    Characterization and performance of a toluene-degrading biofilm developed on pumice stones

    BACKGROUND: Hydrocarbon-degrading biofilms in the treatment of contaminated groundwaters have received increasing attention due to the role played in the so-called "biobarriers". These are bioremediation systems in which a microbial consortium adherent to a solid support is placed across the flow of a contaminated plume, thus promoting biodegradation of the pollutant. RESULTS: A microbial consortium adherent to pumice granules (biofilm) developed from a toluene-enriched microflora in a mini-scale system, following continuous supply of a mineral medium containing toluene, over a 12-month period. Observation by scanning electron microscopy, together with quantification of the biomass attached to pumice, evidenced the presence of abundant exopolymeric material surrounding the cells in the biofilm. Toluene removal monitored during 12-month operation, reached 99%. Identification of the species, based on comparative 16S ribosomal DNA (rDNA) sequence analysis, revealed that Rhodococcus erythropolis and Pseudomonas marginalis were the predominant bacterial species in the microbial consortium.     Arm trajectories and writing strategy in healthy children

    BACKGROUND: Evaluation of elementary writing skills in children is usually obtained with high resolution (and high cost) techniques or with low resolution pen-and-paper tests. In this observational study we tested a quantitative method to obtain normative data to describe arm movement during a writing precursor gesture. METHODS: We recruited 226 healthy children (mean age 9,1 years [range: 6.3 – 11.4 years]), attending primary schools belonging to the “Istituto Comprensivo” of Rivanazzano Terme (Pavia). We asked to drive a cursor through a polygonal path (labyrinth) projected in front of them using a wireless mouse. Dartfish™ video analysis software was used to elaborate images and Excel™, MedCalc™ and Statistica 7™ to analyze values of shoulder, elbow and wrist ranges of motion, arm trajectories, execution times and gesture accuracy. RESULTS: Differences seen in motor strategies, when divided according to attended class, suggest a proximal-distal maturation of motor control. Obtained values were not significantly correlated with variables such as gender, ethnicity or cognitive functioning. CONCLUSIONS: This type of approach to a study of arm movement during childhood represents a valid alternative to other tests, considering that it can differentiate children who perform similarly in the VMI test and is non-invasive, low-cost and easily reproducible

    Itch/β-arrestin2-dependent non-proteolytic ubiquitylation of SuFu controls Hedgehog signalling and medulloblastoma tumorigenesis

    Suppressor of Fused (SuFu), a tumour suppressor mutated in medulloblastoma, is a central player of Hh signalling, a pathway crucial for development and deregulated in cancer. Although the control of Gli transcription factors by SuFu is critical in Hh signalling, our understanding of the mechanism regulating this key event remains limited. Here, we show that the Itch/β-arrestin2 complex binds SuFu and induces its Lys63-linked polyubiquitylation without affecting its stability. This process increases the association of SuFu with Gli3, promoting the conversion of Gli3 into a repressor, which keeps Hh signalling off. Activation of Hh signalling antagonises the Itch-dependent polyubiquitylation of SuFu. Notably, different SuFu mutations occurring in medulloblastoma patients are insensitive to Itch activity, thus leading to deregulated Hh signalling and enhancing medulloblastoma cell growth. Our findings uncover mechanisms controlling the tumour suppressive functions of SuFu and reveal that their alterations are implicated in medulloblastoma tumorigenesis

    Characterising galaxy groups: spectroscopic observations of the Shakhbazyan sample

    Groups of galaxies are the most common cosmic structures. However, due to the poor statistics, projection effects and the lack of accurate distances, our understanding of their dynamical and evolutionary status is still limited. This is particularly true for the so called Shakhbazyan groups (SHK) which are still largely unexplored due to the lack of systematic spectroscopic studies of both their member galaxies and the surrounding environment. In our previous paper, we investigated the statistical properties of a large sample of SHK groups using Sloan Digital Sky Survey data and photometric redshifts. Here we present the follow-up of 5 SHK groups (SHK 10, 71, 75, 80, 259) observed within our spectroscopic campaign with the Telescopio Nazionale Galileo, aimed at confirming their physical reality and strengthening our photometric results. For each of the selected groups we were able to identify between 6 and 13 spectroscopic members, thus confirming the robustness of the photometric redshift approach in identifying real galaxy over-densities. Consistently with the finding of our previous paper, the structures studied here have properties spanning from those of compact and isolated groups to those of loose groups. For what the global physical properties are concerned (total mass, mass-to-light ratios, etc.), we find systematic differences with those reported in the literature by previous studies. Our analysis suggests that previous results should be revisited; we show in fact that, if the literature data are re-analysed in a consistent and homogeneous way, the properties obtained are in agreement with those estimated for our sample.Comment: 31 pages, 25 figures, 10 tables.     Investigating the composition of Potentially Hazardous Asteroids with the NEO-SURFACE survey

    There is a high degree of diversity among the physical properties of the Potentially Hazardous asteroids (PHAs). For these objects, the physical characterization is essential to define a successful mitigation mission, therefore ground-based surveys like NEO-SURFACE could provide a fundamental contribution. Our analysis suggest a prevalence of silicate S-types in the PHA population, which could be due in principle to the high efficiency of the transport mechanisms in the inner main belt, or to an observational bias due to the fact that S-types are brighter. <P /

    Calcareous phytoplankton response to the half century of interannual climatic variability in Santa Barbara Basin (California)

    A high-resolution study of calcareous phytoplankton in a box core from the Santa Barbara Basin (SBB) reveals floral assemblage fluctuations which can be related to climatic and paleoceanographic changes during the last half century (1940-1996). In particular, Gephyrocapsa oceanica production increased during El Niño periods, in response to high temperatures, silica depletion, and increased iron availability. Conversely, Helicosphaera carteri flux increases in conjunction with lower surface temperatures associated with La Niña episodes. Increasing abundances of Florisphaera profunda and Umbilicosphaera sibogae after 1970 reflect a warming trend and increased stratification within the basin associated with the warm phase of the Pacific Decadal Oscillation (PDO). Conversely, increased abundances of Coccolithus pelagicus and Calcidiscus leptoporus before 1970 mark the cold phase of PDO. These coccolithophore production rate data are consistent with instrumental records of surface and thermocline temperatures monitored since 1950.
