    Kinetics for the addition of the p-methoxybenzhydryl cation (AnPhCH+, 10) towards nonconjugated dienes 11 [H2C=C(CH3)-(CH2)n-C(CH3)=CH2] have been determined in CH2Cl2 at −30° to −70°C. Reactivity increases with increasing number of methylene groups separating the two double bonds. For N = 4, reactivity reaches the value for saturated alkyl substituents, and nucleophilic assistance of the second double bond is never observed

    A heterologous prime-boost vaccination strategy using GVAX pancreas vaccine and CRS-207 is showing promise in patients with metastatic pancreatic adenocarcinoma (PDA). GVAX is composed of lethally-irradiated, allogeneic pancreatic cancer cells modified to express GM-CSF and induces a broad response against multiple tumor antigens. GVAX is given with low-dose cyclophosphamide (CY) to inhibit regulatory T cells. CRS-207 is live-attenuated Listeria monocytogenes engineered to express the tumor-associated antigen mesothelin. CRS-207 boosts responses against mesothelin and is unique in its capacity to stimulate both innate and adaptive immunity by activating T cells and NK cells. In a recently completed Phase II study, the CY/GVAX plus CRS-207 combination resulted in statistically-significant improvement of overall survival (OS) compared to CY/GVAX alone (Le, GI ASCO 2014)

    Ferrocenylindium reagents in palladium‐catalyzed cross‐coupling reactions: asymmetric synthesis of planar chiral 2‐aryl oxazolyl and sulfinyl ferrocenes

    [Abstract] The preparation of ferrocenylindium species and palladium‐catalyzed cross-coupling reactions for the synthesis of monosubstituted and planar chiral 1,2‐disubstituted ferrocenes is described. Triferrocenylindium reagents (Fc3In) are efficiently prepared in a one‐pot procedure from ferrocenes by lithiation and transmetallation to indium using InCl3. The palladium‐catalyzed cross‐coupling reactions of Fc3In (40 mol%) with a variety of organic electrophiles (aryl, heteroaryl, benzyl, alkenyl and acyl halides) in THF at 80 °C overnight provided a wide variety of monosubstituted ferrocenes in good to excellent yields. This methodology allowed the stereoselective synthesis of planar chiral 2‐aryl‐1 oxazolylferrocenes and 2‐aryl‐1‐sulfinylferrocenes, which are of interest in asymmetric catalysis.Ministerio de Economía y Competividad; CTQ2015‐68369‐PGalicia.     Consensus guidelines for the use and interpretation of angiogenesis assays

    The formation of new blood vessels, or angiogenesis, is a complex process that plays important roles in growth and development, tissue and organ regeneration, as well as numerous pathological conditions. Angiogenesis undergoes multiple discrete steps that can be individually evaluated and quantified by a large number of bioassays. These independent assessments hold advantages but also have limitations. This article describes in vivo, ex vivo, and in vitro bioassays that are available for the evaluation of angiogenesis and highlights critical aspects that are relevant for their execution and proper interpretation. As such, this collaborative work is the first edition of consensus guidelines on angiogenesis bioassays to serve for current and future reference
