    Synergism between Porcine Reproductive and Respiratory

    This study was conducted to investigate the effects of exposure to porcine reproductive and respiratory syndrome virus, Salmonella choleraesuis, and stress on young swine. Five-week-old segregated early weaned pigs were randomly assigned to one of eight treatments consisting of all possible combinations of three factors: S. choleraesuis (SC) on day zero, porcine reproductive and respiratory syndrome virus (PRRSV) on day three, and dexamethasone (DEX) on days three to seven. DEX was used as a proxy for stress. Treatment differences were seen in performance parameters, levels and duration of SC shedding, level and distribution of SC in tissues, clinical disease, and mortality. The results of this study provided evidence to support field observations that clinical outbreaks of PRRS are the result of interactions among concurrent infections and stressors

    Development of a Fluorescent Microsphere Immunoassay for Detection of Antibodies against Porcine Reproductive and Respiratory Syndrome Virus Using Oral Fluid Samples as an Alternative to Serum-Based Assays

    For effective disease surveillance, rapid and sensitive assays are needed to detect antibodies developed in response to porcine reproductive and respiratory syndrome virus (PRRSV) infection. In this study, we developed a multiplexed fluorescent microsphere immunoassay (FMIA) for detection of PRRSV-specific antibodies in oral fluid and serum samples. Recombinant nucleocapsid protein (N) and nonstructural protein 7 (nsp7) from both PRRSV genotypes (type I and type II) were used as antigens and covalently coupled to Luminex fluorescent microspheres. Based on an evaluation of 488 oral fluid samples with known serostatus, the oral fluid-based FMIAs achieved \u3e92% sensitivity and 91% specificity. For serum samples (n = 1,639), the FMIAs reached \u3e98% sensitivity and 95% specificity. The assay was further employed to investigate the kinetics of the antibody response in infected pigs. In oral fluid, the N protein was more sensitive for the detection of early infection (7 and 14 days postinfection), but nsp7 detected a higher level and longer duration of antibody response (28 days postinfection). In serum, the antibodies specific to nsp7 and N proteins were detected as early as 7 days postinfection, and the responses lasted more than 202 days. This study provides a framework from which a more robust assay could be developed to profile the immune response to multiple PRRSV antigens in a single test. The development of oral fluid-based diagnostic tests will change the way we survey diseases in swine herds and improve our ability to cheaply and efficiently track PRRSV infections in both populations and individual animals

    Predicting the effects of biochar on volatile petroleum hydrocarbon biodegradation and emanation from soil: a bacterial community finger-print analysis inferred modelling approach

    We investigated the response of the dominant bacterial taxa in gravelly sand to the addition of biochar and/or mixtures of volatile petroleum hydrocarbons (VPHs) using denaturing gradient gel electrophoresis (DGGE) and sequencing of cut bands. Biochar addition alone had only weak effects on the soil bacterial community composition in batch study samples, while VPH addition had strong effects. Indirect effects of biochar on soil bacterial communities were apparent in column study samples, where biochar-enhanced sorption affected VPH spreading. Following VPH addition, cell abundance increased by no more than a factor of 2 and several Pseudomonas spp. became dominant in soil with and without biochar. We present a VPH fate model that considers soil bacterial biomass dynamics and a nutrient limited soil biomass carrying capacity. The model simulates an apparent lag phase before the onset of a brief period of intensive VPH biodegradation and biomass growth, which is followed by substantially slower VPH biodegradation, when nitrogen needs to be recycled between decaying and newly formed biomass. If biomass growth is limited by a factor other than the organic pollutant bioavailability, biochar amendment may enhance VPH attenuation in between a VPH source below ground and the atmosphere by reducing the risk of overloading the soil's biodegradation capacity

    A Decade of SN1993J: Discovery of Wavelength Effects in the Expansion Rate

    We have studied the growth of the shell-like radio structure of supernova SN1993J in M81 from September 1993 through October 2003 with very-long-baseline interferometry (VLBI) observations at the wavelengths of 3.6, 6, and 18cm. For this purpose, we have developed a method to accurately determine the outer radius (R) of any circularly symmetric compact radio structure like SN1993J. The source structure of SN1993J remains circularly symmetric (with deviations from circularity under 2%) over almost 4000 days. We characterize the decelerated expansion of SN 1993J through approximately day 1500 after explosion with an expansion parameter m=0.845±0.005m= 0.845\pm0.005 (RtmR \propto t^{m}). However, from that day onwards the expansion is different when observed at 6 and 18cm. Indeed, at 18cm, the expansion can be well characterized by the same mm as before day 1500, while at 6cm the expansion appears more decelerated, and is characterized by another expansion parameter, m6=0.788±0.015m_{6}= 0.788\pm0.015. Therefore, since about day 1500 on, the radio source size has been progressively smaller at 6cm than at 18cm. These findings are in stark contrast to previous reports by other authors on the details of the expansion. In our interpretation the supernova expands with a single expansion parameter, m=0.845±0.005m= 0.845\pm0.005, and the 6cm results beyond day 1500 are due to physical effects, perhaps also coupled to instrumental limitations. Two physical effects may be involved: (a) a changing opacity of the ejecta to the 6cm radiation, and (b) a radial decrease of the magnetic field in the emitting region. (Long abstract cut.     Phase I Hydroxylated Metabolites of the K2 Synthetic Cannabinoid JWH-018 Retain In Vitro and In Vivo Cannabinoid 1 Receptor Affinity and Activity

    K2 products are synthetic cannabinoid-laced, marijuana-like drugs of abuse, use of which is often associated with clinical symptoms atypical of marijuana use, including hypertension, agitation, hallucinations, psychosis, seizures and panic attacks. JWH-018, a prevalent K2 synthetic cannabinoid, is structurally distinct from Δ(9)-THC, the main psychoactive ingredient in marijuana. Since even subtle structural differences can lead to differential metabolism, formation of novel, biologically active metabolites may be responsible for the distinct effects associated with K2 use. The present study proposes that K2's high adverse effect occurrence is due, at least in part, to distinct JWH-018 metabolite activity at the cannabinoid 1 receptor (CB1R).JWH-018, five potential monohydroxylated metabolites (M1-M5), and one carboxy metabolite (M6) were examined in mouse brain homogenates containing CB1Rs, first for CB1R affinity using a competition binding assay employing the cannabinoid receptor radioligand [(3)H]CP-55,940, and then for CB1R intrinsic efficacy using an [(35)S]GTPγS binding assay. JWH-018 and M1-M5 bound CB1Rs with high affinity, exhibiting K(i) values that were lower than or equivalent to Δ(9)-THC. These molecules also stimulated G-proteins with equal or greater efficacy relative to Δ(9)-THC, a CB1R partial agonist. Most importantly, JWH-018, M2, M3, and M5 produced full CB1R agonist levels of activation. CB1R-mediated activation was demonstrated by blockade with O-2050, a CB1R-selective neutral antagonist. Similar to Δ(9)-THC, JWH-018 and M1 produced a marked depression of locomotor activity and core body temperature in mice that were both blocked by the CB1R-preferring antagonist/inverse agonist AM251.Unlike metabolites of most drugs, the studied JWH-018 monohydroxylated compounds, but not the carboxy metabolite, retain in vitro and in vivo activity at CB1Rs. These observations, combined with higher CB1R affinity and activity relative to Δ(9)-THC, may contribute to the greater prevalence of adverse effects observed with JWH-018-containing products relative to cannabis

    O biocarvão (biomassa carbonizada para uso agrícola) tem sido usado como condicionador do solo em todo o mundo, e essa tecnologia é de especial interesse para o Brasil, uma vez que tanto a ?inspiração?, que veio das Terras Pretas de Índios da Amazônia, como o fato de o Brasil ser o maior produtor mundial de carvão vegetal, com a geração de importante quantidade de resíduos na forma de finos de carvão e diversas biomassas residuais, principalmente da agroindústria, como bagaço de cana, resíduos das indústrias de madeira, papel e celulose, biocombustíveis, lodo de esgoto etc. Na última década, diversos estudos com biocarvão têm sido realizados e atualmente uma vasta literatura e excelentes revisões estão disponíveis. Objetivou-se aqui não fazer uma revisão bibliográfica exaustiva, mas sim uma revisão crítica para apontar alguns destaques na pesquisa sobre biochar.
