    Repetitive Segmental Structure of the Transducin β Subunit: Homology with the CDC4 Gene and Identification of Related mRNAs

    Retinal transducin, a guanine nucleotide regulatory protein (referred to as a G protein) that activates a cGMP phosphodiesterase in photoreceptor cells, is comprised of three subunits. We have identified and analyzed cDNA clones of the bovine transducin β subunit that may be highly conserved or identical to that in other G proteins. From the cDNA nucleotide sequence of the entire coding region, the primary structure of a 340-amino acid protein was deduced. The encoded β subunit has a Mr of 37,375 and is comprised of repetitive homologous segments arranged in tandem. Furthermore, significant homology in primary structure and segmental sequence exists between the β subunit and the yeast CDC4 gene product. The Mr 37,375 β subunit polypeptide is encoded by a 2.9-kilobase (kb) mRNA. However, there exists in retina other β-related mRNAs that are divergent from the 2.9-kb mRNA on the basis of oligonucleotide and primer-extended probe hybridizations. All mammalian tissues and clonal cell lines that have been examined contain at least two β-related mRNAs, usually 1.8 and 2.9 kb in length. These results suggest that the mRNAs are the processed products of a small number of closely related genes or of a single highly complex β gene

    Genetic diversity of peanut (Arachis hypogaea L.) and its wild relatives based on the analysis of hypervariable regions of the genome

    BACKGROUND: The genus Arachis is native to a region that includes Central Brazil and neighboring countries. Little is known about the genetic variability of the Brazilian cultivated peanut (Arachis hypogaea, genome AABB) germplasm collection at the DNA level. The understanding of the genetic diversity of cultivated and wild species of peanut (Arachis spp.) is essential to develop strategies of collection, conservation and use of the germplasm in variety development. The identity of the ancestor progenitor species of cultivated peanut has also been of great interest. Several species have been suggested as putative AA and BB genome donors to allotetraploid A. hypogaea. Microsatellite or SSR (Simple Sequence Repeat) markers are co-dominant, multiallelic, and highly polymorphic genetic markers, appropriate for genetic diversity studies. Microsatellite markers may also, to some extent, support phylogenetic inferences. Here we report the use of a set of microsatellite markers, including newly developed ones, for phylogenetic inferences and the analysis of genetic variation of accessions of A. hypogea and its wild relatives. RESULTS: A total of 67 new microsatellite markers (mainly TTG motif) were developed for Arachis. Only three of these markers, however, were polymorphic in cultivated peanut. These three new markers plus five other markers characterized previously were evaluated for number of alleles per locus and gene diversity using 60 accessions of A. hypogaea. Genetic relationships among these 60 accessions and a sample of 36 wild accessions representative of section Arachis were estimated using allelic variation observed in a selected set of 12 SSR markers. Results showed that the Brazilian peanut germplasm collection has considerable levels of genetic diversity detected by SSR markers. Similarity groups for A. hypogaea accessions were established, which is a useful criteria for selecting parental plants for crop improvement. Microsatellite marker transferability was up to 76% for species of the section Arachis, but only 45% for species from the other eight Arachis sections tested. A new marker (Ah-041) presented a 100% transferability and could be used to classify the peanut accessions in AA and non-AA genome carriers. CONCLUSION: The level of polymorphism observed among accessions of A. hypogaea analyzed with newly developed microsatellite markers was low, corroborating the accumulated data which show that cultivated peanut presents a relatively reduced variation at the DNA level. A selected panel of SSR markers allowed the classification of A. hypogaea accessions into two major groups. The identification of similarity groups will be useful for the selection of parental plants to be used in breeding programs. Marker transferability is relatively high between accessions of section Arachis. The possibility of using microsatellite markers developed for one species in genetic evaluation of other species greatly reduces the cost of the analysis, since the development of microsatellite markers is still expensive and time consuming. The SSR markers developed in this study could be very useful for genetic analysis of wild species of Arachis, including comparative genome mapping, population genetic structure and phylogenetic inferences among species

    Effects of Acute Eccentric Exercise on Appetite-Related Hormones and Food Preferences in Men

    Eccentric exercise has been suggested to improve muscle atrophy, muscle function, and insulin sensitivity. The aim of this study was to examine the effect of acute eccentric exercise on appetite-related hormones, food preferences, and food intake. Fourteen moderately active men were recruited to participate in this study (age 24.2 ± 5.5 years; BMI 23.4 ± 3.3 kg/m2; VO2max 48.9 ± 3.1 ml/kg/min). Three different conditions were implemented; no exercise, flat running “inclination 0” and downhill running “inclination –12%.” Appetite-related hormones, subjective appetite sensations, food preference and reward, and ad libitum food intake were measured at pre-, immediately post-, and 24 h post exercise.There were no significant median changes in total ghrelin or pancreatic peptide concentrations between conditions. There were also no median differences in subjective appetite ratings or energy intake between conditions, but the median change in explicit liking of sweet versus savory foods differed significantly between pre-exercise and 24 h post exercise (p = .013). Post-hoc analysis observed a significant difference in the pre-exercise to 24 h post exercise change between front running and downhill running (p = .023), and indicated greater liking of savory foods over sweet foods in downhill running than front running. However, no further differences were seen between conditions for the remaining food preference parameters, suggesting there were no systematic trends in these data. In conclusion, there was no effect of front and downhill running on eating behavior as compared to a nonexercise control condition, but these data need to be replicated in a larger and more heterogeneous sample

    Model Benchmarking and Reference Signals for Angled-beam Shear Wave Ultrasonic NDE Inspections

    NDE modeling and simulation are important tools to support the development and validation of enhanced localization and characterization techniques. Previously, important achievements were made by the USAF to address crack detection in aircraft structures using angled-beam shear wave inspection techniques. However, new work on model benchmarking is needed to move beyond detection and achieve reliable crack characterization. To achieve this goal, simulated studies are needed to verify that models can accurately represent all of the key variables with the inspection of multilayer structures with fastener sites and varying crack conditions. Often with model benchmark studies, the accuracy of the model is evaluated based on the change in response relative to a selected reference signal. During recent simulated and experimental studies, some challenges were discovered concerning the creation and/or selection of a reference signal in a plate with a vertical hole and crack. The focus of this paper is on key findings concerning model benchmarking using CIVA-UT for angled-beam shear wave inspections. The use of a side drilled hole (SDH) in a plate was found to be somewhat problematic as a reference signal for angled beam shear wave inspection. Previously, only a limited number of studies have looked at model benchmarking for angled beam shear wave inspections. Systematic studies were performed with varying SDH depth and size, and varying the ultrasonic probe frequency, focal depth, and probe height. Care must be taken in understanding the precise beam properties with these experiments. One issue is that there is some increased error with the simulation of angled shear wave beams, especially in the near-field. Even more significant, asymmetry in real probes and the inherent sensitivity of signals in the near-field to subtle test conditions were found to provide a greater challenge with achieving model agreement. Through these studies, conditions of good and poor agreement were observed. For some inspection conditions, the skip signal off of the far wall from the side drilled hole can provide a better reference than the direct reflected signal. All in all, these seemingly mundane studies were found to be important with providing guidance on reference signal selection for model benchmarking work on the inspection of fastener sites with cracks

    Mass of the Southern Black Hole in NGC 6240 from Laser Guide Star Adaptive Optics

    NGC 6240 is a pair of colliding disk galaxies, each with a black hole in its core. We have used laser guide star adaptive optics on the Keck II telescope to obtain high-resolution (0.06\sim 0.06") near-infrared integral-field spectra of the region surrounding the supermassive black hole in the south nucleus of this galaxy merger. We use the K-band CO absorption bandheads to trace stellar kinematics. We obtain a spatial resolution of about 20 pc and thus directly resolve the sphere of gravitational influence of the massive black hole. We explore two different methods to measure the black hole mass. Using a Jeans Axisymmetric Multi-Gaussian mass model, we investigate the limit that a relaxed mass distribution produces all of the measured velocity dispersion, and find an upper limit on the black hole mass at 2.0 \pm 0.2 \times 10^9 M_{\sun}. When assuming the young stars whose spectra we observe remain in a thin disk, we compare Keplerian velocity fields to the measured two-dimensional velocity field measured and fit for a mass profile containing a black hole point mass plus a radially-varying spherical component, which suggests a lower limit for the black hole mass of 8.7 \pm 0.3 \times 10^8 M_{\sun}. Our measurements of the stellar velocity dispersion place this AGN within the scatter of the MBHM_{BH}-σ\sigma_{*} relation. As NGC 6240 is a merging system, this may indicate that the relation is preserved during a merger at least until the final coalescence of the two nuclei.Comment: 10 pages, 12 figures; accepted to Ap

    FIRST-2MASS Red Quasars: Transitional Objects Emerging from the Dust

    We present a sample of 120 dust-reddened quasars identified by matching radio sources detected at 1.4 GHz in the FIRST survey with the near-infrared 2MASS catalog and color-selecting red sources. Optical and/or near-infrared spectroscopy provide broad wavelength sampling of their spectral energy distributions that we use to determine their reddening, characterized by E(B-V). We demonstrate that the reddening in these quasars is best-described by SMC-like dust. This sample spans a wide range in redshift and reddening (0.1 < z < 3, 0.1 < E(B-V) < 1.5), which we use to investigate the possible correlation of luminosity with reddening. At every redshift, dust-reddened quasars are intrinsically the most luminous quasars. We interpret this result in the context of merger-driven quasar/galaxy co-evolution where these reddened quasars are revealing an emergent phase during which the heavily obscured quasar is shedding its cocoon of dust prior to becoming a "normal" blue quasar. When correcting for extinction, we find that, depending on how the parent population is defined, these red quasars make up < 15-20% of the luminous quasar population. We estimate, based on the fraction of objects in this phase, that its duration is 15-20% as long as the unobscured, blue quasar phase.Comment: 21 pages, 17 figures plus a spectral atlas.     The physical scale of the far-infrared emission in the most luminous submillimetre galaxies II: evidence for merger-driven star formation

    We present high-resolution 345 GHz interferometric observations of two extreme luminous (L_{IR}>10^{13} L_sun), submillimetre-selected galaxies (SMGs) in the COSMOS field with the Submillimeter Array (SMA). Both targets were previously detected as unresolved point-sources by the SMA in its compact configuration, also at 345 GHz. These new data, which provide a factor of ~3 improvement in resolution, allow us to measure the physical scale of the far-infrared in the submillimetre directly. The visibility functions of both targets show significant evidence for structure on 0.5-1 arcsec scales, which at z=1.5 translates into a physical scale of 5-8 kpc. Our results are consistent with the angular and physical scales of two comparably luminous objects with high-resolution SMA followup, as well as radio continuum and CO sizes. These relatively compact sizes (<5-10 kpc) argue strongly for merger-driven starbursts, rather than extended gas-rich disks, as the preferred channel for forming SMGs. For the most luminous objects, the derived sizes may also have important physical consequences; under a series of simplifying assumptions, we find that these two objects in particular are forming stars close to or at the Eddington limit for a starburst.Comment: 9 pages, 3 Figures, submitted to MNRA

    Probing the role of the cation–π interaction in the binding sites of GPCRs using unnatural amino acids

    We describe a general application of the nonsense suppression methodology for unnatural amino acid incorporation to probe drug–receptor interactions in functional G protein-coupled receptors (GPCRs), evaluating the binding sites of both the M2 muscarinic acetylcholine receptor and the D2 dopamine receptor. Receptors were expressed in Xenopus oocytes, and activation of a G protein-coupled, inward-rectifying K^+ channel (GIRK) provided, after optimization of conditions, a quantitative readout of receptor function. A number of aromatic amino acids thought to be near the agonist-binding site were evaluated. Incorporation of a series of fluorinated tryptophan derivatives at W6.48 of the D2 receptor establishes a cation–π interaction between the agonist dopamine and W6.48, suggesting a reorientation of W6.48 on agonist binding, consistent with proposed “rotamer switch” models. Interestingly, no comparable cation–π interaction was found at the aligning residue in the M2 receptor

    Hdelta-Selected Galaxies in the Sloan Digital Sky Survey I: The Catalog

    [Abridged] We present here a new and homogeneous sample of 3340 galaxies selected from the Sloan Digital Sky Survey (SDSS) based solely on the observed strength of their Hdelta absorption line. These galaxies are commonly known as ``post-starburst'' or ``E+A'' galaxies, and the study of these galaxies has been severely hampered by the lack of a large, statistical sample of such galaxies. In this paper, we rectify this problem by selecting a sample of galaxies which possess an absorption Hdelta equivalent width of EW(Hdelta_max) - Delta EW(Hdelta_max) > 4A from 106682 galaxies in the SDSS. We have performed extensive tests on our catalog including comparing different methodologies of measuring the Hdelta absorption and studying the effects of stellar absorption, dust extinction, emission-filling and measurement error. The measured abundance of our Hdelta-selected (HDS) galaxies is 2.6 +/- 0.1% of all galaxies within a volume-limited sample of 0.05<z<0.1 and M(r*)<-20.5, which is consistent with previous studies of such galaxies in the literature. We find that only 25 of our HDS galaxies in this volume-limited sample (3.5+/-0.7%) show no evidence for OII and Halpha emission, thus indicating that true E+A (or k+a) galaxies are extremely rare objects at low redshift, i.e., only 0.09+/-0.02% of all galaxies in this volume-limited sample are true E+A galaxies. In contrast, 89+/-5% of our HDS galaxies in the volume-limited sample have significant detections of the OII and Halpha emission lines. We find 27 galaxies in our volume-limited HDS sample that possess no detectable OII emission, but do however possess detectable Halpha emission. These galaxies may be dusty star-forming galaxies. We provide the community with this new catalog of Hdelta-selected galaxies to aid in the understanding of these galaxies.Comment: Submitted to PASJ.
