    Surveys of Stylisma Pickeringii var. Pattersonii (Convolvulaceae), its Associated Plant Species, and its Insect Visitors

    Stylisma pickeringii var. pattersonii (Convolvulaceae) is endangered in Illinois and Iowa, and occurs in scattered populations in other states. During 1999 and 2000, two insect species previously unreported from Illinois were observed visiting its flowers. This study was undertaken to survey additional insect visitors, as well as to characterize the plant community where S. pickeringii occurs. The objectives were to survey: 1) floral traits (anthesis and flower density) of S. pickeringii, 2) associated plant species, and 3) insect visitor characteristics. Floral traits were determined and associated plant species surveyed in Mason County (degraded hay field on private property) and Henderson County (dry sand prairie at the Big River State Forest), Illinois. Insects visiting flowers were collected at 10:00 a.m., 12:30 p.m., and 3:00 p.m. during June, July, and August in 2001 and 2002. Individual flowers lasted one day and remained open for 6–8 hours. Peak flowering occurred from early to the middle of July when S. pickeringii was the dominant species in flower. Henderson County contained a greater diversity of native plant species with less bare ground and fewer non-native species than the Mason County site. Fortyseven insect species were observed visiting S. pickeringii flowers. Most frequent visitors were Apis mellifera (Hymenoptera: Apidae), Bombylius fraudulentus (Diptera: Bombyliidae), and Heterostylum croceum (Diptera: Bombliidae). The diversity of visiting insects was higher earlier than later in the day, in July and August than June, and in Henderson than Mason County

    VESPA: software to facilitate genomic annotation of prokaryotic organisms through integration of proteomic and transcriptomic data

    <p>Abstract</p> <p>Background</p> <p>The procedural aspects of genome sequencing and assembly have become relatively inexpensive, yet the full, accurate structural annotation of these genomes remains a challenge. Next-generation sequencing transcriptomics (RNA-Seq), global microarrays, and tandem mass spectrometry (MS/MS)-based proteomics have demonstrated immense value to genome curators as individual sources of information, however, integrating these data types to validate and improve structural annotation remains a major challenge. Current visual and statistical analytic tools are focused on a single data type, or existing software tools are retrofitted to analyze new data forms. We present Visual Exploration and Statistics to Promote Annotation (VESPA) is a new interactive visual analysis software tool focused on assisting scientists with the annotation of prokaryotic genomes though the integration of proteomics and transcriptomics data with current genome location coordinates.</p> <p>Results</p> <p>VESPA is a desktop Javaℱ application that integrates high-throughput proteomics data (peptide-centric) and transcriptomics (probe or RNA-Seq) data into a genomic context, all of which can be visualized at three levels of genomic resolution. Data is interrogated via searches linked to the genome visualizations to find regions with high likelihood of mis-annotation. Search results are linked to exports for further validation outside of VESPA or potential coding-regions can be analyzed concurrently with the software through interaction with BLAST. VESPA is demonstrated on two use cases (<it>Yersinia pestis </it>Pestoides F and <it>Synechococcus </it>sp. PCC 7002) to demonstrate the rapid manner in which mis-annotations can be found and explored in VESPA using either proteomics data alone, or in combination with transcriptomic data.</p> <p>Conclusions</p> <p>VESPA is an interactive visual analytics tool that integrates high-throughput data into a genomic context to facilitate the discovery of structural mis-annotations in prokaryotic genomes. Data is evaluated via visual analysis across multiple levels of genomic resolution, linked searches and interaction with existing bioinformatics tools. We highlight the novel functionality of VESPA and core programming requirements for visualization of these large heterogeneous datasets for a client-side application.     Markers of assimilation of problematic experiences in dementia within the LivDem project

This study aimed to determine whether the Markers of Assimilation of Problematic Experiences in Dementia scale (MAPED) can be used to identify whether the way in which participants talk about dementia changed during the group. All eight sessions of a LivDem group, which were attended by participants were recorded and transcribed. An initial analysis identified 160 extracts, which were then rated using the MAPED system. Inter-rater reliability was 61% and following a resolution meeting, 35 extracts were discarded, leaving 125 extracts with an agreed marker code. All of the participants were identified as producing a speech marker relating to dementia, and these varied between 0 (warding off) to 6 (problem solution). Examples of these markers are provided. The proportion of emergence markers (indicating the initial stages of assimilation) compared to later markers changed significantly between the first four sessions and the final sessions. This difference was still significant even when the markers produced by the most verbal participant, Graham, were excluded. The use of process measures within psychotherapy complements more conventional outcome measures and has both theoretical and clinical implications

    Ultra-low Ultraviolet Radiation in Office Lighting Can Moderate Seasonal Vitamin D Cycle: A Pilot Study

    Background/Aim: Ultraviolet-B (UV-B) radiation initiates vitamin D synthesis in the skin, making sun exposure a major source of vitamin D. We aimed to determine whether office lighting containing ultra-low levels of UV-B radiation could modify the winter decline in vitamin D status in the UK, while being safe and well tolerated. Patients and Methods: Twenty commercial office desk lamps were modified with the addition of UV-B LEDs. Ten hospital office administrative staff received UV-modified lamps with UV-on, and 10 staff received identical placebo lamps with UV switched off, in a double-blind, cross-over pilot study during the winter of 2021/22. Circulating 25-hydroxyvitamin D [25(OH)D] was measured every 4 weeks for 20 weeks: at baseline and during an 8-week trial period, 4-week washout, and a cross-over 8-week trial period. Results: The linear regression combining the complete datasets for phase 1 and 2 of the trial showed that an 8-week UV light intervention significantly increased 25OHD by 7.13 nmol/l with a p-Value=0.02, compared to the placebo group. Similar results were confirmed by cross-over analyses using the datasets of those completing both phases of the trial both with and without using the inverse probability weighing method to handle dropouts. Conclusion: The UV-B-modified lighting was well-tolerated and safe with weekly doses of UV-B of 0.5 - 0.9 Standard Erythema Dose [SED=100 Jm -2 erythema weighted UV radiation] measured at chest level. This ultra-low dosing was effective in reducing the winter decline in vitamin D status

    A two-parameter wind speed algorithm for Ku-band altimeters

    Globally distributed crossovers of altimeter and scatterometer observations clearly demonstrate that ocean altimeter backscatter correlates with both the near-surface wind speed and the sea state. Satellite data from TOPEX/Poseidon and NSCAT are used to develop an empirical altimeter wind speed model that attenuates the sea-state signature and improves upon the present operational altimeter wind model. The inversion is defined using a multilayer perceptron neural network with altimeter-derived backscatter and significant wave height as inputs. Comparisons between this new model and past single input routines indicates that the rms wind error is reduced by 10%–15% in tandem with the lowering of wind error residuals dependent on the sea state. Both model intercomparison and validation of the new routine are detailed, including the use of large independent data compilations that include the SeaWinds and ERS scatterometers, ECMWF wind fields, and buoy measurements. The model provides consistent improvement against these varied sources with a wind-independent bias below 0.3 m s?1. The continuous form of the defined function, along with the global data used in its derivation, suggest an algorithm suitable for operational application to Ku-band altimeters. Further model improvement through wave height inclusion is limited due to an inherent multivaluedness between any single realization of the altimeter measurement pair [?o, HS] and observed near-surface winds. This ambiguity indicates that HS is a limited proxy for variable gravity wave properties that impact upon altimeter backscatter

    Comparative study of healthy older and younger adults shows they have the same skin concentration of vitamin D3 precursor, 7-dehydrocholesterol, and similar response to UVR

    Vitamin D3 synthesis in human skin is initiated by solar ultraviolet radiation (UVR) exposure of precursor 7-dehydrocholesterol (7DHC), but influence of age on the early stage of vitamin D3 metabolism is uncertain. We performed a prospective standardised study in healthy ambulant adults aged ≄65 and ≀40 years examining (1) if baseline skin 7DHC concentration differs between younger and older adults and (2) the impact of older age on serum vitamin D3 response to solar simulated UVR. Eleven younger (18–40 years) and 10 older (65–89 years) adults, phototype I–III, received low-dose UVR (95% UVA, 5% UVB, 1.3 SED) to ~35% of the body surface area. Biopsies were taken for 7DHC assay from unexposed skin, skin immediately and 24 h post-UVR, and blood sampled at baseline, 24 h and 7 d post-UVR for vitamin D3 assay. Samples were analysed by HPLC-MS/MS. Baseline skin 7DHC (mean ± SD) was 0.22 ± 0.07 and 0.25 ± 0.08 ”g/mg in younger versus older adults (no significant difference). Baseline serum vitamin D3 concentration was 1.5 ± 1.5 and 1.5 ± 1.7 nmol/L in younger versus older adults, respectively, and showed a significant increase in both groups post-UVR (no significant differences between age groups). Thus, skin 7DHC concentration was not a limiting factor for vitamin D3 production in older relative to younger adults. This information assists public health guidance on sun exposure/vitamin D nutrition, with particular relevance to the growing populations of healthy ambulant adults ≄65 years

    Quality Assurance of Spectral Ultraviolet Measurements in Europe Through the Development of a Transportable Unit (QASUME)

    QASUME is a European Commission funded project that aims to develop and test a transportable unit for providing quality assurance to UV spectroradiometric measurements conducted in Europe. The comparisons will be performed at the home sites of the instruments, thus avoiding the risk of transporting instruments to participate in intercomparison campaigns. Spectral measurements obtained at each of the stations will be compared, following detailed and objective comparison protocols, against collocated measurements performed by a thoroughly tested and validated travelling unit. The transportable unit comprises a spectroradiometer, its calibrator with a set of calibration lamps traceable to the sources of different Standards Laboratories, and devices for determining the slit function and the angular response of the local spectroradiometers. The unit will be transported by road to about 25 UV stations over a period of about two years. The spectroradiometer of the transportable unit is compared in an intercomparison campaign with six instruments to establish a relation, which would then be used as a reference for its calibration over the period of its regular operation at the European stations. Different weather patterns (from clear skies to heavy rain) were present during the campaign, allowing the performance of the spectroradiometers to be evaluated under unfavourable conditions (as may be experienced at home sites) as well as the more desirable dry conditions. Measurements in the laboratory revealed that the calibration standards of the spectroradiometers differ by up to 10%. The evaluation is completed through comparisons with the same six instruments at their homes sites

    Exact and near backscattering measurements of the linear depolarisation ratio of various ice crystal habits generated in a laboratory cloud chamber

This is an open access article under the CC BY license(http://creativecommons.org/licenses/by/4.0/).Ice clouds were generated in the Manchester Ice Cloud Chamber (MICC), and the backscattering linear depolarisation ratio, ÎŽ, was measured for a variety of habits. To create an assortment of particle morphologies, the humidity in the chamber was varied throughout each experiment, resulting in a range of habits from the pristine to the complex. This technique was repeated at three temperatures: −7 °C, −15 °C and −30 °C, in order to produce both solid and hollow columns, plates, sectored plates and dendrites. A linearly polarised 532 nm continuous wave diode laser was directed through a section of the cloud using a non-polarising 50:50 beam splitter. Measurements of the scattered light were taken at 178°, 179° and 180°, using a Glan–Taylor prism to separate the co- and cross-polarised components. The intensities of these components were measured using two amplified photodetectors and the ratio of the cross- to co-polarised intensities was measured to find the linear depolarisation ratio. In general, it was found that Ray Tracing over-predicts the linear depolarisation ratio. However, by creating more accurate particle models which better represent the internal structure of ice particles, discrepancies between measured and modelled results (based on Ray Tracing) were reduced.Peer reviewe

Many people have low blood levels of vitamin D, especially in winter or if confined indoors, because summer sunshine is the main source of vitamin D for most people. Government vitamin D intake recommendations for the general population are 400 IU (10 ”g) per day for the UK7 and 600 IU (15 ”g) per day for the USA (800 IU (20 ”g) per day for >70 years) and the EU.9 Taking a daily supplement (400 IU /day (10 ”g/day) in the UK) and eating foods that provide vitamin D is particularly important for those self-isolating with limited exposure to sunlight. Vitamin D intakes greater than the upper limit of 4000 IU (100 ”g) per day may be harmful and should be avoided unless under personal medical/clinical advice by a qualified health professional
