    In the title compound, C13H10ClNO, the central C—N bond has considerable double-bond character and the N—O bond indicates a formal negative charge on the O atom. The mol­ecule is stabilized by an intra­molecular C—H⋯O hydrogen bond. The geometry about the C=N bond is Z [C—C—N—O torsion angle = −4.2 (3)°] and the phenyl and benzene rings are trans-oriented around the C=N bond. The phenyl and benzene rings make a dihedral angle of 56.99 (2)°

    Arsenic trioxide (As2O3) is a promising effective chemotherapeutic agent for cancer treatment; however, how and through what molecular mechanisms the oxidative damage of As2O3 is controlled remains poorly understood. Recently, the involvement of dysregulated long noncoding RNA ovarian tumor domain containing 6B antisense RNA1 (lncRNA OTUD6B-AS1) in tumorigenesis is established. Here, for the first time, we characterize the regulation of As2O3 in the oxidative damage against bladder cancer via lncRNA OTUD6B-AS1. As2O3 could activate lncRNA OTUD6B-AS1 transcription in bladder cancer cells, and these findings were validated in a xenograft tumor model. Functional assays showed that lncRNA OTUD6B-AS1 dramatically exacerbated As2O3-mediated oxidative damage by inducing oxidative stress. Mechanistically, As2O3 increased levels of metal-regulatory transcription factor 1 (MTF1), which regulates lncRNA OTUD6B-AS1, in response to oxidative stress. Further, lncRNA OTUD6B-AS1 inhibited mitochondrial NADP+-dependent isocitrate dehydrogenase 2 (IDH2) expression by stabilizing miR-6734-5p, which contributed to cytotoxicity by enhancing oxidative stress. Together, our findings offer new insights into the mechanism of As2O3-induced oxidative damage and identify important factors in the pathway, As2O3/lncRNA OTUD6B-AS1/miR-6734-5p/IDH2, expanding the knowledge of activity of As2O3 as cancer treatment

    Evaluating the Open Source Data Containers for Handling Big Geospatial Raster Data

    Big geospatial raster data pose a grand challenge to data management technologies for effective big data query and processing. To address these challenges, various big data container solutions have been developed or enhanced to facilitate data storage, retrieval, and analysis. Data containers were also developed or enhanced to handle geospatial data. For example, Rasdaman was developed to handle raster data and GeoSpark/SpatialHadoop were enhanced from Spark/Hadoop to handle vector data. However, there are few studies to systematically compare and evaluate the features and performances of these popular data containers. This paper provides a comprehensive evaluation of six popular data containers (i.e., Rasdaman, SciDB, Spark, ClimateSpark, Hive, and MongoDB) for handling multi-dimensional, array-based geospatial raster datasets. Their architectures, technologies, capabilities, and performance are compared and evaluated from two perspectives: (a) system design and architecture (distributed architecture, logical data model, physical data model, and data operations); and (b) practical use experience and performance (data preprocessing, data uploading, query speed, and resource consumption). Four major conclusions are offered: (1) no data containers, except ClimateSpark, have good support for the HDF data format used in this paper, requiring time- and resource-consuming data preprocessing to load data; (2) SciDB, Rasdaman, and MongoDB handle small/mediate volumes of data query well, whereas Spark and ClimateSpark can handle large volumes of data with stable resource consumption; (3) SciDB and Rasdaman provide mature array-based data operation and analytical functions, while the others lack these functions for users; and (4) SciDB, Spark, and Hive have better support of user defined functions (UDFs) to extend the system capability

    Environmental conditions contribute to plant growth and metabolism. This study aimed to determine a suitable environment and climate for large-scale artificial cultivation of an endangered plant, Tetrastigma hemsleyanum, by investigating the seasonal variations influencing the flavonoid biosynthetic selectivity and antioxidant activity of its major metabolites. Under conditions of precipitation (2.0~6.6 mm), temperature (17.5~24.1°C), humidity (67.3~80.2%), and sunshine duration (3.4~5.8 h) from April to May, the total flavonoid content in T. hemsleyanum reached higher levels between 281.3 and 392.8 μg/g. In the second half of April, the production selectivity (PS) of isoorientin (IsoO), orientin (Or), rutin (Rut), isoquercitin (IsoQ), kaempferol-3-O-rutinoside (Km3rut), astragalin (Ast), quercetin (Qu), apigenin (Ap), and kaempferol (Km) were 0.30, 0.06, 0.07, 0.07, 0.00, 0.04, 0.38, 0.05, and 0.03, respectively. Naringenin was dehydrogenated or hydroxylated to initiate two parallel reaction pathways for flavonoid biosynthesis in T. hemsleyanum: path I subsequently generated flavone derivatives including apigenin, luteolin, orientin, and isoorientin, and path II subsequently generated flavonol derivatives including Km, Qu, IsoQ, Rut, Ast, and Km3rut. The reaction selectivity of path II (RPSII) from January 1 to September 30 was considerably higher than that of path I (RPSI), except for March 16-31. In addition, either the content or antioxidant activity of three major metabolites in T. hemsleyanum followed the order of phenolic compounds > polysaccharides > sterols, and exhibited dynamic correlations with environmental factors. Naringenin favored hydroxylation and derived six flavonol compounds from January to September, and favored dehydrogenation and derived three flavone compounds from October to December. In most months of a year, Km preferentially favored hydroxylation rather than glucosylation

    Long-term outcomes of COVID-19 convalescents: An 18.5-month longitudinal study in Wuhan

    Objectives: This study aimed to describe the full scope of long-term outcomes and the ongoing pathophysiological alterations among COVID-19 survivors. Methods: We established a longitudinal cohort of 208 COVID-19 convalescents and followed them at 3.3 (interquartile range [IQR]: 1.3, 4.4, visit 1), 9.2 (IQR: 9.0, 9.6, visit 2), and 18.5 (IQR: 18.2, 19.1, visit 3) months after infection, respectively. Serial changes in multiple physical and psychological outcomes were comprehensively characterized. We, in addition, explored the potential risk factors of SARS-CoV-2 antibody response and sequelae symptoms. Results: We observed continuous improvement of sequelae symptoms, lung function, chest computed tomography (CT), 6-minute walk test, and the Borg dyspnea scale, whereas sequelae symptoms (at least one) and abnormal chest CT patterns still existed in 45.2% and about 30% of participants at 18.5 months, respectively. Anxiety and depression disorders were alleviated for the convalescents, although depression status was sustained for a longer duration. Conclusions: Most COVID-19 convalescents had an overall improved physical and psychological health status, whereas sequelae symptoms, residual lesions on lung function, exercise impairment, and mental health disorders were still observed in a small proportion of participants at 18.5 months after infection. Implementing appropriate preventive and management strategies for the ever-growing COVID-19 population is warranted
