    Antagonistic effect of helpers on breeding male and female survival in a cooperatively breeding bird.

    1. Cooperatively breeding species are typically long lived and hence, according to theory, are expected to maximize their lifetime reproductive success through maximizing survival. Under these circumstances, the presence of helpers could be used to lighten the effort of current reproduction for parents to achieve higher survival. 2. In addition, individuals of different sexes and ages may follow different strategies, but whether male and female breeders and individuals of different ages benefit differently from the presence of helpers has often been overlooked. Moreover, only one study that investigated the relationship between parental survival and the presence of helpers used capture-mark-recapture analyses (CMR). These methods are important since they allow us to account for the non-detection of individuals that are alive in the population but not detected, and thus, the effects on survival and recapture probability to be disentangled. 3. Here, we used multi-event CMR methods to investigate whether the number of helpers was associated with an increase in survival probability for male and female breeders of different ages in the sociable weaver Philetairus socius. In this species, both sexes reduce their feeding rate in the presence of helpers. We therefore predicted that the presence of helpers should increase the breeders' survival in both sexes, especially early in life when individuals potentially have more future breeding opportunities. In addition, sociable weaver females reduce their investment in eggs in the presence of helpers, so we predicted a stronger effect of helpers on female than male survival. 4. As expected we found that females had a higher survival probability when breeding with more helpers. Unexpectedly, however, male survival probability decreased with increasing number of helpers. This antagonistic effect diminished as the breeders grew older. 5. These results illustrate the complexity of fitness costs and benefits underlying cooperative behaviours and how these may vary with the individuals' sex and age.     Maternal effects in relation to helper presence in the cooperatively breeding sociable weaver

    In egg laying species, breeding females may adjust the allocation of nutrients or other substances into eggs in order to maximise offspring or maternal fitness. Cooperatively breeding species offer a particularly interesting context in which to study maternal allocation because helpers create predictably improved conditions during offspring development. Some recent studies on cooperative species showed that females assisted by helpers produced smaller eggs, as the additional food brought by the helpers appeared to compensate for this reduction in egg size. However, it remains unclear how common this effect might be. Also currently unknown is whether females change egg composition when assisted by helpers. This effect is predicted by current maternal allocation theory, but has not been previously investigated. We studied egg mass and contents in sociable weavers ( Philetairus socius ). We found that egg mass decreased with group size, while fledgling mass did not vary, suggesting that helpers may compensate for the reduced investment in eggs. We found no differences in eggs’ carotenoid contents, but females assisted by helpers produced eggs with lower hormonal content, specifically testosterone, androstenedione (A4) and corticosterone levels. Taken together, these results suggest that the environment created by helpers can influence maternal allocation and potentially offspring phenotypes

    Plasma neuregulin 1 as a synaptic biomarker in Alzheimer's disease: a discovery cohort study

    BACKGROUND: Synaptic dysfunction is an early core feature of Alzheimer's disease (AD), closely associated with cognitive symptoms. Neuregulin 1 (NRG1) is a growth and differentiation factor with a key role in the development and maintenance of synaptic transmission. Previous reports have shown that changes in cerebrospinal fluid (CSF) NRG1 concentration are associated with cognitive status and biomarker evidence of AD pathology. Plasma biomarkers reflecting synaptic impairment would be of great clinical interest. OBJECTIVE: To measure plasma NRG1 concentration in AD patients in comparison with other neurodegenerative disorders and neurological controls (NC) and to study its association with cerebrospinal fluid (CSF) core AD and synaptic biomarkers. METHODS: This retrospective study enrolled 127 participants including patients with AD at mild cognitive impairment stage (AD-MCI, n = 27) and at dementia stage (n = 35), non-AD dementia (n = 26, Aβ-negative), non-AD MCI (n = 19), and neurological controls (n=20). Plasma and CSF NRG1, as well as CSF core AD biomarkers (Aβ 42/Aβ 40 ratio, phospho-tau, and total tau), were measured using ELISA. CSF synaptic markers were measured using ELISA for GAP-43 and neurogranin and through immunoprecipitation mass spectrometry for SNAP-25. RESULTS: Plasma NRG1 concentration was higher in AD-MCI and AD dementia patients compared with neurological controls (respectively P = 0.005 and P < 0.001). Plasma NRG1 differentiated AD MCI patients from neurological controls with an area under the curve of 88.3%, and AD dementia patients from NC with an area under the curve of 87.3%. Plasma NRG1 correlated with CSF NRG1 (β = 0.372, P = 0.0056, adjusted on age and sex). Plasma NRG1 was associated with AD CSF core biomarkers in the whole cohort and in Aβ-positive patients (β = -0.197-0.423). Plasma NRG1 correlated with CSF GAP-43, neurogranin, and SNAP-25 (β = 0.278-0.355). Plasma NRG1 concentration correlated inversely with MMSE in the whole cohort and in Aβ-positive patients (all, β = -0.188, P = 0.038; Aβ+: β = -0.255, P = 0.038). CONCLUSION: Plasma NRG1 concentration is increased in AD patients and correlates with CSF core AD and synaptic biomarkers and cognitive status. Thus, plasma NRG1 is a promising non-invasive biomarker to monitor synaptic impairment in AD

    Begging and feeding responses vary with relatedness and sex of provisioners in a cooperative breeder

    Begging behaviour can provide information on offspring hunger levels and be used by parents to adjust food provisioning efforts. In cooperative breeders, helpers also provide care by feeding the young. However, how helpers of different sex and relatedness to the offspring respond to begging behaviour has rarely been studied in cooperatively breeding species, which limits our understanding of the indirect and/or direct benefits that helpers may obtain by responding to offspring demand. Here, we used a cooperatively breeding bird, the sociable weaver, Philetairus socius, to investigate how nest intervisit intervals of breeders and different types of helpers, distinguished by sex and relatedness, varied with acoustic begging. Moreover, we tested whether these different classes of provisioners experienced distinct levels of begging. Our results show that only breeding males, but not breeding females or helpers of any sex and relatedness to the nestlings, returned faster to the nest to feed after experiencing more begging calls. When contrasted directly, we confirmed a statistically supported difference in responses to begging between male and female breeders. Surprisingly, second-order relatives experienced more begging calls than the other classes of more related helpers and breeders. These results show that we might find differences in how provisioners respond to begging levels when classifying group members according to their potential fitness gains. In sociable weavers, the benefits and costs of adjusting feeding efforts to begging seem to differ with sex and life history stage. Experimental and more detailed investigations on begging-feeding interactions are necessary to understand the origin and prevalence of these differences across cooperatively breeding systems.     Association of sleep duration in middle and old age with incidence of dementia

    Sleep dysregulation is a feature of dementia but it remains unclear whether sleep duration prior to old age is associated with dementia incidence. Using data from 7959 participants of the Whitehall II study, we examined the association between sleep duration and incidence of dementia (521 diagnosed cases) using a 25-year follow-up. Here we report higher dementia risk associated with a sleep duration of six hours or less at age 50 and 60, compared with a normal (7h) sleep duration, although this was imprecisely estimated for sleep duration at age 70 (hazard ratios (HR) 1.22 (95% confidence interval 1.01-1.48), 1.37 (1.10-1.72), and 1.24 (0.98-1.57), respectively). Persistent short sleep duration at age 50, 60, and 70 compared to persistent normal sleep duration was also associated with a 30% increased dementia risk independently of sociodemographic, behavioural, cardiometabolic, and mental health factors. These findings suggest that short sleep duration in midlife is associated with an increased risk of late-onset dementia.Peer reviewe

    Egg components and offspring survival vary with group size and laying order in a cooperative breeder

    Prenatal resource allocation to offspring can be influenced by maternal environment and offspring value, and affect offspring survival. An important pathway for flexible maternal allocation is via egg components such as nutrients and hormones. In cooperative breeders, females with helpers may increase resource allocation to eggs-'differential allocation'-or reduce it-'load-lightening'. Yet, helper effects on egg composition have been poorly studied. Moreover, it is unknown how helpers' presence modulates laying order effects on egg content and survival. Here, we investigated how maternal allocation varied with group size and laying order in the cooperatively breeding sociable weaver (Philetairus socius). We estimated interactive effects of helpers and laying order on allocation to egg mass, yolk nutrients-yolk mass, proteins, lipids, carotenoids, vitamin A and vitamin E-and hormones-testosterone, androstenedione, and corticosterone. Results concurred with the 'differential allocation' predictions. Females with more helpers produced later-laid eggs with heavier yolks and more lipids, and laid eggs overall richer in lipids. Proteins, antioxidants, and hormones were not found to vary with helper number. We then analyzed how helper number modulated laying order effects on survival. Females with more helpers did not specifically produce later-laid eggs with higher survival, but eggs laid by females with more helpers were overall more likely to fledge. These findings show that some egg components (yolk mass, lipids) can positively vary according to females' breeding group size, which may improve offspring fitness

    Clinical application of CSF biomarkers for Alzheimer's disease:From rationale to ratios

    Biomarker testing is recommended for the accurate and timely diagnosis of Alzheimer's disease (AD). Using illustrative case narratives we consider how cerebrospinal fluid (CSF) biomarker tests may be used in different presentations of cognitive impairment to facilitate timely and differential diagnosis, improving diagnostic accuracy, providing prognostic information, and guiding personalized management in diverse scenarios. Evidence shows that (1) CSF ratios are superior to amyloid beta (Aβ)1‐42 alone; (2) concordance of CSF ratios to amyloid positron emission tomography (PET) is better than Aβ1‐42 alone; and (3) phosphorylated tau (p‐tau)/Aβ1‐42 ratio is superior to p‐tau alone. CSF biomarkers are recommended for the exclusion of AD as the underlying cause of cognitive impairment, diagnosis of AD at an early stage, differential diagnosis of AD in individuals presenting with other neuropsychiatric symptoms, accurate diagnosis of AD in an atypical presentation, and for clinical trial enrichment. HIGHLIGHTS:  : Cerebrospinal fluid (CSF) Alzheimer's disease (AD) biomarker testing may be underused outside specialist centers. CSF biomarkers improve diagnostic accuracy, guiding personalized management of AD. CSF ratios (amyloid beta [Aβ]1‐42/Aβ1‐40 and phosphorylated tau/Aβ1‐42) perform better than single markers. CSF ratios produce fewer false‐negative and false‐positive results than individual markers. CSF biomarkers should be included in diagnostic work‐up of AD and mild cognitive impairment due to AD

    Cenozoic tectonics of the Western Approaches Channel basins and its control of local drainage systems

    International audienceLe domaine des Approches occidentales de la Manche constitue une zone clé pour caractériser l'évolution post-rift des marges continentales NW européennes associées à la collision Afrique/Europe. Malgré les divers témoins des inversions cénozoïques jalonnant le pourtour de la Manche, la structuration et l'amplitude des mouvements demeurent néanmoins incertaines au sein de la partie méridionale française des Approches occidentales. Il en est de même sur le rôle de l'inversion de la mer du Nord dans la mise en place du Fleuve Manche qui drainait un bassin versant bien supérieur à la Manche actuelle durant les grandes régressions plio-quaternaires et alimentait les éventails sous-marins Celtique et Armoricain en bas de pente. La réalisation des campagnes de sismique-réflexion haute résolution GEOMOC et GEOBREST03 dont les résultats font l'objet de cet article permettent de répondre à ces questions en complétant la connaissance géologique de la Manche. Les nouvelles observations soulignent le diachronisme et le contraste de l'amplitude des mouvements du système de failles associées à l'inversion du bassin d'Iroise. Celle-ci se fait en deux épisodes: un épisode paroxysmal paléogène décomposé en deux phases, éocène (Yprésien probable) et oligocène, et un épisode néogène plus modéré réactivant partiellement les structures impliquées antérieurement. Les déformations se concentrent le long de l'accident nord Iroise (NIF) situé dans le prolongement de la faille Médio-Manche et entraîne localement des plissements de la couverture sédimentaire à l'aplomb des accidents profonds. L'inversion induit ainsi un soulèvement de près de 700 m du plateau médian situé au sud de l'accident nord Iroise. La cartographie isochrone des séquences sismiques identifiées démontre également le contrôle majeur des structures tectoniques sur la mise en place des dépôts néogènes. Le soulèvement de la partie orientale du bassin favorise ainsi la mise en place de vastes prismes progradants d'âge miocène supérieur, et contrôle le développement postérieur du réseau des paléo-vallées constituant l'extrémité occidentale du fleuve Manche. Ce réseau présente une géométrie en baïonnette marquée par de brutaux changements de directions variant de N040 à N070, cette dernière direction caractérisant la plus grande partie des failles néogènes associées au bassin d'Iroise. Les paléo-vallées se seraient développées lors d'une chute du niveau marin au-delà du rebord de plate-forme et la stratigraphie établie à travers cette étude amène à placer le début des incisions au Pliocène (Reurévien ou pré-Tiglien). La chute amplifiée par l'inversion du bassin d'Iroise serait suivie d'un basculement tardif de la plate-forme externe à l'instar des observations réalisées sur de nombreuses marges du pourtour nord atlantique
