    Revisiting Out-of-SSA Translation for Correctness, Code Quality, and Efficiency

    Compared to the previous versions, the only change is correcting an awful typo that made Algorithm 1 wrong. Line 18 is not "if b = loc(pred(b))" but simply "if b = loc(b)".Static single assignment (SSA) form is an intermediate program representation in which many code optimizations can be performed with fast and easy-to-implement algorithms. However, some of these optimizations create situations where the SSA variables arising from the same original variable now have overlapping live ranges. This complicates the translation out of SSA code into standard code. There are three issues to consider: correctness, code quality (elimination of copies), and algorithm efficiency (speed and memory footprint). Briggs et al. proposed patches to correct the initial approach of Cytron et al. A cleaner and more general approach was proposed by Sreedhar et al., along with techniques to reduce the number of generated copies. We propose a new approach based on coalescing and a precise view of interferences, in which correctness and optimizations are separated. Our approach is provably correct and simpler to implement, with no patches or particular cases as in previous solutions, while reducing the number of generated copies. Also, experiments with SPEC CINT2000 show that it is 2x faster and 10x less memory-consuming than the Method~III of Sreedhar et al., which makes it suitable for just-in-time compilation

    International audienceIn the Sahel, the rapid increase of the population during the last decades and the climate variation lead to an important environmental degradation. This work aims to measure the impacts of the human pressure on ecosystem during the six last decades. A diachronic cartography of a 100 km² area close to Niamey was done with aerial photographs (1950 and 1975) and GPS measurements (2009). Results showed that the tiger bush vegetation was completely cleared between 1950 and 2009 while the fallow decreases from 7 % to 1 %. In the sandy valley, the increase of cultivated fields from 20,7 % (1950) to 69,4 % (1975) favoured wind and water erosions which allowed surface soil crusting. Between 1975 and 2009, the bare crusted soil dramatically developed at the expense of the cultivated area which represents only 54,4 % of the studied area. The valleys are going to be overloaded because of high sedimentation rate (> 4 cm per year).Au Sahel, l'explosion démographique de ces dernières décennies et les variations climatiques ont provoqué d'importants changements environnementaux. L'objectif de ce travail est de mesurer les impacts de la pression anthropique sur les écosystèmes dans la région de Niamey au cours des six dernières décennies. L'étude est fondée sur une cartographie diachronique d'une aire de 100 km² située près de Niamey au moyen de photographies aériennes (1950 et 1975) et relevés au GPS (2009). Il est apparu ainsi qu'entre 1950 et 2009, la végétation de la brousse tigrée a été complètement déboisée. Dans les vallées sableuses, les surfaces cultivées ont connu une extension passant de 20,7 % à 69,4 % entre 1950 et 1975. Ceci a favorisé l'emprise des érosions hydrique et éolienne qui ont abouti à une dégradation des terres par encroûtement des sols. Celui-ci est à l'origine de la baisse des surfaces cultivées entre 1975 et 2009 (de 69,4 % à 54,4 %). Dans les bas- fonds, la tendance est au comblement du fait d'un taux de sédimentation de plus de 4 cm par an

    Combined approaches provide an anatomical and transcriptomic fingerprint of maize cell wall digestibility

    Understanding cell wall biosynthesis and degradation in grasses has become a major aim in plant biology. Although independent previous reports have focused on specific features that dictate cell wall digestibility, cytological, biochemical, and gene regulation parameters have never been integrated within the same study. Herein, we applied a combination of state-of-the-art technologies and different scales of observation on two maize lines that are characterized by highly contrasted forage digestibility. Comparative image analysis of internode sections allow to get an anatomical fingerprint associated with high digestibility: a thin peripheral rind of lignified parenchyma, small numerous vascular bundles, and low proportion of PeriVascular Sclerenchyma (PVS). This cell type patterning led to enhanced digestibility when internode sections were treated with Celluclast, a commercially cell wall degrading enzyme. At a lower scale of observation, Laser Capture Microdissection (LCM) followed by thioacidolysis of PVS revealed a higher proportion of Syringyl (S) unit lignins in the low digestible line while the high digestible line was p-Hydroxyphenyl (H)-rich. Moreover, cytological observation of internodes of the two lines point out that this difference in composition is associated with a delayed lignification of PVS. At the same time, comparative transcriptomics on internodes indicated differential expression of several genes encoding enzymes along the phenylpropanoid pathway and known cell wall-associated Transcription Factors (TFs). Together, these results give an integrative view of different factors which could aim in designing a maize silage ideotype and provide a novel set of potential regulatory genes controlling lignification in maize

    Isolation of mineralizing Nestin+ Nkx6.1+ vascular muscular cells from the adult human spinal cord

    <p>Abstract</p> <p>Background</p> <p>The adult central nervous system (CNS) contains different populations of immature cells that could possibly be used to repair brain and spinal cord lesions. The diversity and the properties of these cells in the human adult CNS remain to be fully explored. We previously isolated Nestin<sup>+ </sup>Sox2<sup>+ </sup>neural multipotential cells from the adult human spinal cord using the neurosphere method (i.e. non adherent conditions and defined medium).</p> <p>Results</p> <p>Here we report the isolation and long term propagation of another population of Nestin<sup>+ </sup>cells from this tissue using adherent culture conditions and serum. QPCR and immunofluorescence indicated that these cells had mesenchymal features as evidenced by the expression of Snai2 and Twist1 and lack of expression of neural markers such as Sox2, Olig2 or GFAP. Indeed, these cells expressed markers typical of smooth muscle vascular cells such as Calponin, Caldesmone and Acta2 (Smooth muscle actin). These cells could not differentiate into chondrocytes, adipocytes, neuronal and glial cells, however they readily mineralized when placed in osteogenic conditions. Further characterization allowed us to identify the Nkx6.1 transcription factor as a marker for these cells. Nkx6.1 was expressed in vivo by CNS vascular muscular cells located in the parenchyma and the meninges.</p> <p>Conclusion</p> <p>Smooth muscle cells expressing Nestin and Nkx6.1 is the main cell population derived from culturing human spinal cord cells in adherent conditions with serum. Mineralization of these cells in vitro could represent a valuable model for studying calcifications of CNS vessels which are observed in pathological situations or as part of the normal aging. In addition, long term propagation of these cells will allow the study of their interaction with other CNS cells and their implication in scar formation during spinal cord injury.</p

    Characterisation of a non-pathogenic and non-protective infectious rabbit lagovirus related to RHDV

    The existence of non-pathogenic RHDV strains was established when a non-lethal virus named rabbit calicivirus (RCV) was characterised in 1996 in Italy. Since then, different RNA sequences related to RHDV have been detected in apparently healthy domestic and wild rabbits, and recently a new lagovirus was identified in Australia. We have characterised from seropositive healthy domestic rabbits a non-lethal lagovirus that differs from RHDV in terms of pathogenicity, tissue tropism and capsid protein sequence. Phylogenetic analyses have revealed that it is close to the Ashington strain and to the RCV, but distinct. We proved experimentally that it is infectious but non-pathogenic and demonstrated that, contrary to the other described non-pathogenic lagoviruses, it induces antibodies that do not protect against RHDV. Our results indicate the existence of a gradient of cross-protection between circulating strains, from non-protective, partially protective to protective strains, and highlight the extent of diversity within the genus Lagovirus

    Non-contrast CT markers of intracerebral hematoma expansion : a reliability study

    Objectives: We evaluated whether clinicians agree in the detection of non-contrast CT markers of intracerebral hemorrhage (ICH) expansion. Methods: From our local dataset, we randomly sampled 60 patients diagnosed with spontaneous ICH. Fifteen physicians and trainees (Stroke Neurology, Interventional and Diagnostic Neuroradiology) were trained to identify six density (Barras density, black hole, blend, hypodensity, fluid level, swirl) and three shape (Barras shape, island, satellite) expansion markers, using standardized definitions. Thirteen raters performed a second assessment. Inter and intra-rater agreement were measured using Gwet’s AC1, with a coefficient > 0.60 indicating substantial to almost perfect agreement. Results: Almost perfect inter-rater agreement was observed for the swirl (0.85, 95% CI: 0.78-0.90) and fluid level (0.84, 95% CI: 0.76-0.90) markers, while the hypodensity (0.67, 95% CI: 0.56-0.76) and blend (0.62, 95% CI: 0.51-0.71) markers showed substantial agreement. Inter-rater agreement was otherwise moderate, and comparable between density and shape markers. Inter-rater agreement was lower for the three markers that require the rater to identify one specific axial slice (Barras density, Barras shape, island: 0.46, 95% CI: 0.40-0.52 versus others: 0.60, 95% CI: 0.56-0.63). Inter-observer agreement did not differ when stratified for raters’ experience, hematoma location, volume or anticoagulation status. Intrarater agreement was substantial to almost perfect for all but the black hole marker. Conclusion: In a large sample of raters with different backgrounds and expertise levels, only four of nine non-contrast CT markers of ICH expansion showed substantial to almost perfect inter-rater agreement

    Glucose modifies the effect of endovascular thrombectomy in patients with acute stroke: a pooled-data meta-analysis

    Background and Purpose: Hyperglycemia is a negative prognostic factor following acute ischemic stroke but is not known whether glucose is associated with the effects of endovascular thrombectomy in patients with large vessel stroke. In a pooled-data meta-analysis, we analyzed whether serum glucose is a treatment modifier of the efficacy of endovascular thrombectomy in acute stroke. Methods: Seven randomized trials compared endovascular thrombectomy with standard care between 2010 and 2017 (HERMES Collaboration). 1764 patients with large vessel stroke were allocated to endovascular thrombectomy (n=871) or standard care (n=893). Measurements included blood glucose on admission and functional outcome [modified Rankin Scale (mRS) range: 0-6; lower scores indicating less disability] at 3 months. The primary analysis evaluated whether glucose modified the effect of EVT over standard care on functional outcome, using ordinal logistic regression to test the interaction between treatment and glucose level. Results: Median (IQR) serum glucose on admission was 120 (104-140) mg/dl [6.6mmol/l (5.7-7.7) mmol/l]. Endovascular thrombectomy (EVT) was better than standard care in the overall pooled-data analysis [common odds ratio (acOR), 2.00 (95% CI 1.69–2.38); however, lower glucose levels were associated with greater effects of EVT over standard care. The interaction was nonlinear such that significant interactions were found in subgroups of patients split at glucose &lt; or &gt; 90mg/dl (5.0mmol/l) [(p=0.019 for interaction, acOR 3.81 (95% CI 1.73–8.41) for patients &lt; 90 mg/dl vs 1.83 (95% CI 1.53–2.19) for patients &gt; 90 mg/dl], and glucose &lt; or &gt; 100mg/dl (5.5mmol/l) [(p=0.004 for interaction, acOR 3.17 (95% CI 2.04–4.93) vs acOR 1.72 (95% CI 1.42–2.08)], but not between subgroups above these levels of glucose. Conclusions: Endovascular thrombectomy improved stroke outcomes compared to standard treatment regardless of glucose levels but the treatment effects were larger at lower glucose levels, with significant interaction effects persisting up to 90 to 100mg/dl (5.0-5.5mmol/l). Whether tight control of glucose improves the efficacy of endovascular thrombectomy following large vessel stroke warrants appropriate testing

    The emergence and spread of Plasmodium falciparum resistance to first-line antimalarials creates an imperative to identify and develop potent preclinical candidates with distinct modes of action. Here, we report the identification of MMV688533, an acylguanidine that was developed following a whole-cell screen with compounds known to hit high-value targets in human cells. MMV688533 displays fast parasite clearance in vitro and is not cross-resistant with known antimalarials. In a P. falciparum NSG mouse model, MMV688533 displays a long-lasting pharmacokinetic profile and excellent safety. Selection studies reveal a low propensity for resistance, with modest loss of potency mediated by point mutations in PfACG1 and PfEHD. These proteins are implicated in intracellular trafficking, lipid utilization, and endocytosis, suggesting interference with these pathways as a potential mode of action. This preclinical candidate may offer the potential for a single low-dose cure for malaria
