Genetic variation at MECOM, TERT, JAK2 and HBS1L-MYB predisposes to myeloproliferative neoplasms
Clonal proliferation in myeloproliferative neoplasms (MPN) is driven by somatic mutations in JAK2, CALR or MPL, but the contribution of inherited factors is poorly characterized. Using a three-stage genome-wide association study of 3,437 MPN cases and 10,083 controls, we identify two SNPs with genome-wide significance in JAK2V617F-negative MPN: rs12339666 (JAK2; meta-analysis P=1.27 Ă 10â10) and rs2201862 (MECOM; meta-analysis P=1.96 Ă 10â9). Two additional SNPs, rs2736100 (TERT) and rs9376092 (HBS1L/MYB), achieve genome-wide significance when including JAK2V617F-positive cases. rs9376092 has a stronger effect in JAK2V617F-negative cases with CALR and/or MPL mutations (BreslowâDay P=4.5 Ă 10â7), whereas in JAK2V617F-positive cases rs9376092 associates with essential thrombocythemia (ET) rather than polycythemia vera (allelic Ï2 P=7.3 Ă 10â7). Reduced MYB expression, previously linked to development of an ET-like disease in model systems, associates with rs9376092 in normal myeloid cells. These findings demonstrate that multiple germline variants predispose to MPN and link constitutional differences in MYB expression to disease phenotype
Mutation update for the SATB2 gene
SATB2-associated syndrome (SAS) is an autosomal dominant neurodevelopmental disorder caused by alterations in the SATB2 gene. Here we present a review of published pathogenic variants in the SATB2 gene to date and report 38 novel alterations found in 57 additional previously unreported individuals. Overall, we present a compilation of 120 unique variants identified in 155 unrelated families ranging from single nucleotide coding variants to genomic rearrangements distributed throughout the entire coding region of SATB2. Single nucleotide variants predicted to result in the occurrence of a premature stop codon were the most commonly seen (51/120=42.5%) followed by missense variants (31/120=25.8%). We review the rather limited functional characterization of pathogenic variants and discuss current understanding of the consequences of the different molecular alterations. We present an expansive phenotypic review along with novel genotype-phenotype correlations. Lastly, we discuss current knowledge on animal models and present future prospects. This review should help provide better guidance for the care of individuals diagnosed with SAS
Petrogenesis and geochemical halos of the amphibolite facies, Lower Proterozoic, Kerry Road volcanogenic massive sulfide deposit, Loch Maree Group, Gairloch, NW Scotland
The Palaeoproterozoic Kerry Road deposit is one of the oldest examples of volcanogenic massive sulfide (VMS) mineralization. This small VMS deposit (~500,000 tons grading at 1.2% Cu, 3.5% Zn) is hosted in amphibolite facies mafic-siliciclastic units of the c. 2.0 Ga Loch Maree Group, Scotland. Sulfide mineralization consists of pyrite and pyrrhotite with subordinate chalcopyrite and sphalerite, occurring in disseminated, vein and semi-massive to massive textures.
The deposit was highly deformed and metamorphosed during the c. 1.8â1.7 Ga Laxfordian Orogeny. Textural relationships of deformed sulfide minerals, related to early Laxfordian deformation (D1/D2), indicate initial high pressure-low temperature (100 MPa, 150 °C) conditions before reaching peak amphibolite facies metamorphism, as evident from pyrrhotite crossing the brittle/ductile transition prior to chalcopyrite. Late Laxfordian deformation (D3/D4) is marked by local retrograde greenschist facies at low pressure and temperature (<1.2 MPa, <200 °C), recorded by late red sphalerite remobilization. ÎŽ34S values from all sulfide minerals have a homogeneous mean of 0.8 ± 0.7â° (n = 21), consistent with interaction of hydrothermal fluids in the host oceanic basalt-island arc setting envisaged for deposition of the Loch Maree Group.
Microprobe analyses of amphiboles record evidence of the original alteration halo associated with the Kerry Road deposit, with a systematic Mg- and Si- enrichment from ferrotschermakite (~150 m) to Mg-hornblende (~90 m) to actinolite (0 m) on approach to the VMS deposit. Furthermore, whole rock geochemistry records a progressive enrichment in Si, Cu, Co, and S, and depletion in Al, Ti, V, Cr, Y and Zr with proximity to the VMS system. These elemental trends, together with amphibole geochemistry, are potentially useful exploration vectors to VMS mineralization in the Loch Maree Group, and in similar highly deformed and metamorphosed terranes elsewhere
The parent?infant dyad and the construction of the subjective self
Developmental psychology and psychopathology has in the past been more concerned with the quality of self-representation than with the development of the subjective agency which underpins our experience of feeling, thought and action, a key function of mentalisation. This review begins by contrasting a Cartesian view of pre-wired introspective subjectivity with a constructionist model based on the assumption of an innate contingency detector which orients the infant towards aspects of the social world that react congruently and in a specifically cued informative manner that expresses and facilitates the assimilation of cultural knowledge. Research on the neural mechanisms associated with mentalisation and social influences on its development are reviewed. It is suggested that the infant focuses on the attachment figure as a source of reliable information about the world. The construction of the sense of a subjective self is then an aspect of acquiring knowledge about the world through the caregiver's pedagogical communicative displays which in this context focuses on the child's thoughts and feelings. We argue that a number of possible mechanisms, including complementary activation of attachment and mentalisation, the disruptive effect of maltreatment on parent-child communication, the biobehavioural overlap of cues for learning and cues for attachment, may have a role in ensuring that the quality of relationship with the caregiver influences the development of the child's experience of thoughts and feelings
Incommensurability and Multi-paradigm Grounding in Design Science Research: Implications for Creating Knowledge
International audienceThe problem identification-design-build-evaluate-theorize structure of design science research has been proposed as an approach to creating knowledge in information systems and in broader organizational and social domains. Although the approach has merit, the philosophical foundations of two specific components warrant attention. First, the grounding of design theory on potentially incommensurate kernel theories may produce incoherent design theory. In addition, design theory has no strong logical connection to kernel theories, and so cannot be used to test or validate the contributing kernel theories. Second, the philosophical grounding of evaluation may inadvertently shift from functionally based measures of utility and efficiency, to evaluation based on the pragmatic fulfillment of multidimensional human actions as people encounter information systems, resulting in evaluation errors. Although design and evaluation from a single paradigm is not desirable, sufficient, or representative of design science research, multi-paradigm grounding of design and evaluation must be realized and used consciously by the research community if the design science approach is to remain a legitimate approach to knowledge creation
Thresholds for adding degraded tropical forest to the conservation estate
Logged and disturbed forests are often viewed as degraded and depauperate environments compared with primary forest. However, they are dynamic ecosystems1 that provide refugia for large amounts of biodiversity2,3, so we cannot afford to underestimate their conservation value4. Here we present empirically defined thresholds for categorizing the conservation value of logged forests, using one of the most comprehensive assessments of taxon responses to habitat degradation in any tropical forest environment. We analysed the impact of logging intensity on the individual occurrence patterns of 1,681 taxa belonging to 86 taxonomic orders and 126 functional groups in Sabah, Malaysia. Our results demonstrate the existence of two conservation-relevant thresholds. First, lightly logged forests (68%) of their biomass removed, and these are likely to require more expensive measures to recover their biodiversity value. Overall, our data confirm that primary forests are irreplaceable5, but they also reinforce the message that logged forests retain considerable conservation value that should not be overlooked

Penelitian tugas akhir ini mengkaji tentang Profil Wisatawan di
Museum Radya Pustaka Surakarta. Tujuan dari penelitian ini adalah untuk
mengetahui dari daerah mana saja wisatawan yang berkunjung ke Museum
Radya Pustaka, bagaimana ciri-ciri wisatawan yang berkunjung ke Museum
Radya Pustaka serta harapan-harapan yang diinginkan wisatawan terhadap
Museum Radya Pustaka.
Penelitian dilakukan dengan metode kualitatif. Pengumpulan data
dilakukan melalui wawancara dengan narasumber wisatawan yang berkujung
di Museum Radya Pustaka Surakarta tempat penulis melakukan penelitian,
serta studi pustaka dan studi dokumen guna menambah sumber data.
Hasil penelitian menunjukkan bahwa (1) Sebagian besar wisatawan
yang datang berasal dari Semarang sebesar 32%. (2) Mayoritas wisatawan
yang berkunjung ke Museum Radya Pustaka berusia antara 17-25 tahun dan
kebanyakan dari mereka adalah pelajar atau mahasiswa dengan prosentase
52%. (3) Sebagian besar wisatawan yang datang ke Museum Radya Pustaka
adalah bertujuan untuk melakukan penelitian yaitu sebesar 34%. (4) Harapan
wisatawan yang berkunjung terhadap kelangsungan Museum Radya Pustaka
sebagian besar adalah agar ditingkatkan lagi pengelolaan dan keamanan
museum, agar kejadian hilangnya benda-benda koleksi museum tidak terulang
lagi dikemudian hari.
Kesimpulan dari hasil penelitian ini bahwa wisatawan yang berkujung
ke Museum Radya Pustaka Surakarta mayoritas berasal dari Semarang,
mayoritas berusia 17-25 tahun dan kebanyakan dari mereka adalah berprofesi
sebagai pelajar dan mahasiswa. Kebanyakan wisatawan yang datang bertujuan
untuk melakukan penelitian, serta harapan wisatawan terhadap Museum
Radya Pustaka adalah supaya lebih ditingkatkan lagi pengelolaan dan
keamanan museum

Os M. pretenderam traçar um quadro da HistĂłria da Alimentação, nĂŁo como um novo ramo epistemolĂłgico da disciplina, mas como um campo em desenvolvimento de prĂĄticas e atividades especializadas, incluindo pesquisa, formação, publicaçÔes, associaçÔes, encontros acadĂȘmicos, etc. Um breve relato das condiçÔes em que tal campo se assentou faz-se preceder de um panorama dos estudos de alimentação e temas correia tos, em geral, segundo cinco abardagens Ia biolĂłgica, a econĂŽmica, a social, a cultural e a filosĂłfica!, assim como da identificação das contribuiçÔes mais relevantes da Antropologia, Arqueologia, Sociologia e Geografia. A fim de comentar a multiforme e volumosa bibliografia histĂłrica, foi ela organizada segundo critĂ©rios morfolĂłgicos.
