    Coaching as a Framework for Early Intervention Service Delivery

    Abstract The Division for Early Childhood (DEC) has set forth a list of high leverage practices that have significant impact on children with special needs from birth through early childhood. One of these impactful strategies is building family capacity, which can be effectively addressed using a coaching framework for service delivery. Research in the field has not yet clearly defined coaching terms, definitions or characteristics, which impacts the fidelity of the practice and requires research and consensus moving forward. This body of work seeks to look at current research around the topic of coaching and it’s effectiveness in meeting student and family outcomes. In addition it includes a personal experience using the components of the coaching framework and its coordinating fidelity tool with a family in early intervention.     Electrochemical Properties Of Passive Films On 440c Stainless Steel

    The properties of passive films formed on 440C stainless steel were investigated using advanced electrochemical techniques for the purpose of establishing a method to measure film stability directly in passivation baths. Electrochemical measurements were sensitive to the passive film properties and were able to quantify the effect of cooling rate on passive film stability. The various techniques used--linear polarization, AC impedance, small amplitude cyclic voltammetry (SACV), and coulostatic transient measurements--provided results that exhibited excellent agreement with the AC impedance technique providing the most information about the interfacial reactions. Full electrochemical characterization of the passive film stability required an evaluation of both the polarization resistance and the interfacial capacitance, thus precluding the use of DC measurements. The coulostatic technique demonstrated the most promise as a monitoring tool as it provided a rapid, simple measurement of both the polarization resistance and capacitance

    The Employment and Training Institute collaborated on a project with the Justice Initiatives Institute examining Branch A Milwaukee Municipal Court cases from 2008 to 2013 using records obtained from the Milwaukee Municipal Court and the Milwaukee County Sheriff’s Office and focusing on the population incarcerated for municipal ordinance violations. A majority of those jailed for failure to pay municipal judgments were not employed at the time of booking. Municipal courts often denied Wisconsin residents unable to pay or delinquent in paying court judgments for municipal citations their right to drive for two years -- jeopardizing workers’ employment options and placing them at risk in the criminal justice system if found “driving while suspended. Thousands of low-income Milwaukee teens of color failing to pay fines on municipal violations (even if unrelated to driving) are barred from getting their driver’s license for two years

    A Possible Etiology of the Infertile 46XX Male Subject

    We report on an infertile male patient with the predominant 46XX female karyotype. A testicular biopsy revealed widely separated testicular tubules, absence of sperm formation and large numbers of Leydig cells. Chromosome studies, including measurements of the X chromosomes, showed a significant difference between the lengths of the short arm of the 2 X chromosomes. This information lends support for an X-Y chromosome interchange as the etiology of this syndrome. The clinical features of this rare syndrome and other theories of etiology of XX male subjects are discussed

    Possible Radiation-Induced Damage to the Molecular Structure of Wooden Artifacts Due to Micro-Computed Tomography, Handheld X-Ray Fluorescence, and X-Ray Photoelectron Spectroscopic Techniques

    This study was undertaken to ascertain whether radiation produced by X-ray photoelectron spectroscopy (XPS), micro-computed tomography (μCT) and/or portable handheld X-ray fluorescence (XRF) equipment might damage wood artifacts during analysis. Changes at the molecular level were monitored by Fourier transform infrared (FTIR) analysis. No significant changes in FTIR spectra were observed as a result of μCT or handheld XRF analysis. No substantial changes in the collected FTIR spectra were observed when XPS analytical times on the order of minutes were used. However, XPS analysis collected over tens of hours did produce significant changes in the FTIR spectra

    MeCP2 binds to nucleosome free (linker DNA) regions and to H3K9/H3K27 methylated nucleosomes in the brain

    Methyl-CpG-binding protein 2 (MeCP2) is a chromatin-binding protein that mediates transcriptional regulation, and is highly abundant in brain. The nature of its binding to reconstituted templates has been well characterized in vitro. However, its interactions with native chromatin are less understood. Here we show that MeCP2 displays a distinct distribution within fractionated chromatin from various tissues and cell types. Artificially induced global changes in DNA methylation by 3-aminobenzamide or 5-aza-2′-deoxycytidine, do not significantly affect the distribution or amount of MeCP2 in HeLa S3 or 3T3 cells. Most MeCP2 in brain is chromatin-bound and localized within highly nuclease-accessible regions. We also show that, while in most tissues and cell lines, MeCP2 forms stable complexes with nucleosome, in brain, a fraction of it is loosely bound to chromatin, likely to nucleosome-depleted regions. Finally, we provide evidence for novel associations of MeCP2 with mononucleosomes containing histone H2A.X, H3K9me2 and H3K27me3 in different chromatin fractions from brain cortex and in vitro. We postulate that the functional compartmentalization and tissue-specific distribution of MeCP2 within different chromatin types may be directed by its association with nucleosomes containing specific histone variants, and post-translational modifications
