    Evaluation of surface coating and shrink- wrap packaging on shelf life and quality of mango cultivar ‘Neelum’

    Kerala, the Indian state has the distinction of producing the earliest mangoes in the country, in February and the season extends up to May, coinciding with South West monsoon. Mango cultivar ‘Neelum’, the last commercial variety to attain maturity in the State is hampered by the incidence of fruit fly and anthracnose disease. An attempt was made during 2019-20 to extend the availability of the fruits of this cultivar by giving a surface coating with ‘Nipro Fresh’ wax containing the fungicide, Carbendazim, followed by shrink-wrap packaging in trays made of areca nut leaf sheath, before sanitizing with chlorine (100 ppm) and alum solution (1%). Surface coating with ‘Nipro Fresh’, followed by shrink-wrap packaging of trays containing mangoes, and their subsequent storage in cool chamber at 12-13 °C and 85-90 % relative humidity, extended the shelf life by 54 days, compared to the uncoated and unwrapped samples which had a shelf life of 9 days under ambient conditions. Respiration rate, physiological loss in weight, total soluble solids and carotenoids showed a steady rise while titratable acidity, total phenols and ascorbic acid recorded a declining trend

    Ancestral roles of the Fam20C family of secreted protein kinases revealed in C. elegans.

    Fam20C is a secreted protein kinase mutated in Raine syndrome, a human skeletal disorder. In vertebrates, bone and enamel proteins are major Fam20C substrates. However, Fam20 kinases are conserved in invertebrates lacking bone and enamel, suggesting other ancestral functions. We show that FAMK-1, the Caenorhabditis elegans Fam20C orthologue, contributes to fertility, embryogenesis, and development. These functions are not fulfilled when FAMK-1 is retained in the early secretory pathway. During embryogenesis, FAMK-1 maintains intercellular partitions and prevents multinucleation; notably, temperature elevation or lowering cortical stiffness reduces requirement for FAMK-1 in these contexts. FAMK-1 is expressed in multiple adult tissues that undergo repeated mechanical strain, and selective expression in the spermatheca restores fertility. Informatic, biochemical, and functional analysis implicate lectins as FAMK-1 substrates. These findings suggest that FAMK-1 phosphorylation of substrates, including lectins, in the late secretory pathway is important in embryonic and tissue contexts where cells are subjected to mechanical strain

    Mainstreaming Gender in the Context of Seaweed Farming SHGs in Ramanathapuram: A Pragmatic Evaluation

    A pragmatic examination on the impact of women SHGs in gender mainstreaming was embarked on the 10 seaweed farming SHG units operating at Ramanathapuram district in Tamil Nadu state. An explorative assessment on extent of empowerment, performance level, gender characteristics and economic feasibility analysis was carried out based on socio-economic surveys and personal interviews using pre-tested and structured data gathering protocols with standardized scales and indices. Empowerment Index was quantified based on 8 relevant dimensions constituting it. The male and female counterparts of the families were separately interviewed to evaluate the gender mainstreaming aspects in terms of equity and equality to access to resources, participation profile, decision making aspects, gender need analysis etc. Though majority of activities are male dominated, the female counterparts of the households also have committed role in site selection, seeding, drying, packing etc. The Scales of ‘Performance Assessment’ and ‘Empowerment Index’ developed for this study have upright prospective for future use in other key areas on a sustainable basis. Voids identified in Empowerment Index computation give adequate feedback to authorities to proceed in the accurate direction and to follow apposite changes in strategies of the microenterprise. The gender dimension analysis on mainstreaming aspect gives sensitization on crucial issues like women fisherfolk’s rights and marketing channels for policies and other interventions on gender. The identified interrelationships between the variables an act as catalytic points for group action and empowerment on a maintainable basis. The indicative economics worked out for the economic feasibility analysis of the SHGs advocates that, the unit takes just one year to break even which shows the cost-effectiveness of the seaweed farming enterprise

    Pathobiological Implications of MUC16 Expression in Pancreatic Cancer

    MUC16 (CA125) belongs to a family of high-molecular weight O-glycosylated proteins known as mucins. While MUC16 is well known as a biomarker in ovarian cancer, its expression pattern in pancreatic cancer (PC), the fourth leading cause of cancer related deaths in the United States, remains unknown. The aim of our study was to analyze the expression of MUC16 during the initiation, progression and metastasis of PC for possible implication in PC diagnosis, prognosis and therapy. In this study, a microarray containing tissues from healthy and PC patients was used to investigate the differential protein expression of MUC16 in PC. MUC16 mRNA levels were also measured by RT-PCR in the normal human pancreatic, pancreatitis, and PC tissues. To investigate its expression pattern during PC metastasis, tissue samples from the primary pancreatic tumor and metastases (from the same patient) in the lymph nodes, liver, lung and omentum from Stage IV PC patients were analyzed. To determine its association in the initiation of PC, tissues from PC patients containing pre-neoplastic lesions of varying grades were stained for MUC16. Finally, MUC16 expression was analyzed in 18 human PC cell lines. MUC16 is not expressed in the normal pancreatic ducts and is strongly upregulated in PC and detected in pancreatitis tissue. It is first detected in the high-grade pre-neoplastic lesions preceding invasive adenocarcinoma, suggesting that its upregulation is a late event during the initiation of this disease. MUC16 expression appears to be stronger in metastatic lesions when compared to the primary tumor, suggesting a role in PC metastasis. We have also identified PC cell lines that express MUC16, which can be used in future studies to elucidate its functional role in PC. Altogether, our results reveal that MUC16 expression is significantly increased in PC and could play a potential role in the progression of this disease

    Quantifying sources of variability in infancy research using the infant-directed-speech preference

    Psychological scientists have become increasingly concerned with issues related to methodology and replicability, and infancy researchers in particular face specific challenges related to replicability: For example, high-powered studies are difficult to conduct, testing conditions vary across labs, and different labs have access to different infant populations. Addressing these concerns, we report on a large-scale, multisite study aimed at (a) assessing the overall replicability of a single theoretically important phenomenon and (b) examining methodological, cultural, and developmental moderators. We focus on infants’ preference for infant-directed speech (IDS) over adult-directed speech (ADS). Stimuli of mothers speaking to their infants and to an adult in North American English were created using seminaturalistic laboratory-based audio recordings. Infants’ relative preference for IDS and ADS was assessed across 67 laboratories in North America, Europe, Australia, and Asia using the three common methods for measuring infants’ discrimination (head-turn preference, central fixation, and eye tracking). The overall meta-analytic effect size (Cohen’s d) was 0.35, 95% confidence interval = [0.29, 0.42], which was reliably above zero but smaller than the meta-analytic mean computed from previous literature (0.67). The IDS preference was significantly stronger in older children, in those children for whom the stimuli matched their native language and dialect, and in data from labs using the head-turn preference procedure. Together, these findings replicate the IDS preference but suggest that its magnitude is modulated by development, native-language experience, and testing procedure.     Design and baseline characteristics of the finerenone in reducing cardiovascular mortality and morbidity in diabetic kidney disease trial

    Background: Among people with diabetes, those with kidney disease have exceptionally high rates of cardiovascular (CV) morbidity and mortality and progression of their underlying kidney disease. Finerenone is a novel, nonsteroidal, selective mineralocorticoid receptor antagonist that has shown to reduce albuminuria in type 2 diabetes (T2D) patients with chronic kidney disease (CKD) while revealing only a low risk of hyperkalemia. However, the effect of finerenone on CV and renal outcomes has not yet been investigated in long-term trials. Patients and Methods: The Finerenone in Reducing CV Mortality and Morbidity in Diabetic Kidney Disease (FIGARO-DKD) trial aims to assess the efficacy and safety of finerenone compared to placebo at reducing clinically important CV and renal outcomes in T2D patients with CKD. FIGARO-DKD is a randomized, double-blind, placebo-controlled, parallel-group, event-driven trial running in 47 countries with an expected duration of approximately 6 years. FIGARO-DKD randomized 7,437 patients with an estimated glomerular filtration rate >= 25 mL/min/1.73 m(2) and albuminuria (urinary albumin-to-creatinine ratio >= 30 to <= 5,000 mg/g). The study has at least 90% power to detect a 20% reduction in the risk of the primary outcome (overall two-sided significance level alpha = 0.05), the composite of time to first occurrence of CV death, nonfatal myocardial infarction, nonfatal stroke, or hospitalization for heart failure. Conclusions: FIGARO-DKD will determine whether an optimally treated cohort of T2D patients with CKD at high risk of CV and renal events will experience cardiorenal benefits with the addition of finerenone to their treatment regimen.
