    Periapical status in patients affected by osteoporosis: A retrospective clinical study

    Objectives: To assess the periapical status in patients with osteoporosis (OP) treated with denosumab (D), bisphosphonates (BPs), or not on medication, and to understand if these conditions influence the prevalence and the progression of apical periodontitis (AP).Material and Methods: Seventy-six patients with OP alone or treated with D, or BPs, formed the study group (O), and those from 76 patients matched for age and sex, without diseases, and not taking medications, constituted the control (C) in this retrospective case-control study. The data from the complete clinical and radiographic examination, medical history, decayed, missing, and filled teeth (DMFT), and periapical index score (PAI) were recorded for each patient. Wilcoxon rank test, chi(2), and Student's t test were used as appropriate.Results: The prevalence of AP was similar in O and C. Furthermore, AP was significantly more frequent in root canal-treated teeth in O patients (p = .03).Conclusions: OP does not appear to be associated with the development of AP. Moreover, the increased prevalence of AP in root canal-treated teeth in O patients highlights a possible relationship between the healing dynamics of the disease post-therapy and the patients' medication. A larger sample is needed to confirm these findings

    A bibliometric analysis of the dental scientific literature on COVID-19

    Objectives The rapid production of a large volume of literature during the early phase of the COVID-19 outbreak created a substantial burden for clinicians and scientists. Therefore, this manuscript aims to identify and describe the scientific literature addressing COVID-19 from a dental research perspective, in terms of the manuscript origin, research domain, study type, and level of evidence (LoE). Materials and methods Data were retrieved from Web of Science, Scopus, and PubMed. A descriptive analysis of bibliographic data, collaboration network, and keyword co-occurrence analysis were performed. Articles were further classified according to the field of interest, main research question, type of study, and LoE. Results The present study identified 296 dental scientific COVID-19 original papers, published in 89 journals, and co-authored by 1331 individuals affiliated with 429 institutions from 53 countries. Although 81.4% were single-country papers, extensive collaboration among the institutions of single countries (Italian, British, and Brazilian institutions) was observed. The main research areas were as follows: the potential use of saliva and other oral fluids as promising samples for COVID-19 testing, dental education, and guidelines for the prevention of COVID-19 transmission in dental practice. The majority of articles were narrative reviews, cross-sectional studies, and short communications. The overall LoE in the analyzed dental literature was low, with only two systematic reviews with the highest LoE I. Conclusion The dental literature on the COVID-19 pandemic does not provide data relevant to the evidence-based decision-making process. Future studies with a high LoE are essential to gain precise knowledge on COVID-19 infection within the various fields of Dentistry. Clinical relevance The published dental literature on COVID-19 consists principally of articles with a low level of scientific evidence which do not provide sufficient reliable high-quality evidence that is essential for decision making in clinical dental practice

    Epigenetic Approaches to the Treatment of Dental Pulp Inflammation and Repair: Opportunities and Obstacles

    Concerns over the cost and destructive nature of dental treatment have led to the call for novel minimally invasive, biologically based restorative solutions. For patients with toothache, this has resulted in a shift from invasive root-canal-treatment (RCT) toward more conservative vital-pulp-treatment (VPT) procedures, aimed to protect the pulp and harness its natural regenerative capacity. If the dental pulp is exposed, as long as the infection and inflammation can be controlled, conservative therapies can promote the formation of new tertiary dentine in a stem cell-led reparative process. Crucially, the volume and quality of new dentine is dependent on the material applied; however, currently available dental-materials are limited by non-specific action, cytotoxicity and poor clinical handling. Looking to the future, an improved understanding of the cellular regulators of pulpal inflammation and associated repair mechanisms is critical to predict pulpal responses and devise novel treatment strategies. Epigenetic modifications of DNA-associated proteins and the influences of non-coding RNAs have been demonstrated to control the self-renewal of stem cell populations as well as regulate mineralised tissue development and repair. Notably, the stability of microRNAs and their relative ease of sampling from pulpal blood highlight their potential for application as diagnostic inflammatory biomarkers, while increased understanding of their actions will not only enhance our knowledge of pulpal disease and repair, but also identify novel molecular targets. The potential therapeutic application of epigenetic modifying agents, DNA-methyltransferase-inhibitors (DNMTi) and histone-deacetylase-inhibitors (HDACi), have been shown to promote mineralisation and repair processes in dental-pulp-cell (DPC) populations as well as induce the release of bioactive dentine-matrix-components. Consequently, HDACis and DNMTis have the potential to enhance tertiary dentinogenesis by influencing the cellular and tissue processes at low concentrations with minimal side effects, providing an opportunity to develop a topically placed, inexpensive bio-inductive restorative material. The aim of this review is to highlight the potential role of epigenetic approaches in the treatment of the damaged dental pulp, considering the opportunities and obstacles, such as off-target effects, delivery mechanisms, for the therapeutic use of miRNA as an inflammatory biomarker or molecular target, before discussing the application of HDACi and DNMTi to the damaged pulp to stimulate repair

    Fluxes in H\alpha and Ca II H and K for a sample of Southern stars

    The main chromospheric activity indicator is the S index, which is esentially the ratio of the flux in the core of the Ca II H and K lines to the continuum nearby, and is well studied basically for stars from F to K. Another usual chromospheric proxy is the H\alpha line, which is beleived to be tightly correlated with the Ca II index. In this work we characterize both chromospheric activity indicators, one associated with the H and K Ca II lines and the other with H\alpha, for the whole range of late type stars, from F to M. We present periodical medium-resolution echelle observations covering the complete visual range, which were taken at the CASLEO Argentinean Observatory. These observations are distributed along 7 years. We use a total of 917 flux-calibrated spectra for 109 stars which range from F6 to M5. We statistically study these two indicators for stars of different activity levels and spectral types. We directly derive the conversion factor which translate the known S index to flux in the Ca II cores, and extend its calibration to a wider spectral range. We investigate the relation between the activity measurements in the calcium and hydrogen lines, and found that the usual correlation observed is basically the product of the dependence of each flux with stellar colour, and not the product of similar activity phenomena.Comment: 12 pages, including 11 figures and 2 tables.     Factors that influence the outcome of pulpotomy in permanent teeth

    The promotion of minimally invasive treatments focussed on the maintenance of pulp vitality has become a priority area in Endodontics. These vital pulp treatments (VPT) include partial and full pulpotomy, during which diseased coronal pulp tissue is removed prior to placement of a capping biomaterial and restoration. Traditionally, pulpotomies were confined to the treatment of carious primary and traumatized permanent teeth. However, these treatments have now been proposed as definitive solutions for cariously exposed permanent teeth with mild symptoms or even symptoms indicative of irreversible disease. Until recently, it was recommended that carious exposure of mature permanent teeth be managed by root canal treatment. The promotion of pulpotomy as an alternative treatment has opened up a wave of laboratory and clinical research aimed at improving therapies or evaluating clinical outcomes. In modern evidence‐based endodontics, it is imperative that the outcomes of both partial and full pulpotomy are considered and important prognostic factors identified, so that improvements can be made to aid clinical decision‐making and to direct new research. In this narrative review, the outcomes of partial and full pulpotomy are discussed, before analysis of patient, intraoperative and postoperative factors that influence the outcome of the pulpotomy procedure. The review highlights that although partial and full pulpotomy for the treatment of even pulpal disease are highly successful procedures, this is based on low‐quality evidence with a lack of prospective, comparative trials investigating potential prognostic factors. Based on current evidence, it appears that age, gender, tooth type, root development and intraoperative pulpal haemorrhage do not impact significantly on pulpotomy outcome, whilst others such as caries depth, inflammatory status of the pulp, capping material, level of inflammatory pulpal‐biomarkers and the final restoration integrity do. Other factors, including the influence of exposure type, periodontal condition, pulpal lavage, magnification, operator experience, isolation of the operating field and type of pulpotomy, require further experimental investigation before definitive conclusions can be made relating to the success of the pulpotomy procedure. Finally, there is not only a need for future well‐designed prospective research addressing these issues but also a widening of our understanding of outcome to include patient‐reported as well as clinician‐reported outcomes

    Transcriptional profiling of suberoylanilide hydroxamic acid (SAHA) regulated genes in mineralizing dental pulp cells at early and late time points

    Dental pulp tissue can be damaged by a range of irritants, however, if the irritation is removed and/or the tooth is adequately restored, pulp regeneration is possible (Mjör and Tronstad, 1974 [1]). At present, dental restorative materials limit healing by impairing mineralization and repair processes and as a result new biologically-based materials are being developed (Ferracane et al., 2010 [2]). Previous studies have highlighted the benefit of epigenetic modification by histone deacetylase inhibitor (HDACi) application to dental pulp cells (DPCs), which induces changes to chromatin architecture, promoting gene expression and cellular-reparative events (Duncan et al., 2013 [3]; Paino et al., 2014 [4]). In this study a genome-wide transcription profiling in epigenetically-modified mineralizing primary DPC cultures was performed, at relatively early and late time-points, to identify differentially regulated transcripts that may provide novel therapeutic targets for use in restorative dentistry.     Discovery of a weak magnetic field in the photosphere of the single giant Pollux

    Aims: We observe the nearby, weakly-active single giant, Pollux, in order to directly study and infer the nature of its magnetic field. Methods: We used the new generation spectropolarimeters ESPaDOnS and NARVAL to observe and detect circular polarization within the photospheric absorption lines of Pollux. Our observations span 18 months from 2007-2009. We treated the spectropolarimetric data using the Least-Squares Deconvolution method to create high signal-to-noise ratio mean Stokes V profiles. We also measured the classical activity indicator S-index for the Ca H&K lines, and the stellar radial velocity (RV). Results: We have unambiguously detected a weak Stokes V signal in the spectral lines of Pollux, and measured the related surface-averaged longitudinal magnetic field Bl. The longitudinal field averaged over the span of the observations is below one gauss. Our data suggest variations of the longitudinal magnetic field, but no significant variation of the S-index. We observe variations of RV which are qualitatively consistent with the published ephemeris for a proposed exoplanet orbiting Pollux. The observed variations of Bl appear to mimic those of RV, but additional data for this relationship to be established. Using evolutionary models including the effects of rotation, we derive the mass of Pollux and we discuss its evolutionary status and the origin of its magnetic field. Conclusions: This work presents the first direct detection of the magnetic field of Pollux, and demonstrates that ESPaDOnS and NARVAL are capable of obtaining sub-G measurements of the surface-averaged longitudinal magnetic field of giant stars, and of directly studying the relationships between magnetic activity, stellar evolution and planet hosting of these stars.Comment: 8 pages, 6 figures, accepted for publication in Astronomy and Astrophysic

    Coenzyme Q10 Levels Are Decreased in the Cerebellum of Multiple-System Atrophy Patients

    Background: The objective of this study was to evaluate whether the levels of coenzyme Q10 (CoQ10) in brain tissue of multiple system atrophy (MSA) patients differ from those in elderly controls and in patients with other neurodegenerative diseases. Methods: Flash frozen brain tissue of a series of 20 pathologically confirmed MSA patients [9 olivopontocerebellar atrophy (OPCA) type, 6 striatonigral degeneration (SND) type, and 5 mixed type] was used for this study. Elderly controls (n = 37) as well as idiopathic Parkinson's disease (n = 7), dementia with Lewy bodies (n = 20), corticobasal degeneration (n = 15) and cerebellar ataxia (n = 18) patients were used as comparison groups. CoQ10 was measured in cerebellar and frontal cortex tissue by high performance liquid chromatography. Results: We detected a statistically significant decrease (by 3–5%) in the level of CoQ10 in the cerebellum of MSA cases (P = 0.001), specifically in OPCA (P = 0.001) and mixed cases (P = 0.005), when compared to controls as well as to other neurodegenerative diseases [dementia with Lewy bodies (P<0.001), idiopathic Parkinson's disease (P<0.001), corticobasal degeneration (P<0.001), and cerebellar ataxia (P = 0.001)]. Conclusion: Our results suggest that a perturbation in the CoQ10 biosynthetic pathway is associated with the pathogenesis of MSA but the mechanism behind this finding remains to be elucidated

    The age-mass-metallicity-activity relation for solar-type stars: comparisons with asteroseismology and the NGC 188 open cluster

    Context. The Mount Wilson Ca ii index log is the accepted standard metric of calibration for the chromospheric activity versus age relation for FGK stars. Recent results claim its inability to discern activity levels, and thus ages, for stars older than ~2 Gyr, which would severely hamper its application to date disk stars older than the Sun. Aims. We present a new activity-age calibration of the Mt. Wilson index that explicitly takes mass and [Fe/H] biases into account; these biases are implicit in samples of stars selected to have precise ages, which have so far not been appreciated. Methods. We show that these selection biases tend to blur the activity-age relation for large age ranges. We calibrate the Mt. Wilson index for a sample of field FGK stars with precise ages, covering a wide range of mass and [Fe/H] , augmented with data from the Pleiades, Hyades, M 67 clusters, and the Ursa Major moving group. Results. We further test the calibration with extensive new Gemini/GMOS log ()R'HK) data of the old, solar [Fe/H] clusters, M 67 and NGC 188. The observed NGC 188 activity level is clearly lower than M 67. We correctly recover the isochronal age of both clusters and establish the viability of deriving usable chromospheric ages for solar-type stars up to at least ~6 Gyr, where average errors are ~0.14 dex provided that we explicitly account for the mass and [Fe/H] dimensions. We test our calibration against asteroseismological ages, finding excellent correlation (ρ = + 0.89). We show that our calibration improves the chromospheric age determination for a wide range of ages, masses, and metallicities in comparison to previous age-activity relations
