    Beta-hydroxy-beta-methyl-butyrate blunts negative age-related changes in body composition, functionality and myofiber dimensions in rats

    <p>Abstract</p> <p>Purpose</p> <p>To determine the effects of 16 wk. of beta-hydroxy-beta-methylbutyrate (HMB) administration on age-related changes in functionality and diffusion tensor imaging (DTI) determined myofiber dimensions.</p> <p>Methods</p> <p>Twelve young (44 wk.), 6 middle-aged (60 wk.), 10 old (86 wk.), and 5 very old (102 wk.) male Fisher-344 rat's body composition and grip strength were assessed at baseline. Following, 6 young, 6 middle-aged, 5 old and 5 very old rats were sacrificed for baseline myofiber dimensions and gene transcript factor expression in the soleus (SOL) and gastrocnemius (GAS). The remaining 6 young and 5 old rats were given HMB for 16 wk. and then sacrificed.</p> <p>Results</p> <p>Fat mass increased in the middle-aged control condition (+49%) but not the middle-aged HMB condition. In addition, fat mass declined (-56%) in the old HMB condition but not the old control condition. Normalized strength declined and maintained respectively in the control and HMB conditions from 44 to 60 wk. and increased (+23%) (p < 0.05) from 86 to 102 wk. in only the HMB condition. Declines occurred in myofiber size in all muscles from 44 to 102 wk. in the control condition(-10 to -15%), but not HMB condition. Atrogin-1 mRNA expression in the SOL and GAS muscles was greater in the 102-wk control condition than all other conditions: SOL (+45%) and GAS (+100%). This elevation was blunted by HMB in the 102 wk. old SOL. There was a condition effect in the SOL for myogenin, which significantly increased (+40%) only in the 102-wk. HMB group relative to the 44-wk. group.</p> <p>Conclusions</p> <p>HMB may blunt age-related losses of strength and myofiber dimensions, possibly through attenuating the rise in protein breakdown.</p

    Combined addition of superoxide dismutase, catalase and glutathione peroxidase improves quality of cooled stored stallion semen.

    During cold storage stallion spermatozoa experience undergo oxidative stress, which can impair sperm function and fertilizing capacity. Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) are the main endogenous enzymatic antioxidants in stallion seminal plasma, and counteract reactive oxygen species. Semen dilution reduces the endogenous antioxidant concentrations. The aim of this study was to investigate whether addition of 15 IU/mL each of SOD, CAT, and GPX to diluted stallion semen would ameliorate a reactive oxygen-mediated decrease in semen quality during 72 h of storage at 5 °C. Ejaculates (n = 7) were divided in two aliquots and diluted in INRA 96 without (control) or with addition of antioxidants. Semen analysis was performed at the time of dilution and every 24 h during chilled storage. Antioxidant supplementation completely inhibited the storage-dependent increase in activated caspase 3 (P < 0.05). Concomitantly, the antioxidant-supplemented samples had a greater percentage of viable, motile and rapidly moving sperm than control samples after 72 h storage (P < 0.05). The DNA damage, as evaluated by TUNEL assay and SCSA, increased with storage time (P < 0.05). Antioxidant supplementation did not prevent, but did significantly reduce the increase in DNA strand breakage. The results indicate part of the intrinsic apoptotic pathway leading to effector caspase activation was inhibited, although an activation of molecules with endonuclease activity still occurred. In conclusion, adding equal concentrations of SOD, CAT and GPX to a semen extender suppressed caspase-3 activation and improved preservation of stallion sperm motility and viability during 72 h of storage at 5 °C

    Plant toxin levels in nectar vary spatially across native and introduced populations

    1. Secondary compounds in nectar may function as toxic chemical defences and regulate floral antagonists, but can also mediate plant-pollinator interactions. Despite their ecological importance, few studies have investigated the patterns, causes and consequences of spatial variation in toxic nectar compounds in plant species, and none outside their native range. 2. Grayanotoxin I (GTX I) occurs in nectar of invasive Rhododendron ponticum where it is toxic to honeybees and some solitary bee species. We examined (i) geographic variation in the composition of nectar GTX I and GTX III (which is not toxic to these species) in the native and introduced range of R. ponticum, (ii) how their expression is structured at patch and landscape scales within ranges, and (iii) if climatic and environmental factors underpin spatial patterns. 3. While both GTXs varied within ranges, variation in GTX I, but not GTX III, was detected between ranges. GTX I expression was thus markedly lower or (in 18% of cases) absent from nectar in introduced plants. Spatial autocorrelation was apparent at both patch and landscape scales, and in part related to heat load interception by plants (a function of latitude, aspect and slope) 4. As expression of nectar GTXs was generally robust to environmental variation, and aggregated in space, this trait has the potential to be spatially discriminated by consumers. Given the specificity of change to GTX I, and its differential toxicity to some bee species, we conclude that its expression was likely influenced during invasion by pollinator-mediated selection and/or enemy-release from floral antagonists. 5. Synthesis. As the first demonstration of large-scale geographic variation and spatial structure in toxic nectar compounds, this work deepens our understanding of the chemical ecology of floral interactions in native and introduced species. Spatially explicit studies of nectar secondary compounds are thus required to show how the extent and structure of spatial variation may affect floral ecology. Future development of invasion theory should incorporate a holistic view of plant chemical defence, beyond antagonistic interactions, which integrates the consequences of chemically defended mutualist rewards

    Intestinal stem cells remain viable after prolonged tissue storage

    Intestinal stem cells (ISCs) are responsible for renewal of the epithelium both during normal homeostasis and following injury. As such they have significant therapeutic potential. However, it is unknown whether ISCs can survive tissue storage. We hypothesized that, although the majority of epithelial cells may die, ISCs would remain viable for at least 24 h at 4°C. To explore this hypothesis, jejuni of C57Bl6/J or Lgr5-LacZ mice were removed and either processed immediately or placed in phosphate buffered saline (PBS) at 4°C. Delayed isolations of epithelia were performed after 24, 30, or 48 h storage. At the light microscope level, despite extensive apoptosis of villus epithelial cells, small intestinal crypts remained morphologically intact through 30 h and ISCs were identifiable via Lgr5-LacZ positivity. Electron microscopy showed that ISCs retain high integrity through 24 h. When assessed by flow cytometry, ISCs were more resistant to degeneration than the rest of the epithelium, including neighboring Paneth cells, with higher viability across all time points. Culture of isolated crypts showed no loss of capacity to form complex enteroids after 24 h tissue storage, with efficiencies after 7 days of culture remaining above 80%. By 30 h storage, efficiencies declined but budding capability was retained. We conclude that, with delay in isolation, ISCs remain viable and retain their proliferative capacity. In contrast, the remainder of the epithelium, including the Paneth cells, exhibits degeneration and programmed cell death. If these findings are recapitulated with human tissue, storage at 4°C may offer a valuable temporal window for harvest of crypts or ISCs for therapeutic application

    Differential Macrophage Polarization Promotes Tissue Remodeling and Repair in a Model of Ischemic Retinopathy

    Diabetic retinopathy is the leading cause of visual loss in individuals under the age of 55. Umbilical cord blood (UCB)–derived myeloid progenitor cells have been shown to decrease neuronal damage associated with ischemia in the central nervous system. In this study we show that UCB-derived CD14+ progenitor cells provide rescue effects in a mouse model of ischemic retinopathy by promoting physiological angiogenesis and reducing associated inflammation. We use confocal microscopy to trace the fate of injected human UCB-derived CD14+ cells and PCR with species-specific probes to investigate their gene expression profile before and after injection. Metabolomic analysis measures changes induced by CD14+ cells. Our results demonstrate that human cells differentiate in vivo into M2 macrophages and induce the polarization of resident M2 macrophages. This leads to stabilization of the ischemia-injured retinal vasculature by modulating the inflammatory response, reducing oxidative stress and apoptosis and promoting tissue repair

    In 1989, the United Nation Convention on the Rights of the Child declared, “[state parties] shall take all feasible measures to ensure protection and care of children who are affected by an armed conflict.” In addition to attempting to secure the welfare of children in armed conflict, the Convention went on to ban the recruitment and deployment of children during armed conflict. Despite the vast majority of sovereign nations signing and ratifying this agreement, this treaty, unfortunately, has not prevented children and youth from witnessing, becoming victims of, or participating in political, ethnic, religious, and cultural violence across the past three decades. This chapter offers an “ecological perspective” on the psychosocial consequences of exposure to the trauma of war-related violence and social disruption

    AN ANALYSIS OF THE APTITUDE-TREATMENT INTERACTION BETWEEN CONSULTEE LOCUS-OF-CONTROL AND CONSULTATION MODEL

    An aptitude-treatment interaction was sought to indicate which model of consultation would be most successful with which group of consultees using locus of control as the aptitude measure. Subjects were 300 students of Education with at least one and not more than three credit hours of teaching experience. Subjects were assigned to treatment groups based on their scores on the Adult Nowicki-Strickland Internal-External Control Scale (ANSIE). Extreme groups were formed by drawing from the top (more external) and bottom (more internal) 27% of the score distribution (Kelly, 1939). Two video tapes of the problem-solving and planning phases of consultation were produced. One of the tapes portrayed a collaborative consultation relationship while the other portrayed a directive consultation relationship. Approximately half of the externals and half of the internals viewed the directive video tape. The remaining subjects viewed the tape of the collaborative consultation sessions. All subjects then viewed a second video tape of a classroom situation in which one child was having behavioral difficulty. Following the final tape, subjects responded to four dependent measures. These measures assessed subjects\u27 skills at identifying the problem and developing intervention plans, their ranked expectations that their plans would be successful, and their interpersonal attraction to the consultant in the consultation tape viewed. It was expected that external subjects viewing the directive consultation and internal subjects viewing the collaborative consultation would obtain more successful scores on the dependent measures than their counterparts in unmatched treatments. A multivariate analysis of variance indicated no significant results at any level of analysis. The main effects and interaction effect were measured by obtaining Wilk\u27s Lambda values and performing Rao\u27s F transformations to derive approximate F values. No significant evidence of the benefit of matching consultation approach with consultee locus of control was found. Although futher investigations are needed to determine if other cognitive or personality variables would interact with specific consultation approaches, the current findings do indicate the possibility that directive and collaborative consultation are equally well received by young educators regardless of their locus of control perspectives
