    Induction of humoral immune response to multiple recombinant Rhipicephalus appendiculatus antigens and their effect on tick feeding success and pathogen transmission

    BACKGROUND: Rhipicephalus appendiculatus is the primary vector of Theileria parva, the etiological agent of East Coast fever (ECF), a devastating disease of cattle in sub-Saharan Africa. We hypothesized that a vaccine targeting tick proteins that are involved in attachment and feeding might affect feeding success and possibly reduce tick-borne transmission of T. parva. Here we report the evaluation of a multivalent vaccine cocktail of tick antigens for their ability to reduce R. appendiculatus feeding success and possibly reduce tick-transmission of T. parva in a natural host-tick-parasite challenge model. METHODS: Cattle were inoculated with a multivalent antigen cocktail containing recombinant tick protective antigen subolesin as well as two additional R. appendiculatus saliva antigens: the cement protein TRP64, and three different histamine binding proteins. The cocktail also contained the T. parva sporozoite antigen p67C. The effect of vaccination on the feeding success of nymphal and adult R. appendiculatus ticks was evaluated together with the effect on transmission of T. parva using a tick challenge model. RESULTS: To our knowledge, this is the first evaluation of the anti-tick effects of these antigens in the natural host-tick-parasite combination. In spite of evidence of strong immune responses to all of the antigens in the cocktail, vaccination with this combination of tick and parasite antigens did not appear to effect tick feeding success or reduce transmission of T. parva. CONCLUSION: The results of this study highlight the importance of early evaluation of anti-tick vaccine candidates in biologically relevant challenge systems using the natural tick-host-parasite combination

    Relational Goods and Endurance of Voluntary Associational Participation: The Mapuche Indigenous Case in Santiago de Chile.

    In this article, I analyse the reasons for sustaining membership in voluntary ethnic associations of Mapuche people living in Santiago de Chile. By following a relational goods approach, I suggest that the constructed nature of ethnicity leads the Mapuche to create and join ethnic associations in an urban milieu. This study reveals that the main motivations for sustaining an active associational engagement in Santiago are based on three accounts: identity recovery struggle, leaving an identity legacy and bonding with their ethnic peers; all of these identified as relational goods.     Isothermal Elastohydrodynamic Lubrication Analysis of Heavily Loaded Hypoid Gear Pairs

    A numerical model able to predict the pressure distribution and the film thickness in heavily loaded elliptical EHL contacts is developed and presented in this study. The operating conditions, such as the contact load and the velocities of the mating surfaces, are representative of the corresponding conditions present in automotive differential hypoid gear pair units. The EHL solver presented is able to predict the minimum and central film thickness of the lubricating oil as well as the pressure distribution assuming isothermal and Newtonian conditions. Results are presented for a full quasi-static meshing cycle. A comparison between the numerically calculated values of the central and the minimum film thickness is performed against the corresponding values produced using the Chittenden-Dowson formula. A very good agreement is observed between the values of the central film thickness. However, it is shown that the minimum film thickness values using the Chittenden-Dowson formula can deviate up to 40% compared with the corresponding values which are calculated numerically

    Film Thickness Investigation in Heavily Loaded Hypoid Gear Pair Elastohydrodynamic Conjunctions

    Introduction: Hypoid gear pairs are some of the most highly loaded components of the differential unit in modern automobiles. Prediction of wear rate and generated friction require determination of lubricant film thickness. However, only very few investigations have addressed the issue of thin elastohydrodynamic films in hypoid gear pairs. The main reason for dearth of analysis in this regard has been the need for accurate determination of transient contact geometry and kinematics of interacting surfaces throughout a typical meshing cycle. Furthermore, combined gear dynamics and lubrication analysis of any pairs of simultaneous meshing teeth pairs is required. Simon [1] was among the first to deal with these issues. He used Tooth Contact Analysis (TCA) in order to calculate the instantaneous contact geometry and load for any teeth pair during their meshing cycle. However, in his study, the load carried by the hypoid pair was quite low, making the application of the results limited and not entirely suitable for real life operating conditions of typical hypoid gear pairs of vehicular differentials, which is of interest in the current paper. Xu and Kahraman [2] performed numerical prediction of power losses and consequently the film thickness for highly loaded hypoid gear pairs. However, in their study only the one-dimensional Reynolds equation was employed. Consequently, the effect of lubricant side leakage in the passage through the contact was ignored. A more recent study by Mohammadpour et al. [3] employed realistic gear geometry data (through the use of TCA) for calculation of film thickness time history through mesh. The two-dimensional Reynolds equation, accounting for the side leakage of the lubricant, was solved numerically. It was shown that the side leakage component of the entraining velocity can significantly influence the film thickness. With regard to hypoid gear dynamics, several studies should be mentioned. Wang and Lim [4] studied the dynamic response of hypoid gear pairs under the influence of time varying meshing stiffness. Yang and Lim [5] created a model able to predict the dynamic response of a hypoid gear pair by taking into account the lateral translations of their shafts due to the compliance of the supporting bearings. Karagiannis et al. [6-7] studied the dynamics of automotive differential hypoid gear pairs by taking into account the velocity dependent resistive torque at the gear caused by aerodynamic drag and tyre-road rolling resistance.     Interactions between environmental contaminants and gastrointestinal parasites: novel insights from an integrative approach in a marine predator

    Environmental contaminants and parasites are ubiquitous stressors that can affect animal physiology and derive from similar dietary sources (co-exposure). To unravel their interactions in wildlife, it is thus essential to quantify their concurring drivers. Here, the relationship between blood contaminant residues (11 trace elements and 17 perfluoroalkyl substances) and nonlethally quantified gastrointestinal parasite loads was tested while accounting for intrinsic (sex, age, and mass) and extrinsic factors (trophic ecology inferred from stable isotope analyses and biologging) in European shags Phalacrocorax aristotelis. Shags had high mercury (range 0.65–3.21 ÎŒg g–1 wet weight, ww) and extremely high perfluorooctanoic acid (PFOA) and perfluorononanoic acid (PFNA) residues (3.46–53 and 4.48–44 ng g–1 ww, respectively). Males had higher concentrations of arsenic, mercury, PFOA, and PFNA than females, while the opposite was true for selenium, perfluorododecanoic acid (PFDoA), and perfluooctane sulfonic acid (PFOS). Individual parasite loads (Contracaecum rudolphii) were higher in males than in females. Females targeted pelagic-feeding prey, while males relied on both pelagic- and benthic-feeding organisms. Parasite loads were not related to trophic ecology in either sex, suggesting no substantial dietary co-exposure with contaminants. In females, parasite loads increased strongly with decreasing selenium:mercury molar ratios. Females may be more susceptible to the interactive effects of contaminants and parasites on physiology, with potential fitness consequences

    Profiling microRNAs in individuals at risk of progression to rheumatoid arthritis

    Background: Individuals at risk of rheumatoid arthritis (RA) demonstrate systemic autoimmunity in the form of anti-citrullinated peptide antibodies (ACPA). MicroRNAs (miRNAs) are implicated in established RA. This study aimed to (1) compare miRNA expression between healthy individuals and those at risk of and those that develop RA, (2) evaluate the change in expression of miRNA from "at-risk" to early RA and (3) explore whether these miRNAs could inform a signature predictive of progression from "at-risk" to RA. Methods: We performed global profiling of 754 miRNAs per patient on a matched serum sample cohort of 12 anti-cyclic citrullinated peptide (CCP) + "at-risk" individuals that progressed to RA. Each individual had a serum sample from baseline and at time of detection of synovitis, forming the matched element. Healthy controls were also studied. miRNAs with a fold difference/fold change of four in expression level met our primary criterion for selection as candidate miRNAs. Validation of the miRNAs of interest was conducted using custom miRNA array cards on matched samples (baseline and follow up) in 24 CCP+ individuals; 12 RA progressors and 12 RA non-progressors. Results: We report on the first study to use matched serum samples and a comprehensive miRNA array approach to identify in particular, three miRNAs (miR-22, miR-486-3p, and miR-382) associated with progression from systemic autoimmunity to RA inflammation. MiR-22 demonstrated significant fold difference between progressors and non-progressors indicating a potential biomarker role for at-risk individuals. Conclusions: This first study using a cohort with matched serum samples provides important mechanistic insights in the transition from systemic autoimmunity to inflammatory disease for future investigation, and with further evaluation, might also serve as a predictive biomarker

    The [OIII] emission line luminosity function of optically selected type-2 AGN from zCOSMOS

    We present a catalog of 213 type-2 AGN selected from the zCOSMOS survey. The selected sample covers a wide redshift range (0.15<z<0.92) and is deeper than any other previous study, encompassing the luminosity range 10^{5.5} < Lsun< L[OIII] < 10^{9.1} Lsun. We explore the intrinsic properties of these AGN and the relation to their X-ray emission (derived from the XMM-COSMOS observations). We study their evolution by computing the [OIII]5007A line luminosity function (LF) and we constrain the fraction of obscured AGN as a function of luminosity and redshift. The sample was selected on the basis of the optical emission line ratios, after applying a cut to the signal-to-noise ratio (S/N) of the relevant lines. We used the standard diagnostic diagrams [OIII]/Hbeta versus [NII]/Halpha and ([OIII]/Hbeta versus [SII]/Halpha) to isolate AGN in the redshift range 0.15<z<0.45 and the diagnostic diagram [OIII]/Hbeta versus [OII]/Hbeta to extend the selection to higher redshift (0.5<z<0.92). Combining our sample with one drawn from SDSS, we found that the best description of the evolution of type-2 AGN is a luminosity-dependent density evolution model. Moreover, using the type-1 AGN LF we were able to constrain the fraction of type-2 AGN to the total (type-1 + type-2) AGN population. We found that the type-2 fraction decreases with luminosity, in agreement with the most recent results, and shows signs of a slight increase with redshift. However, the trend with luminosity is visible only after combining the SDSS+zCOSMOS samples. From the COSMOS data points alone, the type-2 fraction seems to be quite constant with luminosity.Comment: 20 pages, 11 figures, accepted for publication in Astronomy and Astrophysic
