    Predicting leptonic CP violation in the light of Daya Bay result

    In the light of the recent Daya Bay result the reactor angle is about 9 degrees, we reconsider the model presented in arXiv:1005.3482 showing that, when all neutrino oscillation parameters are taken at their best fit values of Schwetz et al and the reactor angle to be the central value of Daya Bay, the predicted value of the CP phase is approximately 45 degrees.Comment: 4 pages, 2 figures, update of arXiv:1005.348

    Quark-lepton mass relation and CKM mixing in an A(4) extension of the minimal supersymmetric standard model

    An interesting mass relation between down-type quarks and charged leptons has been recently predicted within a supersymmetric SU(3)(c) circle times SU(2)(L) circle times U(1)(Y) model based on the A(4) flavor symmetry. Here we propose a simple extension which provides an adequate full description of the quark sector. By adding a pair of vectorlike up quarks, we show how the CKM entries V-ub, V-cb, V-td and V-ts arise from deviations of the unitarity. We perform an analysis including the most relevant observables in the quark sector, such as oscillations and rare decays of kaons, B-d and B-s mesons. In the lepton sector, the model predicts an inverted hierarchy for the neutrino masses, leading to a potentially observable rate of neutrinoless double beta decay

    Teaching argumentative synthesis writing through deliberative dialogues: instructional practices in secondary education

    Dialogical argumentation practice contributes positively to argumentative writing skills. Specifically, deliberative dialogues are effective in promoting argument and counterargument integration in students' essays. However, the potential of dialogic activities may be increased if they are combined with instructional practices. The primary objective of this research is to compare the impact of four intervention programs, aimed at improving argumentative synthesis writing from conflicting sources. The four programs resulted from the combination of two instructional components (Explicit Instruction through video modelling—EI, or a Procedural Guideline—G), while Deliberative Dialogues—DD—were a constant element. We conducted a pre-post quasi-experimental study in which 186 Spanish third grade secondary school students (aged 14–15) participated. We evaluated the quality of the syntheses by examining the level of argumentative coverage (the total number of arguments included in the synthesis) and the level of integration (the type and frequency of the argumentative strategies used in the syntheses). The results showed that the effectiveness of the instructional methods varies according to the synthesis quality indicator. Explicit instruction, in combination with deliberative dialogues, was especially helpful in improving the level of integration of syntheses. The procedural guideline, in combination with deliberative dialogues, contributed significantly to the coverage of arguments. The combination of these two elements did not favor the writing of synthesis as expected, probably due to the conditions in which the intervention was carried out. The findings of this study revealed that the coverage of arguments and integration processes are of different nature, follow different learning paths and require different instructional processes.pre-print875 K

    Multistep Fractionation of Microalgal Biomolecules Using Selective Aqueous Two-Phase Systems

    We aim to develop liquid-liquid extraction processes for the fractionation of microalgal components (proteins, pigments, lipids, and carbohydrates). The partitioning behavior of microalgal pigments and proteins in aqueous two-phase systems (ATPS) composed of the polymer polypropylene glycol with molecular weight 400 (PPG 400) + various cholinium based-ionic liquids was studied. A process for fractionation of multiple components from disrupted Neochloris oleoabundans was developed and evaluated. Results show that cholinium dihydrogen phosphate (Ch DHp) allows the fractionation of pigments in the PPG 400-rich phase and proteins in the Ch DHp-rich phase with high selectivity. It was demonstrated that a multiproduct approach can fractionate free glucose, and proteins in the ionic liquid-rich phase, pigments in the polymer-rich phase, while starch and lipids are recovered at the interface.</p

    Analysis of BH3-only proteins upregulated in response to oxygen/glucose deprivation in cortical neurons identifies Bmf but not Noxa as potential mediator of neuronal injury

    Stress signaling in response to oxygen/glucose deprivation (OGD) and ischemic injury activates a group of pro-apoptotic genes, the Bcl-2 homology domain 3 (BH3)-only proteins, which are capable of activating the mitochondrial apoptosis pathway. Targeted studies previously identified the BH3-only proteins Puma, Bim and Bid to have a role in ischemic/hypoxic neuronal injury. We here investigated the transcriptional activation of pro-apoptotic BH3-only proteins after OGD-induced injury in murine neocortical neurons. We observed a potent and early upregulation of noxa at mRNA and protein level, and a significant increase in Bmf protein levels during OGD in neocortical neurons and in the ipsilateral cortex of mice subjected to transient middle cerebral artery occlusion (tMCAO). Surprisingly, gene deficiency in noxa reduced neither OGD- nor glutamate-induced neuronal injury in cortical neurons and failed to influence infarct size or neurological deficits after tMCAO. In contrast, bmf deficiency induced significant protection against OGD- or glutamate-induced injury in cultured neurons, and bmf-deficient mice showed reduced neurological deficits after tMCAO in vivo. Collectively, our data not only point to a role of Bmf as a BH3-only protein contributing to excitotoxic and ischemic neuronal injury but also demonstrate that the early and potent induction of noxa does not influence ischemic neuronal injury

    Vascular endothelial growth factor as a non-invasive marker of pulmonary vascular remodeling in patients with bronchitis-type of COPD

    BACKGROUND: Several studies have indicated that one of the most potent mediators involved in pulmonary vascular remodeling is vascular endothelial growth factor (VEGF). This study was designed to determine whether airway VEGF level reflects pulmonary vascular remodeling in patients with bronchitis-type of COPD. METHODS: VEGF levels in induced sputum were examined in 23 control subjects (12 non-smokers and 11 ex-smokers) and 29 patients with bronchitis-type of COPD. All bronchitis-type patients performed exercise testing with right heart catheterization. RESULTS: The mean pulmonary arterial pressure (mPAP) and pulmonary vascular resistance (PVR) after exercise were markedly increased in all bronchitis-type patients. However, both parameters after exercise with breathing of oxygen was significantly lower than in those with breathing of room air. To attenuate the effect of hypoxia-induced pulmonary vasoconstriction during exercise, we used the change in mPAP or PVR during exercise with breathing of oxygen as a parameter of pulmonary vascular remodeling. Change in mPAP was significantly correlated with VEGF level in induced sputum from patients with chronic bronchitis (r = 0.73, p = 0.0001). Moreover, change in PVR was also correlated with VEGF level in those patients (r = 0.57, p = 0.003). CONCLUSION: A close correlation between magnitude of pulmonary hypertension with exercise and VEGF level in bronchitis-type patients could be observed. Therefore, these findings suggest the possibility that VEGF level in induced sputum is a non-invasive marker of pulmonary vascular remodeling in patients with bronchitis-type of COPD

    Effects of cigarette smoke on endothelial function of pulmonary arteries in the guinea pig

    <p>Abstract</p> <p>Background</p> <p>Cigarette smoking may contribute to pulmonary hypertension in chronic obstructive pulmonary disease by altering the structure and function of pulmonary vessels at early disease stages. The objectives of this study were to evaluate the effects of long-term exposure to cigarette smoke on endothelial function and smooth muscle-cell proliferation in pulmonary arteries of guinea pigs.</p> <p>Methods</p> <p>19 male Hartley guinea pigs were exposed to the smoke of 7 cigarettes/day, 5 days/week, for 3 and 6 months. 17 control guinea pigs were sham-exposed for the same periods. Endothelial function was evaluated in rings of pulmonary artery and aorta as the relaxation induced by ADP. The proliferation of smooth muscle cells and their phenotype in small pulmonary vessels were evaluated by immunohistochemical expression of α-actin and desmin. Vessel wall thickness, arteriolar muscularization and emphysema were assessed morphometrically. The expression of endothelial nitric oxide synthase (eNOS) was evaluated by Real Time-PCR.</p> <p>Results</p> <p>Exposure to cigarette smoke reduced endothelium-dependent vasodilatation in pulmonary arteries (ANOVA p < 0.05) but not in the aorta. Endothelial dysfunction was apparent at 3 months of exposure and did not increase further after 6 months of exposure. Smoke-exposed animals showed proliferation of poorly differentiated smooth muscle cells in small vessels (p < 0.05) after 3 months of exposure. Prolonged exposure resulted in full muscularization of small pulmonary vessels (p < 0.05), wall thickening (p < 0.01) and increased contractility of the main pulmonary artery (p < 0.05), and enlargement of the alveolar spaces. Lung expression of eNOS was decreased in animals exposed to cigarette smoke.</p> <p>Conclusion</p> <p>In the guinea pig, exposure to cigarette smoke induces selective endothelial dysfunction in pulmonary arteries, smooth muscle cell proliferation in small pulmonary vessels and reduced lung expression of eNOS. These changes appear after 3 months of exposure and precede the development of pulmonary emphysema.</p

    The Snail repressor recruits EZH2 to specific genomic sites through the enrollment of the lncRNA HOTAIR in epithelial-to-mesenchymal transition

    The transcription factor Snail is a master regulator of cellular identity and epithelial-to-mesenchymal transition (EMT) directly repressing a broad repertoire of epithelial genes. How chromatin modifiers instrumental to its activity are recruited to Snail-specific binding sites is unclear. Here we report that the long non-coding RNA (lncRNA) HOTAIR (for HOX Transcript Antisense Intergenic RNA) mediates a physical interaction between Snail and enhancer of zeste homolog 2 (EZH2), an enzymatic subunit of the polycomb-repressive complex 2 and the main writer of chromatin-repressive marks. The Snail-repressive activity, here monitored on genes with a pivotal function in epithelial and hepatic morphogenesis, differentiation and cell-type identity, depends on the formation of a tripartite Snail/HOTAIR/EZH2 complex. These results demonstrate an lncRNA-mediated mechanism by which a transcriptional factor conveys a general chromatin modifier to specific genes, thereby allowing the execution of hepatocyte transdifferentiation; moreover, they highlight HOTAIR as a crucial player in the Snail-mediated EMT.Oncogene advance online publication, 25 July 2016; doi:10.1038/onc.2016.260

    Characterization of the SNAG and SLUG Domains of Snail2 in the Repression of E-Cadherin and EMT Induction: Modulation by Serine 4 Phosphorylation

    Snail1 and Snail2, two highly related members of the Snail superfamily, are direct transcriptional repressors of E-cadherin and EMT inducers. Previous comparative gene profiling analyses have revealed important differences in the gene expression pattern regulated by Snail1 and Snail2, indicating functional differences between both factors. The molecular mechanism of Snail1-mediated repression has been elucidated to some extent, but very little is presently known on the repression mediated by Snail2. In the present work, we report on the characterization of Snail2 repression of E-cadherin and its regulation by phosphorylation. Both the N-terminal SNAG and the central SLUG domains of Snail2 are required for efficient repression of the E-cadherin promoter. The co-repressor NCoR interacts with Snail2 through the SNAG domain, while CtBP1 is recruited through the SLUG domain. Interestingly, the SNAG domain is absolutely required for EMT induction while the SLUG domain plays a negative modulation of Snail2 mediated EMT. Additionally, we identify here novel in vivo phosphorylation sites at serine 4 and serine 88 of Snail2 and demonstrate the functional implication of serine 4 in the regulation of Snail2-mediated repressor activity of E-cadherin and in Snail2 induction of EMT
