    Bacterial Ligase D preternary-precatalytic complex performs efficient abasic sites processing at double strand breaks during nonhomologous end joining

    Abasic (AP) sites, the most common DNA lesions are frequently associated with double strand breaks (DSBs) and can pose a block to the final ligation. In many prokaryotes, nonhomologous end joining (NHEJ) repair of DSBs relies on a two-component machinery constituted by the ring-shaped DNA-binding Ku that recruits the multicatalytic protein Ligase D (LigD) to the ends. By using its polymerization and ligase activities, LigD fills the gaps that arise after realignment of the ends and seals the resulting nicks. Here, we show the presence of a robust AP lyase activity in the polymerization domain of Bacillus subtilis LigD (BsuLigD) that cleaves AP sites preferentially when they are proximal to recessive 5'-ends. Such a reaction depends on both, metal ions and the formation of a Watson¿Crick base pair between the incoming nucleotide and the templating one opposite the AP site. Only after processing the AP site, and in the presence of the Ku protein, BsuLigD catalyzes both, the in-trans addition of the nucleotide to the 3'-end of an incoming primer and the ligation of both ends.     Póster presentado en: ICHM (International Commision on the History of Meteorology) Meeting celebrado del 5 al 9 de julio de 2004 en Polling, AlemaniaThe creation of a meteorological observatory in the mountains of Tenerife island was the result of scientific interest, in which historical and political events also played a role. The exceptionality and suitability of the physical and geographic conditions in Tenerife for the study and research of the atmosphere have been known and emphasized since the mid-17th Century. Since then, every area of meteorological importance, from the regime of general circulation of the atmosphere to atmospheric chemistry was the object of observation and studies by scientists visiting the island. The Observatory at Izaña was founded at the beginning of the 20th Century thanks to the triumph of European meteorological science over a diplomatic and political conflict between the Spanish and German governments. Today, after four centuries of scientific interest in Tenerife, the most symbolic characteristic of the Observatory is the universality of its objectives and research projects

    <i>Phaeocystis globosa</i> Virus DNA Polymerase X: a “Swiss Army knife”, Multifunctional DNA polymerase-lyase-ligase for Base Excision Repair

    Phaeocystis globosa virus 16T is a giant virus that belongs to the so-called nucleo-cytoplasmic large DNA virus (NCLDV) group. Its linear dsDNA genome contains an almost full complement of genes required to participate in viral base excision repair (BER). Among them is a gene coding for a bimodular protein consisting of an N-terminal Polβ-like core fused to a C-terminal domain (PgVPolX), which shows homology with NAD+-dependent DNA ligases. Analysis of the biochemical features of the purified enzyme revealed that PgVPolX is a multifunctional protein that could act as a “Swiss army knife” enzyme during BER since it is endowed with: 1) a template-directed DNA polymerization activity, preferentially acting on DNA structures containing gaps; 2) 5′-deoxyribose-5-phosphate (dRP) and abasic (AP) site lyase activities; and 3) an NAD+-dependent DNA ligase activity. We show how the three activities act in concert to efficiently repair BER intermediates, leading us to suggest that PgVPolX may constitute, together with the viral AP-endonuclease, a BER pathway. This is the first time that this type of protein fusion has been demonstrated to be functional

    Seroepidemiological surveys of non vaccine-preventable diseases and their interest in public health

    Seroepidemiological surveys are epidemiological studies carried out by the use of serum tests to detect infection; they can be applied to infections in general and to vaccine-preventable diseases in particular. Among other applications, seroepidemiological studies are useful for determining groups at risk for a specific disease; evaluating transmission mechanisms; and determining population groups who are critical in maintaining the transmission of infectious agents. We analysed the results of seroprevalence studies in Spain on vertically-transmitted non vaccine-preventable diseases (Toxoplasma gondii, Treponema pallidum, cytomegalovirus, herpes simplex virus type 2, and parvovirus human B19), blood-borne diseases (hepatitis C virus, human immunodeficiency virus and Trypanosoma cruzi) and emerging diseases (West Nile virus, Toscana virus, and lymphocytic choriomeningitis virus). We found a reduced seroprevalence against Toxoplasma gondii, Treponema pallidum, cytomegalovirus and human immunodeficiency virus in the Spanish indigenous population. The results found in immigrants reflect the situation of these diseases in their countries of origin and suggest they could have a substantial public health impact in Spain (Trypanosoma cruzi, in association with blood donation). We highlight the circulation in Spain of the West Nile virus and the importance of Toscana virus infection. La seroepidemiología es el estudio epidemiológico que se realiza mediante del uso de pruebas serológicas para detectar infección; es de aplicación a las infecciones en general, y a las enfermedades inmunoprevenibles. Entre otras aplicaciones, los estudios seroepidemiológicos son de utilidad para determinar las poblaciones en riesgo para una enfermedad; para evaluar los mecanismos de transmisión, y para determinar los grupos de población que son críticos para mantener la transmisión de los agentes infecciosos. Se han analizado resultados derivados de estudios de seroprevalencia realizados en España en relación con enfermedades no inmunoprevenibles de transmisión vertical (Toxoplasma gondii, Treponema pallidum, citomegalovirus, virus herpes simple tipo 2, y parvovirus humano B19), enfermedades de transmisión sanguínea (virus de la hepatitis C, virus de la inmunodeficiencia humana y Trypanosoma cruzi) y enfermedades emergentes (Virus del Nilo occidental, virus Toscana, y virus de la coriomeningitis linfocitaria). Se observa reducción en la seroprevalencia en la población autóctona española frente a Toxoplasma gondii, Treponema pallidum, citomegalovirus y virus de la inmunodeficiencia humana. Los patrones obtenidos en población inmigrante reflejan la situación de estas enfermedades en sus países de origen; pudiendo ser causa de situaciones de impacto en la salud pública (Trypanosoma cruzi, en relación con la donación de sangre).     ICT and gamification experiences with CLIL methodology as innovative resources for the development of competencies in compulsory secondary education

    Due to the lack of research in the field of music through CLIL and ICT in Secondary Education in Spain, we propose to merge this subject with an innovative methodology. The general objective is to present a different approach to students in order to increase their motivation for the subject. The specific objectives are to teach music in English by integrating content, cognition and vocabulary through communication and the use of new technologies. This approach makes the design and implementation didactic, audiovisual and multimedia. Cuadernia: digital notebook composed of flash files, videos and audios, and gamification strategies. Quizziz: multiplayer questionnaire of class contents, based on teaching music through a cooperative methodology and gamification, are applied as options with a great educational projection, with information and innovative pedagogical experiences and potential to work together in the classroom with different levels of the subject. The results that follow the Likert scale reflect high student satisfaction with regard to teaching methodology, content acquisition, teacher training, ICT and resources. As a conclusion, of the coincident results between the professor and the students, we would like to implement it in the coming years

To achien un aragonese functional hierarchization we utilice Davies model, using 117 variables fiscal licences).From the continuons hierarchy of real centralities we arrive to a discrete hierarchy of the christallerian type through several steps: thresholds by the means of typified measures, metropolitan area municipal elimination, suppression at an elemental level of negative attraction idex, consideration of centrality mean increments to accept 'continuum' ruptures, chistering of hierarchical levels through improvement of the nearest-neighbour index.Se plantean primero las dificultades que entra&ntilde;a pasar de un modelo jerarquizado continuo a otro discreto y la necesidad de jerarquias discretas para la ordenaci&oacute;n del territorio. Para la jerarquizaci&oacute;n funcional de Arag&oacute;n se parte del modelo de Davies utilizando 117 variables (licencias fiscales).
