    Seroprevalence and Risk Factors of Chlamydia abortus

    Chlamydia abortus, an important pathogen in a variety of animals, is associated with abortion in sheep. In the present study, 1732 blood samples, collected from Tibetan sheep between June 2013 and April 2014, were examined by the indirect hemagglutination (IHA) test, aiming to evaluate the seroprevalence and risk factors of C. abortus infection in Tibetan sheep. 323 of 1732 (18.65%) samples were seropositive for C. abortus antibodies at the cut-off of 1 : 16. A multivariate logistic regression analysis was used to evaluate the risk factors associated with seroprevalence, which could provide foundation to prevent and control C. abortus infection in Tibetan sheep. Gender of Tibetan sheep was left out of the final model because it is not significant in the logistic regression analysis (P>0.05). Region, season, and age were considered as major risk factors associated with C. abortus infection in Tibetan sheep. Our study revealed a widespread and high prevalence of C. abortus infection in Tibetan sheep in Gansu province, northwest China, with higher exposure risk in different seasons and ages and distinct geographical distribution

    Photonic-plasmonic hybrid single-molecule nanosensor measures the effect of fluorescent labels on DNA-protein dynamics

    Current methods to study molecular interactions require labeling the subject molecules with fluorescent reporters. However, the effect of the fluorescent reporters on molecular dynamics has not been quantified because of a lack of alternative methods. We develop a hybrid photonic-plasmonic antenna-in-a-nanocavity single-molecule biosensor to study DNA-protein dynamics without using fluorescent labels. Our results indicate that the fluorescein and fluorescent protein labels decrease the interaction between a single DNA and a protein due to weakened electrostatic interaction. Although the study is performed on the DNA-XPA system, the conclusion has a general implication that the traditional fluorescent labeling methods might be misestimating the molecular interactions

    Identification of genes regulated by Wnt/β-catenin pathway and involved in apoptosis via microarray analysis

    BACKGROUND: Wnt/β-catenin pathway has critical roles in development and oncogenesis. Although significant progress has been made in understanding the downstream signaling cascade of this pathway, little is known regarding Wnt/β-catenin pathway modification of the cellular apoptosis. METHODS: To identify potential genes regulated by Wnt/β-catenin pathway and involved in apoptosis, we used a stably integrated, inducible RNA interference (RNAi) vector to specific inhibit the expression and the transcriptional activity of β-catenin in HeLa cells. Meanwhile, we designed an oligonucleotide microarray covering 1384 apoptosis-related genes. Using oligonucleotide microarrays, a series of differential expression of genes was identified and further confirmed by RT-PCR. RESULTS: Stably integrated inducible RNAi vector could effectively suppress β-catenin expression and the transcriptional activity of β-catenin/TCF. Meanwhile, depletion of β-catenin in this manner made the cells more sensitive to apoptosis. 130 genes involved in some important cell-apoptotic pathways, such as PTEN-PI3K-AKT pathway, NF-κB pathway and p53 pathway, showed significant alteration in their expression level after the knockdown of β-catenin. CONCLUSION: Coupling RNAi knockdown with microarray and RT-PCR analyses proves to be a versatile strategy for identifying genes regulated by Wnt/β-catenin pathway and for a better understanding the role of this pathway in apoptosis. Some of the identified β-catenin/TCF directed or indirected target genes may represent excellent targets to limit tumor growth

    Mechanical durability of superhydrophobic surfaces: the role of surface modification technologies

    Various surface modification technologies have been used to develop superhydrophobic surface, however their durability has been recognized as the major obstacle for the real applications. Here a quantitative investigation was conducted to evaluate the effects of different surface modification methods on the surfaces’ mechanical durability. The superhydrophobic surfaces were prepared by the combination of two surface roughing methods (etching and sandblasting) with chemical modifications with four low surface energy materials: silica sol (SS), octadecanoic acid (OA), heptadecafluoro-1,1,2,2-tetrahydrodecyltrichlorosilane (HDFS) and hexadecyltriethoxysilane (HTS). XPS was used to analyze the elements composition and AFM was used to measure the roughness of the surfaces. The durability of these surfaces was tested by a sandpaper abrasion experiment. The collective results showed that the low surface energy materials had significant effects on the surface roughness, which would then play an important role in the durability of these rough surfaces. The SS modified rough surfaces possessed higher roughness and better durability than the surfaces modified by other three low surface energy materials. SS modified rough surfaces could bear 60 cycles of abrasion with 10 g weights on 1500 CW sandpaper

    Deciphering neo-sex and B chromosome evolution by the draft genome of Drosophila albomicans

    <p>Abstract</p> <p>Background</p> <p><it>Drosophila albomicans </it>is a unique model organism for studying both sex chromosome and B chromosome evolution. A pair of its autosomes comprising roughly 40% of the whole genome has fused to the ancient X and Y chromosomes only about 0.12 million years ago, thereby creating the youngest and most gene-rich neo-sex system reported to date. This species also possesses recently derived B chromosomes that show non-Mendelian inheritance and significantly influence fertility.</p> <p>Methods</p> <p>We sequenced male flies with B chromosomes at 124.5-fold genome coverage using next-generation sequencing. To characterize neo-Y specific changes and B chromosome sequences, we also sequenced inbred female flies derived from the same strain but without B's at 28.5-fold.</p> <p>Results</p> <p>We assembled a female genome and placed 53% of the sequence and 85% of the annotated proteins into specific chromosomes, by comparison with the 12 <it>Drosophila genomes</it>. Despite its very recent origin, the non-recombining neo-Y chromosome shows various signs of degeneration, including a significant enrichment of non-functional genes compared to the neo-X, and an excess of tandem duplications relative to other chromosomes. We also characterized a B-chromosome linked scaffold that contains an actively transcribed unit and shows sequence similarity to the subcentromeric regions of both the ancient X and the neo-X chromosome.</p> <p>Conclusions</p> <p>Our results provide novel insights into the very early stages of sex chromosome evolution and B chromosome origination, and suggest an unprecedented connection between the births of these two systems in <it>D. albomicans</it>.</p

    Palaeoceanographic controls on spatial redox distribution over the Yangtze Platform during the Ediacaran–Cambrian transition

    The Ediacaran–Cambrian interval was an eventful transitional period, when dynamic interactions between the biosphere and its physical environment allowed the Earth System to cross into a new state, characterized by the presence of metazoans, more equable climates and more expansive oxygenation of the oceans. Due to the retreat of widespread sulphidic conditions, redox-sensitive trace-metals could accumulate to a greater extent in ‘black shales’ deposited in localised anoxic/euxinic environments, such as highly productive ocean margins. This study investigates the concentrations of the redox-sensitive trace-metals molybdenum and vanadium in organic-rich sedimentary rocks from seven sections of the Yangtze platform, slope and basin. Iron speciation analyses were carried out in order to distinguish oxic, anoxic-ferruginous and anoxic-sulphidic settings, while sulphur and nitrogen isotope ratios were measured to gain insight into sulphate and nitrate availability, respectively, in the context of changing redox conditions. The data herein demonstrate an overall increase in redox-sensitive trace-metal contents in black shales across the Ediacaran–Cambrian transition, but with marked temporal and spatial variability. Euxinia is evident in South China before 551 Ma in the Ediacaran, and again in the early Cambrian. However, some time-equivalent sections are not enriched in redox-sensitive trace metals, and also exhibit contrasting S-isotope and N-isotope systematics. A more complex configuration of the Yangtze Platform, for example with vast intra-shelf basins, together with changing (generally rising) eustatic sea-level may account for this variability. In this regard, it is proposed that a mid-depth sulphidic wedge, caused by nutrient upwelling over the south-east platform margins, migrated over time (but generally landward), leading to spatially variable redox conditions determined by sea-level, currents and bathymetric constraints. The changing extents of anoxia and euxinia appear to have limited the distribution of emerging Ediacaran and Cambrian ecosystems
