    The Old Bailey proceedings and the representation of crime and criminal justice in eighteenth-century London

    The Proceedings of the Old Bailey, published accounts of felony trials held at London’s central criminal court, were a remarkable publishing phenomenon. First published in 1674, they quickly became a regular periodical, with editions published eight times a year following each session of the court. Despite the huge number of trial reports (some 50,000 in the eighteenth century), the Proceedings, also known as the “Sessions Papers”, have formed the basis of several important studies in social history, dating back to Dorothy George’s seminal London Life in the Eighteenth Century (1925). Their recent publication online, however, has not only made them more widely available, but also changed the way historians consult them, leading to greater use of both quantitative analysis, using the statistics function, and qualitative examination of their language, through keyword searching. In the context of recent renewed interest in the history of crime and criminal justice, for which this is the most important source available in this period, the growing use of the Proceedings raises questions about their reliability, and, by extension, the motivations for their original publication. Historians generally consider the Proceedings to present accurate, if often incomplete, accounts of courtroom proceedings. From this source, along with manuscript judicial records, criminal biographies (including the Ordinary’s Accounts), polemical pamphlets such as Henry Fielding’s Enquiry into the Causes of the Late Increase of Robbers (1751), and of course the satirical prints of William Hogarth, they have constructed a picture of eighteenth-century London as a city overwhelmed by periodic crime waves and of a policing and judicial system which was forced into wide-ranging reforms in order to meet this challenge

    A parametric physical model for the intracluster medium and its use in joint SZ/X-ray analyses of galaxy clusters

    We present a parameterized model of the intra-cluster medium that is suitable for jointly analysing pointed observations of the Sunyaev-Zel'dovich (SZ) effect and X-ray emission in galaxy clusters. The model is based on assumptions of hydrostatic equilibrium, the Navarro, Frenk and White (NFW) model for the dark matter, and a softened power law profile for the gas entropy. We test this entropy-based model against high and low signal-to-noise mock observations of a relaxed and recently-merged cluster from N-body/hydrodynamic simulations, using Bayesian hyper-parameters to optimise the relative statistical weighting of the mock SZ and X-ray data. We find that it accurately reproduces both the global values of the cluster temperature, total mass and gas mass fraction (fgas), as well as the radial dependencies of these quantities outside of the core (r > kpc). For reference we also provide a comparison with results from the single isothermal beta model. We confirm previous results that the single isothermal beta model can result in significant biases in derived cluster properties.Comment: Published in MNRAS. 20 pages. 9 figure

    HST NIR Snapshot Survey of 3CR Radio Source Counterparts II: An Atlas and Inventory of the Host Galaxies, Mergers and Companions

    We present the second part of an H-band (1.6 microns) atlas of z<0.3 3CR radio galaxies, using the Hubble Space Telescope Near Infrared Camera and Multi-Object Spectrometer (HST NICMOS2). We present new imaging for 21 recently acquired sources, and host galaxy modeling for the full sample of 101 (including 11 archival) -- an 87% completion rate. Two different modeling techniques are applied, following those adopted by the galaxy morphology and the quasar host galaxy communities. Results are compared, and found to be in excellent agreement, although the former breaks down in the case of strongly nucleated sources. Companion sources are tabulated, and the presence of mergers, tidal features, dust disks and jets are catalogued. The tables form a catalogue for those interested in the structural and morphological dust-free host galaxy properties of the 3CR sample, and for comparison with morphological studies of quiescent galaxies and quasar host galaxies. Host galaxy masses are estimated, and found to typically lie at around 2*10^11 solar masses. In general, the population is found to be consistent with the local population of quiescent elliptical galaxies, but with a longer tail to low Sersic index, mainly consisting of low-redshift (z<0.1) and low-radio-power (FR I) sources. A few unusually disky FR II host galaxies are picked out for further discussion. Nearby external sources are identified in the majority of our images, many of which we argue are likely to be companion galaxies or merger remnants.     High-throughput sequencing of the paired human immunoglobulin heavy and light chain repertoire.

    Each B-cell receptor consists of a pair of heavy and light chains. High-throughput sequencing can identify large numbers of heavy- and light-chain variable regions (VH and VL) in a given B-cell repertoire, but information about endogenous pairing of heavy and light chains is lost after bulk lysis of B-cell populations. Here we describe a way to retain this pairing information. In our approach, single B cells (>5 × 104 capacity per experiment) are deposited in a high-density microwell plate (125 pl/well) and lysed in situ. mRNA is then captured on magnetic beads, reverse transcribed and amplified by emulsion VH:VL linkage PCR. The linked transcripts are analyzed by Illumina high-throughput sequencing. We validated the fidelity of VH:VL pairs identified by this approach and used the method to sequence the repertoire of three human cell subsets-peripheral blood IgG+ B cells, peripheral plasmablasts isolated after tetanus toxoid immunization and memory B cells isolated after seasonal influenza vaccinatio

    FIRST-2MASS Red Quasars: Transitional Objects Emerging from the Dust

    We present a sample of 120 dust-reddened quasars identified by matching radio sources detected at 1.4 GHz in the FIRST survey with the near-infrared 2MASS catalog and color-selecting red sources. Optical and/or near-infrared spectroscopy provide broad wavelength sampling of their spectral energy distributions that we use to determine their reddening, characterized by E(B-V). We demonstrate that the reddening in these quasars is best-described by SMC-like dust. This sample spans a wide range in redshift and reddening (0.1 < z < 3, 0.1 < E(B-V) < 1.5), which we use to investigate the possible correlation of luminosity with reddening. At every redshift, dust-reddened quasars are intrinsically the most luminous quasars. We interpret this result in the context of merger-driven quasar/galaxy co-evolution where these reddened quasars are revealing an emergent phase during which the heavily obscured quasar is shedding its cocoon of dust prior to becoming a "normal" blue quasar. When correcting for extinction, we find that, depending on how the parent population is defined, these red quasars make up < 15-20% of the luminous quasar population. We estimate, based on the fraction of objects in this phase, that its duration is 15-20% as long as the unobscured, blue quasar phase.Comment: 21 pages, 17 figures plus a spectral atlas.     Local Supermassive Black Holes, Relics of Active Galactic Nuclei and the X-ray Background

    We quantify the importance of mass accretion during AGN phases in the growth of supermassive black holes (BH) by comparing the mass function of black holes in the local universe with that expected from AGN relics, which are black holes grown entirely with mass accretion during AGN phases. The local BH mass function (BHMF) is estimated by applying the well-known correlations between BH mass, bulge luminosity and stellar velocity dispersion to galaxy luminosity and velocity functions. The density of BH's in the local universe is 4.6 (-1.4; +1.9) (h/0.7)^2 10^5 Msun Mpc^-3. The relic BHMF is derived from the continuity equation with the only assumption that AGN activity is due to accretion onto massive BH's and that merging is not important. We find that the relic BHMF at z=0 is generated mainly at z<3. Moreover, the BH growth is anti-hierarchical in the sense that smaller BH's (MBH< 10^7 Msun) grow at lower redshifts (z<1) with respect to more massive one's (z~1-3). Unlike previous work, we find that the BHMF of AGN relics is perfectly consistent with the local BHMF indicating the local BH's were mainly grown during AGN activity. This agreement is obtained while satisfying, at the same time, the constraints imposed from the X-ray background. The comparison with the local BHMF also suggests that the merging process is not important in shaping the relic BHMF, at least at low redshifts (z<3). Our analysis thus suggests the following scenario: local black holes grew during AGN phases in which accreting matter was converted into radiation with efficiencies epsilon = 0.04-0.16 and emitted at a fraction lambda = 0.1-1.7 of the Eddington luminosity.     An X-ray Spectral Survey of Radio-Loud AGN With ASCA

    We present a uniform and systematic analysis of the 0.6-10 keV X-ray spectra of radio-loud active galactic nuclei (AGN) observed by ASCA. The sample 10 BLRGs, 5 QSRs, 9 NLRGs, and 10 RGs. At soft X-rays, about half of the NLRGs and all of the RGs exhibit a thermal plasma component, with a bimodal distribution of temperatures and luminosities, suggesting an origin either in a surrounding cluster or loose group or in a hot corona. At energies above 2 keV, a hard power-law component is detected in 90% of cases. The power-law photon indices and luminosities in BLRGs, QSRs, and NLRGs are similar, consistent with orientation-based unification schemes. Significant excess cold absorption is detected in most NLRGs, but also in some BLRGS and QSRs, which was somewhat unexpected. In contrast to Seyfert galaxies, only one object showss the signature of a warm absorber. The nuclear X-ray luminosity is correlated with the luminosity of the [O III] emission line, the FIR emission at 12 microns, and the lobe radio power at 5 GHz. The Fe K line is detected in 50% of BLRGs, one QSR, and a handful of NLRGs. This sample also includes 6 Weak Line Radio Galaxies (WLRGs). Their spectra WLRGs can be generally decomposed into a soft thermal component with hard absrorbed power-law component, which is significantly flatter than any other radio-loud AGNs. Their intrinsic luminosities are two orders of magnitude lower than in other sources of the sample. An interesting possibility is that WLRGs represent an extreme population of radio galaxies in which the central black hole is accreting at a rate well below the Eddington rate.Comment: To appear in the Astrophysical Journal. 72 pages, including many tables and figures.     Characterization of an unusual bipolar helicase encoded by bacteriophage T5

    Bacteriophage T5 has a 120 kb double-stranded linear DNA genome encoding most of the genes required for its own replication. This lytic bacteriophage has a burst size of ∼500 new phage particles per infected cell, demonstrating that it is able to turn each infected bacterium into a highly efficient DNA manufacturing machine. To begin to understand DNA replication in this prodigious bacteriophage, we have characterized a putative helicase encoded by gene D2. We show that bacteriophage T5 D2 protein is the first viral helicase to be described with bipolar DNA unwinding activities that require the same core catalytic residues for unwinding in either direction. However, unwinding of partially single- and double-stranded DNA test substrates in the 3′–5′ direction is more robust and can be distinguished from the 5′–3′ activity by a number of features including helicase complex stability, salt sensitivity and the length of single-stranded DNA overhang required for initiation of helicase action. The presence of D2 in an early gene cluster, the identification of a putative helix-turn-helix DNA-binding motif outside the helicase core and homology with known eukaryotic and prokaryotic replication initiators suggest an involvement for this unusual helicase in DNA replication initiation

    Substrate Profiling of Tobacco Etch Virus Protease Using a Novel Fluorescence-Assisted Whole-Cell Assay

    Site-specific proteolysis of proteins plays an important role in many cellular functions and is often key to the virulence of infectious organisms. Efficient methods for characterization of proteases and their substrates will therefore help us understand these fundamental processes and thereby hopefully point towards new therapeutic strategies. Here, a novel whole-cell in vivo method was used to investigate the substrate preference of the sequence specific tobacco etch virus protease (TEVp). The assay, which utilizes protease-mediated intracellular rescue of genetically encoded short-lived fluorescent substrate reporters to enhance the fluorescence of the entire cell, allowed subtle differences in the processing efficiency of closely related substrate peptides to be detected. Quantitative screening of large combinatorial substrate libraries, through flow cytometry analysis and cell sorting, enabled identification of optimal substrates for TEVp. The peptide, ENLYFQG, identical to the protease's natural substrate peptide, emerged as a strong consensus cleavage sequence, and position P3 (tyrosine, Y) and P1 (glutamine, Q) within the substrate peptide were confirmed as being the most important specificity determinants. In position P1′, glycine (G), serine (S), cysteine (C), alanine (A) and arginine (R) were among the most prevalent residues observed, all known to generate functional TEVp substrates and largely in line with other published studies stating that there is a strong preference for short aliphatic residues in this position. Interestingly, given the complex hydrogen-bonding network that the P6 glutamate (E) is engaged in within the substrate-enzyme complex, an unexpectedly relaxed residue preference was revealed for this position, which has not been reported earlier. Thus, in the light of our results, we believe that our assay, besides enabling protease substrate profiling, also may serve as a highly competitive platform for directed evolution of proteases and their substrates
