    Severe course of COVID-19 in pregnant woman and newborn’s alloimmune thrombocytopenia — case study and review of the literature

    So far, little is known about the impact of the coronavirus disease 2019 (COVID-19) on pregnancy and data is often inconsistent. Even less information has been published on the management of severe courses of COVID-19 in pregnant women. By writing this article, we aim to share our experience in the treatment of pregnant woman critically ill with COVID-19 and newborn’s condition, FNAIT (Fetal and neonatal alloimmune thrombocytopenia), as well as literature review of this disease. After admission, the woman’s respiratory status rapidly worsened, requiring administration of oxygen and in the end ECMO therapy. At the 9th day of ECMO support, and 28 weeks 3 days of gestation, due to mother’s prognosis and increased feto-placental vascular resistance a decision of Caesarean section was taken. The neonate required intensive care not only due to extreme prematurity but coagulation disorder, alloimmune thrombocytopenia, which we diagnosed a few weeks after delivery.

    Diversity of Peptides Produced by Nodularia spumigena from Various Geographical Regions

    Abstract: Cyanobacteria produce a great variety of non-ribosomal peptides. Among these compounds, both acute toxins and potential drug candidates have been reported. The profile of the peptides, as a stable and specific feature of an individual strain, can be used to discriminate cyanobacteria at sub-population levels. In our work, liquid chromatography-tandem mass spectrometry was used to elucidate the structures of non-ribosomal peptides produced by Nodularia spumigena from the Baltic Sea, the coastal waters of southern Australia and Lake Iznik in Turkey. In addition to known structures, 9 new congeners of spumigins, 4 aeruginosins and 12 anabaenopeptins (nodulapeptins) were identified. The production of aeruginosins by N. spumigena was revealed in this work for the first time. The isolates from the Baltic Sea appeared to be the richest source of the peptides; they also showed a higher diversity in peptide profiles. The Australian strains were characterized by similar peptide patterns, but distinct from those represented by the Baltic and Lake Iznik isolates.     AtOMA1 Affects the OXPHOS System and Plant Growth in Contrast to Other Newly Identified ATP-Independent Proteases in Arabidopsis Mitochondria

    Compared with yeast, our knowledge on members of the ATP-independent plant mitochondrial proteolytic machinery is rather poor. In the present study, using confocal microscopy and immunoblotting, we proved that homologs of yeast Oma1, Atp23, Imp1, Imp2, and Oct1 proteases are localized in Arabidopsis mitochondria. We characterized these components of the ATP-independent proteolytic system as well as the earlier identified protease, AtICP55, with an emphasis on their significance in plant growth and functionality in the OXPHOS system. A functional complementation assay demonstrated that out of all the analyzed proteases, only AtOMA1 and AtICP55 could substitute for a lack of their yeast counterparts. We did not observe any significant developmental or morphological changes in plants lacking the studied proteases, either under optimal growth conditions or after exposure to stress, with the only exception being retarded root growth in oma1-1, thus implying that the absence of a single mitochondrial ATP-independent protease is not critical for Arabidopsis growth and development. We did not find any evidence indicating a clear functional complementation of the missing protease by any other protease at the transcript or protein level. Studies on the impact of the analyzed proteases on mitochondrial bioenergetic function revealed that out of all the studied mutants, only oma1-1 showed differences in activities and amounts of OXPHOS proteins. Among all the OXPHOS disorders found in oma1-1, the complex V deficiency is distinctive because it is mainly associated with decreased catalytic activity and not correlated with complex abundance, which has been observed in the case of supercomplex I + III2 and complex I deficiencies. Altogether, our study indicates that despite the presence of highly conservative homologs, the mitochondrial ATP-independent proteolytic system is not functionally conserved in plants as compared with yeast. Our findings also highlight the importance of AtOMA1 in maintenance of proper function of the OXPHOS system as well as in growth and development of Arabidopsis thaliana

    Kunstgegenstände & Schmuck herzustellen ist ein urmenschliches Bedürfnis. Aus diesem Kernbereich der Schmuckfertigung und des Drechselns sind die vorliegenden Projektideen entstanden. Die in dieser Handreichung vorgestellten Projektideen sind alle von Lehramtsstudierenden im Fach Technik an den Pädagogischen Hochschulen in Schwäbisch Gmünd und Ludwigsburg in offener und selbstgesteuerter Werkstattarbeit seit dem Sommersemester 2021 entstanden. Die Studierenden haben dabei ihre Idee stets fachpraktisch erprobt und fachdidaktisch in den vorliegenden Erklärfilmen und Schritt für Schritt-Anleitungen ausgearbeitet. Der Projektrahmen umfasst das curricular verankerte, handwerklich bedeutsame Arbeiten mit den grundlegenden Werkstoffen Holz, Metall und Kunststoffen sowie den Bereich der Elektrotechnik und des Mikrocontrolling. Die Umsetzung dieser Projektideen soll unterstützend wirken, um individuelle handwerkliche Kompetenzen zu entdecken, erproben und vertiefen sowie technische Interessensbildung und vorberufliche Orientierung in Schulen fördern. Eine Schlüsselrolle für die Anfertigung der Werkstücke nimmt hierbei das Vorführen und Erklären (modellhaftes Vormachen) der notwendigen Fertigungsschritte durch Experten in Form von Erklärfilmen ein. Die zugrundeliegende didaktische Konzeption lehnt sich an die Kognitive Meisterlehre aus der gewerblich-technischen beruflich orientierten Bildung an (Collins et al., 1989). Das Projekt KunstHandWerk wurde durch eine Kooperation zwischen der PH Ludwigsburg und der PH Schwäbisch Gmünd in Zusammenarbeit mit der Gold- und Silberschmiede-Innung Stuttgart-Heilbronn-Reutlingen initiiert und von TRAFO - dem Netzwerk transferorientierter Lehre in Baden-Württemberg gefördert.
