    On the stratigraphic integrity of leaf-wax biomarkers in loess paleosols

    Paleoenvironmental and paleoclimate reconstructions based on molecular proxies, such as those derived from leaf-wax biomarkers, in loess-paleosol sequences represent a promising line of investigation in Quaternary research. The main premise of such reconstructions is the synsedimentary deposition of biomarkers and dust, which has become a debated subject in recent years. This study uses two independent approaches to test the stratigraphic integrity of leaf-wax biomarkers: (i) long-chain n-alkanes and fatty acids are quantified in two sediment-depth profiles in glacial till on the Swiss Plateau, consisting of a Holocene topsoil and the underlying B and C horizons. Since glacial sediments are initially very poor in organic matter, significant amounts of leaf-wax biomarkers in the B and C horizons of those profiles would reflect postsedimentary root-derived or microbial contributions. (ii) Compound-specific radiocarbon measurements are conducted on n-alkanes and n-alkanoic (fatty) acids from several depth intervals in the loess section "Crvenka", Serbia, and the results are compared to independent estimates of sediment age. <br><br> We find extremely low concentrations of plant-wax n-alkanes and fatty acids in the B and C horizons below the topsoils in the sediment profiles. Moreover, compound-specific radiocarbon analysis yields plant-wax <sup>14</sup>C ages that agree well with published luminescence ages and stratigraphy of the Serbian loess deposit. Both approaches confirm that postsedimentary, root-derived or microbial contributions are negligible in the two investigated systems. The good agreement between the ages of odd and even homologues also indicates that reworking and incorporation of fossil leaf waxes is not particularly relevant either

    Diverse soil carbon dynamics expressed at the molecular level

    The stability and potential vulnerability of soil organic matter (SOM) to global change remains incompletely understood due to the complex processes involved in its formation and turnover. Here we combine compound-specific radiocarbon analysis with fraction-specific and bulk-level radiocarbon measurements in order to further elucidate controls on SOM dynamics in a temperate and sub-alpine forested ecosystem. Radiocarbon contents of individual organic compounds isolated from the same soil interval generally exhibit greater variation than those among corresponding operationally-defined fractions. Notably, markedly older ages of long-chain plant leaf wax lipids (n-alkanoic acids) imply that they reflect a highly stable carbon pool. Furthermore, marked 14C variations among shorter- and longer-chain n-alkanoic acid homologues suggest that they track different SOM pools. Extremes in SOM dynamics thus manifest themselves within a single compound class. This exploratory study highlights the potential of compound-specific radiocarbon analysis for understanding SOM dynamics in ecosystems potentially vulnerable to global change

    High-sensitivity measurement of diverse vascular plant-derived biomarkers in high-altitude ice cores

The definitive version was published in Geophysical Research Letters 36 (2009): L13501, doi:10.1029/2009GL037643.Semi-volatile organic compounds derived from burned and fresh vascular plant sources and preserved in high-altitude ice fields were detected and identified through use of recently developed analytical tools. Specifically, stir bar sorptive extraction and thermal desorption coupled with gas chromatography/time-of-flight mass spectrometry allowed measurement of multiple biomarkers in small sample volumes (≤30 ml). Among other compounds of interest, several diterpenoids, which suggest inputs from conifers and conifer burning, were identified in post-industrial era and older Holocene ice from the Sajama site in the Bolivian Andes, but not in a glacial period sample, consistent with aridity changes. Differences in biomarker assemblages between sites support the use of these compounds as regionally constrained recorders of vegetation and climate change. This study represents the first application of these analytical techniques to ice core research and the first indication that records of vegetation fires may be reconstructed from diterpenoids in ice.This project was supported in part by NSF-OCE (0402533), and NSF-EAR (0094475)

    Stable Isotope and Modelling Evidence for CO2 as a Driver of Glacial-Interglacial Vegetation Shifts in Southern Africa

    Atmospheric CO2 concentration is hypothesized to influence vegetation distribution via tree-grass competition, with higher CO2 concentrations favouring trees. The stable carbon isotope (delta C-13) signature of vegetation is influenced by the relative importance of C-4 plants (including most tropical grasses) and C-3 plants (including nearly all trees), and the degree of stomatal closure - a response to aridity in C-3 plants. Compound-specific delta C-13 analyses of leaf-wax biomarkers in sediment cores of an offshore South Atlantic transect are used here as a record of vegetation changes in subequatorial Africa. These data suggest a large increase in C3 relative to C4 plant dominance after the Last Glacial Maximum. Using a process-based biogeography model that explicitly 10 simulates 13 C discrimination, we show that climate change alone cannot explain the observed shift in δ13 C values. The physiological effect of increasing CO2 concentration is decisive, altering the C3/ C4 balance and bringing the simulated and observed δ 13C values into line. It is concluded that CO2 concentration itself was a key agent of tropical vegetation 15 change during the last glacial-interglacial transition. Two additional inferences follow. First, long-term variations in terrestrial δ13 C values are not simply a proxy for regional rainfall as has sometimes been assumed. Such interpretations need to be re-examined. Second, rising CO2 concentration today is likely to be influencing tree-grass competition in a similar way, and thus contributing to the “woody thickening” observed in savannas worldwide. This second inference points to the importance of experiments to determine how vegetation composition in savannas is likely to be influenced by the continuing rise in CO2 concentratio

    Compound-Specific Radiocarbon Analysis by Elemental Analyzer–Accelerator Mass Spectrometry: Precision and Limitations

    [EN]We examine instrumental and methodological capabilities for microscale (10−50μg of C) radiocarbon analysisof individual compounds in the context of paleoclimate and paleoceanography applications, for which relatively high-precisionmeasurements are required. An extensive suite of data for14C-free and modern reference materials processed using differentmethods and acquired using an elemental-analyzer−accelerator-mass-spectrometry (EA-AMS) instrumental setup at ETHZurich was compiled to assess the reproducibility of specific isolation procedures. In order to determine the precision, accuracy,and reproducibility of measurements on processed compounds, we explore the results of both reference materials and threeclasses of compounds (fatty acids, alkenones, and amino acids) extracted from sediment samples. We utilize a MATLAB codedeveloped to systematically evaluate constant-contamination-model parameters, which in turn can be applied to measurementsof unknown process samples. This approach is computationally reliable and can be used for any blank assessment of small-sizeradiocarbon samples. Our results show that a conservative lower estimate of the sample sizes required to produce relativelyhigh-precision14C data (i.e., with acceptable errors of 0.5, a precision of 2% can be achieved for alkenone and fatty acid samples containing≥15 and 10μg of C, respectivel

    Antibiotic susceptibility of Escherichia coli strains in neonatal alpacas with and without diarrhoea

    El estudio tuvo como objetivo determinar la susceptibilidad antibiótica de aislados de Escherichia coli en crías de alpaca con y sin diarrea de las comunidades campesinas de la localidad de Lachocc, departamento de Huancavelica, Perú. Se analizaron hisopados rectales de 95 animales con diarrea y de 60 aparentemente sanos de 1 a 2 meses de edad. El 48.4% (46/95) y el 60% (36/60) de las muestras de animales con y sin diarrea fueron positivas al cultivo de E. coli. Estos aislados fueron sensibles a la mayoría de los antibacterianos, especialmente trimetoprim (98%), gentamicina (95%) y fosfomicina (88%). La excepción fue la nitrofurantoína, cuyas cepas mostraron una baja susceptibilidad (8.3%).The present study aimed to determine the antimicrobial susceptibility of Escherichia coli isolates in neonatal alpacas with and without diarrhea in peasant communities in Lachocc, Huancavelica, Peru. Rectal swabs from alpacas with diarrhoea (n=95) and from apparently normal alpacas (n=60) of 1 to 2 months of age were analysed. The results showed that 48.4% (46/95) and 60% (36/60) of samples from animals with and without diarrhea were positive to E. coli. The definitive version was published in Paleoceanography 19 (2004): PA2012, doi:10.1029/2003PA000927.The radiocarbon contents of various biomarkers extracted from the varve-counted sediments of Saanich Inlet, Canada, were determined to assess their applicability for dating purposes. Calibrated ages obtained from the marine planktonic archaeal biomarker crenarchaeol compared favorably with varve-count ages. The same conclusion could be drawn for a more general archaeal biomarker (GDGT-0), although this biomarker proved to be less reliable due to its less-specific origin. The results also lend support to earlier indications that marine crenarchaeota use dissolved inorganic carbon (DIC) as their carbon source. The average reservoir age offset ΔR of 430 years, determined using the crenarchaeol radiocarbon ages, varied by ±110 years. This may be caused by natural variations in ocean-atmosphere mixing or upwelling at the NE Pacific coast but variability may also be due to an inconsistency in the marine calibration curve when used at sites with high reservoir ages.This work was supported by the Netherlands Organization for Scientific Research (NWO) and NSF grants OCE-9907129 and OCE-0137005 (Eglinton)

    A novel approach for construction of radiocarbon-based chronologies for speleothems

    Robust chronologies are crucial for the correct interpretation of climate proxy records and for detailed reconstructions of palaeoclimate. Stalagmites have garnered strong interest as recorders of past climate in part due to their amenability to U-series dating. However, many stalagmites are not dateable using this technique due to low 238U and/or high detrital Th concentrations (e.g., many tropical cave systems (Adkins et al., 2013)), and occasionally these issues affect stalagmites across wide geographical regions (e.g., large parts of Australia (Green et al. 2013)) complicating the use of stalagmites in these areas. Radiocarbon (14C) offers an alternative method of dating stalagmites, but issues associated with the ‘dead carbon fraction’ (DCF) have historically hindered this approach. Here, a novel 14C-based method for dating stalagmites is presented and discussed. The technique calculates a best-fit growth rate between a time-series of stalagmite 14C data and known atmospheric 14C variability. The new method produces excellent results for stalagmites that satisfy four requirements: i) the absence of long-term secular variability in DCF (i.e., stalagmite DCF varies around a mean value with no long-term trend), ii) stalagmite growth rate does not vary significantly (the technique identifies stalagmites with substantial growth rate variability), iii) the stalagmite record is long enough that measurable 14C decay has occurred, and iv) one ‘anchor’ point exists where the calendar age is known. The model produces good results for a previously U–Th dated stalagmite from Heshang Cave, China, and is then applied to an undated stalagmite from southern Poland. The new method will not replace high-precision U–Th measurements, because the precision of the technique is difficult to quantify. However, it provides a means for dating certain stalagmites undateable by conventional U–Th methods and for refining coarse U–Th chronologies
