Zukunftsstrategien für eine dynamische Westschweizer Landwirtschaft
Die Westschweizer Landwirtschaft ist durch ihre grosse Diversität gekennzeichnet und umfasst in dieser Studie die Kantone, in denen Französisch gesprochen wird (Waadt, Genf, Neuenburg, Wallis, Freiburg, Jura) sowie den Bezirk des Berner Juras. Das Ziel ist, Zukunftsstrategien mit einer Gesamtsicht für diese Landwirtschaft zu erarbeiten. In einem partizipativen Prozess mit den involvierten Akteuren wurde zuerst die Ist-Situation der Land- und Ernährungswirtschaft analysiert um anschliessend Handlungsstrategien für die zukünftige Positionierung der Westschweizer Landwirtschaftsorganisation AGORA abzuleiten. Auf nur vier der 20 untersuchten Fallbeispielbetriebe wird das landwirtschaftliche Einkommen bis 2022 steigen, und zwar mit der Strategie des Flächenwachstums und ohne Milchproduktion. Das Weiterführen der aktuellen Situation ohne Veränderung (Status quo) würde für die 20 Betriebe eine Verschlechterung der finanziellen Situation bedeuten. Die strategische Planung inklusive der Investitionsplanung ist daher von grosser Bedeutung und sollte in Zukunft mehr gefördert werden. Die Westschweiz verfügt zwar über gut entwickelte und etablierte Regionalprodukte (z. Bsp. AOP/IGP) und Wertschöpfungsketten, jedoch könnten die Ausrichtung der Produktion und der Verarbeitung noch gezielter auf die Konsumentenbedürfnisse abgestimmt werden und die Produzenten vermehrt und tiefer in die Wertschöpfungsketten integriert werden, besonders im Industriemilchsektor

Extramitochondrial OPA1 and adrenocortical function
We have previously described that silencing of the mitochondrial protein OPA1 enhances mitochondrial 27
Ca2+ signaling and aldosterone production in H295R adrenocortical cells. Since extramitochondrial OPA1 28
(emOPA1) was reported to facilitate cAMP-induced lipolysis, we hypothesized that emOPA1, via the 29
enhanced hydrolysis of cholesterol esters, augments aldosterone production in H295R cells. A few 30
OPA1 immunopositive spots were detected in �40% of the cells. In cell fractionation studies OPA1/COX 31
IV (mitochondrial marker) ratio in the post-mitochondrial fractions was an order of magnitude higher 32
than that in the mitochondrial fraction. The ratio of long to short OPA1 isoforms was lower in post-mito- 33
chondrial than in mitochondrial fractions. Knockdown of OPA1 failed to reduce db-cAMP-induced phos- 34
phorylation of hormone-sensitive lipase (HSL), Ca2+ signaling and aldosterone secretion. In conclusion, 35
OPA1 could be detected in the post-mitochondrial fractions, nevertheless, OPA1 did not interfere with 36
the cAMP – PKA – HSL mediated activation of aldosterone secretio
dSETDB1 and SU(VAR)3–9 Sequentially Function during Germline-Stem Cell Differentiation in Drosophila melanogaster
Germline-stem cells (GSCs) produce gametes and are thus true “immortal stem cells”. In Drosophila ovaries, GSCs divide asymmetrically to produce daughter GSCs and cystoblasts, and the latter differentiate into germline cysts. Here we show that the histone-lysine methyltransferase dSETDB1, located in pericentric heterochromatin, catalyzes H3-K9 trimethylation in GSCs and their immediate descendants. As germline cysts differentiate into egg chambers, the dSETDB1 function is gradually taken over by another H3-K9-specific methyltransferase, SU(VAR)3–9. Loss-of-function mutations in dsetdb1 or Su(var)3–9 abolish both H3K9me3 and heterochromatin protein-1 (HP1) signals from the anterior germarium and the developing egg chambers, respectively, and cause localization of H3K9me3 away from DNA-dense regions in most posterior germarium cells. These results indicate that dSETDB1 and SU(VAR)3–9 act together with distinct roles during oogenesis, with dsetdb1 being of particular importance due to its GSC-specific function and more severe mutant phenotype
Silent chromatin at the middle and ends: lessons from yeasts
Eukaryotic centromeres and telomeres are specialized chromosomal regions that share one common characteristic: their underlying DNA sequences are assembled into heritably repressed chromatin. Silent chromatin in budding and fission yeast is composed of fundamentally divergent proteins tat assemble very different chromatin structures. However, the ultimate behaviour of silent chromatin and the pathways that assemble it seem strikingly similar among Saccharomyces cerevisiae (S. cerevisiae), Schizosaccharomyces pombe (S. pombe) and other eukaryotes. Thus, studies in both yeasts have been instrumental in dissecting the mechanisms that establish and maintain silent chromatin in eukaryotes, contributing substantially to our understanding of epigenetic processes.
