    Sport, space and segregation Pietermaritzburg, 1900-1980

    Sport is located historically within its political, economic, cultural and social context in order to assess its role in human and spatial relations; and its meaning for various communities. This study aims to measure the impact of dominant ideology (imperialism, segregation and apartheid), the degree to which sport was used instrumentally by the authorities, and the extent to which it was seen as a site of struggle by the oppressed. The main sources for this thesis were archival and published material emanating from the municipality. Before apartheid matured and became more secretive, they were remarkably candid about official aims and objectives. A wide variety of secondary sources was consulted; and interviews conducted. The conclusion is that sport is an appropriate lens through which to view urban history and the relationships that shape it. Driven by a consistent ideological desire for White separateness, sport reflected social hegemony and assumptions about relative competence and ability. Instrumental use of sport by the local authority, apart from a skewed use of economic resources to unite whites and maintain their distance from other communities, was poorly regarded. This was because of the availability of other, coercive means of control and the dispensability of the individuals targeted. Black sportspersons were in effect squatters at impermanent facilities. Black recreation was a challenge to White ideas about the use of urban space. For Africans, the controlled area preferred by the authorities was the beerhall, not the sports field. Effective resistance organised around sport emerged only when the authorities needed Asian and Coloured cooperation to implement group areas and at this point sport became a notable site of struggle. Ultimately sports facilities, originally a symbol of White civilisation and authority, became a potent emblem of the spatial conflict that characterised apartheid. They also reflected the fact that South African urban geography had always been an evolving saga of insiders and outsiders. Sport provided the proponents of White dominance with a theatre for propaganda; while at the same time offering its protagonists a stage upon which to demonstrate their opposition.     During the latter half of the twentieth century, the American food system was transformed by a technological revolution in American agriculture. While these changes provided benefits such as lower-cost food, it also generated concerns that the unconditional embrace of technology would harm rural communities and the environment. Additional concerns were raised about food quality and food safety. Through a case study of a rural Midwestern farming community, this paper examines how direct consumer to producer marketing strategies such as community supported agriculture (CSA) and the public’s current fascination with the heritage of farming may offer a subset of producers and consumers a sustainable alternative to large-scale production agriculture and the “fast” foods resulting from such production

    Stable Isotope Analysis of Human Bone from Ganj Dareh, Iran, Ca. 10,100 calBP

    We report here on stable carbon, nitrogen, and sulfur isotope values from bone collagen of human (n = 20) and faunal (n = 11) remains from the Early Neolithic site of Ganj Dareh, Iran, dating to ca. 10,100 cal. BP. Our focus explores how isotope values of human bone vary by age and sex, and evaluates dietary practices at this site. It also provides a baseline for future studies of subsistence in the early Holocene Central Zagros Mountains, from the site with the first evidence for human ovicaprid management in the Near East. Human remains include individuals of all age groups for dietary reconstruction, as well two Ottoman intrusive burials for temporal and cultural comparison. All analyzed individuals exhibited δ13C and δ15N values consistent with a diet based heavily on C3 terrestrial sources. There is no statistically significant difference between the isotopic compositions of the two sexes, though males appear to show larger variations compared to females. Interesting patterns in the isotopic compositions of the subadults suggested weaning children may be fed with supplements with distinctive δ13C values. Significant difference in sulfur isotope values between humans and fauna could be the earliest evidence of transhumance and could identify one older adult male as a possible transhumant shepherd. Both Ottoman individuals had distinctively different δ13C, δ15N, and δ34S values compared to the Neolithic individuals. This is the first large scale analysis of human stable isotopes from the eastern end of the early Holocene Fertile Crescent. It provides a baseline for future intersite exploration of stable isotopes and insight into the lifeways, health, and processes of neolithisation associated with the origins of goat domestication at Ganj Dareh and the surrounding Central Zagros

    The SLUGGS Survey: Calcium Triplet-based Spectroscopic Metallicities for Over 900 Globular Clusters

    Although the colour distribution of globular clusters in massive galaxies is well known to be bimodal, the spectroscopic metallicity distribution has been measured in only a few galaxies. After redefining the calcium triplet index-metallicity relation, we use our relation to derive the metallicity of 903 globular clusters in 11 early-type galaxies. This is the largest sample of spectroscopic globular cluster metallicities yet assembled. We compare these metallicities with those derived from Lick indices finding good agreement. In 6 of the 8 galaxies with sufficient numbers of high quality spectra we find bimodality in the spectroscopic metallicity distribution. Our results imply that most massive early-type galaxies have bimodal metallicity, as well as colour, distributions. This bimodality suggests that most massive galaxies early-type galaxies experienced two periods of star formation.Comment: 23 pages, 13 figures. Accepted for publication in MNRAS.     Transient tissue priming via ROCK inhibition uncouples pancreatic cancer progression, sensitivity to chemotherapy, and metastasis

    The emerging standard of care for patients with inoperable pancreatic cancer is a combination of cytotoxic drugs gemcitabine and Abraxane, but patient response remains moderate. Pancreatic cancer development and metastasis occur in complex settings, with reciprocal feedback from microenvironmental cues influencing both disease progression and drug response. Little is known about how sequential dual targeting of tumor tissue tension and vasculature before chemotherapy can affect tumor response. We used intravital imaging to assess how transient manipulation of the tumor tissue, or "priming," using the pharmaceutical Rho kinase inhibitor Fasudil affects response to chemotherapy. Intravital Förster resonance energy transfer imaging of a cyclin-dependent kinase 1 biosensor to monitor the efficacy of cytotoxic drugs revealed that priming improves pancreatic cancer response to gemcitabine/Abraxane at both primary and secondary sites. Transient priming also sensitized cells to shear stress and impaired colonization efficiency and fibrotic niche remodeling within the liver, three important features of cancer spread. Last, we demonstrate a graded response to priming in stratified patient-derived tumors, indicating that fine-tuned tissue manipulation before chemotherapy may offer opportunities in both primary and metastatic targeting of pancreatic cancer

    Genomic insights into the origin of farming in the ancient Near East

    We report genome-wide ancient DNA from 44 ancient Near Easterners ranging in time between ~12,000 and 1,400 BC, from Natufian hunter–gatherers to Bronze Age farmers. We show that the earliest populations of the Near East derived around half their ancestry from a ‘Basal Eurasian’ lineage that had little if any Neanderthal admixture and that separated from other non-African lineages before their separation from each other. The first farmers of the southern Levant (Israel and Jordan) and Zagros Mountains (Iran) were strongly genetically differentiated, and each descended from local hunter–gatherers. By the time of the Bronze Age, these two populations and Anatolian-related farmers had mixed with each other and with the hunter–gatherers of Europe to greatly reduce genetic differentiation. The impact of the Near Eastern farmers extended beyond the Near East: farmers related to those of Anatolia spread westward into Europe; farmers related to those of the Levant spread southward into East Africa; farmers related to those of Iran spread northward into the Eurasian steppe; and people related to both the early farmers of Iran and to the pastoralists of the Eurasian steppe spread eastward into South Asia

    Pancreatic cancer genomes reveal aberrations in axon guidance pathway genes.

    Pancreatic cancer is a highly lethal malignancy with few effective therapies. We performed exome sequencing and copy number analysis to define genomic aberrations in a prospectively accrued clinical cohort (n = 142) of early (stage I and II) sporadic pancreatic ductal adenocarcinoma. Detailed analysis of 99 informative tumours identified substantial heterogeneity with 2,016 non-silent mutations and 1,628 copy-number variations. We define 16 significantly mutated genes, reaffirming known mutations (KRAS, TP53, CDKN2A, SMAD4, MLL3, TGFBR2, ARID1A and SF3B1), and uncover novel mutated genes including additional genes involved in chromatin modification (EPC1 and ARID2), DNA damage repair (ATM) and other mechanisms (ZIM2, MAP2K4, NALCN, SLC16A4 and MAGEA6). Integrative analysis with in vitro functional data and animal models provided supportive evidence for potential roles for these genetic aberrations in carcinogenesis. Pathway-based analysis of recurrently mutated genes recapitulated clustering in core signalling pathways in pancreatic ductal adenocarcinoma, and identified new mutated genes in each pathway. We also identified frequent and diverse somatic aberrations in genes described traditionally as embryonic regulators of axon guidance, particularly SLIT/ROBO signalling, which was also evident in murine Sleeping Beauty transposon-mediated somatic mutagenesis models of pancreatic cancer, providing further supportive evidence for the potential involvement of axon guidance genes in pancreatic carcinogenesis
