Muito mais do que um jogo : competências psicológicas para a optimização do desempenho desportivo em jogadores de ténis
Tese de mestrado, Psicologia (Psicologia Clínica e da Saúde - Núcleo de Psicoterapia Cognitiva-Comportamental e Integrativa), 2008, Universidade de Lisboa, Faculdade de Psicologia e de Ciências da EducaçãoO presente trabalho teve como objectivo a criação de um programa de optimização do desempenho desportivo em jogadores de ténis que sejam elegíveis para competir a nível nacional, recorrendo ao desenvolvimento de competências psicológicas. Foi realizada uma revisão de literatura que abrangeu os estudos em que se procuraram identificar as competências fundamentais para a optimização do desempenho. As competências identificadas foram divididas em dois grupos hierarquicamente distintos: as competências avançadas que compreendem a auto- confiança, motivação, regulação da ansiedade, concentração; e as competências básicas que incluem estratégias de regulação da excitação e activação, estabelecimento de objectivos, imagética e auto-verbalizações. O programa foi desenvolvido: a) tendo em conta programas anteriormente realizados com jogadores de ténis e com o objectivo de optimização do desempenho e b) baseando-se nos pressupostos teóricos, modelos e estratégias relacionados com as competências avançadas referidas, recorrendo, para a sua concretização, à implementação do treino de competências básicas.This work aimed to create a mental training program for performance optimization for tennis players that are eligible to compete at the national level. A literature review was provided. This one includes studies in which they sought to identify the essential skills for optimal performance. The skills identified were divided into two hierarchical groups: the advanced skills that embraced self confidence, motivation, anxiety management, concentration; and the basic skills that include arousal regulation strategies, goal-setting, imagery and self-talk. The program was developed: a) taking into account earlier programs carried out with tennis players and with the aim of optimizing the performance and b) based on theoretical assumptions, models and strategies related to these advanced skills, using the implementation of basic skills training for its accomplishment

DNRF107The large family of polypeptide GalNAc-transferases (GalNAc-Ts) controls with precision how GalNAc O-glycans are added in the tandem repeat regions of mucins (e.g., MUC1). However, the structural features behind the creation of well-defined and clustered patterns of O-glycans in mucins are poorly understood. In this context, herein, we disclose the full process of MUC1 O-glycosylation by GalNAc-T2/T3/T4 isoforms by NMR spectroscopy assisted by molecular modeling protocols. By using MUC1, with four tandem repeat domains as a substrate, we confirmed the glycosylation preferences of different GalNAc-Ts isoforms and highlighted the importance of the lectin domain in the glycosylation site selection after the addition of the first GalNAc residue. In a glycosylated substrate, with yet multiple acceptor sites, the lectin domain contributes to orientate acceptor sites to the catalytic domain. Our experiments suggest that during this process, neighboring tandem repeats are critical for further glycosylation of acceptor sites by GalNAc-T2/T4 in a lectin-assisted manner. Our studies also show local conformational changes in the peptide backbone during incorporation of GalNAc residues, which might explain GalNAc-T2/T3/T4 fine specificities toward the MUC1 substrate. Interestingly, we postulate that a specific salt-bridge and the inverse γ-turn conformation of the PDTRP sequence in MUC1 are the main structural motifs behind the GalNAc-T4 specificity toward this region. In addition, in-cell analysis shows that the GalNAc-T4 isoform is the only isoform glycosylating the Thr of the immunogenic epitope PDTRP in vivo, which highlights the relevance of GalNAc-T4 in the glycosylation of this epitope. Finally, the NMR methodology established herein can be extended to other glycosyltransferases, such as C1GalT1 and ST6GalNAc-I, to determine the specificity toward complex mucin acceptor substrates.publishersversionepub_ahead_of_prin
Detection of anti-infliximab antibodies is impacted by antibody titer, infliximab level and IgG4 antibodies: a systematic comparison of three different assays
Background: There is scant information on the accuracy of different assays used to measure anti-infliximab antibodies (ADAs), especially in the presence of detectable infliximab (IFX). We thus aimed to evaluate and compare three different assays for the detection of IFX and ADAs and to clarify the impact of the presence of circulating IFX on the accuracy of the ADA assays.Methods: Blood samples from 79 ulcerative colitis (UC) patients treated with infliximab were assessed for IFX levels and ADAs using three different assays: an in-house assay and two commercial kits, Immundiagnostik and Theradiag. Sera samples with ADAs and undetectable levels of IFX were spiked with exogenous IFX and analyzed for ADAs.Results: The three assays showed 81-96% agreement for the measured IFX level. However, the in-house assay and Immundiagnostik assays detected ADAs in 34 out of 79 samples, whereas Theradiag only detected ADAs in 24 samples. Samples negative for ADAs with Theradiag, but ADA-positive in both the in-house and Immundiagnostik assays, were positive for IFX or IgG4 ADAs. In spiking experiments, a low concentration of exogenous IFX (5 mu g/ml) hampered ADA detection with Theradiag in sera samples with ADA levels of between 3 and 10 mu g/ml. In the Immundiagnostik assay detection interference was only observed at concentrations of exogenous IFX higher than 30 mu g/ml. However, in samples with high levels of ADAs (> 25 mu g/ml) interference was only observed at IFX concentrations higher than 100 mu g/ml in all three assays. Binary (IFX/ADA) stratification of the results showed that IFX+/ADA and IFX-/ADAs + were less influenced by the assay results than the double-positive (IFX+/ADAs+) and double-negative (IFX-/ADAs-) combination.Conclusions: All three methodologies are equally suitable for measuring IFX levels. However, erroneous therapeutic decisions may occur when patients show double-negative (IFX-/ADAs) or double-positive (IFX+/ADAs+) status, since agreement between assays is significantly lower in these circumstances

Background: to evaluate the effects of one week of supplementation with curcumin combined with piperine on physical performance, immune system cell counts, muscle damage, and plasma levels of inflammatory markers after a treadmill running training session. Methods: This study is a double-blind, crossover-balanced clinical trial with a three-week intervention. Sixteen male runners with a mean age of 36 ± 9 years and VO2 max of 60.6 ± 9.03 mL.kg −1 min −1 were recruited and randomly divided into 2 groups: the first group (CPG) was supplemented daily for 7 days with 500 mg of curcumin + 20 mg piperine, and the second group (PG) was supplemented with 540 mg of cellulose. After the 7th day of supplementation, the volunteers participated in the
experimental running protocol, where blood samples were collected before, after, and one hour after exercise for analysis of the number of leukocytes, creatine kinase, and cytokine concentration (IL-2, TNF-α, IFN, IL-6, and IL-10) using flow cytometry. This process was repeated, reversing the supplementation offered to the groups. Results: curcumin and piperine supplementation could not change the physical performance, immune cell counts, and muscle damage; however, the aerobic fatiguing exercise protocol inhibited the elevation of the plasmatic levels of some cytokines. The running exercise protocol could elevate the circulating levels of IL-2 (from 49.7 to 59.3 pg/mL), TNF-α (from 48.5 to 51.5 pg/mL), INF (from 128.8 to 165.0 pg/mL), IL-6 (from 63.1 to 77.3 pg/mL), and IL-10 (from 48.9 to 59.6 pg/mL) 1 h after the end of the running protocol. However, the curcumin and piperine supplementation could inhibit this elevation. Conclusions: curcumin and piperine supplementation had no effect on physical performance, immune cell counts, or muscle damage; however, the supplementation could modulate the kinetics of IL-2, TNF-α, INF, IL-6, and IL-10 1 h after the end of exercise.9E1A-F9DD-3EB8 | Filipe Manuel ClementeN/

Circulation and Transport Processes during an Extreme Freshwater Discharge Event at the Tagus Estuary
During the winter of 2013, the Tagus estuary was under the influence of intense winds and extreme freshwater discharge that changed its hydrodynamics and, consequently, the salt and heat transport. Moreover, the dynamics of the estuary may change due to climate change which will increase the frequency of heat waves and increase the mean sea level. Therefore, it is of utmost importance to study the impact of the future increase in air temperature and mean sea level under extreme events, such as that in the winter of 2013, to ascertain the foreseen changes in water properties transport within the estuary and near coastal zone. Several scenarios were developed and explored, using the Delft3D model suite, considering the results of the CMIP6 report as forcing conditions. Before the event, the mixing region of the estuary presented well-mixed conditions and its marine area a slight stratification. During the event, the estuary was filled with freshwater and the mixing region migrated toward the coast, leading to lower water temperature values inside the estuary. SLR has a higher impact on the salinity and stratification patterns than the air temperature increase. The response of water temperature is directly related to the increase in air temperature. The estuary mouth and the shallow regions will be more prone to changes than the upstream region of the estuary. The projected changes are directly linked to the future CO2 emissions scenarios, being intensive with the highest emission scenario

O principal objetivo deste paper prende-se com uma análise da presença de terrorismo de estado na Prisão de Guantánamo, consistindo numa violação dos direitos humanos, sob a lente da Segurança Humana. Para esse fim, foi elaborada a seguinte pergunta de investigação que servirá de auxílio para conduzir a investigação:
De que forma o terrorismo de Estado na prisão de Guantánamo constituiu uma violação dos direitos humanos?
Foi ainda elaborada uma delimitação temporal com início em 2002, após a criação da prisão, e cujo término será
em 2022 para que seja possível fazer um paralelo com a atualidade do status deste estabelecimento prisional.Na sequência da Resolução 1325 (2000) das Nações Unidas sobre Mulheres, Paz e Segurança (MPS), que apelou aos Estados para integrarem uma perspetiva de género nas suas políticas de segurança e construção da paz, a União Europeia (UE) tem dados alguns passos no sentido de integrar as questões de género na sua política externa e de segurança. Apesar destes desenvolvimentos empíricos, a literatura sobre segurança no espaço europeu não tem prestado a atenção devida aos processos através dos quais as questões de género e a agenda MPS podem contribuir para a construção da UE como um ator global. Este artigo procura aprofundar o debate sobre global actorness numa perspetiva de género, tendo em conta as seguintes questões: (1) que dimensões são relevantes para conceptualizar a UE como um actor global? (2) como é que a igualdade de género e a agenda MPS estão integradas na política externa e de segurança da UE? (3) como é que a UE moldou e promoveu a agenda MPS na arena global? Para responder a estas questões, partimos do modelo conceptual de global actorness de Bretherton e Vogler (2006) e na literatura sobre as dimensões de género da política externa e de segurança da União Europeia.Improvement of security governance on climate security is critical if fast-paced threats caused by the emission
of greenhouse gases are to be dealt with. This article envisions climate security as a global public good, moving the debate beyond traditional security discussions and refocusing attention upon climate security governance for its effective provision. Additionally, having in mind the recognition granted to sub-state authorities under the Paris Agreement, it suggests that the provision of security governance in matters of climate change requires a multilevel governance (MLG) approach whereby sub-state actors become inescapable agents of climate security governance. In doing so, it is our wish to move the debate forward in two complementary ways: first, by expanding the notion of security in order to perceive climate security as a global public good and second, by shedding light on the reinforced role of sub-state authorities under the Paris agreement using the framework of MLGEste artigo analisa a noção de ataque constante do art.º 49.º do I PA às CG e a sua adequabilidade em relação às ciberoperações militares. também de ameaças e ataques. A cibersegurança dos Estados pode-se encontrar em risco, na falta de formação pertinente, recursos financeiros ou cooperação eficaz.
O presente trabalho analisa de que maneira Portugal se encontra preparado para enfrentar as ameaças cibernéticas. Para isso, são apresentadas as principais tendências em cibercriminalidade em Portugal e os mecanismos de lidar com os desafios. Sugere-se que os ciberincidentes agravam as vulnerabilidades estatais, criam alguma desconfiança nas instituições nacionais. Porém, simultaneamente, impulsionam os avanços tecnológicos em ciberproteção.
