Speaking of international cooperation on many occasions we generate some associated with concepts far removed from what really is image. What we will capture in this experience it is not intended to alarm or create scenarios manipulated or magnify poverty, lack of resources or human misery ... only pretend to be faithful to reality as far and near at the same time as we tell. Two nurses and a professor at the University of Alicante (Spain) developed in Turkana (Kenya) a draft Community intervention in which public participation is central to its development. The main objective of the intervention is to identify major health needs and how people can respond to them independently and responsibly. The project is initiated by identifying community leaders through whom are involved in the community to begin the process of health education which responds to the health needs of the latter. After almost a year of intervention have been achieved key objectives related to hygiene, nutrition and care during pregnancy and childbirth.
Serotonin is involved in the psychostimulant and hypothermic effect of 4-methylamphetamine in rats.
4-Methylamphetamine (4-MA) has recently emerged as a designer drug of abuse in Europe and it is consumed always with amphetamine. There have been reported some deaths and non-fatal intoxications related to 4-MA. We investigated the changes in locomotor activity and body temperature after 4-MA administration to male Sprague-Dawley rats. Our experiments were carried out at a normal or high ambient temperature. 4-MA (2.5-10 mg/Kg, given subcutaneously) increased, in a dose-dependent manner, the horizontal locomotor activity that was significantly reduced by ketanserin, p-cholorophenylalanine (pCPA) or haloperidol, but not by pindolol. In addition, we have studied the effect of 4-MA on core body temperature by means of an implanted electronic thermograph, enabling continuous measurement of body temperature. We observed a dose-dependent hypothermic response to 4-MA that reached a maximum 45 min after a single injection. We also evidenced slight tachyphylaxis to the hypothermic effect when 4-MA was administered four times in a 2 h interval. The pre-treatment of animals with pCPA or pindolol, but not with ketanserin, fully abolished the hypothermic effect of 4-MA. With all that, we conclude that hypothermia induced by 4-MA is due to the release of 5-HT which activates postsynaptic 5-HT1A receptors
Extended-spectrum beta-lactamaseproducing bacteria in a Tertiary Care Hospital in Madrid: Epidemiology, risk factors and antimicrobial susceptibility patterns
Introduction: Extended-spectrum beta-lactamase (ESBL) producing bacteria have been increasingly reported
as causal agents of nosocomial infection worldwide. Resistance patterns vary internationally, and even locally,
from one institution to the other. We investigated the clinical isolates positive for ESBL-producing bacteria in
our institution, a tertiary care hospital in Madrid (Spain), during a 2-year period (2007 2008).
Methods: Clinical and microbiological data were retrospectively reviewed. Two hundred and nineteen patients
were included in the study.
Results: Advanced age, diabetes, use of catheters, previous hospitalization and previous antibiotic treatment
were some of the risk factors found among patients. Escherichia coli was the most frequent isolate, and
urinary tract the most common site of isolation. Internal Medicine, Intensive Care Unit (ICU) and General
Surgery presented the highest number of isolates. There were no outbreaks during the study period. Antibiotic
patterns showed high resistance rates to quinolones in all isolates. There was 100% sensitivity to carbapenems.
Conclusion: Carbapenems continue to be the treatment of choice for ESBL-producing bacteria. Infection
control measures are of great importance to avoid the spread of these nosocomial infection
A copper switch for inducing CRISPR/Cas9-based transcriptional activation tightly regulates gene expression in Nicotiana benthamiana
[EN] Background
CRISPR-based programmable transcriptional activators (PTAs) are used in plants for rewiring gene networks. Better tuning of their activity in a time and dose-dependent manner should allow precise control of gene expression. Here, we report the optimization of a Copper Inducible system called CI-switch for conditional gene activation in Nicotiana benthamiana. In the presence of copper, the copper-responsive factor CUP2 undergoes a conformational change and binds a DNA motif named copper-binding site (CBS).
Results
In this study, we tested several activation domains fused to CUP2 and found that the non-viral Gal4 domain results in strong activation of a reporter gene equipped with a minimal promoter, offering advantages over previous designs. To connect copper regulation with downstream programmable elements, several copper-dependent configurations of the strong dCasEV2.1 PTA were assayed, aiming at maximizing activation range, while minimizing undesired background expression. The best configuration involved a dual copper regulation of the two protein components of the PTA, namely dCas9:EDLL and MS2:VPR, and a constitutive RNA pol III-driven expression of the third component, a guide RNA with anchoring sites for the MS2 RNA-binding domain. With these optimizations, the CI/dCasEV2.1 system resulted in copper-dependent activation rates of 2,600-fold and 245-fold for the endogenous N. benthamiana DFR and PAL2 genes, respectively, with negligible expression in the absence of the trigger.
Conclusions
The tight regulation of copper over CI/dCasEV2.1 makes this system ideal for the conditional production of plant-derived metabolites and recombinant proteins in the field. This work has been funded by EU Horizon 2020 Project Newcotiana Grant 760331, PID2019-108203RB-100 Plan Nacional I + D, Spanish Ministry of Economy and Competitiveness and 'ERACoBioTech' Project SUSPHIRE Grant No. 722361. Evaluation and comparison of two different cartilage-sparing techniques in 356 otoplasties performed in children
Background: Prominent ear is the most common deformity of the external ear. Although hundreds of surgical otoplasty techniques have been described, none of them has proved enough advantage over others to become the gold standard in this field. In this study, we evaluated the results of a cohort of 188 patients operated with two different cartilagesparing otoplasty techniques.Patients and methods: A total of 356 otoplasties were performed in 188 patients, between January 2012 and November 2016. Two different techniques were utilized in two different groups: a modified-Mustardé otoplasty and a modified-Furnas technique. Postoperative follow-ups were performed at week 1, and at 1 and 6 months and at 1 year. Complications were recorded and compared between both techniques. The success rate was measured according to McDowells’ criteria. Results: A total of 356 ears were operated in 188 patients (85 men; 103 women; mean age 9.97; range: 6–15 years). In all, 105 patients underwent modified-Mustarde otoplasty, and 83 were operated utilizing a modified-Furnas technique. Success rates ranged from 90 to 100% depending on the technique utilized. No significative differences wereobserved in the incidence of complications and success rates. Conclusion: None of the compared otoplasty techniques showed better results than the other. Notwithstanding, otoplasty shows to be an effective treatment with high success rates (independent of the technique utilized) for patients with prominent ears. Keywords: antihelix, concha, Furnas, Mustarde, otoplasty, prominent ear
Microclimatic studies in Paranhos water galleries, Porto (Portugal)
Sanz Rubio, Enrique et al.-- 4 pages, 3 figures, 4 references.-- Proceedings of the International Congress on Science and Technology for the Conservation of Cultural Heritage, held 2-5 october, Santiago de Compostela, España.Paranhos constitutes one of the main water galleries excavated in the granite substratum
of Porto City (Portugal) to gather the groundwater for public use during the past six
centuries. The interest of this water gallery is double: a) Cultural Heritage and scientific value,
b) Underground geoturism. A multidisciplinary research to study the microclimatology, geochemistry,
mineralogy and geomicrobiology of the Paranhos catchwork galleries is currently in
progress. The characterization of the main environmental parameters (air-rock temperature,
relative humidity, CO2, 222Rn, etc.) in the internal atmosphere would allow the establishment of:
a) Fluid-rock interactions and development of a physical-chemical model of air-water-rock, as a
base to define their relations with unusual mineral formations and microbial communities b) Microclimatic
parameters and determination of effective radioactive dose for visitors based on
222Rn studies, as potential use as underground geotourism site under safe conditions. This research was supported by the Spanish Project CGL2011-25690.Peer reviewe
