over plate assays using Laminaria japonica fucoidan as only carbon source, testing three nitrogen sources (urea, peptone, and sodium nitrate). The selected fungal strains were subsequently used in submerged fermentations, which were performed for (1) selection of the strains able to growth over fucoidan medium and (2) media selection, testing the synergy of fucoidan with other sugars for inducing high enzyme titles. Radial expansion and hyphae parameters were observed for Aspergillus niger PSH, Mucor sp. 3P, and Penicillium purpurogenum GH2 grown only over fucoidan-urea medium. A. niger PSH showed the maximum enzymatic activity values, which were significantly different (p<0.05) from those achieved by the other selected fungi. Sucrose addition to fucoidan media proportioned the highest fucoidanase activity values for this fungal strain.     Les pectinases, enzymes d'un intérêt commercial certain pour l'agroindustrie, sont produites essentiellement par les moisissures en fermentation en milieu solide (FMS) et liquide (FML). La sélection d'une souche hyperproductrice en FMS a été obtenue par un criblage de 248 souches sur pulpe de café. Une technique d'analyse d'images a permis une caractérisation précise des souches suivant des traits morphologiques. Suite à une irradiation aux ultra violets des souches adaptées, soit à la FMS soit à la FML, ont été sélectionnées. Les mutants adaptés à la FMS produisent cinq fois plus de pectinases que les mêmes souches en FML et vice versa. Les données expérimentales montrent des différences au niveau des protéines biosynthétisées par une même souche placée soit en FMS soit en FML. (Résumé d'auteur
