Multiwavelength optical observations of chromospherically active binary systems V. FF UMa (2RE J0933+624): a system with orbital period variation
This is the fifth paper in a series aimed at studying the chromospheres of
active binary systems using several optical spectroscopic indicators to obtain
or improve orbital solution and fundamental stellar parameters. We present here
the study of FF UMa (2RE J0933+624), a recently discovered, X-ray/EUV selected,
active binary with strong H_alpha emission. The objectives of this work are, to
find orbital solutions and define stellar parameters from precise radial
velocities and carry out an extensive study of the optical indicators of
chromospheric activity. We obtained high resolution echelle spectroscopic
observations during five observing runs from 1998 to 2004. We found radial
velocities by cross correlation with radial velocity standard stars to achieve
the best orbital solution. We also measured rotational velocity by
cross-correlation techniques and have studied the kinematic by galactic space-
velocity components (U, V, W) and Eggen criteria. Finally, we have determined
the chromospheric contribution in optical spectroscopic indicators, from Ca II
H & K to Ca II IRT lines, using the spectral subtraction technique. We have
found that this system presents an orbital period variation, higher than
previously detected in other RS CVn systems. We determined an improved orbital
solution, finding a circular orbit with a period of 3.274 days. We derived the
stellar parameters, confirming the subgiant nature of the primary component and
obtained rotational velocities (vsini), of 33.57 km/s and 32.38 km/s for the
primary and secondary components respectively. From our kinematic study, we can
deduce its membership to the Castor moving group. Finally, the activity study
has given us a better understanding of the possible mechanisms that produce the

Effects of salt stress on plant growth, abscisic acid and salicylic acid in own-rooted cultivars of Vitis vinifera L.
Aim of study: In most areas of vineyards worldwide, cultivars are frequently grafted on specific rootstocks to avoid Daktulosphaira vitifoliae pest attack. Nevertheless, the absence of this pest in Canary Islands allowed the chance to conserve and cultivate traditional or new own-rooted genotypes without the requirement of the rootstocks. To investigate the responses of own-rooted genotypes of Vitis vinifera L. to salt stress conditions, ‘Castellana Negra’ (‘CN’) and ‘Negramoll’ (‘Ne’) were used with the aim to characterize their morphological and physiological responses.Area of study: Canary Islands, Spain.Material and methods: The effects of NaCl stress on growth, abscisic acid (ABA), salicylic acid (SA) and proline were assessed in ‘CN’ and ‘Ne’ under greenhouse conditions.Main results: In ‘CN’, the decrease of leaf number in stressed plants was lower and started eleven days later than in ‘Ne’. Salt stress also reduced stomatal conductance (gs), although such decrease took place earlier in ‘CN’ than in ‘Ne’. ABA and SA concentrations in ‘CN’ leaves were 2-fold higher than those of ‘Ne’. Salt stress increased leaf ABA and SA content in both genotypes, compared to control. In conclusion, ABA and SA appear to be involved in grapevines responses to salinity and suggest that exogenous SA could be useful to mitigate the stress impacts.Research highlights: ‘CN’ exhibited a better response than ‘Ne’ through the delay of salt injury establishment, and the dissimilar responses between ‘CN’ and ‘Ne’ seem to be associated to the higher accumulation of ABA and SA under salt stress

Identification of Interleukin-27 (IL-27)/IL-27 Receptor Subunit Alpha as a Critical Immune Axis for In Vivo HIV Control
Intact and broad immune cell effector functions and specific individual cytokines have been linked to HIV disease outcome, but their relative contribution to HIV control remains unclear. We asked whether the proteome of secreted cytokines and signaling factors in peripheral blood can be used to discover specific pathways critical for host viral control. A custom glass-based microarray, able to measure >600 plasma proteins involved in cell-to-cell communication, was used to measure plasma protein profiles in 96 HIV-infected, treatment-naive individuals with high (>50,000) or low (600 soluble proteins, our data highlight the importance of Th17 cells and Wnt/β-catenin signaling in HIV control and especially identify the IL-27/IL-27 receptor subunit alpha (IL-27RA) axis as a predictor of plasma viral load and proviral copy number in the peripheral blood. These data may provide important guidance to therapeutic approaches in the HIV cure agenda
Identification of interleukin-27 (IL-27)/IL-27 receptor subunit alpha as a critical immune axis for in vivo hiv control
Intact and broad immune cell effector functions and specific individual cytokines have been linked to HIV disease outcome, but their relative contribution to HIV control remains unclear. We asked whether the proteome of secreted cytokines and signaling factors in peripheral blood can be used to discover specific pathways critical for host viral control. A custom glass-based microarray, able to measure >600 plasma proteins involved in cell-to-cell communication, was used to measure plasma protein profiles in 96 HIV-infected, treatment-naive individuals with high (> 50,000) or low (<10,000 HIV RNA copies/ml) viral loads. Univariate and regression model analysis demonstrate that plasma levels of soluble interleukin-27 (IL-27) are significantly elevated in individuals with high plasma viremia (P < 0.0001) and are positively correlated with proviral HIV-DNA copy numbers in peripheral blood mononuclear cells (PBMC) (Rho = 0.4011; P = 0.0027). Moreover, soluble IL-27 plasma levels are negatively associated with the breadth and magnitude of the total virus-specific T-cell responses and directly with plasma levels of molecules involved in Wnt/beta-catenin signaling. In addition to IL-27, gene expression levels of the specific IL-27 receptor (IL27RA) in PBMC correlated directly with both plasma viral load (Rho = 0.3531; P = 0.0218) and the proviral copy number in the peripheral blood as an indirect measure of partial viral reservoir (Rho = 0.4580; P = 0.0030). These results were validated in unrelated cohorts of early infected subjects as well as subjects before and after initiation of antiretroviral treatment, and they identify IL-27 and its specific receptor as a critical immune axis for the antiviral immune response and as robust correlates of viral load and proviral reservoir size in PBMC.
IMPORTANCE The detailed knowledge of immune mechanisms that contribute to HIV control is a prerequisite for the design of effective treatment strategies to achieve HIV cure. Methylation regulation of Antiviral host factors, Interferon Stimulated Genes (ISGs) and T-cell responses associated with natural HIV control
GWAS, immune analyses and biomarker screenings have identified host factors associated with in vivo HIV-1 control. However, there is a gap in the knowledge about the mechanisms that regulate the expression of such host factors. Here, we aimed to assess DNA methylation impact on host genome in natural HIV-1 control. To this end, whole DNA methylome in 70 untreated HIV-1 infected individuals with either high (>50,000 HIV-1-RNA copies/ml, n = 29) or low (<10,000 HIV-1-RNA copies/ml, n = 41) plasma viral load (pVL) levels were compared and identified 2,649 differentially methylated positions (DMPs). Of these, a classification random forest model selected 55 DMPs that correlated with virologic (pVL and proviral levels) and HIV-1 specific adaptive immunity parameters (IFNg-T cell responses and neutralizing antibodies capacity). Then, cluster and functional analyses identified two DMP clusters: cluster 1 contained hypo-methylated genes involved in antiviral and interferon response (e.g. PARP9, MX1, and USP18) in individuals with high viral loads while in cluster 2, genes related to T follicular helper cell (Tfh) commitment (e.g. CXCR5 and TCF7) were hyper-methylated in the same group of individuals with uncontrolled infection. For selected genes, mRNA levels negatively correlated with DNA methylation, confirming an epigenetic regulation of gene expression. Further, these gene expression signatures were also confirmed in early and chronic stages of infection, including untreated, cART treated and elite controllers HIV-1 infected individuals (n = 37). These data provide the first evidence that host genes critically involved in immune control of the virus are under methylation regulation in HIV-1 infection. These insights may offer new opportunities to identify novel mechanisms of in vivo virus control and may prove crucial for the development of future therapeutic interventions aimed at HIV-1 cure
Ultracool dwarf benchmarks with \emph{Gaia} primaries

Intracoronary administration of allogeneic adipose tissue-derived mesenchymal stem cells improves myocardial perfusion but not left ventricle function, in a translational model of acute myocardial infarction
Background-¿Autologous adipose tissue-derived mesenchymal stem cells (ATMSCs) therapy is a promising strategy to improve post-myocardial infarction outcomes. In a porcine model of acute myocardial infarction, we studied the long-term effects and the mechanisms involved in allogeneic ATMSCs administration on myocardial performance. Methods and Results-¿Thirty-eight pigs underwent 50 minutes of coronary occlusion; the study was completed in 33 pigs. After reperfusion, allogeneic ATMSCs or culture medium (vehicle) were intracoronarily administered. Follow-ups were performed at short (2 days after acute myocardial infarction vehicle-treated, n=10; ATMSCs-treated, n=9) or long term (60 days after acute myocardial infarction vehicle-treated, n=7; ATMSCs-treated, n=7). At short term, infarcted myocardium analysis showed reduced apoptosis in the ATMSCs-treated animals (48.6 6% versus 55.9 5.7% in vehicle; P=0.017); enhancement of the reparative process with up-regulated vascular endothelial growth factor, granulocyte macrophage colony-stimulating factor, and stromal-derived factor-1a gene expression; and increased M2 macrophages (67.2 10% versus 54.7 10.2% in vehicle; P=0.016). In long-term groups, increase in myocardial perfusion at the anterior infarct border was observed both on day-7 and day-60 cardiac magnetic resonance studies in ATMSCs-treated animals, compared to vehicle (87.9 28.7 versus 57.4 17.7 mL/min per gram at 7 days; P=0.034 and 99 22.6 versus 43.3 14.7 22.6 mL/min per gram at 60 days; P=0.0001, respectively). At day 60, higher vascular density was detected at the border zone in the ATMSCs-treated animals (118 18 versus 92.4 24.3 vessels/mm2 in vehicle; P=0.045). Cardiac magnetic resonance-measured left ventricular ejection fraction of left ventricular volumes was not different between groups at any time point. Conclusions-¿In this porcine acute myocardial infarction model, allogeneic ATMSCs-based therapy was associated with increased cardioprotective and reparative mechanisms and with better cardiac magnetic resonance-measured perfusion. No effect on left ventricular volumes or ejection fraction was observed

unknown with very few finds. Underwater research has focused on naval archaeology and, until very recently, no attempt had been made to look for prehistoric underwater sites. In the past decade,new research projects have been launched to explore selected areas on the Cantabrian shelf and offshore of Gibraltar.
