Logikbasierte Optimierungsverfahren für die Bedarfsprognose
Kunden der Premiumklasse können sich beim Neuwagenkauf eine individuelle Konfiguration auf Basis mehrerer hundert Ausstattungsoptionen zusammenstellen. Die exorbitant hohe Anzahl möglicher Konfigurationsvarianten wird dabei in Boolescher Algebra dokumentiert.
Im Fall der Materialbedarfsplanung werden Verbauraten zukünftiger Sonderausstattungen prognostiziert. Das grundsätzliche Problem, die Prognosen konsistent zu den angebotenen Konfigurationsvarianten zu gestalten, ist verwandt mit dem Entscheidungsproblem Probabilistic Satisfiability (PSAT). PSAT wurde Mitte des 19. Jahrhundert von George Boole formuliert und verknüpft die Aussagenlogik mit der Wahrscheinlichkeitsrechnung. Dennoch konnten erst Ende des 20. Jahrhunderts größere Instanzen mit Hilfe von linearer Optimierung gelöst werden.
Auf Basis von PSAT werden in dieser Arbeit mathematisch exakte Methoden entwickelt, mit denen ein Planer von Verbauraten den durch die Produktdokumentation beschriebenen Raum der möglichen Konfigurationsvarianten vollständig berücksichtigen kann. Die Methoden ermöglichen es, einen zur Produktdokumentation konsistenten Bedarfsplan zu entwickeln. Die Teilebedarfsprognose unterstützend können außerdem die durch einen Bedarfsplan induzierten Verbauraten aller Bauteile evaluiert werden, die mit der Produktdokumentation logisch verknüpft sind
(Q)SAR Modelling of Nanomaterial Toxicity - A Critical Review
There is an increasing recognition that nanomaterials pose a risk to human health, and that the novel engineered nanomaterials (ENMs) in the nanotechnology industry and their increasing industrial usage poses the most immediate problem for hazard assessment, as many of them remain untested. The large number of materials and their variants (different sizes and coatings for instance) that require testing and ethical pressure towards non-animal testing means that expensive animal bioassay is precluded, and the use of (quantitative) structure activity relationships ((Q)SAR) models as an alternative source of hazard information should be explored. (Q)SAR modelling can be applied to fill the critical knowledge gaps by making the best use of existing data, prioritize physicochemical parameters driving toxicity, and provide practical solutions to the risk assessment problems caused by the diversity of ENMs. This paper covers the core components required for successful application of (Q)SAR technologies to ENMs toxicity prediction, and summarizes the published nano-(Q)SAR studies and outlines the challenges ahead for nano-(Q)SAR modelling. It provides a critical review of (1) the present status of the availability of ENMs characterization/toxicity data, (2) the characterization of nanostructures that meets the need of (Q)SAR analysis, (3) the summary of published nano-(Q)SAR studies and their limitations, (4) the in silico tools for (Q)SAR screening of nanotoxicity and (5) the prospective directions for the development of nano-(Q)SAR models
Menthol-Loaded Lipid Nanoparticles for Consumer-Care: Development & Optimized Characterization
Menthol findet aufgrund seines angenehmen Geschmacks, Kühleffekts und Geruchs
breite Verwendung. Da Menthol eine sehr geringe Wasserlöslichkeit aufweist,
ist eine herkömmliche flüssige Formulierung mit Menthol nur möglich, wenn es
in Öl oder in Ethanol gelöst ist. Eine solche Formulierung eignet sich jedoch
nicht für Kinder oder für Menschen, die Ethanol vermeiden. Außerdem können
Formulierungen, die Menthol in hohen Konzentrationen enthalten, die
Schleimhaut reizen. Aus diesen Gründen war das erste Ziel dieser Arbeit die
Entwicklung einer leicht anwendbaren, flüssigen Formulierung ohne Ethanol,
welche eine verlängerte Freisetzung von Menthol ermöglicht, um Reizungen zu
vermeiden. Als Trägersystem wurden nanostrukturierte Lipidträger (NLC)
ausgewählt. Die optimale Zusammensetzung der Lipid-Nanopartikel wurde durch
ein Lipid-Screening mittels DSC und durch ein Tensid-Screening mittels
Kontaktwinkelbestimmung ermittelt. Die entwickelten Formulierungen wurden
unter Verwendung einer Heißhochdruckhomogenisierung hergestellt. Ein
systematischer Vergleich der Produktionsparameter ergab, dass der optimale
Herstellungsprozess mit drei Homogenisierungszyklen bei einem Druck von 500
bar und mittels Creme-Schmelzverfahren erfolgt. Dieses Verfahren führt zu
stabilen Lipidnanopartikeln im Submikrometerbereich mit enger
Partikelgrößenverteilung und mit ZP-Werten über -40 mV. Die Partikel blieben
auch nach Langzeitlagerung (über 1 Jahr) und Versprühen stabil. Die Ergebnisse
der membranfreien in vitro-Freisetzungsstudie und der in vivo-
Freisetzungsstudie zeigten eine verlängerte Freisetzung von Menthol im
Vergleich zu herkömmlichen mentholhaltigen Mundspülungen. Das
Kristallisationsverhalten der entwickelten Lipidnanopartikel ist durch eine
geringe Menge der niedrigschmelzenden Modifikation gekennzeichnet. Daher kann
eine Instabilität während der Lagerung aufgrund der polymorphen Transformation
ausgeschlossen werden. Das sanfte Herstellungsverfahren bei einer niedrigeren
Temperatur kann die Wirksamkeit der Verkapselung von Wirkstoffen mit einem
relativ hohen Dampfdruck wie z.B. Menthol erhöhen. Im zweiten Teil der Arbeit
wurden ausgewählte Prozesse der Entwicklung, Charakterisierung und Lagerung
von Lipidnanopartikeln systematisch untersucht und optimiert. Für das Lipid-
Screening wurden die herkömmliche DSC-Methode, die mikroskopische Methode und
eine neu entwickelte Suppositorien-Methode verglichen. Die Mikroskop- und die
Suppositorien-Methode erwiesen sich als einfache und effiziente Methoden zur
Vorhersage der physikalischen Stabilität von Lipidnanopartikel-Dispersionen.
Bzgl. dem Tensid-Screening via Goniometer wurde gezeigt, dass der
Kontaktwinkel stark von der Probenvorbereitung abhängt. Für die
Partikelgrößenanalyse durch Laserdiffraktometrie wurde festgestellt, dass es
andere, bisher unbekannte Fehlerquellen gibt, z.B. das Instrument für die
Probenahme, die Rührgeschwindigkeit und die Verwendung von Beschallung. Diese
Fehlerquellen können zu falschen Ergebnissen führen, nach ihrem vollständigen
Verständnis aber auch vorteilig zur Vorhersage der Probenstabilität zu einem
sehr frühen Entwicklungszustand genutzt werden. Bei der dynamischen
Differenzkalorimetrie wurde gezeigt, dass die DSC-Ergebnisse durch den
Wassergehalt der Probe beeinflusst werden. Daher sollte für die Bestimmung der
Enthalpien eine auf den Lipidgehalt basierende Berechnung erfolgen. Für den in
vitro Occlusionstest nach de Vringer wurde gezeigt, dass der Test in einer
üblichen Klimakammer kostengünstig durchgeführt werden kann, indem eine
gesättigte Lösung von Natriumbromid verwendet wird, die indirekt die
Feuchtigkeit reguliert. Abschließend wurde gezeigt, dass auch die primären
Verpackungsmaterialien und die Lagertemperatur die physikalische und chemische
Stabilität der verpackten Produkte beeinflussen. Wenn langfristige
physikalische Stabilitätstests durchgeführt werden, sollte Glas Typ 1
verwendet werden. Für kurzfristige physikalische Stabilitätstests können
weniger kostspielige Materialien, z.B. HDPE oder PP verwendet werden.Menthol is widely used because of its minty taste, cooling effect and pleasant
odour. Since menthol has very low water solubility, a liquid formulation is
only possible, if it is dissolved in oily medium or in ethanol. However, such
a formulation is not suitable for children or for people, who avoid ethanol.
Additionally, formulations containing menthol in high concentrations could
irritate the mucous membrane. For these reasons the aim of the first part of
this work was to develop an easy to apply, liquid formulation without ethanol
and prolonged release of menthol to avoid irritation. As carriers systems for
menthol nanostructured lipid carriers (NLC) were selected. With the objective
of being used in consumer care products the optimal composition of the lipid
nanoparticles were determined by a lipid screening using the DSC method and
with a surfactant screening by using contact angle determination. The
developed formulations were produced by using hot high-pressure
homogenization. A systematic comparison of the production parameters revealed
that the best production process included three homogenization cycles at a
pressure of 500 bar and a cream melting process. This process leads to stable
solid particles in the submicron range with narrow particle size distribution
and with ZP-values higher than -40 mV. The particles remain stable after long
term storage (over 1 year) and after spraying with a noozle. The results of
both, a membrane-free in vitro release study and an in vivo release study,
indicate a prolonged release of menthol compared with conventional menthol-
containing mouthwash. The crystallization behavior of the lipid nanoparticles
in the developed formulation is characterized by a small amount of the low-
melting modification. Hence instability during storage due to the polymorphic
transformation can be excluded. The gentle production method at a lower
temperature can improve the encapsulation efficacy of materials with
relatively high vapour pressure, e.g. such as menthol In the second part of
the work selected processes for the development, characterization, and storage
of the lipid particles were systematically studied and optimized The lipid
screening method was optimized by comparing the conventional DSC method, and a
microscopic method and the newly developed “suppository” method. The
microscope and the suppository method were shown to be an easy and efficient
method for predicting the physical stability of lipid nanoparticle
dispersions. With regard to the surfactant screening via goniometer it was
shown that the contact angle results strongly depend on the method used for
the sample preparation. For the particle size analysis by laser diffractometry
it was found that there are other, previously unknown pitfalls, e.g.
instrument for sampling, the stirring speed and the use of sonication. The
pitfalls can lead to uncorrect results, but can be used – if understood – to
predict the stability of the samples at a very early state of the development
Within the scope of differential scanning calorimetry it was shown that the
DSC results are influenced by the water content of a sample. Therefore, for
the calculation of enthalpies, methods based on lipid content only should be
preferred. For the in vitro Occlusion test after de Vringer, it has been shown
that the test can be cost-effectively performed in a usual climate chamber by
using a saturated solution of sodium bromide which regulates indirectly the
humidity. Finally, it was shown that the primary packaging materials and the
storage temperature affect the physical and chemical stability of the packaged
products. If long-term physical stability tests are performed only glass type
1 type packaging material should be used. For short term-physical stability
tests less costly materials, e.g. HDPE or PP can be used
Breakpoints around the HOXD cluster result in various limb malformations
Background: Characterisation of disease associated balanced chromosome rearrangements is a
promising starting point in the search for candidate genes and regulatory elements.
Methods: We have identified and investigated three patients with limb abnormalities and breakpoints
involving chromosome 2q31. Patient 1 with severe brachydactyly and syndactyly, mental retardation,
hypoplasia of the cerebellum, scoliosis, and ectopic anus, carries a balanced t(2;10)(q31.1;q26.3)
translocation. Patient 2, with translocation t(2;10)(q31.1;q23.33), has aplasia of the ulna, shortening of
the radius, finger anomalies, and scoliosis. Patient 3 carries a pericentric inversion of chromosome 2,
inv(2)(p15q31). Her phenotype is characterised by bilateral aplasia of the fibula and the radius, bilateral
hypoplasia of the ulna, unossified carpal bones, and hypoplasia and dislocation of both tibiae.
Results: By fluorescence in situ hybridisation, we have mapped the breakpoints to intervals of
approximately 170 kb or less. None of the three 2q31 breakpoints, which all mapped close to the
HOXD cluster, disrupted any known genes.
Conclusions: Hoxd gene expression in the mouse is regulated by cis-acting DNA elements acting over
distances of several hundred kilobases. Moreover, Hoxd genes play an established role in bone
development. It is therefore very likely that the three rearrangements disturb normal HOXD gene regulation
by position effects
Disruption of Netrin G1 by a balanced chromosome translocation in a girl with Rett syndrome
We have identified a girl with characteristic features of Rett syndrome (RTT) who carries a de novo balanced translocation involving chromosomes 1 and 7. Both breakpoints were mapped by fluorescence in situ hybridization with selected genomic clones from the regions of interest. Southern blot hybridisations, utilizing probes derived from breakpoint spanning BACs, detected several aberrant fragments specific for the patient. Sequence analysis of the cloned junction fragment indicated that on chromosome 1 the predominantly brain-expressed Netrin G1 (NTNG1) gene is disrupted, whereas on chromosome 7 there was no indication for a truncated gene. The chromosome 1 breakpoint lies within the 3' part of NTNG1 and affects alternatively spliced transcripts, suggesting that the phenotype in this patient is the result of disturbed NTNG1 expression. In silico translation of the NTNG1 splice variants predicted protein isoforms with different C-termini: one membrane bound through a glycosylphosphatidylinositol anchor and the other soluble. The membrane-bound protein isoform would be affected by the breakpoint, whereas the soluble form would remain intact. Our results suggest that the central nervous system is sensitive to NTNG1 expression levels and that NTNG1 is a novel candidate disease gene for RTT
Disruption of the Serine/Threonine Kinase 9 Gene Causes Severe X-Linked Infantile Spasms and Mental Retardation
X-linked West syndrome, also called “X-linked infantile spasms” (ISSX), is characterized by early-onset generalized seizures, hypsarrhythmia, and mental retardation. Recently, we have shown that the majority of the X-linked families with infantile spasms carry mutations in the aristaless-related homeobox gene (ARX), which maps to the Xp21.3-p22.1 interval, and that the clinical picture in these patients can vary from mild mental retardation to severe ISSX with additional neurological abnormalities. Here, we report a study of two severely affected female patients with apparently de novo balanced X;autosome translocations, both disrupting the serine-threonine kinase 9 (STK9) gene, which maps distal to ARX in the Xp22.3 region. We show that STK9 is subject to X-inactivation in normal female somatic cells and is functionally absent in the two patients, because of preferential inactivation of the normal X. Disruption of the same gene in two unrelated patients who have identical phenotypes (consisting of early-onset severe infantile spasms, profound global developmental arrest, hypsarrhythmia, and severe mental retardation) strongly suggests that lack of functional STK9 protein causes severe ISSX and that STK9 is a second X-chromosomal locus for this disorder
