    Prevention of Herpesviridae Infections by Cationic PEGylated Carbosilane Dendrimers

    Infections caused by viruses from the Herpesviridae family produce some of the most prevalent transmitted diseases in the world, constituting a serious global public health issue. Some of the virus properties such as latency and the appearance of resistance to antiviral treatments complicate the development of effective therapies capable of facing the infection. In this context, dendrimers present themselves as promising alternatives to current treatments. In this study, we propose the use of PEGylated cationic carbosilane dendrimers as inhibitors of herpes simplex virus 2 (HSV-2) and human cytomegalovirus (HCMV)infections. Studies of mitochondrial toxicity, membrane integrity, internalization and viral infection inhibition indicated that G2-SN15-PEG, G3-SN31-PEG, G2-SN15-PEG fluorescein isothiocyanate (FITC) labeled and G3-SN31-PEG-FITC dendrimers are valid candidates to target HSV-2 and HCMV infections since they are biocompatible, can be effectively internalized and are able to significantly inhibit both infections. Later studies (including viral inactivation, binding inhibition, heparan sulphate proteoglycans (HSPG)binding and surface plasmon resonance assays) confirmed that inhibition takes place at first infection stages. More precisely, these studies established that their attachment to cell membrane heparan sulphate proteoglycans impede the interaction between viral glycoproteins and these cell receptors, thus preventing infection. Altogether, our research confirmed the high capacity of these PEGylated carbosilane dendrimers to prevent HSV-2 and HCMV infections, making them valid candidates as antiviral agents against Herpesviridae infections

    Non-Linear Optical Properties of Er3+–Yb3+-Doped NaGdF4 Nanostructured Glass–Ceramics

    Transparent oxyfluoride glass–ceramics containing NaGdF4 nanocrystals were prepared by melt-quenching and doped with Er3+ (0.5 mol%) and different amounts of Yb3+ (0–2 mol%). The selected dopant concentration the crystallization thermal treatments were chosen to obtain the most efficient visible up-conversion emissions, together with near infrared emissions. The crystal size increased with dopant content and treatment time. NaGdF4 NCs with a size ranging 9–30 nm were obtained after heat treatments at Tg + 20–80 °C as confirmed by X-ray diffraction and high-resolution transmission electron microscopy. Energy dispersive X-ray analysis shows the incorporation of rare earth ions into the NaGdF4 nanocrystals. Near-infrared emission spectra, together with the up-conversion emissions were measured. The optical characterization of the glass–ceramics clearly shows that Er3+ and Yb3+ ions are incorporated in the crystalline phase. Moreover, visible up-conversion emissions could be tuned by controlling the nanocrystals size through appropriated heat treatment, making possible a correlation between structural and optical properties.This research was funded by Spanish National projects MAT2017-87035-C2-1-P/2-P (AEI/FEDER, UE), Basque Country University PPG17/07 and GIU17/014 and Basque Government PIBA2018-24.     Dealing With the Pandemic of COVID-19 in Portugal: On the Important Role of Positivity, Experiential Avoidance, and Coping Strategies

    The global COVID-19 pandemic crisis has caused an unprecedented impact on most areas of people’s lives. Thus, framed within the scope of Existential Positive Psychology (PP2.0), this study aimed at assessing the psychological distress of adults living in Portugal during the first national lockdown, how they are coping with stress, as well to contribute to a deeper understanding about the role that positivity, experiential avoidance, and coping strategies have in psychological distress and well-being. For this purpose, 586 Portuguese adults (73% females) ranging between 18 and 78 years old (M = 38.96, SD = 12.20) completed an online survey during the initial phase of the pandemic crisis in Portugal. Findings suggest that experiential avoidance was the strongest predictor of a negative response (depression, anxiety, stress, loneliness, and negative emotions), whereas positivity was a better predictor of psychological well-being and lower levels of depression. Additionally, self-blame, behavioral disengagement, and emotional venting were strong risk factors for psychological distress, whereas positive reframing, planning, and acceptance were associated with more positive outcomes. These findings highlight the critical role of experiential avoidance on individuals’ psychological distress and the essential contribution of positive life orientation in promoting flourishing. By offering a better understanding of the complex navigation through the dialectics between positive and negative life features, this study provides important and useful cues for psychological interventions directed at promoting a more positive and adaptive human functioning even through such potential adverse and painful life events

    Update on and future perspectives for the diagnosis of alpha-1 antitrypsin deficiency in Brazil

Severe AATD predisposes individuals to COPD and liver disease. Early diagnosis is essential for implementing preventive measures and limiting the disease burden. Although national and international guidelines for the diagnosis and management of AATD have been available for 20 years, more than 85% of cases go undiagnosed and therefore untreated. In Brazil, reasons for the underdiagnosis of AATD include a lack of awareness of the condition among physicians, a racially diverse population, serum AAT levels being assessed in a limited number of individuals, and lack of convenient diagnostic tools. The diagnosis of AATD is based on laboratory test results. The standard diagnostic approach involves the assessment of serum AAT levels, followed by phenotyping, genotyping, gene sequencing, or combinations of those, to detect the specific mutation. Over the past 10 years, new techniques have been developed, offering a rapid, minimally invasive, reliable alternative to traditional testing methods. One such test available in Brazil is the A1AT Genotyping Test, which simultaneously analyzes the 14 most prevalent AATD mutations, using DNA extracted from a buccal swab or dried blood spot. Such advances may contribute to overcoming the problem of underdiagnosis in Brazil and elsewhere, as well as being likely to increase the rate detection of AATD and therefore mitigate the harmful effects of delayed diagnosis.This study received financial support from Grifols S.A., Barcelona, Spain

    A multidisciplinary approach to sexual behavior profiles: The SEX360 model

    The diversity of sexual behaviors is driven by multiple determinants, including physiological, cultural, educational, and sociological factors. However, the definition of sexual behavior profiles has been barely addressed from a comprehensive point of view. We aimed to develop a multidisciplinary questionnaire for defining individual sexual behavior profiles. The questionnaire was developed by a panel of experts with research experience in the fields of urology, gynecology, psychology, anthropology, and sexology. The list of items was defined in a focus group session and was based on four categories—family-oriented, loving, recreational, and functional—resulting from the combination of two axes: traditional versus non-traditional and sexual benefit versus extra-sexual benefit. Real-time Delphi dynamics was used to assign a weight to each question and a bias to the corresponding responses. The final questionnaire included 50 items considered relevant for describing sexual profiles; the final questionnaire was named SEX360. Of the 50 items included in SEX360, 14 were considered essential for computing the final score; 9 of them were associated with 2 categories, 4 of them with 4 categories, and 1 of them with 3. Nine items referred to the category “family-oriented”, 10 to “loving”, 8 to “recreational”, and 9 to “functional”. The weights assigned to each question ranged from 3.00 to 4.33, and the centers of gravity ranged from 1 to 4. The questionnaire proposed shows the existence of a vast diversity of sexual behavior profiles and may serve as a tool for sexual behavior research.Peer ReviewedPostprint (author's final draft

    Potential health claims of durum and bread wheat flours as functional ingredients

    Wheat is an important cereal with a key role in human nutrition. In this study, dietary fiber (DF) and arabinoxylans of di erent durum (Triticum turgidum ssp. Durum L.) and bread (Triticum aestivum L.) wheat flours were analyzed in order to point out their potential nutritional and health claims allege according to the current European regulation (Regulation (EU) No 432/2012). Moreover, other bioactive compounds (phenolics and tocopherols) were quantified as a first approach to their phytochemical composition in the analyzed wheat varieties. DF was analyzed following AOAC enzymatic-gravimetric methods; arabinoxylans and total phenols were quantified by colorimetric methods; tocopherols were determined by HPLC; antioxidant activity was evaluated using three di erent in vitro assays. Insoluble DF was the prevailing fraction in all analyzed samples. Water extractable arabinoxylans were higher in durum wheat flours. Whole flours contained higher total phenolics compounds. Alpha-tocopherol was the major isoform. Whole flours showed higher antioxidant properties.     Identification of Rickettsia species in ticks from ruminants in Lebanon

    Rickettsia are tick-borne emerging pathogens recognized as important agents of human tick-borne diseases worldwide. Aim of this study was the identification and characterization of Spotted Fever Group (SFG) rickettsiae in ticks from ruminants in Lebanon. The study concerned 88 ticks collected in June 2014 from 30 Lebanese farms of ruminants and identified according morphological keys. Total DNA was extracted and used to identify and characterize Rickettsia spp. DNA through PCR and sequencing of fragments of 17 kDa protein, ompA, ompB, gltA, atpA, dnaK, dnaA, recA and 16S rRNA. Five different tick species were found: Hyalomma anatolicum (n=6), Rhipicephalus annulatus (n=56), Rhipicephalus bursa (n=1), Rhipicephalus sanguineus (n=6) and Rhipicephalus turanicus (n=19). A prevalence of 68.2% was found at the first screening performed amplyfing Rickettsia spp. 16S rRNA. Among these Rickettsia positive samples, 17 were identified at the species level and 43 as SFG rickettsiae based on the multigene genotyping strategy. ‘Candidatus Rickettsia barbariae’, an emerging member of the rickettsial SFG, was identified in 9 samples. R. massiliae, R. aeschlimannii and R. africae were identified in four, three and one tick, respectively, confirming the presence of SFG Rickettsia spp. involved in human diseases. SFG rickettsiae with public health relevance were found in ticks collected in Lebanon, where the widespread distribution of tick vectors and possible livestock animal hosts in contact with humans may favor transmission to humans.     Dating historical droughts from religious ceremonies, the international pro pluvia rogation database

    Climate proxy data are required for improved understanding of climate variability and change in the pre-instrumental period. We present the first international initiative to compile and share information on pro pluvia rogation ceremonies, which is a well-studied proxy of agricultural drought. Currently, the database has more than 3500 dates of celebration of rogation ceremonies, providing information for 153 locations across 11 countries spanning the period from 1333 to 1949. This product provides data for better understanding of the pre-instrumental drought variability, validating natural proxies and model simulations, and multi-proxy rainfall reconstructions, amongst other climatic exercises.     Identification of new sources of resistance to RHBV- rice hoja blanca virus

    With the aim to find new sources of resistance to rice hoja blanca (white leaf) disease, transmitted by the insect Tagosodes orizicolus, 660 genotypes were evaluated under greenhouse and field conditions. Seven resistant genotypes were identified, and genomic studies were performed to demonstrate that the resistance in these sources is genetically different from that of Fedearroz 2000, which is currently the variety with the most resistance to hoja blanca. These new resistance sources constitute a resource that can be used to sustainably extend hoja blanca disease management throughout all of the rice-growing regions of tropical America. This is the first report of hoja blanca resistance in indica rice and different from that of Fedearroz 2000.Con el objetivo de encontrar nuevas fuentes de resistencia a la enfermedad de la hoja blanca del arroz, transmitida por el insecto Tagosodes orizicolus, se evaluaron 660 genotipos en condiciones de invernadero y campo. Se identificaron siete genotipos con resistencia a la enfermedad y se realizaron estudios del genoma para evidenciar que eran genéticamente diferentes a Fedearroz 2000, la variedad de mejor comportamiento ante el virus, en el momento. Estas nuevas fuentes de resistencia constituyen un recurso que puede utilizarse para extender un manejo sostenible de la enfermedad, en todas las regiones productoras de arroz en América tropical.
