    A geometric constraint over k-dimensional objects and shapes subject to business rules

    This report presents a global constraint that enforces rules written in a language based on arithmetic and first-order logic to hold among a set of objects. In a first step, the rules are rewritten to Quantifier-Free Presburger Arithmetic (QFPA) formulas. Secondly, such formulas are compiled to generators of k-dimensional forbidden sets. Such generators are a generalization of the indexicals of cc(FD). Finally, the forbidden sets generated by such indexicals are aggregated by a sweep-based algorithm and used for filtering. The business rules allow to express a great variety of packing and placement constraints, while admitting efficient and effective filtering of the domain variables of the k-dimensional object, without the need to use spatial data structures. The constraint was used to directly encode the packing knowledge of a major car manufacturer and tested on a set of real packing problems under these rules, as well as on a packing-unpacking problem

    A Homeotic Shift Late in Development Drives Mimetic Color Variation in a Bumble Bee

    Natural phenotypic radiations, with their high diversity and convergence, are well-suited for informing how genomic changes translate to natural phenotypic variation. New genomic tools enable discovery in such traditionally nonmodel systems. Here, we characterize the genomic basis of color pattern variation in bumble bees (Hymenoptera, Apidae, Bombus), a group that has undergone extensive convergence of setal color patterns as a result of Müllerian mimicry. In western North America, multiple species converge on local mimicry patterns through parallel shifts of midabdominal segments from red to black. Using genome-wide association, we establish that a cis-regulatory locus between the abdominal fate-determining Hox genes, abd-A and Abd-B, controls the red–black color switch in a western species, Bombus melanopygus. Gene expression analysis reveals distinct shifts in Abd-B aligned with the duration of setal pigmentation at the pupal–adult transition. This results in atypical anterior Abd-B expression, a late developmental homeotic shift. Changing expression of Hox genes can have widespread effects, given their important role across segmental phenotypes; however, the late timing reduces this pleiotropy, making Hox genes suitable targets. Analysis of this locus across mimics and relatives reveals that other species follow independent genetic routes to obtain the same phenotypes

    Key Role of Polyphosphoinositides in Dynamics of Fusogenic Nuclear Membrane Vesicles

    The role of phosphoinositides has been thoroughly described in many signalling and membrane trafficking events but their function as modulators of membrane structure and dynamics in membrane fusion has not been investigated. We have reconstructed models that mimic the composition of nuclear envelope precursor membranes with naturally elevated amounts of phosphoinositides. These fusogenic membranes (membrane vesicle 1(MV1) and nuclear envelope remnants (NER) are critical for the assembly of the nuclear envelope. Phospholipids, cholesterol, and polyphosphoinositides, with polyunsaturated fatty acid chains that were identified in the natural nuclear membranes by lipid mass spectrometry, have been used to reconstruct complex model membranes mimicking nuclear envelope precursor membranes. Structural and dynamic events occurring in the membrane core and at the membrane surface were monitored by solid-state deuterium and phosphorus NMR. “MV1-like” (PC∶PI∶PIP∶PIP2, 30∶20∶18∶12, mol%) membranes that exhibited high levels of PtdIns, PtdInsP and PtdInsP2 had an unusually fluid membrane core (up to 20% increase, compared to membranes with low amounts of phosphoinositides to mimic the endoplasmic reticulum). “NER-like” (PC∶CH∶PI∶PIP∶PIP2, 28∶42∶16∶7∶7, mol%) membranes containing high amounts of both cholesterol and phosphoinositides exhibited liquid-ordered phase properties, but with markedly lower rigidity (10–15% decrease). Phosphoinositides are the first lipids reported to counterbalance the ordering effect of cholesterol. At the membrane surface, phosphoinositides control the orientation dynamics of other lipids in the model membranes, while remaining unchanged themselves. This is an important finding as it provides unprecedented mechanistic insight into the role of phosphoinositides in membrane dynamics. Biological implications of our findings and a model describing the roles of fusogenic membrane vesicles are proposed

    Stretch-Activated Piezo1 Channel in Endothelial Cells Relaxes Mouse Intrapulmonary Arteries

    In intrapulmonary artery (IPA), endothelial cells (EC) respond to mechanical stimuli by releasing vasoactive factors to set the vascular tone. Piezo1, a stretch-activated calcium permeable channel is a sensor of mechanical stress in EC. The present study was undertaken to investigate the implication of Piezo1 in the endothelium-dependent regulation of IPA tone and its potential involvement in pulmonary hypertension, the main disease of this circulation. IPA tone was quantified by means of a myograph in control Piezo1+/+ mouse and in mouse lacking endothelial Piezo1 (EC-Piezo1-/-). Endothelial intracellular calcium concentration ([Ca2+]i) and nitric oxide (NO) production were measured, in mouse or human EC, with fluo-4 and DAF-fm probes, respectively. Immunofluorescence labeling and patch-clamp experiments revealed the presence of Piezo1 channels in EC. Yoda1, a Piezo1 agonist, induced an endothelium-dependent relaxation that was significantly reduced in pulmonary arteries in EC-Piezo1-/- compared to Piezo1+/+ mouse. Yoda1 as well as mechanical stimulation (by osmotic stress) increased [Ca2+]i in mouse or human EC. Consequently, both stimuli increased the production of NO. NO and [Ca2+]i increases were reduced in EC from Piezo1-/- mouse or in the presence of Piezo1 inhibitors. Furthermore, deletion of Piezo1 increased alpha-adrenergic mediated contraction. Finally, in chronically hypoxic mice, a model of pulmonary hypertension, Piezo1 still mediated arterial relaxation and deletion of this channel did not impair the development of the disease. The present study thus demonstrates that endothelial Piezo1 contributes to intrapulmonary vascular relaxation by controlling endothelial [Ca2+]i and NO production and that this effect is still present in pulmonary hypertension

    Including debris cover effects in a distributed model of glacier ablation

    Distributed glacier melt models generally assume that the glacier surface consists of bare exposed ice and snow. In reality, many glaciers are wholly or partially covered in layers of debris that tend to suppress ablation rates. In this paper, an existing physically based point model for the ablation of debris-covered ice is incorporated in a distributed melt model and applied to Haut Glacier d’Arolla, Switzerland, which has three large patches of debris cover on its surface. The model is based on a 10 m resolution digital elevation model (DEM) of the area; each glacier pixel in the DEM is defined as either bare or debris-covered ice, and may be covered in snow that must be melted off before ice ablation is assumed to occur. Each debris-covered pixel is assigned a debris thickness value using probability distributions based on over 1000 manual thickness measurements. Locally observed meteorological data are used to run energy balance calculations in every pixel, using an approach suitable for snow, bare ice or debris-covered ice as appropriate. The use of the debris model significantly reduces the total ablation in the debris-covered areas, however the precise reduction is sensitive to the temperature extrapolation used in the model distribution because air near the debris surface tends to be slightly warmer than over bare ice. Overall results suggest that the debris patches, which cover 10% of the glacierized area, reduce total runoff from the glacierized part of the basin by up to 7%

    Poor glycemic control in type 2 diabetes enhances functional and compositional alterations of small, dense HDL3c

    High-density lipoprotein (HDL) possesses multiple biological activities; small, dense HDL3c particles displaying distinct lipidomic composition exert potent antiatherogenic activities which can be compromised in dyslipidemic, hyperglycemic insulin-resistant states. However, it remains indeterminate (i) whether such functional HDL deficiency is related to altered HDL composition, and (ii) whether it originates from atherogenic dyslipidemia, dysglycemia, or both. In the present work we analyzed compositional characteristics of HDL subpopulations and functional activity of small, dense HDL3c particles in treatment-naïve patients with well-controlled (n=10) and poorly-controlled (n=8) type 2 diabetes (T2D) and in normolipidemic age- and sex-matched controls (n=11). Our data reveal that patients with both well- and poorly-controlled T2D displayed dyslipidemia and low-grade inflammation associated with altered HDL composition. Such compositional alterations in small, dense HDL subfractions were specifically correlated with plasma HbA1c levels. Further analysis using a lipidomic approach revealed that small, dense HDL3c particles from T2D patients with poor glycemic control displayed additional modifications of their chemical composition. In parallel, antioxidative activity of HDL3c towards oxidation of low-density lipoprotein was diminished. These findings indicate that defective functionality of small, dense HDL particles in patients with T2D is not only affected by the presence of atherogenic dyslipidemia, but also by the level of glycemic control, reflecting compositional alterations of HDL.     The SPARSE model for the prediction of water stress and evapotranspiration components from thermal infra-red data and its evaluation over irrigated and rainfed wheat

    Evapotranspiration is an important component of the water cycle, especially in semi-arid lands. A way to quantify the spatial distribution of evapotranspiration and water stress from remote-sensing data is to exploit the available surface temperature as a signature of the surface energy balance. Remotely sensed energy balance models enable one to estimate stress levels and, in turn, the water status of continental surfaces. Dual-source models are particularly useful since they allow derivation of a rough estimate of the water stress of the vegetation instead of that of a soil–vegetation composite. They either assume that the soil and the vegetation interact almost independently with the atmosphere (patch approach corresponding to a parallel resistance scheme) or are tightly coupled (layer approach corresponding to a series resistance scheme). The water status of both sources is solved simultaneously from a single surface temperature observation based on a realistic underlying assumption which states that, in most cases, the vegetation is unstressed, and that if the vegetation is stressed, evaporation is negligible. In the latter case, if the vegetation stress is not properly accounted for, the resulting evaporation will decrease to unrealistic levels (negative fluxes) in order to maintain the same total surface temperature. This work assesses the retrieval performances of total and component evapotranspiration as well as surface and plant water stress levels by (1) proposing a new dual-source model named Soil Plant Atmosphere and Remote Sensing Evapotranspiration (SPARSE) in two versions (parallel and series resistance networks) based on the TSEB (Two-Source Energy Balance model, Norman et al., 1995) model rationale as well as state-of-the-art formulations of turbulent and radiative exchange, (2) challenging the limits of the underlying hypothesis for those two versions through a synthetic retrieval test and (3) testing the water stress retrievals (vegetation water stress and moisture-limited soil evaporation) against in situ data over contrasted test sites (irrigated and rainfed wheat). We demonstrated with those two data sets that the SPARSE series model is more robust to component stress retrieval for this cover type, that its performance increases by using bounding relationships based on potential conditions (root mean square error lowered by up to 11 W m−2 from values of the order of 50–80 W m−2), and that soil evaporation retrieval is generally consistent with an independent estimate from observed soil moisture evolution
