    Cumulative incidence and risk factors for Limber Tail in the Dogslife Labrador Retriever cohort

    Limber tail is a condition that typically affects larger working breeds causing tail limpness and pain, resolving without veterinary intervention. It is poorly understood and the disease burden has not been well characterised. Data collected from owners of the Dogslife cohort of Labrador Retrievers have been used to describe incidents and a case-control study was undertaken to elucidate risk factors with 38 cases and 86 controls. The cumulative incidence of unexplained tail limpness was 9.7 per cent. Swimming is not a necessary precursor for limber tail, but it is a risk factor (OR=4.7) and working dogs were more susceptible than non-working dogs (OR=5.1). Higher latitudes were shown to be a risk factor for developing the condition and the case dogs were more related to each other than might be expected by chance. This suggests that dogs may have an underlying genetic predisposition to developing the condition. This study is the first, large-scale investigation of limber tail and the findings reveal an unexpectedly high illness burden. Anecdotally, accepted risk factors have been confirmed and the extent of their impact has been quantified. Identifying latitude and a potential underlying genetic predisposition suggests avenues for future work on this painful and distressing condition

    Asymmetric recurrent laryngeal nerve conduction velocities and dorsal cricoarytenoid muscle electromyographic characteristics in clinically normal horses

    The dorsal cricoarytenoid (DCA) muscles, are a fundamental component of the athletic horse’s respiratory system: as the sole abductors of the airways, they maintain the size of the rima glottis which is essential for enabling maximal air intake during intense exercise. Dysfunction of the DCA muscle leads to arytenoid collapse during exercise, resulting in poor performance. An electrodiagnostic study including electromyography of the dorsal cricoarytenoid muscles and conduction velocity testing of the innervating recurrent laryngeal nerves (RLn) was conducted in horses with normal laryngeal function. We detected reduced nerve conduction velocity of the left RLn, compared to the right, and pathologic spontaneous activity (PSA) of myoelectrical activity within the left DCA muscle in half of this horse population and the horses with the slowest nerve conduction velocities. The findings in this group of horses are consistent with left sided demyelination and axonal loss, consistent with Recurrent Laryngeal Neuropathy (RLN), a highly prevalent degenerative disorder of the RLn in horses that predominantly affects the left side. The detection of electromyographic changes compatible with RLN in clinically unaffected horses is consistent with previous studies that identified “subclinical” subjects, presenting normal laryngeal function despite neuropathologic changes within nerve and muscle confirmed histologically

    Innovation and strategy: Does it make a difference! A linear study of micro & SMEs

    Innovation is known to be used by some businesses to help them differentiate their trading position within a specific locale. Research has also proven that by adopting a strategy, aims can be achieved and turnover increased. This study will endeavour to use these two key areas to evaluate how, they are used and to what extent they can improve business performance. The study was carried using 145 Micro and SME business, located within the rural county of Lincolnshire. The businesses were taken from specific locations on a random basis which included rural, remote rural and urban settings. Initial findings found that businesses situated within remote areas performed better than those within the other test areas, leading to the conclusion that they use innovation as a strategy to outperform their business rivals

    Spider Silk Constructs Enhance Axonal Regeneration and Remyelination in Long Nerve Defects in Sheep

    BACKGROUND: Surgical reapposition of peripheral nerve results in some axonal regeneration and functional recovery, but the clinical outcome in long distance nerve defects is disappointing and research continues to utilize further interventional approaches to optimize functional recovery. We describe the use of nerve constructs consisting of decellularized vein grafts filled with spider silk fibers as a guiding material to bridge a 6.0 cm tibial nerve defect in adult sheep. METHODOLOGY/PRINCIPAL FINDINGS: The nerve constructs were compared to autologous nerve grafts. Regeneration was evaluated for clinical, electrophysiological and histological outcome. Electrophysiological recordings were obtained at 6 months and 10 months post surgery in each group. Ten months later, the nerves were removed and prepared for immunostaining, electrophysiological and electron microscopy. Immunostaining for sodium channel (NaV 1.6) was used to define nodes of Ranvier on regenerated axons in combination with anti-S100 and neurofilament. Anti-S100 was used to identify Schwann cells. Axons regenerated through the constructs and were myelinated indicating migration of Schwann cells into the constructs. Nodes of Ranvier between myelin segments were observed and identified by intense sodium channel (NaV 1.6) staining on the regenerated axons. There was no significant difference in electrophysiological results between control autologous experimental and construct implantation indicating that our construct are an effective alternative to autologous nerve transplantation. CONCLUSIONS/SIGNIFICANCE: This study demonstrates that spider silk enhances Schwann cell migration, axonal regrowth and remyelination including electrophysiological recovery in a long-distance peripheral nerve gap model resulting in functional recovery. This improvement in nerve regeneration could have significant clinical implications for reconstructive nerve surgery

    Qualitative Behaviour Assessment of dogs in the shelter and home environment and relationship with quantitative behaviour assessment and physiological responses

    Qualitative Behaviour Assessment (QBA) was utilised to examine the behavioural expression of dogs in different housing environments and the results were compared to measurements of quantitative behaviour and physiology. Firstly, quantitative behavioural and physiological differences were investigated between dogs in 3 housing environments (short-term shelter confinement, ≀ 4 days, n = 10; long-term shelter confinement, > 30 days, n = 9; and domestic living situations, n = 10). Each dog’s behaviour was recorded over a 4 h period using an ethogram consisting of 21 behaviour categories. Dogs in both short (SD) and long (LD) term confinement displayed higher frequencies of paw-lifting (P < 0.001), displacement behaviour (digging and/or drinking P < 0.01), vocalisation (P < 0.05) and locomotory activity (P < 0.001) compared to dogs maintained as family pets (PD). Salivary cortisol concentrations did not differ amongst groups (H = 0.55, P = 0.76). Secondly, quantitative behaviour and QBA were combined to investigate differences among these same 29 dogs when filmed for 1 min in both their Home Environment and a standardised Novel Environment. QBA of these video clips was made by 10 observers utilising Free-Choice-Profiling methodology. Generalised Procrustes Analysis was used to calculate a consensus profile and three main dimensions of dog expression in both Environments. The observers repeated dog scores on these dimensions with high accuracy (P < 0.001). Observers perceived dogs as more ‘relaxed/content’ in the Home Environment (H = 17.86, P < 0.0001), and more ‘calm/relaxed’ in the Novel Environment (H = 13.58, P < 0.001), than SD and LD dogs. In the Novel Environment, LD dogs were perceived as more ‘inquisitive/curious’ (H = 5.97, P < 0.05), and SD dogs as more ‘curious/cautious’ (H = 6.82, P < 0.05), than the other groups. Quantitative assessment of the 1 min Home and Novel Environment video clips were analysed using Principle Component Analysis (PCA), generating two main factors explaining 88% and 76% of the variation respectively. PCA factor 1 (‘rest’) and QBA Dimension 1 (‘relaxed/content’) correlated (P < 0.0001) in the Home Environment’. In the Novel Environment PCA factor 1 (‘stand’, ‘sniff’) correlated with QBA Dimension 1 (‘clam/relaxed’) and PCA factor 2 (‘sniff’, ‘walk’) correlated with QBA Dimension 2 (‘curious/inquisitive’). There was no correlation between QBA dimensions and cortisol concentrations. In sum, these results indicate that a combined quantitative/qualitative assessment facilitates the interpretation of behavioural variances resulting from housing differences and supports utilising QBA for the assessment of dog behavioural expression
