    Electrophoresis in the Comet Assay

    The comet assay is a sensitive technique to measure lesions in DNA, based on electrophoretic separation of DNA from cells embedded in agarose. Movement of DNA fragments is determined by the potential (V/cm), the time, and the viscosity of the medium (agarose). There is historically considerable confusion as to other factors, that is, current, liquid depths, circulation of the liquid, and temperature. Lack of standardization of electrophoresis including suboptimal power supplies and electrophoresis tanks causes considerable variations within and between laboratories. Ring trials have not been able to clearly identify the cause(s) of variation. Comparison of comet data from cohorts of human blood lymphocytes is used in the COST project hCOMET to identify early biomarkers of the disease. This calls for standardization of analysis. We performed measurements of electric potentials in a tank using multiple electrodes. Variations (time/position) were reduced by circulating electrophoresis liquid at 10% (volume) per min; this also stabilized the temperature. Circulation was accompanied by only slightly reduced variation in DNA damage among 384 irradiated cell samples electrophoresed concomitantly. In conclusion, comparing data between laboratories and cohorts must give emphasis to electrophoresis conditions. Results should be specified with respect to voltage (V/cm), time, and agarose concentration. We expect that suitable correction factors for these parameters may reduce inter-laboratory variations in comet data, allowing more precise comparison of results from different human cohorts

    Maternal seafood consumption in highly educated women is reduced in pregnancy: A pilot study

    Oily fish is a unique natural source to vitamin D and marine n-3 fatty acids. The aim of the present study was to investigate from where pregnant women get dietary advices and whether they eat seafood during pregnancy or not. A self-selected convenience sample (n = 43) of highly educated women responded to a self-administrated web based questionnaire. The women reduced their intake of both lean (p < 0.013) and oily (p < 0.003) fish during pregnancy, indicating that the women were not fully aware of the beneficial effects of seafood intake during pregnancy

    Lifestyle and metabolic factors affect risk for meningioma in women: a prospective population-based study (The Cohort of Norway)

    BackgroundMeningioma is the most common primary brain tumor, with a clear preponderance in women. Obesity is considered a risk factor for the development of meningioma. Obesity is also the clinical hallmark of metabolic syndrome, characterized by glucose intolerance, dyslipidemia, and hypertension. Lifestyle and metabolic factors directly impact overweight and obesity and are therefore potential risk factors for meningioma development. The aim of this study is to assess lifestyle and metabolic factors for meningioma risk in women.MethodsThe Cohort of Norway (CONOR) is a nationwide health survey, conducted between 1994 and 2003, including anthropometric measures, blood tests, and health questionnaires. Linkage to the National Cancer Registry enabled the identification of intracranial meningioma during follow-up until December 2018.ResultsA total of 81,652 women were followed for a combined total of 1.5 million years, and 238 intracranial meningiomas were identified. Increasing levels of physical activity (HR 0.81; 95% CI 0.68–0.96; p trend <0.02) and parity (HR 0.83; 95% CI 0.71–0.97; p trend <0.03) were negatively associated with meningioma risk. Diabetes mellitus or glucose intolerance increased the risk for meningioma (HR 2.54; 95% CI 1.60–4.05). Overweight and obesity were not associated with meningioma risk, nor was metabolic syndrome. However, participants without metabolic dysfunction had a reduced meningioma risk, while participants with all five metabolic factors present had a 4-fold risk increase for meningioma (HR 4.28; 95% CI 1.34–13.68).ConclusionLifestyle factors seem to significantly influence meningioma risk. However, disentangling the complex associations and interactions between factors for meningioma risk will be a challenging task for future studies

    Assessing the carcinogenic potential of low-dose exposures to chemical mixtures in the environment: the challenge ahead.

    Lifestyle factors are responsible for a considerable portion of cancer incidence worldwide, but credible estimates from the World Health Organization and the International Agency for Research on Cancer (IARC) suggest that the fraction of cancers attributable to toxic environmental exposures is between 7% and 19%. To explore the hypothesis that low-dose exposures to mixtures of chemicals in the environment may be combining to contribute to environmental carcinogenesis, we reviewed 11 hallmark phenotypes of cancer, multiple priority target sites for disruption in each area and prototypical chemical disruptors for all targets, this included dose-response characterizations, evidence of low-dose effects and cross-hallmark effects for all targets and chemicals. In total, 85 examples of chemicals were reviewed for actions on key pathways/mechanisms related to carcinogenesis. Only 15% (13/85) were found to have evidence of a dose-response threshold, whereas 59% (50/85) exerted low-dose effects. No dose-response information was found for the remaining 26% (22/85). Our analysis suggests that the cumulative effects of individual (non-carcinogenic) chemicals acting on different pathways, and a variety of related systems, organs, tissues and cells could plausibly conspire to produce carcinogenic synergies. Additional basic research on carcinogenesis and research focused on low-dose effects of chemical mixtures needs to be rigorously pursued before the merits of this hypothesis can be further advanced. However, the structure of the World Health Organization International Programme on Chemical Safety 'Mode of Action' framework should be revisited as it has inherent weaknesses that are not fully aligned with our current understanding of cancer biology

    DNA damage in circulating leukocytes measured with the comet assay may predict the risk of death

    The comet assay or single cell gel electrophoresis, is the most common method used to measure strand breaks and a variety of other DNA lesions in human populations. To estimate the risk of overall mortality, mortality by cause, and cancer incidence associated to DNA damage, a cohort of 2,403 healthy individuals (25,978 person-years) screened in 16 laboratories using the comet assay between 1996 and 2016 was followed-up. Kaplan-Meier analysis indicated a worse overall survival in the medium and high tertile of DNA damage (p < 0.001). The effect of DNA damage on survival was modelled according to Cox proportional hazard regression model. The adjusted hazard ratio (HR) was 1.42 (1.06-1.90) for overall mortality, and 1.94 (1.04-3.59) for diseases of the circulatory system in subjects with the highest tertile of DNA damage. The findings of this study provide epidemiological evidence encouraging the implementation of the comet assay in preventive strategies for non-communicable diseases
