    Separation of proteins via pH parametric pumping with electric field

    A new semi-continuous parametric pumping process for the separation of protein mixtures has been developed, based on cyclic variation of pH and electric field. The model system used for this process consisted of a mixture of human serum albumin and human hemoglobin in contact with CM Sepharose cation exchanger. Experimental results show that the protein separation via the pH parapump with electric field is two to three times greater than the results which are obtained in batch chromatography or via either single-column or multi-column parapumps with cyclic variation of pH and ionic strength. The separation in the new process depends on selective adsorption of one protein onto the ion exchanger, and also on a difference in the migration velocities of the two proteins in the presence of an electric field. Protein A (hemoglobin) was stripped from the top stream and enriched in the bottom stream by adsorption at low pH and desorption at high pH. The top stream was thereby enriched in Protein B (albumin) relative to Protein A. Protein B was then stripped from the bottom stream by applying an electric field across the parapump during the desorption stage of each cycle of the process. Various modes of operation and a number of experimental parameters were considered. A low ionic strength organic buffer (0.05M Tris-Maleate + NaOH) was selected in order to maximize field strength with minimum heat generation, and the optimum fluid displacement for this buffer was determined experimentally. The effect of the electric field on the adsorption and desorption concentration waves and pH profiles was examined for different bulk velocities and alternate field polarities. These results were applied to the final process. A mathematical model was developed to verify the experimental data, based on finite mass transfer and constant electric mobility. The adsorption isotherm was assumed to be linear in the region of interest. The transport equations were solved by the finite difference method and also by the Stop & Go model. A pH wave lag was necessarily incorporated into the mathematical model. The pH wave velocity was assumed to be constant. The model agrees well with the experiemental data. Protein separation was optimized mathematically as a function of top or bottom feed rates and as a function of total production rate. Various parametric pumping processes have been reported based on temperature, pressure, and pH. The use of electric field polarity as a thermodynamic variable in a parapumping process is a promising new concept in protein separation technology

    Cis,cis-dipropenyl ether was prepared by potassium t-butoxide catalyzed rearrangement of diallyl ether in diglyme solution. Pyrolysis was used in an attempt to produce an isomeric dipropenyl ether mixture with a high percentage of the less stable trans,trans isomer; 21.6% trans,trans-dipropenyl ether was produced by pyrolysis at 395°C, but formation of an azeotropic byproduct in significant quantities negated the feasibility of this synthesis. A mixture of 45.7% cis,cis-, 43.6% cis,trans-, and 10.7% trans,trans-dipropenyl ether was produced by mercuric acetate catalyzed isomerization at 80°C, and the mixture was separated on a Nester/Faust spinning band column of two hundred theoretical plates. Physical properties, including boiling point, density, index of refraction, and proton and carbon-13 chemical shifts were determined for all three isomers. Liquid-phase Raman spectra and liquid-phase, vapor-phase, and CCl4 solution infrared spectra were taken on the three dipropenyl ethers and vibrational frequency assignments were made. Conformations have been established based on the vibrational and NMR spectra. The most stable conformation for cis,cis-dipropenyl ether is the planar C2v form, but a non-planar conformer exists at higher temperatures, in the liquid-phase, and in solution. Trans,trans-dipropenyl ether is slightly more stable in the planar Cs conformation with the planar C2v conformer present at a slightly lower population level. Two Cs conformations are found for cis,trans-dipropenyl ether; the linear coupled Cs planar form is more stable than the bent Cs planar conformation

    Tato bakalářská práce se zabývá návrhem konstrukčního řešení protézy zápěstí umožňující polohovatelnost a aretaci a následnou realizací funkčního prototypu. Konstrukční řešení je navrženo tak, aby bylo inovativní. S ohledem na rešerši je volen takový rozsah pohybu a počet poloh aretačního mechanismu, aby byla zajištěna funkčnost protézy zápěstí. Velikost protézy zápěstí je dimenzována pro ruku dospělého muže. Pro návrh konstrukčního řešení byl využit program Autodesk Inventor Proffesional 2012, výkresová dokumentace byla zpracována s pomocí AutoCAD Mechanical 2012. Jednotlivé díly prototypu byly vytvořeny pomocí technologie rapid prototyping z ABS plastu, případně obráběním ocelových polotovarů.This bachelor thesis deals with the structural design of the wrist prosthesis allowing positionability and lock and subsequent implementation of a functional prototype. The structural design is made to be innovative. With respect to the research, the range of movement and the number of positions of the locking mechanism are selected to ensure the functionality of the prosthetic wrist. The size of wrist prosthesis is designed for an adult male hand. For the structural design Autodesk Inventor Proffesional 2012 was used. Drawings were prepared using AutoCAD Mechanical 2012. Individual components of the prototype are made of ABS plastic, using rapid prototyping technology, or steel machining blanks.

    Review of gender, socioeconomic status, and ethnic background considerations reported in active school travel intervention studies

    Introduction: Trends over the last half century have shown that fewer children today are engaging in active modes of travel to and from school than in previous generations. Interventions promoting active school travel (AST) can be effective at reversing these trends and increasing rates of AST among children. This paper aims to explore (1) how studies of AST interventions have considered or framed multiple equity factors, namely those related to gender, socioeconomic status (SES), and ethnic background, in the design and evaluation of AST interventions, and (2) to what extent studies of AST interventions report these equity considerations in their analyses, outcomes, programming, and discussions. Methods: Six databases were used (BIOSIS Previews, GeoBase, PubMed, SCOPUS, SPORTDiscus, and Web of Science) to obtain AST literature published between 2010 and 2019 from around the world. The inclusion criteria applied in this review included a focus on an AST intervention for children (aged 5–19 years), quantitative results, and a primary outcome related to AST. A total of 69 papers were included in the review. Results: Across AST intervention studies, equity considerations were scarcely reported. Among the explicit mentions, gender and socioeconomic status were reported most often compared to ethnic background. Some papers reported differential effects among groups. Gender, SES, and ethnic background were, when reported, generally considered as variables that were controlled for within study samples. Conclusions: Suggestions to address equity within studies of AST interventions include reporting equity considerations made throughout the intervention and its evaluation in the published article. Using and reporting equity frameworks to develop AST interventions and conducting sub-group analyses during evaluation allows for greater assessment of equity effects and improved utility and applicability of the AST intervention. Ensuring the integration of equity into AST literature is needed to support evidence-based and equitable practices
