    New properties of a bioinspired pyridine benzimidazole compound as a novel differential staining agent for endoplasmic reticulum and Golgi apparatus in fluorescence live cell imaging

    Indexación: Scopus.In this study, we explored new properties of the bioinspired pyridine benzimidazole compound B2 (2,4-di-tert-butyl-6-(3H-imidazo[4,5-c]pyridine-2-yl)phenol) regarding its potential use as a differential biomarker. For that, we performed 1D 1HNMR (TOCSY), UV-Vis absorption spectra in different organic solvents, voltammetry profile (including a scan-rate study), and TD-DFT calculations that including NBO analyses, to provide valuable information about B2 structure and luminescence. In our study, we found that the B2 structure is highly stable, where the presence of an intramolecular hydrogen bond (IHB) seems to have a crucial role in the stability of luminescence, and its emission can be assigned as fluorescence. In fact, we found that the relatively large Stokes Shift observed for B2 (around 175 nm) may be attributed to the stability of the B2 geometry and the strength of its IHB. On the other hand, we determined that B2 is biocompatible by cytotoxicity experiments in HeLa cells, an epithelial cell line. Furthermore, in cellular assays we found that B2 could be internalized by passive diffusion in absence of artificial permeabilization at short incubation times (15 min to 30 min). Fluorescence microscopy studies confirmed that B2 accumulates in the endoplasmic reticulum (ER) and Golgi apparatus, two organelles involved in the secretory pathway. Finally, we determined that B2 exhibited no noticeable blinking or bleaching after 1 h of continuous exposure.     Fibroma cemento-osificante gingival mandibular: presentación de un caso

    Presentamos el caso clínico de una mujer con un fibroma cemento- osificante gigante dependiente de la encía mandibular de 20 años de evolución. El fibroma cemento-osificante es un tumor poco frecuente, clasificado dentro de las lesiones fibroóseas. Aparece generalmente en hueso y en raras ocasiones afecta a tejidos gingivales, como ocurre en nuestro caso. Es una neoplasia de crecimiento lento y bien delimitada, lo que le confiere un carácter benigno. Con los hallazgos histológicos es difícil diferenciar el fibroma cemento-osificante de otras lesiones tales como el osteoblastoma, el osteosarcoma de bajo grado y particularmente de la displasia fibrosa. Para obtener un diagnóstico preciso es fundamental guiarnos además por la información clínica y radiológica y así llevar a cabo un tratamiento oportuno, que condicione un pronóstico excelente.We report a case of a woman presenting a giant cementoossifying fibroma depending of the mandibular gingivae. The evolution of the process was 20 years. Cemento-ossifying fibroma is a relatively rare tumour classified between fibroosseous lesions. This lesion appears within the bone although in some occasions it involves the gingivae soft tissues, as the case we present. It is a slow-growing and well-defined tumorous lesion, because of this, it is considered as a benign lesion. The histologic findings alone may be similar to other pathologies such as osteoblastoma, low-grade osteosarcoma and particularly to fibrous dysplasia. An accurate diagnosis requires careful clinical, radiological and histological correlation in order to make an optimal treatment and an excellent outcome

    Ionic liquids as tribological performance improving additive for in-service and used fully-formulated diesel engine lubricants

    In recent years, several papers have been published that investigate the potential use of ionic liquids (ILs) as additives in lubricants. However, corrosive attack of ILs on lubricated metal surfaces and low miscibility of ILs in the non-polar oils are major obstacles to maintaining an optimum lubrication performance level. High miscibility and no corrosive behaviour of Trihexyltetradecyl phosphonium bis(2,4,4-tri-methylpentyl) phosphinate and Trihexyltetradecyl phosphonium bis(2-ethylhexyl) phosphate, as lubricating oil additives have recently been described in literature. This article presents work on using these phosphonium based ILs as an additive in the fully formulated diesel engine lubricants. This approach could allow the used lubricants to recover their tribological performance for further use at the end of service life. This extension of service life has the potential to generate significant economic and environmental benefits. Also it will add to the much needed knowledge about the effect of interaction between ILs and existing additives in engine-aged lubricants on the tribological performance of ring-liner tribo-system of diesel engines. Results revealed an improvement in friction and antiwear performance of used lubricant by addition of both ILs. However an increase in wear was noted for new (fresh) and in-service lubricant samples. An interesting interference between existing lubricant additives and added ILs in a boundary film formation process has been observed

    Growth and puberty in a 2-year open-label study of lisdexamfetamine dimesylate in children and adolescents with attention-deficit/hyperactivity disorder

    Background: Stimulant medications for the treatment of attention-deficit/hyperactivity disorder have a history of safe and effective use; however, concerns exist that they may adversely affect growth trajectories in children and adolescents. Objective: The objective of this study was to evaluate the longer-term effects of lisdexamfetamine dimesylate on weight, height, body mass index and pubertal development in children and adolescents with attention-deficit/hyperactivity disorder. Methods: Children and adolescents aged 6–17 years with attention-deficit/hyperactivity disorder took open-label lisdexamfetamine dimesylate (30, 50 or 70 mg/day) in this open-label 2-year safety and efficacy study. Safety evaluations included treatment-emergent adverse events, measurement of weight, height and body mass index, and selfreported pubertal status using Tanner staging. Results: The safety analysis population comprised all enrolled participants (N = 314) and 191 (60.8%) completed the study. Weight decrease was reported as a treatmentemergent adverse event in 63 participants (20.1%) and two participants (0.6%) discontinued the study as a result of treatment-emergent adverse events of weight decrease

    Promoting FAIRness in marine data at Centro Nacional Instituto Español de Oceanografía

    The Spanish Institute of Oceanography is responsible, among other aspects, for scienti c and technical advice for the Government's sheries policy as well as for the protection and sustainability of the marine environment. In this task, it generates a large amount of oceanographic data characterized by its spatial dispersion during acquisition as well as by its di erent typology. The purpose of both the National Oceanographic Data Center and the GIS team is to safeguard data and to disclose what data exists and where, how and when it has been acquired and, in addition, to provide access to that data through the collaboration with di erent international data infrastructures like EMODnet or SeaDataNet. To this end, the data and metadata are subjected to quality control and formatted for integration into a national Spatial Data Infrastructure (SDI). This SDI has a GeoNetwork catalogue with ~ 1750 oceanographic campaigns, together with (meta)data and services that are continuously being revised and incorporated. All this with the ultimate goal of making the data increasingly FAIR

    Efficacy of lisdexamfetamine dimesylate throughout the day in children and adolescents with attention-deficit/hyperactivity disorder:results from a randomized, controlled trial

    Lisdexamfetamine dimesylate (LDX) is a long-acting, prodrug stimulant therapy for patients with attention-deficit/hyperactivity disorder (ADHD). This randomized placebo-controlled trial of an optimized daily dose of LDX (30, 50 or 70 mg) was conducted in children and adolescents (aged 6–17 years) with ADHD. To evaluate the efficacy of LDX throughout the day, symptoms and behaviors of ADHD were evaluated using an abbreviated version of the Conners’ Parent Rating Scale-Revised (CPRS-R) at 1000, 1400 and 1800 hours following early morning dosing (0700 hours). Osmotic-release oral system methylphenidate (OROS-MPH) was included as a reference treatment, but the study was not designed to support a statistical comparison between LDX and OROS-MPH. The full analysis set comprised 317 patients (LDX, n = 104; placebo, n = 106; OROS-MPH, n = 107). At baseline, CPRS-R total scores were similar across treatment groups. At endpoint, differences (active treatment − placebo) in least squares (LS) mean change from baseline CPRS-R total scores were statistically significant (P < 0.001) throughout the day for LDX (effect sizes: 1000 hours, 1.42; 1400 hours, 1.41; 1800 hours, 1.30) and OROS-MPH (effect sizes: 1000 hours, 1.04; 1400 hours, 0.98; 1800 hours, 0.92). Differences in LS mean change from baseline to endpoint were statistically significant (P < 0.001) for both active treatments in all four subscales of the CPRS-R (ADHD index, oppositional, hyperactivity and cognitive). In conclusion, improvements relative to placebo in ADHD-related symptoms and behaviors in children and adolescents receiving a single morning dose of LDX or OROS-MPH were maintained throughout the day and were ongoing at the last measurement in the evening (1800 hours)
