Evidence for the convergence of ÎČ-adrenergic and muscarinic signalling systems at a post-receptor site
AbstractThe ÎČ-adrenergic agonist isoproterenol stimulates inositol trisphosphate (IP3) formation and cytosolic Ca2+ ([Ca2+]i) mobilization in rat parotid acini via a cAMP-dependent process. Atropine, a muscarinic antagonist, inhibited these isoproterenol responses without affecting isoproterenol-induced amylase secretion or peak [Ca2+]i and IP3 responses elicited by α1-adrenergic stimulation with epinephrine. Atropine had no effect on isoproterenol-induced [Ca2+]i responses in a cell line which lacked muscarinic receptors and did not alter ÎČ-adrenoreceptor ligand binding. These results suggest that the inhibition by atropine results from a post-receptor effect on cAMP-mediated stimulation of phosphatidylinositol 4,5 bisphosphate (PIP2) hydrolysis
Carrier - envelope phase-tagged imaging of the controlled electron acceleration from SiO2 nanospheres in intense few-cycle laser fields
Waveform-controlled light fields offer the possibility of manipulating
ultrafast electronic processes on sub-cycle timescales. The optical lightwave
control of the collective electron motion in nanostructured materials is key
to the design of electronic devices operating at up to petahertz frequencies.
We have studied the directional control of the electron emission from 95 nm
diameter SiO2 nanoparticles in few-cycle laser fields with a well-defined
waveform. Projections of the three-dimensional (3D) electron momentum
distributions were obtained via single-shot velocity-map imaging (VMI), where
phase tagging allowed retrieving the laser waveform for each laser shot. The
application of this technique allowed us to efficiently suppress background
contributions in the data and to obtain very accurate information on the
amplitude and phase of the waveform-dependent electron emission. The
experimental data that are obtained for 4 fs pulses centered at 720 nm at
different intensities in the range (1â4) Ă 1013 W cmâ2 are compared to quasi-
classical mean-field Monte-Carlo simulations. The model calculations identify
electron backscattering from the nanoparticle surface in highly dynamical
localized fields as the main process responsible for the energetic electron
emission from the nanoparticles. The local field sensitivity of the electron
emission observed in our studies can serve as a foundation for future research
on propagation effects for larger particles and field-induced material changes
at higher intensities
Setting our sights on infectious diseases
In May 2019, the Wellcome Centre for Anti-Infectives Research (WCAIR) at the University of Dundee, UK, held an international conference with the aim of discussing some key questions around discovering new medicines for infectious diseases and a particular focus on diseases affecting Low and Middle Income Countries. There is an urgent need for new drugs to treat most infectious diseases. We were keen to see if there were lessons that we could learn across different disease areas and between the preclinical and clinical phases with the aim of exploring how we can improve and speed up the drug discovery, translational, and clinical development processes. We started with an introductory session on the current situation and then worked backward from clinical development to combination therapy, pharmacokinetic/pharmacodynamic (PK/PD) studies, drug discovery pathways, and new starting points and targets. Wine and other alcohol consumption and risk of ovarian cancer in the California Teachers Study cohort
OBJECTIVE: Whether alcohol consumption influences ovarian cancer risk is unclear. Therefore, we investigated the association between alcohol intake at various ages and risk of ovarian cancer. METHODS: Among 90,371 eligible members of the California Teachers Study cohort who completed a baseline alcohol assessment in 1995â1996, 253 women were diagnosed with epithelial ovarian cancer by the end of 2003. Multivariate Cox proportional hazards regression analysis was performed to estimate relative risks (RRs) and 95% confidence intervals (CIs). RESULTS: Consumption of total alcohol, beer, or liquor in the year prior to baseline, at ages 30â35 years, or at ages 18â22 years was not associated with risk of ovarian cancer. Consumption of at least one glass per day of wine, compared to no wine, in the year before baseline was associated with increased risk of developing ovarian cancer: RR = 1.57 (95% CI 1.11â2.22), P(trend) = 0.01. The association with wine intake at baseline was particularly strong among peri-/post-menopausal women who used estrogen-only hormone therapy and women of high socioeconomic status. CONCLUSIONS: Alcohol intake does not appear to affect ovarian cancer risk. Constituents of wine other than alcohol or, more likely, unmeasured determinants of wine drinking were associated with increased risk of ovarian cancer
Phyto-oestrogens and breast cancer chemoprevention
Phytoestrogens are polyphenol compounds of plant origin that exhibit a structural similarity to the mammalian steroid hormone 17ÎČ-oestradiol. In Asian nations the staple consumption of phyto-oestrogen-rich foodstuffs correlates with a reduced incidence of breast cancer. Human dietary intervention trials have noted a direct relationship between phyto-oestrogen ingestion and a favourable hormonal profile associated with decreased breast cancer risk. However, these studies failed to ascertain the precise effect of dietary phyto-oestrogens on the proliferation of mammary tissue. Epidemiological and rodent studies crucially suggest that breast cancer chemoprevention by dietary phyto-oestrogen compounds is dependent on ingestion before puberty, when the mammary gland is relatively immature. Phyto-oestrogen supplements are commercially marketed for use by postmenopausal women as natural and safe alternatives to hormone replacement therapy. Of current concern is the effect of phyto-oestrogen compounds on the growth of pre-existing breast tumours. Data are contradictory, with cell culture studies reporting both the oestrogenic stimulation of oestrogen receptor-positive breast cancer cell lines and the antagonism of tamoxifen activity at physiological phyto-oestrogen concentrations. Conversely, phyto-oestrogen ingestion by rodents is associated with the development of less aggressive breast tumours with reduced metastatic potential. Despite the present ambiguity, current data do suggest a potential benefit from use of phyto-oestrogens in breast cancer chemoprevention and therapy. These aspects are discussed
Fine-Scale Mapping of the 4q24 Locus Identifies Two Independent Loci Associated with Breast Cancer Risk
Background: A recent association study identified a common variant (rs9790517) at 4q24 to be associated with breast cancer risk. Independent association signals and potential functional variants in this locus have not been explored.
Methods: We conducted a fine-mapping analysis in 55,540 breast cancer cases and 51,168 controls from the Breast Cancer Association Consortium.
Results: Conditional analyses identified two independent association signals among women of European ancestry, represented by rs9790517 [conditional P = 2.51 Ă 10â4; OR, 1.04; 95% confidence interval (CI), 1.02â1.07] and rs77928427 (P = 1.86 Ă 10â4; OR, 1.04; 95% CI, 1.02â1.07). Functional annotation using data from the Encyclopedia of DNA Elements (ENCODE) project revealed two putative functional variants, rs62331150 and rs73838678 in linkage disequilibrium (LD) with rs9790517 (r2 â„ 0.90) residing in the active promoter or enhancer, respectively, of the nearest gene, TET2. Both variants are located in DNase I hypersensitivity and transcription factorâbinding sites. Using data from both The Cancer Genome Atlas (TCGA) and Molecular Taxonomy of Breast Cancer International Consortium (METABRIC), we showed that rs62331150 was associated with level of expression of TET2 in breast normal and tumor tissue.
Conclusion: Our study identified two independent association signals at 4q24 in relation to breast cancer risk and suggested that observed association in this locus may be mediated through the regulation of TET2.
Impact: Fine-mapping study with large sample size warranted for identification of independent loci for breast cancer risk
Proceedings of the 11th Annual Deep Brain Stimulation Think Tank: pushing the forefront of neuromodulation with functional network mapping, biomarkers for adaptive DBS, bioethical dilemmas, AI-guided neuromodulation, and translational advancements
The Deep Brain Stimulation (DBS) Think Tank XI was held on August 9â11, 2023 in Gainesville, Florida with the theme of âPushing the Forefront of Neuromodulationâ. The keynote speaker was Dr. Nico Dosenbach from Washington University in St. Louis, Missouri. He presented his research recently published in Nature inn a collaboration with Dr. Evan Gordon to identify and characterize the somato-cognitive action network (SCAN), which has redefined the motor homunculus and has led to new hypotheses about the integrative networks underpinning therapeutic DBS. The DBS Think Tank was founded in 2012 and provides an open platform where clinicians, engineers, and researchers (from industry and academia) can freely discuss current and emerging DBS technologies, as well as logistical and ethical issues facing the field. The group estimated that globally more than 263,000 DBS devices have been implanted for neurological and neuropsychiatric disorders. This year's meeting was focused on advances in the following areas: cutting-edge translational neuromodulation, cutting-edge physiology, advances in neuromodulation from Europe and Asia, neuroethical dilemmas, artificial intelligence and computational modeling, time scales in DBS for mood disorders, and advances in future neuromodulation devices
