    Ischemic preconditioning protects against cardiac ischemia reperfusion injury without affecting succinate accumulation or oxidation.

    Ischemia-reperfusion (IR) injury occurs when blood supply to an organ is disrupted and then restored, and underlies many disorders, notably myocardial infarction and stroke. While reperfusion of ischemic tissue is essential for survival, it also initiates cell death through generation of mitochondrial reactive oxygen species (ROS). Recent work has revealed a novel pathway underlying ROS production at reperfusion in vivo in which the accumulation of succinate during ischemia and its subsequent rapid oxidation at reperfusion drives ROS production at complex I by reverse electron transport (RET). Pharmacologically inhibiting ischemic succinate accumulation, or slowing succinate metabolism at reperfusion, have been shown to be cardioprotective against IR injury. Here, we determined whether ischemic preconditioning (IPC) contributes to cardioprotection by altering kinetics of succinate accumulation and oxidation during IR. Mice were subjected to a 30-minute occlusion of the left anterior descending coronary artery followed by reperfusion, with or without a protective IPC protocol prior to sustained ischemia. We found that IPC had no effect on ischemic succinate accumulation with both control and IPC mice having profound increases in succinate compared to normoxia. Furthermore, after only 1-minute reperfusion succinate was rapidly metabolised returning to near pre-ischemic levels in both groups. We conclude that IPC does not affect ischemic succinate accumulation, or its oxidation at reperfusion

    Needs and Problems of the Youth in Croatia: A Review of the Application of "Questionnaire of Problems"

    U okviru istraživanja “Potrebe i problemi mladih” provedenog u sklopu 11. Ljetne psihologijske škole na stratificiranom uzorku mladih Republike Hrvatske (n=1905) primijenjen je “Upitnik problema” kojim se ispitivala izraženost problema podijeljenih u 11 skupina - (1) briga za budućnost, (2) nedostatak društvenih sadržaja, (3) problemi vezani uz školovanje, (4) specifični problemi lokalne zajednice, (5) nemogućnost osamostaljenja, (6) nezadovoljstvo samim sobom, (7) problemi u intimnim odnosima, (8) egzistencijalna kriza, (9) problemi vezani uz ovisnosti, (10) problemi odnosa u obitelji i (11) nezadovoljstvo odnosima s vršnjacima. Najizraženijim skupinama problema na cjelokupnom uzorku pokazale su se sljedeće tri skupine: “briga za budućnost”, “nedostatak društvenih sadržaja” i “problemi vezani uz školovanje”. Premda je usporedba rezultata s obzirom na dob, spol, obrazovno usmjerenje, regionalnu podjelu i zahvaćenost ratom ukazala na određene razlike u izraženosti pojedinih skupina problema, utvrđeno je da niti jedna od navedenih varijabli nema bitniji utjecaj na hijerarhiju prioritetnih problema.Within the survey “Needs and Problems of the Youth” which was conducted during the 11th Summer School of Psychology on a stratified sample of young people in Croatia (n=1905), the “Questionnaire of Problems” was applied. The survey investigated the prominence of problems divided into 11 groups – (1) concern about future, (2) lack of social events and opportunities, (3) educational problems, (4) specific problems of local communities, (5) problems with gaining independence, (6) personal dissatisfaction, (7) problems with intimate relations, (8) existential crisis, (9) problems connected with addictions, (10) problems with family relations, and (11) dissatisfaction with peer relations. The sample presented the following three as the most prominent groups of problems: "concern about future”, “lack of social events and options” and “educational problems”. Although comparison of results with regard to age, sex, vocation, regional distribution and war experiences showed a variation in prominence of certain groups of problems, we found no evidence that the variables significantly affected the hierarchy of priority problems

    Archaeological microgravimetric prospection inside don church (Valencia, Spain)

    The microgravimetric surveying technique is applicable to the detection of shallow subsurface structures if a lateral density contrast is presented, and thus, it is a valid technique for archaeological prospection. In this paper, this technique has been revealed to be an efficient tool for archaeological studies, such as those performed inside Don Church (18th century), located in the urban area of Alfafar town, Valencia (Spain), where a buried crypt, suggested by different boreholes drilled during the second restoration process in 1993, is expected. Details of the site’s characteristics, topographic survey procedures, microgravimetric field operations, data collection and gravity reduction operations (where the inner building effect of walls, pillars and the altar is confirmed as one of the most important) are also presented.     Atividade antimicrobiana de compostos isolados e derivados semissintéticos de Miconia ferruginata

    This study evaluated the antimicrobial activity of isolated compounds and semisynthetic derivatives from Miconia ferruginata (Melastomataceae) against five microorganisms: Staphylococcus aureus (ATCC 25923), Escherichia coli (ATCC 25922), Bacillus subtilis (ATCC 6623), Pseudomonas aeruginosa (ATCC 15442), and Candida albicans (ATCC 10231). The isomeric mixture of ursolic and oleanolic acids was active against S. aureus (MIC = 250 μg mL-1) and against E. coli, B. subtilis, and P. aeruginosa (MIC = 500 μg mL-1). The flavone 5,6,7-trihydroxy-4’-methoxyflavone and the methyl esters, semisynthetic derivatives of a mixture of ursolic and oleanolic acids, showed no activity against the tested microorganisms. These results suggest that the carboxyl group present in the triterpenes may contribute to antimicrobial activity.Este estudo descreve a avaliação da atividade antimicrobiana de compostos isolados e derivados semissintéticos de Miconia ferruginata (Melastomataceae) contra cinco microorganismos: Staphylococcus aureus (ATCC 25923), Escherichia coli (ATCC 25922), Bacillus subtilis (ATCC 6623), Pseudomonas aeruginosa (ATCC 15442) and Candida albicans (ATCC 10231). A mistura isomérica dos ácidos ursólico e oleanólico foi ativa contra S. aureus (CIM = 250 μg mL-1) e contra E. coli, B. subtilis e P. aeruginosa (CIM = 500 μg mL-1). A flavona 5,6,7-trihidroxi-4’-metóxiflavona e os ésteres metílicos, derivados semissintéticos da mistura dos ácidos ursólico e oleanólico, não apresentaram atividade contra os microrganismos testados. Estes resultados sugerem que o grupo carboxila presente nos triterpenos podem contribuir para a atividade antimicrobiana

    Reduction of Tinnitus Severity by the Centrally Acting Muscle Relaxant Cyclobenzaprine: An Open-Label Pilot Study

    Tinnitus, the phantom perception of sounds, is a highly prevalent disorder. Although a wide variety of drugs have been investigated off label for the treatment of tinnitus, there is no approved pharmacotherapy. We report an open-label exploratory pilot study to assess the effect of muscle relaxants acting on the central nervous system on tinnitus patients. Cyclobenzaprine at high (30 mg) and low doses (10 mg), orphenadrine (100 mg), tizanidine (24 mg) and eperisone (50 mg) were administered to a maximum of 20 patients per group over a 12-week period. High-dose cyclobenzaprine resulted in a significant reduction in the Tinnitus Handicap Inventory (THI) score between baseline and week 12 in the intention-to-treat sample. On the other hand, other treatments were not effective. These results were confirmed in an explorative analysis where baseline corrected THI and Clinical Global Impression scores at week 12 were compared between groups. The present open trial presents a new promising pharmacotherapy for tinnitus that should be validated in placebo-controlled double-blind trials

    Low-level Laser Therapy to the Mouse Femur Enhances the Fungicidal Response of Neutrophils against Paracoccidioides brasiliensis

    Neutrophils (PMN) play a central role in host defense against the neglected fungal infection paracoccidioidomycosis (PCM), which is caused by the dimorphic fungus Paracoccidioides brasiliensis (Pb). PCM is of major importance, especially in Latin America, and its treatment relies on the use of antifungal drugs. However, the course of treatment is lengthy, leading to side effects and even development of fungal resistance. the goal of the study was to use low-level laser therapy (LLLT) to stimulate PMN to fight Pb in vivo. Swiss mice with subcutaneous air pouches were inoculated with a virulent strain of Pb or fungal cell wall components (Zymosan), and then received LLLT (780 nm; 50 mW; 12.5 J/cm2; 30 seconds per point, giving a total energy of 0.5 J per point) on alternate days at two points on each hind leg. the aim was to reach the bone marrow in the femur with light. Non-irradiated animals were used as controls. the number and viability of the PMN that migrated to the inoculation site was assessed, as well as their ability to synthesize proteins, produce reactive oxygen species (ROS) and their fungicidal activity. the highly pure PMN populations obtained after 10 days of infection were also subsequently cultured in the presence of Pb for trials of protein production, evaluation of mitochondrial activity, ROS production and quantification of viable fungi growth. PMN from mice that received LLLT were more active metabolically, had higher fungicidal activity against Pb in vivo and also in vitro. the kinetics of neutrophil protein production also correlated with a more activated state.     Genomic Analyses Show Extremely Perilous Conservation Status of African and Asiatic cheetahs (Acinonyx jubatus).

    We live in a world characterized by biodiversity loss and global environmental change. The extinction of large carnivores can have ramifying effects on ecosystems like an uncontrolled increase in wild herbivores, which in turn can have knock-on impacts on vegetation regeneration and communities. Cheetahs (Acinonyx jubatus) serve important ecosystem functions as apex predators; yet, they are quickly heading towards an uncertain future. Threatened by habitat loss, human-wildlife conflict and illegal trafficking, there are only approximately 7100 individuals remaining in nature. We present the most comprehensive genome-wide analysis of cheetah phylogeography and conservation genomics to date, assembling samples from nearly the entire current and past species' range. We show that their phylogeography is more complex than previously thought, and that East African cheetahs (A. j. raineyi) are genetically distinct from Southern African individuals (A. j. jubatus), warranting their recognition as a distinct subspecies. We found strong genetic differentiation between all classically recognized subspecies, thus refuting earlier findings that cheetahs show only little differentiation. The strongest differentiation was observed between the Asiatic and all the African subspecies. We detected high inbreeding in the Critically Endangered Iranian (A. j. venaticus) and North-western (A. j. hecki) subspecies, and show that overall cheetahs, along with snow leopards, have the lowest genome-wide heterozygosity of all the big cats. This further emphasizes the cheetah's perilous conservation status. Our results provide novel and important information on cheetah phylogeography that can support evidence-based conservation policy decisions to help protect this species. This is especially relevant in light of ongoing and proposed translocations across subspecies boundaries, and the increasing threats of illegal trafficking
