    Insights: An exploration of experiences with schizophrenia and schizoaffective disorder

    This qualitative study explores the experiences of nine people with diagnoses of schizophrenia or schizoaffective disorder, living in the area of Guelph, Ontario. The stigma and poor prognosis traditionally attached to schizophrenia and schizoaffective disorder rage on. The profound disruption caused by the symptoms of schizophrenia and schizoaffective disorder and the social repercussions of receiving these diagnoses, contribute to the individual\u27s disconnection from family and community. Major alterations in the person\u27s social roles and sense of self follow. Stigma and discrimination compound the injury to self-esteem to the extent that a person internalizes them. Yet, however grim the situation appears, in all of the participants\u27 stories there is a sense of improvement in health toward recovery. Participants have reconnected with others, and are rebuilding their lives and sense of selves. In this thesis I find there are several possible roles for clinical and community development focused practitioners, including assisting people in accessing resources; encouraging people to connect with others; helping with skills to manage symptoms; and ending stigma and discrimination

    Canadian Arctic Tide Measurement Techniques and Results

    About 10 years ago the Canadian Hydrographic Service recognized the need for a planned approach to completing tide and current surveys of the Canadian Arctic Archipelago in order to meet the requirements of marine shipping and construction industries as well as the needs of environmental studies related to resource development. Therefore, a program of tidal surveys was begun which has resulted in a data base of tidal records covering most of the Archipelago. In this paper the problems faced by tidal surveyors and others working in the harsh Arctic environment are described and the variety of equipment and techniques developed for short, medium and long-term deployments are reported. The tidal characteristics throughout the Archipelago, determined primarily from these surveys, are briefly summarized. It was also recognized that there would be a need for real time tidal data by engineers, surveyors and mariners. Since the existing permanent tide gauges in the Arctic do not have this capability, a project was started in the early 1980’s to develop and construct a new permanent gauging system.     Abstract We reported previously that the core oligosaccharide region of the lipopolysaccharide (LPS) is essential for optimal adhesion of Actinobacillus pleuropneumoniae, an important swine pathogen, to respiratory tract cells. Rough LPS and core LPS mutants of A. pleuropneumoniae serotype 1 were generated by using a mini-Tn10 transposon mutagenesis system. Here we performed a structural analysis of the oligosaccharide region of three core LPS mutants that still produce the same O-antigen by using methylation analyses and mass spectrometry. We also performed a kinetic study of proinflammatory cytokines production such as interleukin (IL)-6, tumor necrosis factor-α, IL1-β, MCP-1, and IL8 by LPS-stimulated porcine alveolar macrophages, which showed that purified LPS of the parent strain, the rough LPS and core LPS mutants, had the same ability to stimulate the production of cytokines. Most interestingly, an in vitro susceptibility test of these LPS mutants to antimicrobial peptides showed that the three core LPS mutants were more susceptible to cationic peptides than both the rough LPS mutant and the wild type parent strain. Furthermore, experimental pig infections with these mutants revealed that the galactose (Gal I) and d,d-heptose (Hep IV) residues present in the outer core of A. pleuropneumoniae serotype 1 LPS are important for adhesion and overall virulence in the natural host, whereas deletion of the terminal GalNAc-Gal II disaccharide had no effect. Our data suggest that an intact core-lipid A region is required for optimal protection of A. pleuropneumoniae against cationic peptides and that deletion of specific residues in the outer LPS core results in the attenuation of the virulence of A. pleuropneumoniae serotype 1

    Stormwater Pond Maintenance, and Wetland Management for Phosphorus Retention

    (c) 1036202Reduction in phosphorus is critical because phosphate, a dissolved form of phosphorus, sustains algal and cyanobacteria growth and causes a wide range of water-quality impairments in the ponds and downstream waters including algal blooms, excess floating plants, taste, and odor problems. Many stormwater ponds and wetlands that treat stormwater appear to be less effective than expected or originally intended in phosphorus retention, a key function of these ponds in urban environments. There is evidence that many old ponds are releasing phosphorus from bottom sediments at high rates and likely exporting phosphorus to downstream surface water bodies. A major outcome of this project is a pond Assessment Tool to assess the risk of high phosphorus concentrations in ponds and sediment release of phosphorus. The tool is based on 20 ponds with detailed water quality and phosphorus release measurements and a meta-analysis of 230 ponds in the Twin Cities metro area. Other outcomes included a working definition of a constructed stormwater pond and a wetland treating stormwater in the framework of water-body regulations, the development of recommendations for stormwater pond maintenance and wetland management, and an update to the sections on the constructed stormwater ponds section of the 2009 Stormwater Maintenance BMP Guide

    Elemental spatial and temporal association formation in left temporal lobe epilepsy

    The mesial temporal lobe (MTL) is typically understood as a memory structure in clinical settings, with the sine qua non of MTL damage in epilepsy being memory impairment. Recent models, however, understand memory as one of a number of higher cognitive functions that recruit the MTL through their reliance on more fundamental processes, such as “self-projection” or “association formation”. We examined how damage to the left MTL influences these fundamental processes through the encoding of elemental spatial and temporal associations. We used a novel fMRI task to image the encoding of simple visual stimuli, either rich or impoverished, in spatial or spatial plus temporal information. Participants included 14 typical adults (36.4 years, sd. 10.5 years) and 14 patients with left mesial temporal lobe damage as evidenced by a clinical diagnosis of left temporal lobe epilepsy (TLE) and left MTL impairment on imaging (34.3 years, sd. 6.6 years). In-scanner behavioral performance was equivalent across groups. In the typical group whole-brain analysis revealed highly significant bilateral parahippocampal activation (right > left) during spatial associative processing and left hippocampal/parahippocampal deactivation in joint spatial-temporal associative processing. In the left TLE group identical analyses indicated patients used MTL structures contralateral to the seizure focus differently and relied on extra-MTL regions to a greater extent. These results are consistent with the notion that epileptogenic MTL damage is followed by reorganization of networks underlying elemental associative processes. In addition, they provide further evidence that task-related fMRI deactivation can meaningfully index brain function. The implications of these findings for clinical and cognitive neuropsychological models of MTL function in TLE are discussed

    The Insertion and Transport of Anandamide in Synthetic Lipid Membranes Are Both Cholesterol-Dependent

    International audienceBackground: Anandamide is a lipid neurotransmitter which belongs to a class of molecules termed the endocannabinoids involved in multiple physiological functions. Anandamide is readily taken up into cells, but there is considerable controversy as to the nature of this transport process (passive diffusion through the lipid bilayer vs. involvement of putative proteic transporters). This issue is of major importance since anandamide transport through the plasma membrane is crucial for its biological activity and intracellular degradation. The aim of the present study was to evaluate the involvement of cholesterol in membrane uptake and transport of anandamide.Methodology/Principal Findings: Molecular modeling simulations suggested that anandamide can adopt a shape that is remarkably complementary to cholesterol. Physicochemical studies showed that in the nanomolar concentration range, anandamide strongly interacted with cholesterol monolayers at the air-water interface. The specificity of this interaction was assessed by: i) the lack of activity of structurally related unsaturated fatty acids (oleic acid and arachidonic acid at 50 nM) on cholesterol monolayers, and ii) the weak insertion of anandamide into phosphatidylcholine or sphingomyelin monolayers. In agreement with these data, the presence of cholesterol in reconstituted planar lipid bilayers triggered the stable insertion of anandamide detected as an increase in bilayer capacitance. Kinetics transport studies showed that pure phosphatidylcholine bilayers were weakly permeable to anandamide. The incorporation of cholesterol in phosphatidylcholine bilayers dose-dependently stimulated the translocation of anandamide.Conclusions/Significance: Our results demonstrate that cholesterol stimulates both the insertion of anandamide into synthetic lipid monolayers and bilayers, and its transport across bilayer membranes. In this respect, we suggest that besides putative anandamide protein-transporters, cholesterol could be an important component of the anandamide transport machinery. Finally, this study provides a mechanistic explanation for the key regulatory activity played by membrane cholesterol in the responsiveness of cells to anandamide

    Observation of Orbitally Excited B_s Mesons

    We report the first observation of two narrow resonances consistent with states of orbitally excited (L=1) B_s mesons using 1 fb^{-1} of ppbar collisions at sqrt{s} = 1.96 TeV collected with the CDF II detector at the Fermilab Tevatron. We use two-body decays into K^- and B^+ mesons reconstructed as B^+ \to J/\psi K^+, J/\psi \to \mu^+ \mu^- or B^+ \to \bar{D}^0 \pi^+, \bar{D}^0 \to K^+ \pi^-. We deduce the masses of the two states to be m(B_{s1}) = 5829.4 +- 0.7 MeV/c^2 and m(B_{s2}^*) = 5839.7 +- 0.7 MeV/c^2.Comment: Version accepted and published by Phys. Rev. Let

    Transcriptional regulation of bone formation by the osteoblast-specific transcription factor Osx

    Bone formation is a complex developmental process involving the differentiation of mesenchymal stem cells to osteoblasts. Osteoblast differentiation occurs through a multi-step molecular pathway regulated by different transcription factors and signaling proteins. Osx (also known as Sp7) is the only osteoblast-specific transcriptional factor identified so far which is required for osteoblast differentiation and bone formation. Osx knock-out mice lack bone completely and cartilage is normal. This opens a new window to the whole research field of bone formation. Osx inhibits Wnt pathway signaling, a possible mechanism for Osx to inhibit osteoblast proliferation. These reports demonstrate that Osx is the master gene that controls osteoblast lineage commitment and the subsequent osteoblast proliferation and differentiation. This review is to highlight recent progress in understanding the molecular mechanisms of transcriptional regulation of bone formation by Osx

    Limitations of Water Resources Infrastructure for Reducing Community Vulnerabilities to Extremes and Uncertainty of Flood and Drought

    Debate and deliberation surrounding climate change has shifted from mitigation toward adaptation, with much of the adaptation focus centered on adaptive practices, and infrastructure development. However, there is little research assessing expected impacts, potential benefits, and design challenges that exist for reducing vulnerability to expected climate impacts. The uncertainty of design requirements and associated government policies, and social structures that reflect observed and projected changes in the intensity, duration, and frequency of water-related climate events leaves communities vulnerable to the negative impacts of potential flood and drought. The results of international research into how agricultural infrastructure features in current and planned adaptive capacity of rural communities in Argentina, Canada, and Colombia indicate that extreme hydroclimatic events, as well as climate variability and unpredictability are important for understanding and responding to community vulnerability. The research outcomes clearly identify the need to deliberately plan, coordinate, and implement infrastructures that support community resiliency.Fil: McMartin, Dena W..
