Había una gran diferencia entre parroquias e incluso dentro de ellas. Estos objetivos hay que enmarcarlos dentro de la política reformista e ilustrada que se realizó durante el reinado de Carlos III

    Adipose-Derived Stem Cells in Crohn's Rectovaginal Fistula

    Therapeutic options for recto-vaginal fistula in the setting of Crohn's disease are limited and many data are available in the literature. The manuscript describes the history of a patient who has been the pioneer of our Clinical Trials in treating this disease in fistulizing Crohn's disease environment. We believe it is the first time that a patient with this disease has been treated by adipose-derived stem cells in allogeneic form. The conclusion of our study with Mary is that the use of mesenchymal stem cells derived from adipose tissue is secure, either in autologous or allogeneic form. Furthermore, we have proved that if we use multi-dose and multiple applications on a patient, it does not produce any adverse effect, which confirms us the safety of using these cells in patients at least in the fistulizing Crohn's disease environment

    Low-cost irradiance sensors for irradiation assessments inside tree canopies.

    The solar irradiation that a crop receives is directly related to the physical and biological processes that affect the crop. However, the assessment of solar irradiation poses certain problems when it must be measured through fruit inside the canopy of a tree. In such cases, it is necessary to check many test points, which usually requires an expensive data acquisition system. The use of conventional irradiance sensors increases the cost of the experiment, making them unsuitable. Nevertheless, it is still possible to perform a precise irradiance test with a reduced price by using low-cost sensors based on the photovoltaic effect. The aim of this work is to develop a low-cost sensor that permits the measurement of the irradiance inside the tree canopy. Two different technologies of solar cells were analyzed for their use in the measurement of solar irradiation levels inside tree canopies. Two data acquisition system setups were also tested and compared. Experiments were performed in Ademuz (Valencia, Spain) in September 2011 and September 2012 to check the validity of low-cost sensors based on solar cells and their associated data acquisition systems. The observed difference between solar irradiation at high and low positions was of 18.5% ± 2.58% at a 95% confidence interval. Large differences were observed between the operations of the two tested sensors. In the case of a-Si cells based mini-modules, an effect of partial shadowing was detected due to the larger size of the devices, the use of individual c-Si cells is recommended over a-Si cells based mini-modules

    P73 plays a role in erythroid differentiaion through GATA1 induction

    The TP73 gene gives rise to transactivation domain-p73 isoforms (TAp73) as well as ΔNp73 variants with a truncated N terminus. Although TAp73α and -Β proteins are capable of inducing cell cycle arrest, apoptosis, and differentiation, ΔNp73 acts in many cell types as a dominant-negative repressor of p53 and TAp73. It has been proposed that p73 is involved in myeloid differentiation, and its altered expression is involved in leukemic degeneration. However, there is little evidence as to which p73 variants (TA or ΔN) are expressed during differentiation and whether specific p73 isoforms have the capacity to induce, or hinder, this differentiation in leukemia cells. In this study we identify GATA1 as a direct transcriptional target of TAp73α. Furthermore, TAp73α induces GATA1 activity, and it is required for erythroid differentiation. Additionally, we describe a functional cooperation between TAp73 and ΔNp73 in the context of erythroid differentiation in human myeloid cells, K562 and UT-7. Moreover, the impaired expression of GATA1 and other erythroid genes in the liver of p73KO embryos, together with the moderated anemia observed in p73KO young mice, suggests a physiological role for TP73 in erythropoiesis.Peer reviewe

    Cryo-EM and single-particle analysis with Scipion

    Cryo-electron microscopy has become one of the most important tools in biological research to reveal the structural information of macromolecules at near-atomic resolution. In single-particle analysis, the vitrified sample is imaged by an electron beam and the detectors at the end of the microscope column produce movies of that sample. These movies contain thousands of images of identical particles in random orientations. The data need to go through an image processing workflow with multiple steps to obtain the final 3D reconstructed volume. The goal of the image processing workflow is to identify the acquisition parameters to be able to reconstruct the specimen under study. Scipion provides all the tools to create this workflow using several image processing packages in an integrative framework, also allowing the traceability of the results. In this article the whole image processing workflow in Scipion is presented and discussed with data coming from a real test case, giving all the details necessary to go from the movies obtained by the microscope to a high resolution final 3D reconstruction. Also, the power of using consensus tools that allow combining methods, and confirming results along every step of the workflow, improving the accuracy of the obtained results, is discussed

    Herschel FIR counterparts of selected Ly-alpha emitters at z~2.2. Fast evolution since z~3 or missed obscured AGNs?

    Ly-alpha emitters (LAEs) are seen everywhere in the redshift domain from local to z~7. Far-infrared (FIR) counterparts of LAEs at different epochs could provide direct clues on dust content, extinction, and spectral energy distribution (SED) for these galaxies. We search for FIR counterparts of LAEs that are optically detected in the GOODS-North field at redshift z~2.2 using data from the Herschel Space Telescope with the Photodetector Array Camera and Spectrometer (PACS). The LAE candidates were isolated via color-magnitude diagram using the medium-band photometry from the ALHAMBRA Survey, ancillary data on GOODS-North, and stellar population models. According to the fitting of these spectral synthesis models and FIR/optical diagnostics, most of them seem to be obscured galaxies whose spectra are AGN-dominated. From the analysis of the optical data, we have observed a fraction of AGN or composite over source total number of ~0.75 in the LAE population at z~2.2, which is marginally consistent with the fraction previously observed at z=2.25 and even at low redshift (0.2<z<0.45), but significantly different from the one observed at redshift ~3, which could be compatible either with a scenario of rapid change in the AGN fraction between the epochs involved or with a non detection of obscured AGN in other z=2-3 LAE samples due to lack of deep FIR observations. We found three robust FIR (PACS) counterparts at z~2.2 in GOODS-North. This demonstrates the possibility of finding dust emission in LAEs even at higher redshifts.Comment: 11 pages (including Appendices), 6 figures.     Castillejo del Bonete es un complejo tumular situado en el borde meridional de la Meseta IbĂ©rica, ocupado en fechas calcolĂ­ticas y de la Edad del Bronce, vinculado a la Cultura de las Motillas. Materiales arqueolĂłgicos muy diversos han sido recuperados asociados a las arquitecturas del lugar (tĂșmulos, corredores, potentes muros, etc.). Se presenta un avance de la investigaciĂłn paleoecolĂłgica sobre las colecciones de carbĂłn, polen y microvertebrados. AdemĂĄs se presentan cuentas de piedra y madera, colgantes de concha, material lĂ­tico, la colecciĂłn cerĂĄmica, nuevas metalografĂ­as e industria metĂĄlica y botones de marfil.     Algorithmic robustness to preferred orientations in single particle analysis by CryoEM

    The presence of preferred orientations in single particle analysis (SPA) by cryo-Electron Microscopy (cryoEM) is currently one of the hurdles preventing many structural analyses from yielding high-resolution structures. Although the existence of preferred orientations is mostly related to the grid preparation, in this technical note, we show that some image processing algorithms used for angular assignment and three-dimensional (3D) reconstruction are more robust than others to these detrimental conditions. We exemplify this argument with three different data sets in which the presence of preferred orientations hindered achieving a 3D reconstruction without artifacts or, even worse, a 3D reconstruction could never be achievedWe acknowledge support from “la Caixa” Foundation (Fellowship LCF/BQ/DI18/11660021.     New Mutations in Chronic Lymphocytic Leukemia Identified by Target Enrichment and Deep Sequencing

    Chronic lymphocytic leukemia (CLL) is a heterogeneous disease without a well-defined genetic alteration responsible for the onset of the disease. Several lines of evidence coincide in identifying stimulatory and growth signals delivered by B-cell receptor (BCR), and co-receptors together with NFkB pathway, as being the driving force in B-cell survival in CLL. However, the molecular mechanism responsible for this activation has not been identified. Based on the hypothesis that BCR activation may depend on somatic mutations of the BCR and related pathways we have performed a complete mutational screening of 301 selected genes associated with BCR signaling and related pathways using massive parallel sequencing technology in 10 CLL cases. Four mutated genes in coding regions (KRAS, SMARCA2, NFKBIE and PRKD3) have been confirmed by capillary sequencing. In conclusion, this study identifies new genes mutated in CLL, all of them in cases with progressive disease, and demonstrates that next-generation sequencing technologies applied to selected genes or pathways of interest are powerful tools for identifying novel mutational changes

    <p>Abstract</p> <p>Background</p> <p>Although there have been many studies on the p73 gene, some of its functions still remain unclear. There is little research on the relationship between p73 gene transcription and its protein expression and the response to certain drugs such as oxaliplatin and cetuximab, which are drugs currently used in colorectal cancer.</p> <p>The purpose of this study was to evaluate the impact of TAp73 expression on oxaliplatin and cetuximab-based chemotherapy in colorectal cancer cell lines with different K-Ras and B-Raf mutational status.</p> <p>Methods</p> <p>TAp73 was analyzed in three colorectal tumor cell lines HT-29, SW-480 and Caco-2. mRNA TAp73 was determined using Real time PCR; TAp73 protein by immunoblotting and cell viability was analyzed by the MTT method.</p> <p>Results</p> <p>We found that mRNA and TAp73 protein were decreased in cells treated with oxaliplatin (in monotherapy or combined with cetuximab) when B-Raf is mutated. This was statistically significant and was also associated with higher cell viability after the treatment.</p> <p>Conclusions</p> <p>Here, for the first time we report, that there is a signaling loop between B-Raf activation and p73 function.</p> <p>Low expression of TAp73 in colorectal cancer cell lines with mutated B-Raf may be involved in the lack of response to oxaliplatin in monotherapy or combined with cetuximab.</p
