Funding Resilient Infrastructure in New Jersey: Attitudes Following a Natural Disaster
Recent major natural disasters in New Jersey have demonstrated the need to increase the resilience of transportation infrastructure. This research examines public attitudes toward revenue sources that can be dedicated to protecting vulnerable areas, most notably the transportation linkages on which the state depends. A statewide survey was conducted to gather data approximately four months following Superstorm Sandy, the costliest natural disaster in the state’s history. The authors’ objective was to sample public attitudes while the impacts of the disaster were still fresh. They found little support for temporary tax increases to improve resiliency, with the most positive support for taxing visitors (i.e., a hotel and recreational tax) and for a 30-year bond measure (i.e., taxing the future). This observation seemingly contradicts broad support for investing in new infrastructure, as well as maintaining and protecting existing infrastructure. Multivariate analysis to understand the underlying attitudes toward raising revenue found that more left-leaning or communitarian attitudes are associated with more support for gasoline, income, or sales taxes devoted to mitigating vulnerability. Those who supported investment in transit and protecting infrastructure also were more likely to support these taxes. There was no parallel finding of factors associated with taxing visitors or issuing bonds
An Actin-Based Wave Generator Organizes Cell Motility
Although many of the regulators of actin assembly are known, we do not understand how these components act together to organize cell shape and movement. To address this question, we analyzed the spatial dynamics of a key actin regulator—the Scar/WAVE complex—which plays an important role in regulating cell shape in both metazoans and plants. We have recently discovered that the Hem-1/Nap1 component of the Scar/WAVE complex localizes to propagating waves that appear to organize the leading edge of a motile immune cell, the human neutrophil. Actin is both an output and input to the Scar/WAVE complex: the complex stimulates actin assembly, and actin polymer is also required to remove the complex from the membrane. These reciprocal interactions appear to generate propagated waves of actin nucleation that exhibit many of the properties of morphogenesis in motile cells, such as the ability of cells to flow around barriers and the intricate spatial organization of protrusion at the leading edge. Application of multiplexed ion mobility spectrometry towards the identification of host protein signatures of treatment effect in pulmonary tuberculosis.
RationaleThe monitoring of TB treatments in clinical practice and clinical trials relies on traditional sputum-based culture status indicators at specific time points. Accurate, predictive, blood-based protein markers would provide a simpler and more informative view of patient health and response to treatment.ObjectiveWe utilized sensitive, high throughput multiplexed ion mobility-mass spectrometry (IM-MS) to characterize the serum proteome of TB patients at the start of and at 8 weeks of rifamycin-based treatment. We sought to identify treatment specific signatures within patients as well as correlate the proteome signatures to various clinical markers of treatment efficacy.MethodsSerum samples were collected from 289 subjects enrolled in CDC TB Trials Consortium Study 29 at time of enrollment and at the end of the intensive phase (after 40 doses of TB treatment). Serum proteins were immunoaffinity-depleted of high abundant components, digested to peptides and analyzed for data acquisition utilizing a unique liquid chromatography IM-MS platform (LC-IM-MS). Linear mixed models were utilized to identify serum protein changes in the host response to antibiotic treatment as well as correlations with culture status end points.ResultsA total of 10,137 peptides corresponding to 872 proteins were identified, quantified, and used for statistical analysis across the longitudinal patient cohort. In response to TB treatment, 244 proteins were significantly altered. Pathway/network comparisons helped visualize the interconnected proteins, identifying up regulated (lipid transport, coagulation cascade, endopeptidase activity) and down regulated (acute phase) processes and pathways in addition to other cross regulated networks (inflammation, cell adhesion, extracellular matrix). Detection of possible lung injury serum proteins such as HPSE, significantly downregulated upon treatment. Analyses of microbiologic data over time identified a core set of serum proteins (TTHY, AFAM, CRP, RET4, SAA1, PGRP2) which change in response to treatment and also strongly correlate with culture status. A similar set of proteins at baseline were found to be predictive of week 6 and 8 culture status.ConclusionA comprehensive host serum protein dataset reflective of TB treatment effect is defined. A repeating set of serum proteins (TTHY, AFAM, CRP, RET4, SAA1, PGRP2, among others) were found to change significantly in response to treatment, to strongly correlate with culture status, and at baseline to be predictive of future culture conversion. If validated in cohorts with long term follow-up to capture failure and relapse of TB, these protein markers could be developed for monitoring of treatment in clinical trials and in patient care
Science Objectives and Early Results of the DEEP2 Redshift Survey
The DEIMOS spectrograph has now been installed on the Keck-II telescope and
commissioning is nearly complete. The DEEP2 Redshift Survey, which will take
approximately 120 nights at the Keck Observatory over a three year period and
has been designed to utilize the power of DEIMOS, began in the summer of 2002.
The multiplexing power and high efficiency of DEIMOS enables us to target 1000
faint galaxies per clear night. Our goal is to gather high-quality spectra of
\~60,000 galaxies with z>0.75 in order to study the properties and large scale
clustering of galaxies at z ~ 1. The survey will be executed at high spectral
resolution, , allowing us to work
between the bright OH sky emission lines and to infer linewidths for many of
the target galaxies (for several thousand objects, we will obtain rotation
curves as well). The linewidth data will facilitate the execution of the
classical redshift-volume cosmological test, which can provide a precision
measurement of the equation of state of the Universe. This talk reviews the

The Cosmic Ultraviolet Baryon Survey (CUBS) I. Overview and the diverse environments of Lyman limit systems at z<1
We present initial results from the Cosmic Ultraviolet Baryon Survey (CUBS).
CUBS is designed to map diffuse baryonic structures at redshift z<~1 using
absorption-line spectroscopy of 15 UV-bright QSOs with matching deep galaxy
survey data. CUBS QSOs are selected based on their NUV brightness to avoid
biases against the presence of intervening Lyman Limit Systems (LLSs) at
zabs~ 17.2 over a total
redshift survey pathlength of dz=9.3, and a number density of n(z)=0.43 (-0.18,
+0.26). Considering all absorbers with log N(HI)/cm^-2 > 16.5 leads to
n(z)=1.08 (-0.25, +0.31) at z<1. All LLSs exhibit a multi-component structure
and associated metal transitions from multiple ionization states such as CII,
CIII, MgII, SiII, SiIII, and OVI absorption. Differential chemical enrichment
levels as well as ionization states are directly observed across individual
components in three LLSs. We present deep galaxy survey data obtained using the
VLT-MUSE integral field spectrograph and the Magellan Telescopes, reaching
sensitivities necessary for detecting galaxies fainter than 0.1L* at d<~300
physical kpc (pkpc) in all five fields. A diverse range of galaxy properties is
seen around these LLSs, from a low-mass dwarf galaxy pair, a co-rotating
gaseous halo/disk, a star-forming galaxy, a massive quiescent galaxy, to a
galaxy group. The closest galaxies have projected distances ranging from d=15
to 72 pkpc and intrinsic luminosities from ~0.01L* to ~3L*. Our study shows
that LLSs originate in a variety of galaxy environments and trace gaseous

Urine Injury Biomarkers and Risk of Adverse Outcomes in Recipients of Prevalent Kidney Transplants: The Folic Acid for Vascular Outcome Reduction in Transplantation Trial
Recipients of kidney transplants (KTR) are at increased risk for cardiovascular events, graft failure, and death. It is unknown whether urine kidney injury biomarkers are associated with poor outcomes among KTRs. We conducted a post hoc analysis of the Folic Acid for Vascular Outcome Reduction in Transplantation (FAVORIT) Trial using a case-cohort study design, selecting participants with adjudicated cardiovascular events, graft failure, or death. Urine neutrophil gelatinase–associated lipocalin (NGAL), kidney injury molecule-1 (KIM-1), IL-18, and liver–type fatty acid binding protein (L-FABP) were measured in spot urine samples and standardized to urine creatinine concentration. We adjusted for demographics, cardiovascular risk factors, eGFR, and urine albumin-to-creatinine ratio. Patients had 291 cardiovascular events, 257 graft failure events, and 359 deaths. Each log increase in urine NGAL/creatinine independently associated with a 24% greater risk of cardiovascular events (adjusted hazard ratio [aHR], 1.24; 95% confidence interval [95% CI], 1.06 to 1.45), a 40% greater risk of graft failure (aHR, 1.40; 95% CI, 1.16 to 1.68), and a 44% greater risk of death (aHR, 1.44; 95% CI, 1.26 to 1.65). Urine KIM-1/creatinine and IL-18/creatinine independently associated with greater risk of death (aHR, 1.29; 95% CI, 1.03 to 1.61 and aHR, 1.25; 95% CI, 1.04 to 1.49 per log increase, respectively) but not with risk of cardiovascular events or graft failure. Urine L-FABP did not associate with any study outcomes. In conclusion, among prevalent KTRs, higher urine NGAL, KIM-1, and IL-18 levels independently and differentially associated with greater risk of adverse outcomes
The DEEP2 Galaxy Redshift Survey: Design, Observations, Data Reduction, and Redshifts
We describe the design and data sample from the DEEP2 Galaxy Redshift Survey,
the densest and largest precision-redshift survey of galaxies at z ~ 1
completed to date. The survey has conducted a comprehensive census of massive
galaxies, their properties, environments, and large-scale structure down to
absolute magnitude M_B = -20 at z ~ 1 via ~90 nights of observation on the
DEIMOS spectrograph at Keck Observatory. DEEP2 covers an area of 2.8 deg^2
divided into four separate fields, observed to a limiting apparent magnitude of
R_AB=24.1. Objects with z < 0.7 are rejected based on BRI photometry in three
of the four DEEP2 fields, allowing galaxies with z > 0.7 to be targeted ~2.5
times more efficiently than in a purely magnitude-limited sample. Approximately
sixty percent of eligible targets are chosen for spectroscopy, yielding nearly
53,000 spectra and more than 38,000 reliable redshift measurements. Most of the
targets which fail to yield secure redshifts are blue objects that lie beyond z
~ 1.45. The DEIMOS 1200-line/mm grating used for the survey delivers high
spectral resolution (R~6000), accurate and secure redshifts, and unique
internal kinematic information. Extensive ancillary data are available in the
DEEP2 fields, particularly in the Extended Groth Strip, which has evolved into
one of the richest multiwavelength regions on the sky. DEEP2 surpasses other
deep precision-redshift surveys at z ~ 1 in terms of galaxy numbers, redshift
accuracy, sample number density, and amount of spectral information. We also
provide an overview of the scientific highlights of the DEEP2 survey thus far.
This paper is intended as a handbook for users of the DEEP2 Data Release 4,
which includes all DEEP2 spectra and redshifts, as well as for the

On the Link Between Central Black Holes, Bar Dynamics, and Dark Matter Halos in Spiral Galaxies
The discovery of a relationship between supermassive black hole (SMBH) mass
and spiral arm pitch angle (P) is evidence that SMBHs are tied to the overall
secular evolution of a galaxy. The discovery of SMBHs in late-type galaxies
with little or no bulge suggests that an underlying correlation between the
dark matter halo concentration and SMBH mass (MBH) exists, rather than between
the bulge mass and MBH. In this paper we measure P using a two-dimensional fast
fourier transform and estimate the bar pattern speeds of 40 barred spiral
galaxies from the Carnegie-Irvine Galaxy Survey. The pattern speeds were
derived by estimating the gravitational potentials of our galaxies from Ks-band
images and using them to produce dynamical simulation models. The pattern
speeds allow us to identify those galaxies with low central dark halo
densities, or fast rotating bars, while P provides an estimate of MBH. We find
that a wide range of MBH exists in galaxies with low central dark matter halo
densities, which appears to support other theoretical results. We also find
that galaxies with low central dark halo densities appear to follow more
predictable trends in P versus de Vaucouleurs morphological type (T) and bar
strength versus T than barred galaxies in general. The empirical relationship
between MBH and total gravitational mass of a galaxy (Mtot) allows us to
predict the minimum Mtot that will be observationally measured of our fast bar

B-Type Natriuretic Peptide and Cardiac Troponin I Are Associated With Adverse Outcomes in Stable Kidney Transplant Recipients
Approximately 200,000 kidney transplant recipients are living in the US; they are at increased risk for cardiovascular and other adverse outcomes. Biomarkers predicting these outcomes are needed. Using specimens collected during the FAVORIT (Folic Acid for Vascular Outcome Reduction In Transplantation) trial, we determined whether plasma levels of B-type natriuretic peptide (BNP) and cardiac troponin I are associated with adverse outcomes in stable kidney transplant recipients
Which veterans enroll in a VA health information exchange program?
Objective: To characterize patients who voluntarily enrolled in an electronic health information exchange (HIE) program designed to share data between Veterans Health Administration (VHA) and non-VHA institutions.
Materials and Methods: Patients who agreed to participate in the HIE program were compared to those who did not. Patient characteristics associated with HIE enrollment were examined using a multivariable logistic regression model. Variables selected for inclusion were guided by a health care utilization model adapted to explain HIE enrollment. Data about patients’ sociodemographics (age, gender), comorbidity (Charlson index score), utilization (primary and specialty care visits), and access (distance to VHA medical center, insurance, VHA benefits) were obtained from VHA and HIE electronic health records.
Results: Among 57 072 patients, 6627 (12%) enrolled in the HIE program during its first year. The likelihood of HIE enrollment increased among patients ages 50–64, of female gender, with higher comorbidity, and with increasing utilization. Living in a rural area and being unmarried were associated with decreased likelihood of enrollment.
Discussion and Conclusion: Enrollment in HIE is complex, with several factors involved in a patient’s decision to enroll. To broaden HIE participation, populations less likely to enroll should be targeted with tailored recruitment and educational strategies. Moreover, inclusion of special populations, such as patients with higher comorbidity or high utilizers, may help refine the definition of success with respect to HIE implementation
