    Exponential sum approximations for t−βt^{-\beta}

    Given β>0\beta>0 and δ>0\delta>0, the function t−βt^{-\beta} may be approximated for tt in a compact interval [δ,T][\delta,T] by a sum of terms of the form we−atwe^{-at}, with parameters w>0w>0 and a>0a>0. One such an approximation, studied by Beylkin and Monz\'on, is obtained by applying the trapezoidal rule to an integral representation of t−βt^{-\beta}, after which Prony's method is applied to reduce the number of terms in the sum with essentially no loss of accuracy. We review this method, and then describe a similar approach based on an alternative integral representation. The main difference is that the new approach achieves much better results before the application of Prony's method; after applying Prony's method the performance of both is much the same.Comment: 18 pages, 5 figures.     A stability index for detonation waves in Majda's model for reacting flow

    Using Evans function techniques, we develop a stability index for weak and strong detonation waves analogous to that developed for shock waves in [GZ,BSZ], yielding useful necessary conditions for stability. Here, we carry out the analysis in the context of the Majda model, a simplified model for reacting flow; the method is extended to the full Navier-Stokes equations of reacting flow in [Ly,LyZ]. The resulting stability condition is satisfied for all nondegenerate, i.e., spatially exponentially decaying, weak and strong detonations of the Majda model in agreement with numerical experiments of [CMR] and analytical results of [Sz,LY] for a related model of Majda and Rosales. We discuss also the role in the ZND limit of degenerate, subalgebraically decaying weak detonation and (for a modified, ``bump-type'' ignition function) deflagration profiles, as discussed in [GS.1-2] for the full equations.Comment: 36 pages, 3 figure

    Heat fluxes under the ceiling induced by wall fires with various burner aspect ratios in a channel

    A detailed experimental investigation of wall fires in a channel was conducted to study the heat fluxes under the ceiling. Various burner aspect ratios and fire heat release rates were employed to simulate different wall fire scenarios. The effect of source-ceiling height was also examined. The results show that the distribution of heat flux under the ceiling from fires on rectangular burners was significantly influenced by the burner aspect ratio. As the burner aspect ratio increased, the heat flux under the ceiling at a given position perpendicular to the side wall increased. It was found that the existing heat flux correlation developed for a square burner could not capture such influence as it did not include the burner aspect ratio. A new predictive model based on the equivalent burner diameter concept was proposed incorporating the burner aspect ratio and was shown to predict well the heat flux for all the cases with different heat release rates, burner aspect ratios and source-ceiling heights. The model was also validated against available data in the literature which were not used in its derivation. Further analysis was also conducted for the temperature contours constructed from the temperature measurements under the ceiling

    Detected troponin elevation is associated with high early mortality after lung resection for cancer

    BACKGROUND: Myocardial infarction can be difficult to diagnose after lung surgery. As recent diagnostic criteria emphasize serum cardiac markers (in particular serum troponin) we set out to evaluate its clinical utility and to establish the long term prognostic impact of detected abnormal postoperative troponin levels after lung resection. METHODS: We studied a historic cohort of patients with primary lung cancer who underwent intended surgical resection. Patients were grouped according to known postoperative troponin status and survival calculated by Kaplan Meier method and compared using log rank. Parametric survival analysis was used to ascertain independent predictors of mortality. RESULTS: From 2001 to 2004, a total of 207 patients underwent lung resection for primary lung cancer of which 14 (7%) were identified with elevated serum troponin levels within 30 days of surgery, with 9 (64%) having classical features of myocardial infarction. The median time to follow up (interquartile range) was 22 (1 to 52) months, and the one and five year survival probabilities (95% CI) for patients without and with postoperative troponin elevation were 92% (85 to 96) versus 60% (31 to 80) and 61% (51 to 71) versus 18% (3 to 43) respectively (p < 0.001). T stage and postoperative troponin elevation remained independent predictors of mortality in the final multivariable model. The acceleration factor for death of elevated serum troponin after adjusting for tumour stage was 9.19 (95% CI 3.75 to 22.54). CONCLUSION: Patients with detected serum troponin elevation are at high risk of early mortality with or without symptoms of myocardial infarction after lung resection

    ADCC Develops Over Time during Persistent Infection with Live-Attenuated SIV and Is Associated with Complete Protection against SIV(mac)251 Challenge

    Live-attenuated strains of simian immunodeficiency virus (SIV) routinely confer apparent sterilizing immunity against pathogenic SIV challenge in rhesus macaques. Understanding the mechanisms of protection by live-attenuated SIV may provide important insights into the immune responses needed for protection against HIV-1. Here we investigated the development of antibodies that are functional against neutralization-resistant SIV challenge strains, and tested the hypothesis that these antibodies are associated with protection. In the absence of detectable neutralizing antibodies, Env-specific antibody-dependent cell-mediated cytotoxicity (ADCC) emerged by three weeks after inoculation with SIVDeltanef, increased progressively over time, and was proportional to SIVDeltanef replication. Persistent infection with SIVDeltanef elicited significantly higher ADCC titers than immunization with a non-persistent SIV strain that is limited to a single cycle of infection. ADCC titers were higher against viruses matched to the vaccine strain in Env, but were measurable against viruses expressing heterologous Env proteins. In two separate experiments, which took advantage of either the strain-specificity or the time-dependent maturation of immunity to overcome complete protection against SIV(mac)251 challenge, measures of ADCC activity were higher among the SIVDeltanef-inoculated macaques that remained uninfected than among those that became infected. These observations show that features of the antibody response elicited by SIVDeltanef are consistent with hallmarks of protection by live-attenuated SIV, and reveal an association between Env-specific antibodies that direct ADCC and apparent sterilizing protection by SIVDeltanef

    Evaluating the use of HILIC in large-scale, multi dimensional proteomics:horses for courses?

    AbstractDespite many recent advances in instrumentation, the sheer complexity of biological samples remains a major challenge in large-scale proteomics experiments, reflecting both the large number of protein isoforms and the wide dynamic range of their expression levels. However, while the dynamic range of expression levels for different components of the proteome is estimated to be ∼107–8, the equivalent dynamic range of LC–MS is currently limited to ∼106. Sample pre-fractionation has therefore become routinely used in large-scale proteomics to reduce sample complexity during MS analysis and thus alleviate the problem of ion suppression and undersampling. There is currently a wide range of chromatographic techniques that can be applied as a first dimension separation. Here, we systematically evaluated the use of hydrophilic interaction liquid chromatography (HILIC), in comparison with hSAX, as a first dimension for peptide fractionation in a bottom-up proteomics workflow. The data indicate that in addition to its role as a useful pre-enrichment method for PTM analysis, HILIC can provide a robust, orthogonal and high-resolution method for increasing the depth of proteome coverage in large-scale proteomics experiments. The data also indicate that the choice of using either HILIC, hSAX, or other methods, is best made taking into account the specific types of biological analyses being performed

    Application of a Sensitive and Reproducible Label-Free Proteomic Approach to Explore the Proteome of Individual Meiotic-Phase Barley Anthers

    <p>Meiosis is a highly dynamic and precisely regulated process of cell division, leading to the production of haploid gametes from one diploid parental cell. In the crop plant barley (Hordeum vulgare), male meiosis occurs in anthers, in specialized cells called meiocytes. Barley meiotic tissue is scarce and not easily accessible, making meiosis study a challenging task. We describe here a new micro-proteomics workflow that allows sensitive and reproducible genome-wide label-free proteomic analysis of individual staged barley anthers. This micro-proteomic approach detects more than 4,000 proteins from such small amounts of material as two individual anthers, covering a dynamic range of protein relative abundance levels across five orders of magnitude. We applied our micro-proteomics workflow to investigate the proteome of the developing barley anther containing pollen mother cells in the early stages of meiosis and we successfully identified 57 known and putative meiosis-related proteins. Meiotic proteins identified in our study were found to be key players of many steps and processes in early prophase such as: chromosome condensation, synapsis, DNA double-strand breaks or crossover formation. Considering the small amount of starting material, this work demonstrates an important technological advance in plant proteomics and can be applied for proteomic examination of many size-limited plant specimens. Moreover, it is the first insight into the proteome of individual barley anther at early meiosis.     A bootstrap method for sum-of-poles approximations

    A bootstrap method is presented for finding efficient sum-of-poles approximations of causal functions. The method is based on a recursive application of the nonlinear least squares optimization scheme developed in (Alpert et al. in SIAM J. Numer. Anal. 37:1138–1164, 2000), followed by the balanced truncation method for model reduction in computational control theory as a final optimization step. The method is expected to be useful for a fairly large class of causal functions encountered in engineering and applied physics. The performance of the method and its application to computational physics are illustrated via several numerical examples
